Streptococci are Gram-positive, facultative anaerobes and have no catalase activities. Among mutans streptococci containing ${\alpha}-type$ hemolytic activity, S. mutans is a causative agent for dental caries. As well as acid production yielding the demineralization of tooth enamel, adherence and colonization of S. mutans to the teeth are also important for its virulence. These early colonization are accomplished by the bacterial fibrillar protein, Antigen I/II (Ag I/II) and glucosyltransferase (GTF). Therefore, Ag I/II and GTF are reasonable targets for the development of vaccine against S. mutans GS-5. The ag I/II and gtfD genes from S. mutans GS-5 were cloned and sequenced. Sequence analyses showed the nucleotides sequence of cloned genes had high homology to the sequences previously reported. The sequence alignment of 280 nucleotides between the cloned Ag I/II and the available sequence of the corresponding S. mutans GS-5 showed the perfect match. Comparing with the sequence of gtfD from S. mutans UA159, the corresponding nucleotide sequence of S. mutans GS-5 showed some mismatches and the mismatches introduced changes in four residues out of 105 amino acids, yielding four missense mutations.
The aim of this in vitro study was to analyze the composition of human tooth enamel in terms of three components, Ca, P, and F after demineralization and remineralization in acid buffer solution. A total of 8 human premolars without any defects and cracks were selected and buccal and lingual sides of the teeth were cleaned with an ultrasonic device and pumice without fluoride 5$\times$5mm windows were opened, and other areas were completely covered with 3-coats of nail varnish to prevent from being in contact with demineralized and remineralized solutions. After demineralization process, each tooth was sectioned into two slices, highly polished one of them with$\gamma$-alumina, and then analyzed the composition of the demineralized tooth with EPMA(electron probe micro-analyzer). The other slices were put into the remineralized solution for 10 days, polished, and analyzed in the same manner. These data were statistically analyzed with one sample t-test(p<0.05). The results were as follows. 1. Normal tooth enamel consists of 49.76% Ca, 39.80% P, and 0.28% F. 2. After demineralization, percentage of Ca and P ratio were decreased by about 5.57 and 5.07% respectively. Percentage of F ratio was also decreased by about 0.01%, which was not statistically significant. 3. After remineralization, percentage of Ca, P increased about by 4.47 and 4.35% respectively Percentage of F decreased by about 0.01%, which was not statistically significant. In conclusion, remineralized solution used in our study has the potential to induce the uptake the Ca and P into the pore sites of the demineralized enamel. But, in the oral cavity. there were rapid temperature change, organic matrix that inhibits the movement of the ions, and limitation of continuous contact with this remineralized solution. Therefore, further in vivo study is necessary.

The purpose of this study was to investigate the inhibitory effect of Er:YAG laser against the intraoral acid producing bacterium of S. mutans. Bacterial pellet containing S. mutans KCTC 3065 was irradiated by Er:YAG laser having a $650\;{\mu}m$ diameter beam by non-contact mode. Irradiated parameters were 50mJ, 10Hz and exposure time were 1s, 3s, 5s, 7s, 9s respectively. We obtained the following results of relative growth rate and acid-producing ability of S. mutans by culturing for 48hrs. 1. The growth rate of S. mutans was decreased in the group of laser irradiation compared to the control group(P<0.01). 2. The growth rate at laser irradiation group of 7s, 9s irradiation time was decreased significantly compared to the laser irradiation group of 1s, 3s, 5s irradiation time, until 12 hours(P<0.05). After 24 hours, all groups of laser irradiation were not found to be statistically different in each other. 3. The acid-producing ability of S. mutans was inhibited for a certain duration by irradiation of laser. In summary, the growth rate and acid producing ability of S. mutans decreased according to laser irradiation. This effect was directly related to the amount of irradiation time. These results suggested that Er:YAG laser had an growth inhibition effect on S. mutans.

