Development of RAPD Marker Related to Brown Planthopper Resistance Gene Derived from Rice Cultivar, Cheongcheongbyeo (청청벼에서 유래한 벼멸구 저항성관련 RAPD Marker의 개발)

  • This study was carried out to select DNA markers closely linked to brown planthopper (BPH) resistance gene originated from a rice cultivar 'Cheongcheong­byeo'. For the mapping of resistant gene to BPH, a doubled-haploid (DH) population was developed by anther culture of $F_1$ plants from a cross 'Cheongcheong­byeo/Nagdongbyeo'. In BPH bioassay and marker screen­ing for the DH population, the segregation of resistant and susceptible plants to BPH fitted to a 1:1 ratio. A total of 310 RAPDs of 520 markers showed polymorphism in parental survey using 'Cheongcheongbyeo' and 'Nag­dongbyeo'. In the analysis of relationship between BPH resistance and marker pattern for 40 DH lines, the OPE16 produced a specific dominant fragment, 700 bp, which was closely linked with BPH resistance gene of 'Cheong­cheongbyeo'. Based on the linkage analysis using 7 markers, BPH resistance of 'Cheongcheongbyeo' was mapped on chromosome 12, which was closely linked with $OPE16_{700}$ at a distance of 4.6 cM.

Some Aspects of Population Dynamics of Rice Leafhoppers in Korea (한국에 있어서 벼멸구$\cdot$매미충의 개체군동태에 관하여)

  • Regional differences in economic importance of the four species of rice leafhoppers, viz., Laodelphax striatellus, Nephotettix cincticeps, Nilaparvata Iugens and Sogatella furcifera, were analyzed and discussed based on population surveys conducted in Suweon and in southern and south-western coastal areas. The economic importance of L. striatellus and N. cincticeps, which are found throughout the year in Korea, seems to be less in middle regions. This is believed due to effective natural control through severe winter climate, cultural practices in rice and barley growing, and natural enemies such as spiders. However, the economic importance of these two species is significantly greater in southern regions with a less severe winter climate, and where cultural practices in rice and barley favour synchronization between the life cycles of insects and the seasonal succession of host plants. with regard to N. lugens and S. furcifera, which cannot overwinter in Korea, initial populations are the result of long distance migration across the East China Sea. Weather elements related to their migration result in more abundant immigrants of these two species in southern than in middle regions. Thus, different policies are needed for the integrated control of rice leafhoppers in middle and southern Korea.

The Effects of the Application Time of Buprofezin 25% WP on the Density of the Brown Planthopper(Nilaparvata lugens Stal) (Buprofezin 25% WP의 처리시기와 벼멸구(Nilaparvata lugens Stal) 밀도억제효과)

  • These studies were conducted to investigate the biological effects of buprofezin 25% WP on the adult of the brown planthopper (BPH) and its control effects on the BPH population, when buprofezin was treated on different days after the introduction of the BPH adult to the potted rice. Also age structures of the BPH population at the time of buprofezin treatment were observed. The results were as follows: Buprofezin (7.0 g a.i./l0 a) could reduce the longevity and fecundity of the newly emerged female but did not affect the biology of older female, and it did not affect the biology of the male. The larval densities at the time of buprofezin treatment was reduced to nearly zero after treatment. The higher the residual effect and the proportion of larvae at the time of buprofezin treatment were, the higher its con¬trol effect on the BPH population was. In case of buprofezin 7.0 g a.i./l0a treatment, the time interval of treatment that could show the highest control effect was about 10 days (from 18 days after the adult introduction to the adult emergence days of next generation).

  • This study was carried out to analyst the damage due to the brown planthopper, Nilapalvata lugens Stal, at different stages(maximum tillering, booting, heading, and milking) of rice. The results are as follows: The population density of the migrating and the first generations of the brown planthoppers seemed not to be large enough to cause significant effects on the plant height and the number of tillers, while the second and the third generations caused significant reduction in 1000 grain weight(Yl) and the filled grain percentage$(Y_2)$. They were very closely related to the population density(X) of tile second generation of the BPH; $$Y1(g)=25.602-0.0213X(r=-0.5205^{**})$$ $$Y2(\%)=76.97-0.1370X(r=-0.4820^{**})$$ The relationship between rice yield (Yg) and the accumulated total density (X) of BPH per hill by the time of harvest; $$Y(g)=24.694-0.0576X(r=-0.6959^{**})$$ Since the source of the outbreak BPH is the first generation, it nay conclude that the detailed information on the population status of the first generation; density, age components, ratio of the brachypterous form, are very important in relation to management of the pest.

