Development of a Species Identification Method for the Egg and Fry of the Three Korean Bitterling Fishes (Pisces: Acheilognathinae) using RFLP (Restriction Fragment Length Polymorphism) Markers (제한절편 길이 다형성(RFLP) 분자마커를 이용한 납자루아과 담수어류 3종의 난과 치어 종 동정 기법 개발)

  • This study aimed to develop a species identification method for the egg and fry of the three Korean bitterling fishes (Pisces: Acheilognathinae), including Acheilognathus signifer, Acheilognathus yamatsutae and Rhodeus uyekii based on the PCR-based Restriction Fragment Length Polymorphism (RFLP) markers. We conducted a field survey on the Deokchicheon River from the North Han River basin, where the three Acheilognathinae species co-occur, and also analyzed the existing sequence dataset available from the GenBank. We found coexistence of the three species at the study site. The egg and fry were obtained from the host mussels (Unio douglasiae sinuolatus) by hand from May to June 2015 and in May 2017. To develop PCR-based RFLP markers for species identification of the three Acheilognathinae fish species, restriction enzymes pinpointing species-specific single nucleotide variation (SNV) sites in mitochondrial DNA COI (cytochrome oxidase I) and cyt b (cytochrome b) genes were determined. Genomic DNA was extracted from the egg and fry and RFLP experiments were carried out using restriction enzymes Apal I, Stu I and EcoR V for A. signifer, A. yamatsutae and R. uyekii, respectively. Consequently, unambiguous discrimination of the three species was possible, as could be seen in DNA band patterns from gel electrophoresis. Our developed PCR-based RFLP markers will be useful for the determination of the three species for the young and would assist in studying the spawning patterns and reproductive ecology of Acheilognathinae fishes. Furthermore, we believe the obtained information will be of importance for future maintenance, management and conservation of these natural and endangered species.

  • The purpose of this study was to evaluate the performance of and to derive future maintenance-management measures of the constructed alternative habitat for the Kaloula borealis at the University of Seoul, examining the period between 2015-2017. The research was constructed in 2014 and in a $191m^2$ area. The performance evaluation was divided into maintaining the habitat of the target species, maintaining the population and reproduction rates of the target species, maintaining the habitat of the wild species, the resilience of natural ecosystems, and the harmony with the surrounding environment. In terms of maintaining the habitat of the target species, soil collected from the existing habitat of the Kaloula borealis and was the depth was increased to 30cm in the alternative habitat. An artificial water supply was required every year during the supporting the spawning and hatching of other amphibians along with the Kaloula borealis. The sources of water of the alternative habitat were both rain and tap water, as it cannot be maintained naturally. Additionally, the Kaloula borealis thrived because it inhabited the research site and the average temperature was $26.2^{\circ}C$ from April-June, which is when the Kaloula borealis spawns. In terms of maintaining the population and reproduction rates of the Kaloula borealis, they were evaluated to have stable rates of reproduction. In terms of maintaining the habitat of the wild species, studies on vegetation and the structure of the characteristics of prey or predators will be needed. Also, alien species, such as Humulus japonicus and Bidens frondosa needed to be removed to maintain the wetland ecosystem of the wild species. In the assessment of the resilience of the natural ecosystems, the mud was monitored, noting the changes in the depth of water, with steps taken to reduce the leakage of water. The mud collected from the Haneul Pond wetland, which is located around the research site was piled up. Also, partial mowing management and the inducement of a natural vegetation colony was required for vegetation management. It was also necessary to create porous spaces, such as old trees and tree branches to create a habitat with hiding places and feeding and spawning places for small organisms. In terms of the harmony with the surrounding environment, the following threat factors needed to be managed: amphibian roadkill by vehicles and pedestrians and artificial draining due to nearby user access. Based on the monitoring results, alternative habitat management measures presented the promoting various waterside structures, in which amphibians can spawn and hide in, managing the water environment consistently, managing the vegetation, focused on the habitat of the wild species, and managing the surrounding environment for the habitat. The creation of an alternative habitat should be managed through monitoring, reflecting the characteristics of the changes in the site. Also continuing efforts are also needed to improve the habitat of the target species.

Effect of Sucrose and Supplementary Substances on the Germination Ecology and the Seedling Growth of Native Bletilla striata (자생 자란의 발아생태와 유식물 생육에 미치는 당과 첨가물의 영향)

