Efficacy of Suppression of Phytophthora Blight of Red Pepper Caused by Phytophthora capsici by Treatment with Antagonistic Trichoderma harzianum DYMC in Greenhouse (온실에서 길항미생물 Trichoderma hazianum DYMC 처리에 의한 고추 역병 억제 효과)

  • To control Phytophthora blight of red pepper biologically caused by Phytophthora capsici, we developed Trichoderma harzianum DYMC for commercial product. DYMC was storage at room temperature and was investigated their population every 3 months for 1 year. For investigating the dynamic population of T. harzianum in the pot soils, we applied powder and suspension applications with DYMC, and then investigated for 95 days. The efficacy of powder and suspension applications of DYMC for control of Phytophthora blight of red pepper and plant growth were investigated for 50 days in greenhouse experiment. The population of T. harzianum was decreased at the room temperature for 1 year but there was not statistically significance. After soil treated in the pot with DYMC, the population of Trichoderma spp. was the highest when DYMC powder at 5 g was applied to mix with pot soil, and the population was deceased significantly among treatment means as time goes by ($R^2=0.76$, F=10.5960, P=<.0001). Incidence of Phytophthora blight of, red pepper was significantly reduced among treatment means on 50th day after treated with DYMC ($R^2=0.82$, P=16.4758, P=<.0001). Disease control value was the highest at 62.5% when DYMC powder at 5 g was applied to mix with pot soil. No significant difference (P=0.05) of effects of plant and root growth showed by treated with DYMC on 60th day, except stem. Mixing the application of DYMC powder with soil to control Phytophthora blight of red pepper was greater than suspension application to dilute with water. DYMC could be used as an effective biocontrol agent to control Phythophthora blight of red pepper.

Changes of Mating Type Distribution and Fungicide-resistance of Phytophthora infestans Collected from Gangwon Province (강원지역 감자 역병균 Phytophthora infestans의 교배형 및 약제저항성 변화)

  • Potato late blight caused by Phytophthora infestans was the most constrain disease at potato cultivation areas. The mating type distribution and fungicides response of P. infestans were investigated to elucidate the changes of pathogen from Gangwon province. On the fungal isolates in 2006, 58.7% were A1 mating type and 41.3% were A2 mating type. In 2007, A1 mating type isolates increased to 93.3% and A2 mating type isolates were collected from Jinbu areas as much as 6.7%. About 234 isolates analysed for metalaxyl response, the results was resistance 73.7%, intermediate 18.8% and sensitive 7.5% in 2006. And it was resistance 59.4%, intermediate 4.0% and sensitive 36.6% in 2007. It meant that mating type distribution and fungicide response were very different over the collection sites. Minimal inhibition concentration (MIC) of dimethomorph examined with 126 isolates in 2006 and 106 isolates in 2007. MIC over $1.0\;{\mu}g/ml$ was 56.3% in 2006 and it was 3.8% in 2007. The average $EC_{50}$ value of P. infestans was $0.37\;{\mu}g/ml$ in 2006, but it decreased to $0.12\;{\mu}g/ml$ in 2007. Fungicides response and pathogenesis of P. infestans should be monitored continuously to enhance the chemical efficacy at potato fields.

Determination of Application Rate of Composted Pig Manure for Wetland Rice (논토양에서 돈분톱밥퇴비 시용량 결정에 관한 연구)