Effects of Carbofuran Soil Incorporation on the Early Occurring Rice Insect Pests and the Brown Planthopper (본논초기해충군과 벼멸구에 미치는 Carbofuran 토양혼화처리의 영향)

  • Control effects of carbofuran soil incorporation just before transplanting on the early occur-ring rice insect pests were studied in the rice field. Also, its residual effects on the brown planthopper (BPH) were studied by pot experiments. Carbofurn soil incorporation in late May was much more effective in controlling the early occurring rice insect pests such as small brown planthopper, green leafhopper and rice stem borer than carbofuran broadcasting or diazinon (3G) + BPMC (50% EC) treatment in mid June. Residual effects of carbofuran soil incorportaion on the female adult BPH decreased ca. a half one month after treatment. However, its residual effects on the next generation of the BPH lasted much longer; control effects on the next generation were above 90 and 70% on 37 and 46 days after treatment, respectively. From these results, it is thought that carbofuran soil incorporation just before rice transplanting can be a good control method to the early occurring rice insect pests and the brown planthopper immigrating during June. Also, it can suppress immigrating BPH popula¬tion moderately in early July.

  • his study was conducted to Investigate the occurrence of small brown planthopper (SBPH), LaodeIpahx striatellus Fallen, and the ind~cence 01 rice virus diseases by d~fferent seeding dates in dryseeded rice. The occurrerlce of SBPH was the highest an nce seeded in May 11, followed byMay 1, May 21 and May 31. However. SBPH dld not occur on the rice seeded in June 10 duelo the rice was emerged after peak occurrence of first generailon adult of SBPH. The occurrenceof the first generation adult of SBPH was May 11 and it was 3-day earlier than that of infantnce transplanting. The ~nd~cencofe rice vlrus diseases, rice stripe vilus (RSV) and rice black-streakeddwarf virus (RBSDV), was the most severe at seeding date May 11. and then followed by May1, May 21 and May 31 Thus the incidence of "ce virus diseases was closely related with thedensity of the first generat~on adult of SBPH Rice yield was decreased by later seeding dates, although rice yield from May 1 to May 21 was relatively stable and hqii compared with laterseeding dates. Accordingly, there was slgniilcant relatlonsh~p between rice yield and seeding datesice yield and seeding dates

The Absorption and Metabolism of Fenobucarb and Carbofuran by Susceptible and Carbamate Insecticide-selected Strains of the Brown Planthopper (Nilaparvata lugens Stal) (Fenobucarb 및 Carbofuran의 저항성 벼멸구 체벽 투과량과 체내대사에 관한 연구)

  • Cuticular penetration and detoxication as mechanisms of resistance to the carbamate insecticides in fenobucarb-selected($R_{f}$) and carbofuran-selected($R_{c}$) strains of the brown planthopper (N. lugens Stal) were investigated. Rates of penetration were not significantly different in the susceptible and resistant strains. However, total amount of excretion of the $R_{f}$ and $R_{c}$ strains were much larger than that of the susceptible strain. Fenovucarb and carbofuran were in vivo metabolizd much faster in the $R_{f}$ strain than in the susceptible strain. OSBP(o-sec-butyl phenol) and 3-ketocarbofuran phenol were invitro the major metabolites of fenobucarb and carbofuran in the brown planthopper, respectively. Total amount of the two major metabolites were produced abotu 2 times larger in the $R_{f}$ and $R_{c}$ strains compared to the susceptible strain. OSBP and 3-ketocarbofuran phenol were not so toxic to the brown planthopper ($LD_{50}$ >100 $\mu\textrm{g}$/g hopper). Based on our data, detoxication plays a large role in resistance to fenobucarb and carbofuran in the resistant strain of BPH, although several resistance factors maybe involved.

Studies on the control of brown planthoppers with Padan 4G and several factors governing the insect mortality (Padan 입제(4G)의 벼멸구 방제효과 및 살충효과에 미치는 몇가지 요인에 관한 연구)

  • A series of experiments were carried out in the laboratory and fields to reevaluate the effects of Padan (cartap) to the brown planthopper (BPH). Nilapanata lugens. The $LD_{50}\;land\;LC_{50}$ values for the female and male BPH were determined by the topical application and seedling- dipping/root -soaking methods. The values were differed with the sex and test methods, and the BPH mortality was greatly increased with a rise in temperatures $(25-35^{\circ}C)$. In a viewpoint of honeydew excretion and offsprings produced, there was no any possibility in BPH resurgence at the sublethal exposures of Padan. The BPH mortality to Padan 4 G was greatly low in the pot tests compared with those to diazinon and carbofuran, but in the paddy fields the efficacy of Padan 4G was nearly reversed. A single application of Padan 4G at the rate of 4kg/10a dramatically suppressed the BPH populations in the paddy fields, and the control effect was much more accelerated in the drained paddy field than in the submerged paddy field.

