Textural and Geochemical Characteristics of Ferromanganese Crusts from the Lomilik and Litakpooki Seamounts, Marshall Islands, West Pacific (서태평양 마샬제도 Lomilik와 Litakpooki 해저산 망간각의 조직 및 지화학적 특성)

  • Six ferromanganese crusts from the Lomilik and Litatfooki seamounts in the Marshall Islands were analyzed for texture, geochemistry and stratigraphy to delineate the paleoceanographic conditions. The crusts can be divided into three layers; 1) outermost massive layer (Layer 1), 2) middle porous Fe-oxides rich layer infllled with biointemal clasts (Layer 2), and 3) innermost massive layer cemented and/or replaced by carbonate fluoapatite (CFA) (Layer 3). The Layer 1 contains higher Mn, Co, Ni, and Mg than other two layers, and the Layer 2 was relatively more enriched in Fe, Al, Ti, Ba, Cu, and Zn. However, the Layer 3 shows higher Ca and P and lower Mn, Fe, Co, and Ni contents than overlying two layers. Based on the Co-chronometry, the crusts are postulated to have begun to grow from 56-31 Ma (early Eocene to Oligocene). The boundaries between layers 1 and 2, and layers 2 and 3 are dated to be 7-3 Ma and 26-14 Ma, respectively. High contents of Ca and P in Layer 3 clearly indicate that the layer had been phosphatized prior to the formation of Layer 2. Considering the well-preserved mjcrostructures in Layer 3, it is unlike that the crusts themselves were recrystallized in suboxic condition. Also, the lower Co concentrations in Layer 3 may imply that the Co supply was not constant during the formation of Layer 3. Layer 2, characterized by the porous texture, grew over Layer 3 during 26-9 Ma. Internal biogenic sediments including foraminifera within the original cavities and the enrichment of organophillic elements such as Ba, Cu, and Zn, suggest that Layer 2 have below high production regions. Also, high content of allumino silicate components may indicate increased terrigeneous input during the formation of Layer 2. The Layer 2. The Layer 1 has been subjected to little diagenetic influence since the Pliocene.

  • The starting point of the research is the relation between capital restructuring and urban restructuring. The economic restructuring, which has been caused by the economic crisis in the early 1970s in the United States has brought a spatial restructuring at different geographic scales. The degree of the success of urban restructuring is contingent to the local economic and political environments. The local contingent factor such as local politics should not be neglected for investigating the restructuring process. Through the case study of Chicago, the research provides two inconsistencies in applying the structural approach to the local level: first, the lack of the theoretical link between crisis and restructuring; and second, the crucial importance of local politics in shaping urban development.

Longitudinal flowcytometric measurement of respiratory burst activity of neutrophils in patients with pneumonia (폐렴경과 중 순환 호중구의 Respiratory Burst 활성도 변화)

  • Background : Recognition and ingestion of opsonized microorganisms by neutrophils induces the burst of oxidative metabolic activity. Products of the respiratory burst activity provide powerful oxygen dependent killing mechanism. Measurement of respiratory burst activity has been a major indicator of the functional capacity of neutrophils. We determined the respiratory burst activity of neutrophils in patients with pneumonia and observed the changes during the clinical course of pneumonia. Methods: The EDTA blood was drawn from 24 normal controls and same numbers of pneumonia patients. The respiratory burst activity(with the production of $H_2O_2$ which changes nonfluorescent DCF-DA to green fluorescent DCF) in the non-stimulated state and the stimulated state with fMLP and PMA of neutrophils was measured by flowcytometry at day 1, 3, 5, 7 and 9 of admission. Results: The respiratory burst activity of neutrophils was mildly increased by stimulation with fMLP. But there was no statistical significance between normal control and patients with pneumonia. The respiratory burst activity of neutrophils was markedly increased by stimulation with PMA in both groups. There was a significant difference in response to PMA between normal control and patients with pneumonia. The production of hydrogen peroxide from neutrophils was decreased during early course of pneumonia and it was recuperated gradually to normal level in 9 days. Conclusion : Hydrogen peroxide production from neutrophils was suppressed during early course of pneumonia and restored after treatment. It is suggested that the production of oxygen radical in response to PMA stimulation from each neutrophils is decreased rather than increased during the early course of pneumonia.