  • A study was conducted for the determination of application rate of pig-dung composted with sawdust (referred as pig manure hereafter) for wetland rice. The study involved the field experiments with the cultivation of rice under different rates of application of pig manure in combination of different rates of chemical fertilizers, in a wetland rice soil. The field experiment involved following treatments : (I) Without fertilizer, (II) Standard rate of chemical fertilizers based on soil testing($98-73-71kg\;ha^{-1}$ as $N-P_2O_5-K_2O$), (III) $2.1Mg\;ha^{-1}$ of pig manure $>+80-37kg\;ha^{-1}$ of $N- K_2O$ as chemical fertilizer(Less $N-P_2O_5-K_2O$ contained in the compost), (IV) $4.2Mg\;ha^{-1}$ of pig manure+ $62-3kg\;ha^{-1}$ of $N-K_2O$as chemical fertilizer(Less $N-P_2O_5-K_2O$ contained in the compost), (V) $10Mg\;ha^{-1}$ of pig manure+ Treatment(II), (VI) $20Mg\;ha^{-1}$ of pig manure +Treatment(II). Number of tillers in treatment (I) were higher than other treatments in tillering and panicle formation stage. After heading stage, treatments (V) and (VI) have higher number of tillers, but treatment (III) and (IV) have fewer number of tillers during all growing stage. Uptake of NPK in rice plants was higher in treatment (VI), but the efficiency of N, P and K was higher in treatment (I), (III) and (IV). The yield of unhulled rice were in order of tretments (VI)>(V)>(IV)>(II)>(III), although the difference was not statistically significant. Inorganic nitrogen, available P and exchangable K contents in soil were highest at tillering stage in all treatments and became low from panicle formation to harvest stage. Available P in soil was increased by the application of pig manure upto 20 cm depth. Exchangeable cation contents in 40 to 60 cm soil depth was much higher in treatment (VI) than in other treatments. Treatment (V) and (VI) showed much higher losses of N. $P_2O_5 $ and $K_2O$ than other treatment. Though treatment (VI) tended yield higher than in other treatments, showed lodging and occurrence of leaf and neck blast in this treatment. Yield of unhulled rice in treatment (IV) was not significant statistically and reduced nutritional losses. It is conclude that treatment (IV) seems to be the most reasonable one for the application of pig manure in combination of chemical fertilizers.

Evaluation of No-tillage Rice Cover Crop Cropping Systems for Organic Farming (벼 유기농업을 위한 무경운 피복작물 작부체계 평가)

  • The objectives of this study were to evaluate no-tillage cover crop cropping systems for organic farming in paddy. The experiment was performed at Ihyeon series (silt loam: 9.1% sand, 73.0% silt and 17.9% clay) which affected by different management practices. Planthopper population per 20 plant was significantly higher 65.3 for conventional tillage with chemical compounds (control) compared with 3.4~9.6 for no-tillage treatments (without rice straw or green manure, amended with rice straw, hairy vetch, rape, rye, and Chinese milk vetch). Also, disease severity of sheath blight was significantly higher 10.5% for control compared to 0.7~2.9% for no-tillage treatments. Four weed species, namely Monochoria vaginalis, Ludwigia prostrata, Rotala indica, and Aneilema keisak occurred in no-tillage paddy, whereas Monochoria vaginalis occurred in control only. The pH, available phosphate, and microbial biomass C in paddy were steeply decreased in response to submerging, but increased at first heading stage. Soil $NH_4$-N content at first heading stage was significantly higher in no-tillage treatments compared with control. The grain yield was significantly higher in 4.30 Mg $ha^{-1}$ for control than other treatments. Meanwhile, rice productivity was significantly higher in 2.69 Mg $ha^{-1}$ for no-tillage amended with Chinese milk vetch compared to other no-tillage treatments. The number of panicle per plant, grain number perpanicle, and percent ripened grain were highly related for increasing the yield of rice. These results show that Chinese milk vetch was optimum cover crop for organic farming in no-tillage paddy.

Early Matured Pear Cultivar 'Supergold' with High Quality and Greenish-white Skin for Overseas Trade (조생종 고품질 녹백색 배 '슈퍼골드')

  • Pear cultivar 'Supergold' (Pyrus pyrifolia var. culta Nakai) was originated from the cross between 'Chuwhangbae' and 'Manpungbae' with the aims of improving the fruit quality of 'Chuwhangbae' cultivar at Pear Research Station of National Institute of Horticultural & Herbal Science, Rural Development Administration in 1994. 'Supergold' was preliminarily selected in 2002 and named in 2008. The tree shows a vigorous growth habit and semi-spread characters like as 'Manpungbae'. Furthermore, it has sufficient flowers and carries abundant pollen grains, so it can also be used as a pollinator. 'Supergold' is highly resistant to black leaf spot (Alternaria kikuchiana) in the field condition. The optimum harvest time is around Sep. 11th, which is ahead of 'Whangkeumbae' about 5 days in the harvest period. The fruit shape is oblate and fruit skin color is greenish-white at harvesting time. The average weight of fruit is 570 g, and the soluble solids content is $13.6\;^{\circ}Brix$. The flesh is very soft and juicy, and renders good eating quality. Shelf life is about 6 months under the cold storage condition. To determine the self-incompatibility (SI) genotype of 'Supergold' pear cultivar, it was crossed with other cultivars of which SI genotypes have already known. The result of cross-pollinations of 'Supergold' with other cultivars showed relatively high rates of fruit set from 64.5% to 91.0%, except for the cross with pollens of 'Nijisseiki' that represented only 28.8% of fruiting rate. Although sometimes the stigma of 'Supergold' crossed with 'Hayatama', 'Chojuro', and 'Nijisseiki' showed malformed pollen tube tips, 'Supergold' is generally supposed to have cross-compatibility with all other pollen donor cultivars. It is considered that the S-allele of 'Supergold' is $S_3S_4$, which is based on the result of PCR-RFLP.