Effects of Root Zone Applications of Some Systemic Insecticides for Control of the Brown Planthopper, Nilaparvata lugens ($St{\aa}l$) (Homoptera: Delphacidae) (벼멸구에 대한 여러 가지 침투성 살충제의 근부처리 효과)

  • Laboratory and greenhouse experiments were conducted to evaluate the effect of root zone application of some systemic insecticides for control of the brown planthopper (BPH), Nilaparvata lugens, as one of the management option to minimize the adverse effects and maximize the efficacy of insecticide application. Five systemic insecticides, namely, carbofuran, carbosulfan, diazinon, ethoprophos and imidacloprid, as granular formulation were placed in the root zone and measured the mortality, fecundity, and nymphal survivorship of the planthopper. Diazinon and ethoprophos did not show the significant mortality of the BPH. When the BPH were inoculated at the day of carbofuran treatment, adult mortality was shown almost 100% at seven days after treatment and BPH nymphs were remained very few on rice at 25 days after treatment. When carbofuran were placed 10 days before the BPH inoculation, it showed almost 100% adult mortality after one day, and no nymphs were found until 25 day after inoculation. Efficacy of carbofuran on BPH when applied in 10 day-old rice was higher than in 30 day-old rice. These results indicated that the root zone application of carbofuran can control N. lugens effectively with less adverse effect to the natural enemies inhabited on rice plants.

Study on the Long-term Forecasting of Brown Planthopper Outbreaks (벼멸구 발생의 장기예찰을 위한 기초적 연구)

  • Since the outbreak of the brown planthopper (Nilaparvata lugens) in 1915 caused tremendous losses in rice production, one of the more effective method of prevention of such a disaster could be the establishment of longterm forecasting system, In 1916 the author indicated there was a correlation between sunspot activities and brown planthopper and the white back planthopper outbreaks. However, the examples seem to be too small size to state a definite correlation. The purpose of the present study IS to revi~w the history of the brown planthopper outbreaks, and to establish a more effective forcasting system. The present forcasting methods are based on light trap catches of adults which already migrate into this country from mainland China. The regular cycle of 11.2 years of sunspot activity began in 1710, and was continued to present. To gather more records of brown planthopper, the author checked 'Joseon Wangjo Silrok' and analized the so-called 'Hwang' 'Hwang-chung' and 'Chung' which have multiple meanings, together with 'Samguk Sagi' 'Goryo Sa' and 'Munheon Bigo.' The results obtained by the about from review of these old literature citations revealed that ten species of insect and unknown species were involved: i. e., pine moth (Dendrolimus spectabilis), army worm (Mythimna separata), brown planthopper (Nilarvata lugens), white-back planthopper (Sogatella furcifera), migratory locust (Locutsa migratoria), rice stem borer (Chilo suppressalis,), mole cricket (Gryllotalpa africana), rice-plant weevil (Echinocnemus squameus), cut worm (Euxoa segetum), and mulberry pyralid Margaronia pyloalis) The suspected incidence of planthopper in old records expressed by 'Hwang' or 'Chung' revealed a total or 25 out of 37 in 'Samguk sagi,' 21 out of 49 in 'Goryo sa,' 9 of 73 in 'Wanjo-silrog,' and none of 8 in 'Munheon bigo' were planthoppers. Therefore, a total of 36 out of 167 records of insect incidence in the old literature can be possibly attributed to planthoppers. The brown planthopper and white-back planthopper migrate together to Korea every year from mainland China, However, the number of each species are differ by year. In 1975 outbreak the brown planthopper was dominant; and the white-back planthopper prevailed in 1946 and 1977 outbreaks, During the course of this study, the author was able to add a new record of outbreak of planthop per. In 1916 the white-back planthopper outbreak caused serious losses in Chungcheong-namdo and Jeonla-namdo, with losses estimated as high as 160 and 190 thousand seok (23.2 and 27.5 thousand M/T), in Naju and Secheon county, respectively. Since 1912, major outbreaks of brown planthopper or white-back planthopper have been recored 5 times. These occurrences coincide and well matched the period of minimum number of sunspots, With these authenticated records of planthoppers, the author believes there is a close correlation between brown planthopper and white-back planthopper outbreaks in Korea and sunspot activities. Therefore, in years of low number of sunspots, we should watch for and expect outbreaks of these. insects. At this time, it will be necessary to provide all possible prevention measures.