The objective of the study was to apply the vibration technique to reduce the viscosity of bonding adhesives and thereby compare the bond strength and resin penetration into dentinal tubules achieved with those gained using the conventional technique. Eighty-eight noncarious extracted human permanent molar teeth were sectioned to remove the coronal enamel and were embedded in 1-inch PVC pipe with acrylic resin. The occlusal surfaces were placed so that the tooth and the embedding medium were at the same level to form one flat surface, and the samples were subsequently polished with silicon carbide abrasive papers. The samples were randomly assigned to 4 groups(n=22). On Group 1 and 2, Single Bond(3M-ESPE, St. Paul, USA) was used, and on Group 3 and 4, One-Step(Bisco Inc., Schaumburg, USA) was used, and each was applied according to its manufacturer's instructions. For Group 2 and Group 4, vibration was applied with ultrasonic scaler for 10 seconds, and the adhesive was light-cured for 10 seconds. Resin composite was condensed on to the prepared surface in two increments using a mold kit(Ultradent Products Inc., USA) and each was light-cured for 40 seconds. After 24 hours in tap water at room temperature the specimens were thermocycled, and shear bond strengths were measured with a universal testing machine(Instron 4465, Canton, USA). To investigate infiltration patterns of the adhesive materials, the surface of specimen was examined with scanning electron microscope. The results were as follows. 1. The shear bond strengths of vibration groups(Group 2, Group 4) were significantly greater than those of the non-vibration groups(Group 1, Group 3)(p<0.05). 2. The shear bond strengths of Single Bond and One-Step were not significantly different (p>0.05). 3. The vibration groups showed greater number of resin tags in tubules and lateral branches under SEM.
Objective: The aim of this study was to investigate the experimental brush wear pattern of a light cured surface sealant, Biscover (Bisco, Schaumburg, IL), and to evaluate its cariostatic effect. Methods: Caries-free human premolars were used for the Biscover coating group (n = 90) and the control group (n = 10). The Biscover coating group was randomly assigned to nine subgroups of 10 each and the control group was assigned to two subgroups of 5 each according to the number of brushing strokes. An experimental 3-body wear test was conducted under different strokes of wear test. Tooth-brushing was accomplished with movement of each brush head set at a frequency of 100 rpm under a force of 1.5N, Surface roughness was tested before, and after Biscover coating, and after brushing. Then, each of the 10 teeth of both groups were placed in artificial caries inducing solution for 7 days. All tooth surfaces were assessed using scanning electron microscopy. Results: Biscover coated surfaces showed a smoother texture than enamel surfaces. The roughness was increased after experimental brushing and after 10,800 brushing strokes, the whole layer of Biscover wore out. However, teeth in the Biscover coating group had a cariostatic effect in cariogenic conditions. Conclusions: We suggest that white lesions in orthodontic patients can be suppressed by topical applications of Biscover.

The purpose of this study was to clarify the palatal arch length, width and height in the primary and permanent dentition. Samples were consisted of normal occlusions both in the primary dentition(50 males and 50 females) and in the permanent dentition(50 males and 50 females). With their upper plaster casts were used and through 3-dimensional laser scanning(3D Scanner, DS4060, LDI, U.S.A.), cloud data, polygonization, section curve and loft surface, fit and horizontal plane were based to measure the palatal arch length, width and height(Surfacer 10.0, Imageware, U.S.A.). T-tests were applied for the statistical analyze of the data. The results were as follows : 1. In the measurement values, the values of the male were higher than those of the female except primary anterior palatal height. There were not only statistically significant differences in anterior palatal width(p<0.05) and posterior palatal width(p<0.01) in primary dentition but palatal width(p<0.05), anterior palatal length(p<0.01), middle and posterior palatal length(p<0.05) in permanent dentition between male and female. 2. In the indices of palate, there were statistically significant differences in height-length index(p<0.05) and width-length index(p<0.01) between male and female in primary dentition. In permanent dentition, there was statistically difference between male and female. 3. In the measurement values, posterior palatal width was increased most greatly. Posterior palatal height, anterior palatal width and anterior palatal length were followed by descending order. On the other hand, anterior palatal height and posterior palatal length were decreased. 4. In the indices of palate, the height-length index, the width-length index and posterior height-width index were increased, but the others were decreased.