  • In year 2003, a soybean(Glycine max) sample showing severe dwarfing symptom was collected from a farmers' field in Cheongsong in Korea. The results from the diagnosis of the sample by RT-PCR revealed that it was infected by Soybean dwarf virus(SbDV), SbDV-L81. This study could be the first report of the occurrence of the virus in Korea. To further characterize the virus, the partial nucleotide sequence of the genomic RNA of SbDV-L81 was determined by RT-PCR using species-specific primers. The sequences were analyzed and subsequently compared to previously characterized strains of SbDV based on the pattern of symptom expression and vector specificities. The intergenic region between ORF 2 and 3 and the coding regions of ORF 2, 3 and 4 were relatively similar to those of dwarfing strains(SbDV-DS and DP) rather than those of yellowing strains(SbDV-YS and YP). Likewise, the result from the analysis of 5'-half of the coding region of ORF5 indicated that SbDV-L81 was closely related to strains(SbDV-YP and DP) transmitted by Acyrthosiphon pisum. These data from the natural symptom and the comparisons of five regions of nucleotide sequences of SbDV suggested that SbDV-L81 might be closely related SbDV-DP.

Screening of Resistant Chinese Cabbage Cultivar against Clubroot (Plasmodiophora brassicae) for Cultivation in Highland (고랭지 배추재배를 위한 뿌리혹병 저항성 품종선발)

  • This study was conducted to select the cultivars and strains which are suitable in cultivation of the Chinese cabbage having good horticultural characteristics and the resistance to clubroot caused by Plasmodiophora brassicae, which is a widespread disease that causes serious problems in many Brassica growing areas. Thirty four cultivars and nineteen strains of the Chinese cabbage were planted in the contaminated fields in highland and investigated the wilting symptom due to the initial disease occurrence and high temperature. Disease occurrence rate and severity of clubroot and the horticultural characteristics were also examined. According to the results, Bulamplus, Kangryeokyeoleumbaechu among cultivars of Chinese cabbage, SP-05, SP-02, CR DB 50, and CR DB 29 among strains of Chinese cabbage were resistant to hot weather by showing delayed above-ground symptom (wilting) of clubroot in 30 days after transplanting. In case of below-ground clubroot rate, Daetong, super CR Kimi 85 and super CR Hiroki among the cultivars of Chinese cabbage were resistant to clubroot in 60 days after transplanting. When fresh weight of Chinese cabbage among cultivars were compared, super CR Hiroki of over 2,500 g was the highest among other cultivars. Although the resistant and susceptible reactions of Chinese cabbage cultivars and strains against clubroot disease were somewhat different from the above-ground symptoms (wilting) of clubroot in 30 days and below-ground symptoms of clubroot in 60 days, we recommend that super CR Hiroki, CR Kimi 85, CR Janggun among cultivars of Chinese cabbage and CR DB 50 and CR DB 29 among strains of Chinese cabbage will be more suitable in cultivation of Chinese cabbage in highland.

Seed Longevity of Rice Germplasm in the National Agrobiodiversity Center (종자은행 보존 벼 유전자원의 생태형별 종자수명)