  • In this experiment American Summer Pairman apple was selected as test fruit in order to study the effect of sub-atmospheric pressure storage in the apple. keeping a certain pressure condition, a new sub-atmospheric pressure system was designed and constructed in which fruits were stored at $25^{\circ}C$ under two different atmospheric pressure conditions such as Normal Atmospheric Pressure (NAP) and Sub-Atmospheric Pressure (SAP). Moreover, they were divided into plots of 5% and 0% of $CO_2$, on the basis of gas composition. Under these conditions, the amount of respiration and ethylene evolution, and the changes of intercellular gas composition and organic components were investigated throughout the storage. The results obtained are as follows: (1) The intercellular gas was exhausted so rapidly by the SAP treatment that the gas equilibrium in the tissues reached within 5 minutes. (2) The amount of respiration was found to be higher in plots of NAP than SAP, and under the conditions, controlling $CO_2$ content, plots of $CO_2$ 5% were lower in the amount of respiration than $CO_2$ 0%. The climateric rise was revealed more slowly in plots of the SAP than NAP. These results indicate that the SAP treatment was an efficient method for controlling the respiration of fruits. Furthermore, these results were also realized with the results of the respiratory quotient (R.Q) and intercellular gas composition. (3) Evolution of ethylene, the ripening hormone in plant, was shown the similar tendency to the climacteric pattern of respiration; at the stage of climacteric maximum, the maximun amount of ethylene was found earlier in plots of NAP than SAP, and post climacteric stage was prolonged in the plots of the SAP compared to those of the NAP. The ethylene concentration in tissue appeared lower in plots of the SAP than NAP, which might suggest that the SAP treatment was caused to restriction of ethylene evolution. (4) Effects of the SAP treatment mentioned hitherto were proved also with the test of the external appearance such as changes of color and freshness, firmness, rotting and weight loss. (5) In the investigation of organic components, vitamin C and organic acids varied less in plots of SAP than NAP. Specially, it was remarkable that the loss of malic acid was least decreased by the SAP treatment. These effects of the SAP treatment were distinctive in the changes of the ratios of malic and citric acid to total acid, and the ratios of free sugar to free acid.

Antioxidant capacity and Raw 264.7 macrophage anti-inflammatory effect of the Tenebrio Molitor (갈색거저리(Tenebrio Molitor)의 항산화능과 Raw 264.7 대식세포의 항염증 효과)

  • The purpose of this paper is to investigate potential anti-inflammatory and anti-oxidant effects of Tenebrio molitor. Macrophage cell response by outside stimulation leads expression of pro-inflammatory cytokines, such as tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), interleukin-6 (IL-6), $interleukin-1{\beta}$ ($IL-1{\beta}$), and trigger expression of genes which are affected by inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), resulting in formation of inflammatory factors like nitric oxide (NO) and Prostaglandin $E_2$ (PGE2). Cell viability was determined by MTT assay. In order to investigate anti-inflammatory agents, the inhibitory effects on the production of lipopolysaccharide (LPS)-induced NO in RAW 264.7 cells were examined. T. Molitor significantly decreased the production of NO in a dose-dependent manner, and also reduced the expression of iNOS, a COX-2 protein. As a result, the levels of protein such as $PGE_2$, iNOS, COX-2 and MARKs were significantly reduced compared to non-treated group in T. Molitor water extract (TDW) treated group. Also, antioxidant effect of T. Molitor were investigated using DPPH, ABTS+ and superoxide anion radical scavenging activity tests in cell-free system. Antioxidant activity of T. molitor was found low in the DPPH radical scavenging test while high in the ABTS+ and superoxide anion radical scavenging activity tests. These results show that TDW could be an effective anti-pro-inflammatory and anti-oxidant agent.

