Studies on the Naringinase Produced by Aspergillus nidulans (Part I) Purification and Characteristics of Naringinase (Aspergillus nidulans 가 생산하는 Naringinase 관한 연구 (제2보) Naringinase의 정제 및 물리화학적 성질)

  • Naringinase extracted from the culture media of Aspergillus nidulans was purified, and the activity was proved to be stronger by 781 fold in the part of precipitation with ammonium sulfate, and column chromatography using DEAE-Sephadex A-25 and Sephadex G-100. Two fractions which had the same enzyme activity were isolated by the purification. Both fractions showed the highest enzymes activity under the reaction conditions of pH 5.0 and 40$^{\circ}C$. Molecular weight of fraction I and fraction II were estimated as 78,000 and 26,000 respectively. This indicated that fraction I would be trimer of fraction II. The enzyme was inhibited by glucose and rhammose, and the Km value was calculated to be 2.3${\times}$ 10$\^$-6/ g/ml.

  • Naringinase from Atpergillus nidulans was immobillized on DEAE-Sephadex A-25 and its characteristics were studied. The optimal conditions for the preparation of the immobilized enzyme were as follow; optimal pH, incubation time and the suitable amount of enzyme were 6.0, 30 min. and 110 units per gram of the dried ion exchage resin, respectively. The optimal pH of the immobilized enzyme was higher than that of the native enzyme. The optimal temperature increased from 4$0^{\circ}C$ to 5$0^{\circ}C$. The heat and pH stability of the immobillized enzyme were better than those of the native enzyme. No significant difference in the Michaelis constant was detected. Activation energy of the immobilized enzyme was 7.96 Kcal/mole, and the apparent Michaelis rate equation was used to describe the action of this material. The degree of hydrolysis was dependant on the flow rate at low rate of perfusion through the column. As the flow rate increased, the value of the apparent Km decreased.

  • SiOC film was deposited by the chemical vapor deposition using BTMSM and oxygen mixed precursor. The characteristic of SiOC film varied with increasing of the gas flow rate ratios. The dielectric constant was obtained by C-V measurement using the structure of metal/SiOC film/Si. The space effect due to the steric hindrance between alkyl group at terminal bond of Si-$CH_3$ made the pores, and increased the thickness. However, the SiOC film due to the lowering of the polarization decreased the thickness and then decreased the dielectric constant. After annealing process, the dielectric constant decreased because of the evaporation of the OH or $H_2O$ sites. The thickness was related to the lowering of the dielectric constant by the reduction of the polarization and the thickness decreased with the decrease of the dielectric constant. The refractive index was in inverse proportion to thickness. The trends of the thickness and refractive index did not change after annealing.

  • Naringinase produced from Aspergillus nidulans was immobilized in acrylamide gel by the entrapping method and its characteristics were studied. Optimum acrylamide concentration was 10%, but N.N'-methylene bisacrylamide concentration had no influence on the final enzyme gel activity. The suitable amount of enzyme dissolved in the polymerization reaction mixture was 126 units/ml. Optimum pH of immobilized enzyme was 5.0 which was the same as that of free enzyme. However, immobilized enzyme showed a higher optumum reaction temperature, markedly increased pH and temperature stability. In a packed-column reactor, the observed reaction rate was increased proportionally to flow rate up to 5ml/min., but independent above 6ml/min.. Activation energy of the immobilized enzyme was 13.01 Kcal/mole, and the energy required for the thermal inactivation was 39.4 Kcal/mole. The apparent Km for 100 mesh gel was $7.23{\times}10^{-3}$ mole.

Rheological Properties of Doen-Jang(Korea Fermented Soybean Paste) Suspensions (된장 현탁액의 리올로지적 성질)

  • The rheological properties of soybean paste suspensions as affected by concentrations and heating were studied. All suspensions of soybean paste showed thioxotropic plastic behavior. The concentrated suspensions below the ratio of water to soybean paste, 1.25 exhibited Figure-eight phenomena which were presented by two Intersections of the up curve with the dorm curve of shear rate. The dilute suspension (soybean paste: water=1:2) showed H type hysteresis loop at low temperature. But increasing the heating temperature above $60^{\circ}C$, the hysteresis loop exhibited a Figure eight phenomenon. Consistency index of the suspensions increased remarkably by heating and showed the highest value around isoelectric point of the protein. It was suggested that the proteinous nitrogen might be the main component taking part in viscous behavior under different pH and heat treatment.

