Acute Oral, Pulmonary and Intravenous Toxicity/Pathogenicity Testing of Burkholderia pyrrocinia CAB08106-4 of in Rats (랫드를 이용한 Burkholderia pyrrocinia CAB08106-4의 급성경구, 호흡기, 정맥독성/병원성시험)

  • Burkholderia pyrrocinia CAB08106-4 has an anti-bacterial effect on Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp.. It is an environmentally friendly microbial product that prevents and controls a variety of phytopathogens involving Garlic White Rot caused by Sclereotium cepivorum and Sclereotium sp.. The aim of this study was to assess and to compare the pathogenicity of Burkholderia pyrrocinia CAB08106-4 by single exposure of rats through several routes such as oral, intranasal and intravenous. For the acute toxicity / pathogenicity study, the animals were sacrificed on days 3, 7, 14 and 21, and macroscopically observed their organs to examine the numbers of internally-retained pesticidal microbes. Clinical examinations were performed daily during administration period, and body weight gain was evaluated. In the study, no clinical sign, weight gain and mortality were observed in relation to the administration of test article. The significant changes of internal/external microbes by test article were not detected. The pathological findings in relation to the administration of the test article in the necropsy were not observed. It could be concluded that the microorganism was not toxic or pathogenic in rats via oral, intranasal and intravenous route.

Effect of Gamma Irradiation on the Expression of Gene Endoding Metalloprotease in Vibrio vulnificus (감마선 조사가 vibrio vulnificus의 Metalloprotease 유전자 발현에 미치는 영향)

  • To check the microbiological safety with respect to increased virulence of surviving pathogens after irradiation, in this study, the transcriptional change of vvp gene encoding metalloprotease, which is one of the typical virulence factors of Vibrio mulnificus, was monitored by real-time PCR during the course of growth cycle after reinoculation of irradiated Vibrio. When V. vulnificus was exposed to a dose of 0.5 and 1 kGy, the lag period before growth resumption of sub-cultures became longer than non-irradiated counterpart as increase of irradiation dose. In the case of non-irradiated culture, the transcription of vvp was significantly activated at 15 h after inoculation, when bacterial growth reached the stationary phase, and the highest level of pretense activity (686 U/mL) was measured at the same time. Interestingly, vvp expression of irradiated Vibrio was turned up earlier than non-irradiated Vibrio during the mid log phase of growth, whereas these rapid induction of vvp expression from irradiated cells didn't result in an increase of metalloprotease production. When Vibrio was irradiated at 0.5 and 1 kGy, the protease activities peaked at 18 h after inoculation and the levels of activities were lower 1.2- and 1.4-fold, respectively, compared to the non-irradiated counterpart. Results from this study indicate that gamma radiation is not likely to activate the virulence ability of surviving Vibrio.

Effects of Biologically Active Materials Prepared for Several Minerals and Plants on the Growth of Rumen Microbes (무기물성 및 식물성 생리활성 물질이 반추위 미생물의 성장에 미치는 영향)

  • In order to know the effects of scoria, germanium, charcoal, ginger, stevia, and CLA(Conjugated Linoleic Acid) as biologically active materials on pathogenic microbes and rumen anaerobic microbes, the growth rate of pathogens (including Escherichia coli O157, Salmonella paratyphi, Listeria monocytogenes and Staphylococcus aureus) and in vitro lumen microbial growth, gas production, ammonia concentration, carboxymethyl-cellulase (CMCase) activity, and microbial populations were investigated. The growth of pathogenic microbes was inhibited by the supplement of 0.10% ginger. Ginger had powerful antimicrobial properties on all the pathogens used in this experiments. Additionally in the antibacterial assay by paper disc method, we could observe the clear zone of similar area with the positive control(antibiotics) for E. coli as applied with the 10% stevia or the 10% CLA only. The supplements of ginger, stevia and CLA in vitro rumen fermentation inhibited populations of rumen bacteria and protozoa. Particularly supplement of ginger resulted in remarkable reduction of the protozoa population, which means it might serve as a source inhibiting material of methane creation in the rumen.

  • The purpose of this study was to evaluate the effect of dietary ceramic powder on laying performance, Pathogenic bacterial (E. coli, salmonella) counts in caecal contents and excreta, malodorous substances $(NH_3,\;H_2S,\;VFA)$ in excreta and fatty acid composition of e99 yolk in laying hens. Three hundred sixty layers at 34 weeks of age were divided into three groups of 120 hens each and each group was fed diets containing 0 (control), 0.4, or 0.8 ceramic powder for six weeks. It is concluded that $0.4\%$ ceramic powder supplementation of diets decreased of E. coli, salmonella counts in the intestine and emission of $NH_3$ and VFA gas from excreta, and improved the laying Performance and ratio of egg yolk fatty acids contents.