Bioactive Utility of the Extracts from Rhus verniciflua Stokes (RVS) : Biological Function of the Extracts from RVS (옻나무 추출물의 생리활성 이용에 대한 연구 : 옻나무 추출물의 생물학적 기능)

  • Antioxidative effects of the water or ethanol extracts from Rhus verniciflua Stokes (RVS) were measured by protection against hydroxyl radicals in mouse brain tissue culture. In the water extracts from RVS, cell viabilities were estimated 60.0, 66.0, 72.0, 84.0 and 90.0% at addition of 1, 2, 4, 7 and $10{\mu}L$, respectively, compared with GO (20 mU/mL) alone. The cell viability in the ethanol extracts was similarly with water extracts. In the antitumor effects, the results showed that percentages of the HeLa cell death were approximately 24% for 12 hrs, 57% for 48 hrs at addition of 10%/well ethanol extracts respectively. To know inhibition of tumor growth, in vivo, mice (BALB/c) were inoculated with 0.25 mL CT-26 $(1{\times}10^6\;cells/mL)$ subcutaneously. After the generation of tumor, the results of RVS extracts (ethanol, water) injection showed generally that the tumor size in BALB/c was reduced. For physicochemical characterization of the RVS extracts, purified substances of water or ethanol extracts were analized with SDS-PAGE and ICP spectrometer. In electrophoresis, gel showed 2 bands (210, 230 KDa). The results of ICP verified that RVS extracts contain $Cu^{2+}$ in both samples. Conclusively, this substance might be a laccase which has a biological effective function, as a natural bioactive substance.

Changes of Fermentation Characteristics and Sensory Evaluation of Kimchi on Different Storage Temperature (숙성온도를 달리한 김치의 발효 및 관능특성)

  • To investigate the fermentation characteristics of Kimchi which was made at $12{\pm}1^{\circ}C$ and stored at $17{\pm}1^{\circ}C$ and $4{\pm}1^{\circ}C$, pH, total acid, total microbes, lactic acid bacteria cell count, dissolved $CO_2$content, reducing sugars and temperature at the center of Kimchi jar were measured and to know how much Japanese like to have Korean traditional Kimchi, Kimchi samples with 1.0, 2.0 and 3.0% salt contents were prepared and surved to panelist at $4{\pm}1^{\circ}C$. Sensory evaluation of Kimchi in terms of appearance, texture, carbonated mouthfeel, salty taste, sour taste, peppery taste and overall acceptability was done by scoring system with maximum 7 points and was analyzed statistically by SAS program. The results of fermentation of Kimchi which was made at $12{\pm}1^{\circ}C$ and stored at $17{\pm}1^{\circ}C\;and\;4{\pm}1^{\circ}C$ were as follows: pH and total acid content of Kimchi which was stored at $17{\pm}1^{\circ}C$ for 4 days were almost same as those of Kimchi at $4{\pm}1^{\circ}C$ for 48 days. Cell counts of total microbes and lactic acid bacteria in Kimchi which was stored at $17{\pm}1^{\circ}C$ for 2 days and at $4{\pm}1^{\circ}C\;for\;9\;days\;were\;1.5{\times}10^9\;cells/mL\;and\;6.3{\times}10^8\;cells/mL,\;2.0{\times}10^8\;cells/mL\;and\;8.7{\times}10^7\;cells/mL$, respectively. Time and temperature at the center of Kimchi jar during fermentation was took 25 and 35 hours to get to $17^{\circ}C\;and\;4^{\circ}C$ from initial temperature, respectively. Japanese who had experiences in eating Kimchi liked Korean traditional Kimchi with 2.0% of salt content. Sensory evaluation of Japanese consumer on peppery taste and overall acceptability of Kimchi showed significant difference (P<0.05).

Effect of a Herb Mixture (HIM-I) on the Protection of the Hematopoietic-Immune System and Self-renewal Tissues against Radiation Damage (면역조혈계 및 재생조직의 방사선 손상에 대한 생약복합물(HIM-I)의 방호 효과)

  • In previous studies, we evaluated the effect of the 6 energy-tonic or blood-building prescriptions of traditional oriental medicine, and observed that Si-Wu-Tang and Bu-Zhong-Yi-Qi-Tang showed high activity in the protection of the gastrointestinal and hematopoietic organs in irradiated mice. But any of these prescriptions did not show a high activity in the activation of the immune cells. We performed this study to design an herb mixture which protects the self-renewal tissues and also promotes recovery of the immune system against radiation damage. In order to meet all the requirements, we designed a new mixture of 3 edible herbs listed in Korean Food Code. The mixture of Angelim gigas radix, Cnidium officinale rhizoma and Paeonia japonica radix was decocted with hot water, and the activities of the water extract (HIM-I) were evaluated. HIM-1 stimulated the immune cells in a much higher extent than the traditional prescriptions, and promoted dramatically the growth of bone marrow stem cells in vitro. Also, HIM-1 protected digestive and hematopoietic organs against radiation as effectively as the 2 prescriptions, Si-Wu-Tang and Bu-Zhong-Yi-Qi-Tang. On the other hand, it showed high in vitro antioxidative activity that might be considered as a mechanism of the protective effects against radiation. Although the detailed mechanisms of those effects remain to be elucidated, these results indicated that HIM-I might be a useful agent for protection and recovery of body from various risk factor as well as radiation, especially since it is a relatively nontoxic natural product.