  • The growth characteristics of Methylomonas methanolica YUFE 101, isolated from sewage samples, have been studied. conclusions of the study were; (1) Optimum cultivation pH and temperature are 6.3 and 32.5$^{\circ}C$ respectively (2) The specific oxygen uptake rate was 332 $\mu$ι/mg-dry weight/hr. (3) The maximum specific growth rate was 0.19 h $r^{-1}$ and celluar yield was 0.43 g-dry cell/g-methanol in batch culture, (4) The maximum biomass productivity achieved was 0.21 g-dry cel1/ι/hr at a dillution rate of 0.1 h $r^{-1}$ during continuous cultivation. (5) The contents of crude protein and total nucleic acid in the dry cell were 73 % and 12 % respectively.

  • Fungi which produce red pigment were isolated from tapioca chips, Korean Koji, air, and plant leaves. Among the Fungi, T-1 strain was selected as test strain. This strain was identified as a Monatcus sp. by the morphological propeties. Various culture conditions, and physical and physiological characteristics of red pigment were studied. According to the studies of culture conditions, optimum condition was found to be pH 6.5; 4 days of incubation; temperature, 32~33 c: 3.5% of Tapioca chips powder as carbon source, 0.2% of sodium nitrate as nitrogen source and 100 ml of medium in the 500 ml Erlenmyer flask at a rotary shaker (rpm180) as aeration condition. Also effective levels of vitamins, amino acids and inorganic compounds was found to be l$\mu\textrm{g}$/ml of folic acid and niacin; 0.3% of L-arginine, L-glutamic acid and L-proline; and 0.001% of manganese dioxide giving good results.

  • Formulation and production performance of proposed high nutriton-low cost (HNLC) supplementary foods for children in Korea were studied for Wonseong County Comprehensive Nutrition Program. The basic formulas recommendable from the experimental results on the nutritional value, organoleptical qualities and mechanical performance using MFM-KIST extrusion cooking system were considered as CSS-3, CSS-4 and BSS-4. Initial priority of application soybean was with CSS-4(68% corn flour, defatted soybean flour 20%, sesame 2%, corn oil 4%, sugar 4%, salt 1%, vitamins, mineral and other additives 1%). All these formulas as on rioted snack type-HNLC supplementary foods were found in the acceptable range of mechanical, organoleptical and economical point of view.

Prediction of Sucrose Hydrolysis Rate using Equivalent Time at A Reference Temperature under Regular Temperature Fluctuations (규칙적인 온도변화에서 표준온도 상당시간을 이용한 Sucrose 가수분해속도의 예측)

  • A simple approximate model using equivalent time at a reference temperature ($t_{eq}$) was derived to predict quality changes caused by temperature fluctuations. The validity and effectiveness of this model have been assessed with experimental data of sucrose hydrolysis. Kinetic parameters of sucrose hydrolysis were estimated by one step method using equivalent time at a reference temperature with linearly increasing temperature profile. Sucrose hydrolysis was a first order reaction, and the activation energy was 25.84 kcal/mol. The extent of sucrose hydrolysis of liquid model system under accelerated test with sinusoidal temperature fluctuations were determined. The proposed model yielded accurate prediction with the correlation coefficient in the range of $0.92{\sim}0.99$.

The Effect of Organic Acids on Dewatering Efficiency of Soybean Milk Residue by Hydraulic Press (유기산이 두유박의 압착 탈수율에 미치는 영향)

  • The efficiency of dewatering of soybean milk residue was improved by hydraulic pressing after pH adjustment to 4.5 with organic acids such as acetic and lactic acids. Water content of raw soybean milk residue was reduced from 80% to 72% by pressing after pH adjustment, while only to 78% by the conventional hydraulic press. The water content of the residue after pH adjustment was further reduced to 63% by hydraulic pressing with ohmic heating. The pH adjustment facilitated separation of cake from the filter cloth and reduced the solid content of the expressed liquid from 10 to 3%.