Patients with fixed orthodontic appliances frequently have increased levels of plaque accumulation leading to the possibility of gingivitis or enamel decalcification. Although many methods may be helpful in reducing dental plaque formation, the optimal mechanical removal of plaque is the most important factor during orthodontic treatment. The purpose of this study was to evaluate the efficacy of an electric toothbrush (with a specially designed orthodontic brush head) compared to a manual toothbrush in controlling plaque and gingivitis for patients with fixed orthodontic appliances. Oral hygiene status was measured in thirty-four patients using a plaque index, a gingival index and a bleeding index, before and four weeks after the attachment of fixed orthodontic appliances. Patients were randomly divided into two groups: electric and manual toothbrush groups. Oral hygiene instruction was given according to the type of toothbrush used. The Braun Oral-B D9511 with Braun Oral-B Ortho OD 15-1 brush head was used as the electric toothbrush while the Butler G.U.M. 124 was given as the manual toothbrush. After four md eight weeks, oral hygiene status was measured again. Through a comparison between the electric and the manual toothbrush groups, the following results were obtained. 1. All oral hygiene indices showed an increasing tendency after four weeks of fixed orthodontic appliance. 2. All indices presented a decreasing tendency four and eight weeks after oral hygiene instruction. 3. In case of the gingival index and bleeding index, the decreasing tendency did not show a statistically significant difference between the electric and the manual toothbrush groups. 4. The decreasing tendency of plaque index presented a statistically significant difference between the two groups, showing that the electric toothbrush was more effective in terms of oral hygiene. These findings suggest that an electric toothbrush is useful to orthodontic patients with fixed appliances.

Fluoride has been used widely for the prevention of dental caries. Many dental professionals applicated acidulated phosphate fluoride(APF) gel for the purpose of prevention of dental caries in recent years. However, the topical application of fluoride varnish is growing and getting much recognition in many parts of Europe and Scandinavia. The main purpose of this study was to compare the effect of fluoride varnish and acidulated phosphate fluoride gel on artificial caries lesion. The artificial caries lesion was caused on the sound bovine enamel and divided 60 specimens into three groups each containing twenty for the purpose of study. No application was done on group 1, which acted as control group. Group 2 was treated with the topical application of fluoride varnish and removed after 1 hour Group 3 was treated with APF gel and removed after a minute. The cycle of remineralization/demineralization went on in vitro and the microhardness was measured for the each group after 5 and 15days. The following results were obtained : 1. According to the results after 5 days, the microhardness of groups 2 and 3 were significantly higher than group 1, the control group(p<0.05) Similar results was also noticed after 15 days(p<0.05) 2. In comparison of microhardness between groups 2 and 3 after 5days, there were no significant differences between them. The results after 15 days was also similar 3. Much difference in microhardness wasn't present in groups 2 and 3 after 5 and 15days. However, the microhardness of the group 1 dropped significantly in the result of 15 days and 5 days(p<0.05).

The purpose of this study was to examine differences in microleakage of enamel-sealant interface when all-in-one adhesives were used compared with conventional acid etching and single-bottle adhesive system. Seventy-five extracted permanent third molars were randomly divided into 5 groups and treated with only Etching, $Adper^{TM}\;Single\;Bond^{TM}\;Adper^{TM}\;Prompt^{TM}$ L-Pop, AQ-bond and One-up Bond F each. After sealant application, the samples were thermocycled and the degree of microleakage was determined. The results were as follows : 1. Group 3, 4, 5 using the all-in-one adhesive system showed significantly higher microleakage score than Group 1(p<0.05). 2. The lowest mean microleakage score was Group 2(0.41) followed by Group 1(1.05) Group 3(1.65), Group 4(2.85) and Group 5(3.05). 3. Among Groups using all-in-one adhesives, Group 3 showed significantly lower mean microleakage score than the other groups(p<0.05) but showed significantly higher mean score than both Group 1 and Group 2(p<0.05). 4. There was no statistically significant difference(p>0.05) between Group 4 and Group 5. 5. In SEM examination, all the groups used all-in-one adhesive showed shorter resin tags than Group 1.
