Disinfection efficacy of slightly acidic electrolyzed water (SlAEW) against some fresh vegetables (미산성 차아염소산수의 신선 채소류에 대한 살균 유효성)

  • In the present study, disinfection efficacy of slightly acidic electrolyzed water [SlAEW, 30 ppm of effective chlorine at $20{\pm}1^{\circ}C$, oxidation-reduction potential (ORP) $562{\pm}23mV$, pH 6.4] on 4 kinds of vegetables (lettuce leaf, endive leaf, perilla leaf and kale leaf) was evaluated to obtain a microbial reduction characteristics which are necessary to design a process control for non-thermal sterilization of fresh vegetables. Active chlorine, residual chlorine, microbial counts and residual microbial counts, which are the key factors in the non-thermal sterilization process were measured by dipping them in SlAEW three times for 30 minutes in order to analyze the relationship between factors. Total microbial count was decreased mostly during the first 10 minutes of washing, and the limit value that can be reduced by immersion treatment was 3 log CFU/g for the total microbial count surviving in 4 kinds of vegetables. The total number of microorganism that can be reduced by washing in SIAEW for 10 min was found to be about 2 log CFU/g on average. In addition, the active chlorine decreased in the initial 10 minutes in 2.2 ppm, 2.0 ppm, 1.7 ppm and 2.5 ppm in lettuce, perilla leaf, endive leaf and kale leaf, respectively, and about 50-80% of the chlorine was reduced in the initial 10 min appear.

Changes in Quality of Soybean Sprouts Washed with Electrolyzed Water during Storage (전기분해수 처리에 따른 콩나물의 저장 중 품질변화)

  • The storage quality of soybean sprouts washed with various electrolyzed waters was investigated. The washing solutions consisted of tap water (TW), sodium hypochlorite electrolyzed water (SHEW), and slightly acidic electrolyzed water (SAEW). The number of bacteria on the soybean sprouts after 5 min of exposure to TW, SHEW, and SAEW resulted in >0.5, 2.0, and 2.0 log CFU/g reductions, respectively. At both $4^{\circ}C$ and $25^{\circ}C$, the number of bacteria, weight loss ratio, and b value rapidly increased, and pH and L value rapidly decreased in soybean sprouts washed with TW. Whereas, the number of bacteria, pH, weight loss ratio, and color slowly changed in the soybean sprouts washed with SHEW and SAEW. Consequently, these results indicate that washing with electrolyzed water is an effective means of maintaining the quality and enhancing the shelf-life of soybean sprouts; both SHEW and SAEW reduced bacterial growth without affecting the other properties of soybean sprouts during storage.

Studies on Photoprotection of Walnut Veneer Exposed to UV Light (자외선 노출에 의한 Walnut 베니어의 광 변색 방지 연구)

  • The purpose of this study was to evaluate the effect of several chemical treatments to prevent photodegradation of wood veneer by external UV (Ultraviolet) light. Of woods, walnut veneer is selected as a raw material for this study since it is known as a luxurious wood with dark color giving an esthetic effect. Alcohol-benzene, hydrogen peroxide ($H_2O_2$) and sodium hypochlorite (NaClO) solution were used for investigate the effect on color stabilization. Despite the removal of the extractive compounds, which is known as a discoloration component, a significant color change of walnut wood veneer was observed. Meanwhile, the veneers treated by 20 and 30% $H_2O_2$ solution at $75^{\circ}C$ for 1 h also showed the no positive effect of color stability exposed to UV light although they have a bleaching effect on wood veneer. Besides, it was difficult to maintain the original color of walnut veneer due to the elution of the extractive compounds. On the other hands, the veneer treated by NaClO solution indicated the good performance on color stability despite of the intensive UV light test. However, when the concentration exceeds 3%, surface roughness and fiber damage occurred simultaneously. Therefore, the walnut species should be treated with proper concentration when sodium hypochlorite is applied to the veneer.

Disinfection Methods of Pruning Scissor for Preventing Transmission of Fire Blight (과수 화상병 전염 차단을 위한 전정 가위 소독 방법)

  • Pathogens of fire blight can survive for a long time on pruning tools, etc., and fire blight can be spread through agricultural implements. In particular, in Korea, pruning is frequently performed to remove flowers, immature fruits, and succulent shoots, and this farm work is known to be a major factor of the spread of fire blight. Therefore, in this study, in order to completely sterilize pathogens on pruning shears, we verified the disinfection effect of disinfectants distributed domestically and used them to identify an effective disinfection method. When disinfecting by immersion after inoculating Erwinia amylovora TS3128 on the scissor blade, 70% ethanol and 1% and 0.2% sodium hypochlorite sterilized immediately 10 sec after immersion, while 30% chlorine dioxide showed little disinfection effect. When disinfecting by spraying, 70% ethanol sterilized bacteria 1 sec after spraying, but 1% and 0.2% sodium hypochlorite disinfected bacteria after 10 and 60 sec, respectively. After cutting the naturally disease occurring branches five and 100 times, the bacteria were not hardly sterilized in immersion treatment in 70% ethanol for 30 sec but perfectly disinfected over 60 sec. Considering these results, pruning shears should be disinfected by frequently immersing them in 70% ethanol for at least 60 sec during pruning work in the field.

