  • Objectives : The purpose of this study was to investigate the anti-inflammatory effects of extract from Trogopterorum Faeces (TF) on the RAW 264.7 cells. Methods : To prove the TF's anti-inflammatory effects, we investigated nitric oxide (NO) production and own cell viability. We examined the cytokine productions on lipopolysacchride (LPS)-induced RAW 264.7 cells and also cellular regulatory mechanisms. Results : TF does not have any cytotoxic effect. TF reduced LPS-induced NO production, interleukin (IL)-1b, IL-6, IL-10 and tumor necrosis factor-a (TNF-a) in RAW 264.7 cells. TF inhibited the activation of mitogen-activated protein kinases (MAPKs) such as p38, extracelluar signal-regulated kinase (ERK 1/2) and c-Jun NH2-terminal kinase (JNK) and also the degradation of inhibitory kappa B a (Ik-Ba) in the LPS-stimulated RAW 264.7 cells. TF reduced the serum levels of IL-1b, IL-6, TNF-a. The survival rate of LPS-induced endotoxin shock was increased by TF administration. Conclusions : TF down-regulated LPS-induced NO and cytokines production, which could provide a clinical basis for anti-inflammatory properties.

  • Objective : Portulaca oleracea (PO) has been used as an important traditional medicine for inflammatory and bacterial diseases in East Asia. However, the protective effects of PO on acute pancreatitis (AP) is not well-known. Therefore, this study was performed to identify the anti-inflammatory and prophylactic effects of PO on cerulein-induced AP. Methods : AP was induced in mice via intraperitoneal injection of supramaximal concentrations of the stable cholecystokinin analogue cerulein ($50{\mu}g/kg$) given every hour for 6 times. Water extracts of PO (100, 300, or 500 mg/kg) was administrated intra-peritoneally 1 h prior to the first injection of cerulein. The mice were killed at 6 h after the final cerulein injection. Pancreas and lung were rapidly removed for morphologic and histochemical examination, myeloperoxidase (MPO) assay. Blood samples were taken to determine serum amylase and lipase activities. Results : Administration of PO significantly inhibited pancreatic weight/body weight ratio, pancreas and lung histological injury. And MPO activity which indicates neutrophil infiltration was inhibited by PO extracts on cerulein-induced pancreatitis. In addition, PO administration inhibited digestive enzymes such as serum amylase and lipase activity on cerulein-induced pancreatitis. Conclusion : Our results could suggest that pre-treatment of PO reduces the severity of cerulein-induced AP, thereby, PO could be used as a protective agent against AP. Also, this study could give a clinical basis that PO could be a drug or agent to prevent AP.

  • Objectives : The purpose of present study was to investigate the vasorelaxant activities and mechanisms of action of the ethanol extract of P. yedoensis leaf (PYL) on isolated rat aortic rings. Methods : Dried P. yedoensis leaves were extracted 3 times with 100% ethanol for 3 h in a reflux apparatus. Isolated rat aortic rings were suspended in organ chambers containing 10 ml Krebs-Henseleit (K-H) solution. The rings were maintained at $37^{\circ}C$ and aerated with a mixture of 95% $O_2$ and 5% $CO_2$. Changes in their tension were recorded via isometric transducers connected to a data acquisition system. Results : PYL relaxed the contraction of aortic rings induced by phenylephrine (PE, 1 ${\mu}M$) or KCl (60 mM) in a concentration dependent manner. However, the vasorelaxant effects of PYL on endothelium-denuded aortic rings were lower than endothelium-intact aortic rings. And the vasorelaxant effects of PYL on endothelium-intact aortic rings were reduced by pre-treatment with $N{\omega}$-Nitro-L-arginine methyl ester (10 ${\mu}M$), methylene blue (10 ${\mu}M$), 1-H-[1,2,4]-oxadiazolo-[4,3-${\alpha}$]-quinoxalin-1-one (10 ${\mu}M$), tetraethylammonium (5 mM). In addition, PYL inhibited the contraction induced by extracellular $Ca^{2+}$ in endothelium-denuded aortic rings pre-contracted by PE or KCl in $Ca^{2+}$-free K-H solution. Conclusions : These results suggest that PYL exerts its vasorelaxant effects via the activation of Nitric Oxide (NO) formation by means of L-arginine and NO-cGMP pathways and via the blockage of receptor operated calcium channels, voltage dependent calcium channels and calcium-activated potassium channels.

  • Objectives : Obesity is a chronic metabolic disease caused by disorder of energy balance and lipid metabolism. This study was conducted by histopathology and histomorphometry to investigate the anti-obesity effects of mixed water extract of Plantaginis Semen & Poria (CJB) on liver, epididymal fat pads and pancreas zymogen granules in obese rats induced with high fat diet. Method : Male Sprague-Dawley rats to be divided four groups were fed into four different treatments: normal (NOR) diet, high-fat (HF) diet, HF diet+CJB (100 mg/kg and 300 mg/kg, P.O.) for 8 weeks. The weekly body weights were measured in four experimental groups, respectively. Also histopathological and histomorphometrical changes of liver, epididymal fat pads and pancreas zymogen granules were observed in normal control and obese rats, respectively. Results : Adminstration of CJB significantly reduced body weights compared to those of HF group for experimental period. After 8 weeks, liver weights in the CJB groups were lower than those of HF group. In addition, HF diet related steatohepatitis, adipocyte hypertrophy, exocrine disturbances (decreases of pancreatic zymogen granules) were also dose-dependently inhibited by treatment of test material, CJB 100 and CJB 300 as compared with HF group, respectively. Conclusion : Based on the results, it is considered that CJB will be showed hepatoprotective and anti-obese effects, may be directly and/or indirectly mediated by pancreatic zymogen granules because they dose-dependently inhibited steatohepatitis, hypertrophy of adipocytes and decreases of pancreatic zymogen granules induced by HF diet supply, respectively.

  • Objectives : This study is aimed at establishing the discriminative criteria for Adenophorae Radix (AT; Sasam), Codonopsis Lanceolatae Radix (CL; Yangyu) and Adenophorae Remotiflori Radix (AR; Jeni). Because of confusable publications and the similitude of herb shape, CL and AR have been used as AT in the markets. Methods : We firstly discriminated and confirmed AT, CL and AR through observing the external morphology of original plants. Then, external-internal morphological features of the medicinal herbs were studied. A stereoscope was used to determine macroscopic features of dried herbal medicines and a microscope was conducted to examine internal features with the samples, which were embedded in paraffin and stained. Results : 1. The original plant of CL was discriminated with the stem shape in comparison with others. AT and AR were classified by the leaf position of original plant. 2. In the macroscopic morphological study of medicinal herbs, CL had lumps, which seem like a wart. Also, AT and AR were different in the density of cross section. 3. In the microscopic morphological study of the medicinal herbs, the phloem, fissures and the arrangement of xylem were selected as discriminative criteria for 3 kinds of herbs. With these contents, we suggested the identification keys, including further details. Conclusions : These results, especially identification keys, will be helpful to distinguish 3 kinds of herbs, referred to as a 'Sasam'. Moreover, macroscopic and microscopic methods used in this article would be applicable tools for the discrimination of other herbs.