Comparison of Temperature-dependent Development Model of Aphis gossypii (Hemiptera: Aphididae) under Constant Temperature and Fluctuating Temperature (실내 항온과 온실 변온조건에서 목화진딧물의 온도 발육비교)

  • The developmental time period of Aphis gossypii was studied in laboratory (six constant temperatures from 15 to $30^{\circ}C$ with 50~60% RH, and a photoperiod of 14L:10D) and in a cucumber plastic house. The mortality of A. gossypii in the laboratory was high in the 2nd (20.0%) and 3rd stage(13.3%) at low temperature but high in the 3rd (26.7%) and 4th stage (33.3%) at high temperatures. Mortality in the plastic house was high in the 1st and 2nd stage but there was no mortality in the 4th stage at low temperature. The total developmental period was longest at $15^{\circ}C$ (12.2 days) in the laboratory and shortest at $28.5^{\circ}C$ (4.09 days) in the plastic house. The lower threshold temperature at the total nymphal stage was $6.8^{\circ}C$ in laboratory. The thermal constant required to reach the total nymphal stage was 111.1DD. The relationship between the developmental rate and temperature fit the nonlinear model of Logan-6 which has the lowest value for the Akaike information criterion(AIC) and Bayesian information criterion(BIC). The distribution of completion of each development stage was well described by the 3-parameter Weibull function ($r^2=0.89{\sim}0.96$). This model accurately described the predicted and observed outcomes. Thus it is considered that the model can be used for predicting the optimal spray time for Aphis gossypii.

An Ecological Comparison of Benthic Macroinvertebrate Community in Downstream Region of Large Dams (대형댐 하류지역 저서성 대형무척추동물 군집의 생태학적 비교)

  • Benthic macroinvertebrates were investigated in Yongdam-dam and Hapcheon-dam resions from June to October 2011, Korea. Yongdam-dam and Hapcheon-dam are geographically contiguous, but they are classified as other water system. Experiment site(YE-1, 2, 3; HE-1, 2, 3) which is thought to be affected by dam, control site(YC-1, 2; HC-1, 2) which is thought not to be affected by dam were selected. Species composition, macroinvertebrate communities, composition of the functional feeding groups, habitat orientied groups and community stability were assessed Yongdam-dam and Hapcheon-dam regions. Ten sites were selected for quantitative(Surber sampler $30cm{\times}30cm$) of benthic macroinvertebrates. As a results, a total of 6,369 individuals including 69 species, 33 families, 12 orders, 6 classes and 4 phyla were recognized in Yongdam-dam region. Also, a total of 5,728 individuals including 81 species, 44 families, 13 orders, 5 classes and 4 phyla were recognized in Hapcheon-dam region. Dominance index was 0.27~0.50(mean${\pm}$SD $0.38{\pm}0.09$), diversity index was 2.22~2.97($2.67{\pm}0.29$), evenness index was 0.63~0.76($0.72{\pm}0.06$) and richness index was 4.43~7.06($5.69{\pm}0.99$) in Yongdam-dam region. Dominance index was 0.40~0.81($0.59{\pm}0.18$), diversity index was 1.40~2.39($2.00{\pm}0.43$), evenness index was 0.38~0.68($0.56{\pm}0.13$) and richness index was 4.04~5.80($4.95{\pm}0.70$) in Hapcheon-dam region. In the functional feeding groups, filtering-collectors and gathering-collectors were the highest in the whole sites. In the habitat orientied groups, burrowers, clingers and swimmers were considerably occupied in all sites. As a result of community stability analysis, experiment sites has been identified much as species high resistance and resilience to environmental changes in Yongdam-dam. Control sites has been identified much as species low resistance and resilience to environmental changes in Hapcheon-dam. Species belonging to the Igroup is considered to be important in the river ecosystem stability of large dams downstream areas.