  • The purpose of this study was to know the seed longevity of rice (Oryza sativa L.) germplasm for effective viability monitoring. The longevity was determined via germination tests of 3,066 accessions of rice germplasm from the National Agrobiodiversity Center, Rural Development Administration, Korea. The rice germplasm accessions have been conserved at a mid-term storage ($4^{\circ}C$, 30% RH) in plastic bottle containing dehydrated (blue) silica-gel and long-term storage ($-18^{\circ}C$, 35% RH) in hermetically sealed metal can on either sides for 25~26.5 years. The final germination percentages of 3,066 rice germplasm accessions of $6.5{\pm}1.0%$ seed moisture content with 94% initial germination stored at $4^{\circ}C$ for 26.5 years declined to 47% while at $-18^{\circ}C$ for 25 years maintained high germinability as 93%. Germination time courses, which represent the average performance of rice ecotypes stored at $4^{\circ}C$ and 30% RH, were fitted regression equation, to calculate the time at which germination characteristically declined to 50% ($P_{50}$). These $P_{50}$ values of Indica, Japonica, Javanica and Tongil type in rice were 39.9, 22.9, 25.4 and 31.8 years, respectively. The rice germplasm stored at $4^{\circ}C$ could be clustered in 4 groups using quartile of final germination after 26.5 years storage. The seed longevity ($P_{50}$) of each group was estimated by regression equation of changed germination percentages according to storage periods. The $P_{50}$ values of group I, group II, group III and group IV were 21.1, 23.6, 30.0 and 75.7 years.

Effects of Dietary Silkworm Droppings on Quality Characteristics of Pork Loin (잠분(蠶糞) 급여가 돈육의 품질 특성에 미치는 영향)

  • This study investigated the effects of silkworm droppings (SWD) added diet feeding on quality characteristics of pork loin meat Pigs were divided into 7 treatment groups (10 pigs/group) and subjected to one of seven treatment diets $(0,\;1\%,\;2\%\;and\; 3\%\;SWD\;for\;4 weeks,\;1\%\;2\%\;3\%$ SWD for 8 weeks, SWD diets; total fed diets) before slaughter. Pork loin were collected from the animals (110kg body weight) slaughtered at a commercial slaughterhouse. Pork loin meat were packaged aerobically and then stored at $4^{\circ}C$ for 2, 5, 8, or 12 days. Samples were analyzed for general composition, texture characteristics and physico-chemical properties. pH value was not significantly different between the control and SWD treatment groups. pH of control and SWD treatment groups were increased as the storage period passed General composition, cooking loss and texture was not significantly different between the control and SWD treatment groups. WHC of SWD treatment group was higher than that of control group. WHC of control and SWD treatment groups were significantly decreased as the storage period passed (p<0.05). Meat and fat color (CIE $L^{\ast}$, $a^{\ast}$, $b^{\ast}$) were no significant differences between the control and SWD treatment groups. $L^{\ast}$ and $b^{\ast}$ value of meat color were decreased as the storage period passed There was a not significantly difference in shear force value among control and SWD treatment groups. Shear force value was decreased significantly during storage in all treatment groups. In all results, dietary SWD-supplementation was not affected in general components, texture characteristics and physico-chemical properties. It is suggested that dietary SWD-supplementation could produce of high quality pork.

Effect of Phosphorous Acid on Control of Phytophthora Blight of Red Pepper (고추 역병에 대한 아인산 (phosphorous acid)의 방제 효과)

  • Control effects of phosphorous acid were investigated on Phytophthora blight of red pepper plants in greenhouse and field. In vitro test, the mycelial growth of Phytophthora capsici was inhibited by the phosphorous acid more than 97% at 1,000 ${\mu}g$ and 10,000 ${\mu}g$ a.i./mL in the liquid and solid culture, respectively. At concentration of 10 ${\mu}g$ a.i./mL of phosphorous acid, in the liquid culture the mycelial growth of P. capsici was inhibited 46.2%, however inhibited only 4.9% on the soild culture. Zoosporangial formation was also inhibited 89.1% by phosphorous acid at 100 ${\mu}g$ a.i./mL. Phosphorous acid affected more zoosporangial formation of P. capsici than its mycelial growth. At the concentrations of 10,000, 1,000 and 100 ${\mu}g$ a.i./mL of phosphorous acid, germination of zoosporangia was inhibited 100, 84.3 and 44.2%, respectively. Mycelial growth and zoosporangial formation of P. capsici were little affected at the concentration of 10 ${\mu}g$ a.i./mL of phosphorous acid. Cermination of zoosporangia was also little affected at this concentration, however growth of the germ tubes was inhibited and the abnormal mycelial growth was observed. Phosphorous acid suppressed the incidence of Phytophthora blight of red pepper plants up to $77.0{\sim}62.0%$, in greenhouse. Phosphorous acid suppressed the incidence of Phytophthora blight of red pepper plants up to 54.0% at the conventional culture in field. Treatments of phosphorous acid increased up to 113% in height, 135% in number of fruit, and 129% in weight of fruit.