The effect of cleaning methods on bond strength of zirconia after saliva contamination (타액으로 오염된 지르코니아 수복물의 접착강도에 세척 방법들이 미치는 영향)

  • This study evaluated the effects of various cleaning methods on the shear bond strength of zirconia ceramics after saliva contamination. Eighty zirconia disk specimens were divided into 8 groups. All groups were treated with one coat of MDP primer. All specimens (except the negative control) were contaminated with human saliva on the zirconia surface. The positive control went through the bonding procedure immediately after contamination without any cleaning procedure. With the exception of control groups, the remaining six groups were rinsed with water and either applied with MDP recoating (WATER+MDP) or without MDP recoating (WATER). While some were cleaned with a Ivoclean with MDP recoating (IVOCLEAN+MDP) or not applied with MDP recoating(IVOCLEAN), others were cleaned with a 1% NaOCl solution with MDP recoating (NaOCl+MDP) or without MDP recoating (NaOCl). The shear bond strength of all specimens were measured after being stored in distilled water at $37^{\circ}C$ for 24 hours. The data was analyzed statistically by an analysis of ANOVA, Tukey's post hoc test and Student's t-test was used to compare the shear bond strength according to the re-coating of MDP after the cleaning procedure. The positive control group showed the lowest shear bond strength value, and the WATER group and NaOCl group showed no significant difference when compared to the positive control group. The IVOCLEAN group showed significantly higher shear bond strength when compared to Water group and NaOCl group but not with the group of negative control. After rinsing with water or the NaOCl solution, the comparison of the single coating of MDP and re-coating of MDP showed different shear bond strengths but there was no significant difference to the negative control. After rinsing with Ivoclean, there was no significant difference to the negative control regardless of the recoating of MDP. In conclusion, the shear bond strength was affected by the cleansing procedure and Ivoclean was found to be effective regardless of the re-coating of MDP. When water or the NaOCl solution is used to remove surface contaminants, the re-coating of MDP provides a positive effect on cementation.

Ohmic Thawing System을 이용한 해동기법이 냉동 돈육 품질에 미치는 영향

  • 본 연구는 Ohmic heating system을 이용하여 돈육을 해동하여 해동에 의한 식육의 손상을 최소화하고 신선육과 유사한 해동육을 얻기 위해 최적 해동 속도에 대한 기초 자료를 마련하고자 실시하였다. 각각의 ohmic power intensity(AC, 0, 10, 20, 30, 40 Volt)에 따른 돈육의 해동속도는 기하학적으로 중심부 변화에서 가장 빠른 해동속도는 40V에서 1,582 cm/h로 산출되었고 OV의 0.307cm/h에 비하여 약 5배 정도 빠르게 나타났으며 power intensity가 증가할수록 해동속도가 증가하였다. 이와 같이 ohmic intensity(X: volt)와 해동속도(Y: cm/h)변화를 수학적으로 나타내었는데 다음과 같다; lnY=-0.8971+$1.0345{\cdot}X$ $R^2=0.9968$. 각각의 ohmic power와 비교에서 대조구인 신선육의 보수력이 가장 높았고 해동시료인 처리구간 비교에서 power가 상대적으로 높은 30V, 40V에서 보수력이 가장 좋게 나타났다. Cooking loss에서는 power intensity에 따른 유의적인 변화는 발견되지 않았다. Colo에서는 b-값의 경우 ohmic power intensity가 증가함에 따라 다소 감소하는 경향을 보여 주었다. 해동시 power intensity가 증가할수록 pH는 다소 낮아지는 경향을 보여주었다. 또한 육의 TBA가는 이와 반대로 ohmic thawing은 육의 TBA가를 증가시키는 요인으로 작용하였는데, power intensity에 따른 변화는 유의적인 차이를 보여주지 않았다. 그러나 VBN가에서는 신선육에 비해 해동육의 VBN은 증가하였지만 해동방법에 따른 차아는 발견되지 않았다.

무전해 도금방식을 이용한 PET 필름 위 선택적 Ni-Cu 박막의 특성분석

  • 최근 이동통신 LED 에너지 자동차 산업분야에서 제품의 고기능화 고성능화를 위한 신소재 개발 및 친환경적인 신공정 개발에 있어, PI 또는 PET와 같은 유연성 소재 위에 선택적 패턴 도금 기술, 고기능성 나노/복합 도금 등이 주목 받고 있다. 또한 전 세계적으로 유해물질의 수 출입 규제 움직임이 강력하게 제기되고 있다. 본 연구에서는 유연성 소재인 PET 위에 친환경적 방법으로 구리를 선택적으로 도금하기 위한 실험을 진행하였다. 준비된 PET 필름 위에 Ag paste를 Screen Printing법을 이용하여 Ag 전극을 패턴하고, 그 위에 무전해 도금방식을 이용하여 Ni과 Cu가 도금 되도록 하였다. Ni 무전해 도금은 pH6.5, 65도에서 시행되었으며, Cu 무전해 도금은 환경규제물질인 포름알데히드 대신에 차아인산나트륨을 사용하여 70도에서, 중성근처의 pH 농도(pH7과 pH8)에서 시행되었다. 이들 다층 박막에 대해 X-ray diffraction (XRD), SEM (Scanning Electron Microscope), XPS (X-ray Photoelectron Spectroscopy) 등을 이용하여 물리-화학적/전기적 특성들을 이용하여 조사 분석하였다.

  • 무전해 도금이란 외부의 전원을 사용하지 않고 환원제를 이용하여 금속 피막을 석출시키는 방법이다. 이러한 무전해 도금은 최근에 와서 인쇄회로기판(printed circuit board)등 다양한 전자부품에 적용되고 있다. 최근 급격한 금 가격 상승으로 인해 전자부품용에 사용되는 금의 사용량을 감소시키거나 또는 금을 대체할 수 있는 도금 층에 관한 연구가 활발하게 진행되고 있다. 금을 대체할 수 있는 즉 비용 경감의 가능성이 있는 도금 재료로는 팔라듐이 유력하다. 팔라듐은 금의 약 1/2가격으로 우수한 내식성 및 낮은 접촉저항을 가지기 때문에 접점재료로서 오래전부터 사용되어져왔다. 무전해 팔라듐-인 도금의 착화제, 환원제, 경도, 접촉저항에 대한 연구는 많이 있지만 도금속도에 미치는 인자에 대해서는 명확하지 않은 것이 많다. 따라서 본 연구는 무전해 팔라듐-인 도금의 석출석도에 미치는 에틸렌디아민 농도의 영향에 대하여 조사하였다. 실험방법으로는 환원제로 차아인산을 사용하고 착화제로는 에틸렌디아민을 사용하여 문전해 팔라듐-인 도금액을 제조하였다. 에틸렌디아민의 농도는 각각 5ml/L, 7.5ml/L, 10ml/L, 12ml/L로 하였다. pH는 7.5, 온도는 $45^{\circ}C$로 하여 30분 동안 도금을 실시하였다. 무전해 팔라듐-인 도금속도는 정밀저울로 무게를 측정하였고 ICP-OES을 사용하여 도금층의 농도를 분석, XRF를 사용하여 성분분석과 XRD를 사용해 결정회절을 분석하였다. 또한 SEM을 사용하여 단면 관찰을 하였으며 석출속도에 미치는 에틸렌디아민의 영향을 전기화학 분극곡선을 통해서 고찰을 시도해 보았다.

  • In this study, we investigated the characteristics of NO oxidation using un-divided electrolyzed seawater as oxidant. The concentration of available chlorine and the temperature of electrolyzed seawater are increased with electrolysis time in the closed-loop constant current electrolysis system. While NO gas flow through bubbling reactor which is filled with electrolyzed seawater, the oxidation rate of NO to $NO_2$ is increased with the concentration of available chlorine and the temperature. $NO_2$, generated by oxidation reaction, is dissolved in electrolyzed seawater and existed as $HNO_3{^-}$ ion.

  • 층층둥굴레(Polygonatum stenophyllum Maxim.)는 백합과(Liliaceae) 둥굴레속(Polygonatum)에 속하는 다년생초본이다. 둥굴레속의 근경은 점액질이 풍부하고 전분질, 아미노산, 알카로이드 등의 성분을 다량 함유하고 있어, 식품학적으로도 우수한 것으로 보고되었다. 그리고 새순과 여린줄기는 식약청 식품원재료서비스에 등록된 식용가능 식물자원이다. 본 연구에 사용된 층층둥굴레는 경기도 황학산수목원에서 2년, 5년생과 종자를 분양받아 종자는 기내에서 발아시킨 후 실험에 사용하였다. 야생의 개체를 무균적으로 기내도입하기 위해 차아염소산나트륨의 농도를 0~1.5%까지 달리하여 멸균한 결과, 유의적 차이는 없으나 1~1.5%처리가 비오염율이 20% 내외로 조사되었다. 층층둥굴레 캘러스 유도에 미치는 옥신류 호르몬의 영향을 알아보기 위해 멸균된 근경을 절단하고 옥신류 호르몬의 종류와 농도가 각각 다르게 처리된 MS배지에 치상하여 배양한 결과, 2,4-D 0.5mg/L 처리구에서 캘러스 유도율이 87%로 가장 높게 조사 되었다. 이러한 결과는 층층둥굴레의 기내배양 방법을 통해 효과적으로 모본과 동일한 층층둥굴레의 대량생산 체계를 만들 수 있는 참고자료로 활용될 수 있고 더불어 관련 산업계의 소재다양화 측면에서 유용하게 사용될 것으로 보여진다.