  • 본연구는 조경소재로 이용가능성이 크지만 현재 자생지가 파괴되어 복원이 요구되고 있는 야생자란의 대량번식을 위해 무균배양시 배지 내 담함량의 변화와 펩톤, 트립톤 등의 첨가가 종자발아와 계대배양 후 유식물의 생육에 미치는 영향을 알아보고자 실시하였다. 배지 내 펩톤과 트립톤의 첨가는 발아에 영향을 주지는 않았지만, 당의 함량은 그 농도가 10g/L까지 증가함에 따라 발아율을 높였다. 또한 발아 후 유식물의 생육시 당의 첨가는 뿌리의 생육을 두드러지게 향상시켰으며, 생체중도 거의 2~3배정도 많았다. 하이포넥스 배지(대조구)에서는 높은 발아율을 보였지만 유식물의 생육은 트립톤 첨가배지(2g/L)에서 많았는데 엽수, 뿌리수, 엽장 근장, 생체중 등이 모두 다른 처리구의 2~3배에 이르는 초기생육을 보였다. 계대배양 이후의 생육상은 펩톤 첨가배지에서 가장 많은 생육량을 보였는데 특히 엽장과 엽폭 그리고 근장이 다른 처리구보다 월등히 높은 경향을 나타냈다. 생체중도 한 개체당 0.18g으로 가장 높게 나타나 펩톤의 첨가가 계대배양 이후의 생육을 크게 촉지시키는 것으로 나타났다. 결론적으로 하이포넥스 배지에 트립톤 2g/L를 첨가하였을 때 발아율과 유식물의 생육이 다른 배지에 대해 매우 양호한 것으로 나타나 자생 자란의 종자발아용 배지로 가장 적당한 것으로 사료된다. 그리고 이후 계대배양시에는 펩톤의 첨가 (3g/L)가 유식물의 생육을 가장 촉진시키는 것으로 나타났다.

Breeding Behavior of Black Vulture In Erdenesant, Mongolia (몽골 Erdenesant 지역의 독수리 번식행동)

  • We studied the behavior of black vultures breeding in Erdenesant, Mongolia through time budget observation. We observed a pair of black vulture for 115 days from April 30 to August 22, 2005, of which 15 days were before hatching and 100 days of brood rearing. The egg hatched on May $14^{th}$. Incubation was done by both a male and female vultures, but the period covered by the male vulture(56.82%) was longer than that by the female one(34.62%). In the early days of brood rearing, time spent by the female vulture at the nest took 54.9 % and it was 19.27% for the male. In the middle of brood rearing period, just inaction and preening were noticed, as they watched their chicks for a long time without sheltering chick under the parent's body. Late brood rearing period was characterized by less chick care and adults mostly stayed in the nest only when to feed the chicks. During breeding time, both the male and the female vulture fed only the chicks and did not give food to each other. During rearing period, the male vulture fed the chick more often than female.

  • 녹조현상은 부영양화된 호수나 유속이 느린 하천에서 부유성의 조류(식물 플랑크톤)가 대량 증식하여 수면에 집적하게 되고 물의 색을 현저하게 녹색으로 변화시킴으로써 발생된다. 최근에는 이러한 녹조 현상이 광역화, 독성화, 장기화의 특성을 띠며 빈번히 발생되고 있다. 녹조현상은 독소를 발생시키는 남조류에 의해 수생식물에 악영향을 주는 것으로 알려져 있다. 예를 들면 독소에 의한 가축에의 영향, 생태계 파괴로 인한 생태학적인 문제, 산소결핍으로 인한 물고기 및 각종 수중생물 폐사 등의 심각한 문제를 야기한다. 또한 조류는 식수에서 맛과 냄새를 유발할 뿐 아니라 Microcystin-LR과 같은 유해한 독소를 배출하여 공중 보건을 위협한다. 이에 식수원으로 사용되는 하천의 조류 번식에 따른 대응방안 마련이 절실히 요구된다. 유입되는 조류로 부터의 정수처리 설비의 처리 부하를 줄이기 위해서는 취수시스템과 연계한 고속 전처리 조류 제거 시스템을 개발이 필요하다. 기존의 전기응집부상공정(Electro-Coagulation and Flotation, ECF)은 화학 약품(응집제) 투여량이 적은 이점이 있지만 비교적 긴 전기 분해 시간이 필요하여 기존 정수처리 시스템과 연계성에 있어 한계가 있다. 이에 본 연구는 전기 분해 시간을 줄여 유입된 조류를 수 초 내에 응집하여 1분 이내에 조류를 분리하는 초고속 조류 전처리 기술을 개발하였다. 개발된 기술의 현장적용 및 실험 결과, 응집과정이 없이도 Chlo-a는 약 45 %의 제거 효율을 나타났다. 또한 응집제의 투입 및 전극에 의한 부상시스템에 의해 Chlo-a가 약 80 %로 제거되는 것으로 나타나 빈번하게 발생되는 조류로부터 안정적인 물 공급을 위한 전처리 공정으로 활용이 가능할 것으로 판단된다.

Comparison of the Reproductive Characteristics of Sweetfish Plecoglossus altivelis in the Main Streams of Jeju Island (제주도 주요 하천에 서식하는 은어(Plecoglossus altivelis)의 번식 특성 비교)

  • We investigated the reproductive characteristics of the sweetfish Plecoglossus altivelis, including changes in the gonadosomatic index (GSI), frequency of gonad developmental stages, and abundance of drifting larvae, in three streams (Gangjeong, Yeonoi, and Ongpo) on Jeju Island from May 2004 to December 2005. The GSI values of female P. altivelis in all Jeju streams began to increase in September and reached a maximum in October and November. Peak GSI values in males occurred in Gangjeong from October to November, in Yeonoi from November to January, and in Ongpo from September to October. The gonadal development of P. altivelis was classified into four stages: growth (March to October), maturity (September to December in females; July to December in males), spawning (September to January), and degeneration (October to March in females; after November in males). Drifting larvae were collected from October to January. These results suggest that the main spawning activity of P. altivelis in Jeju streams occurs from October to November. The information about the reproductive characteristics of P. altivelis obtained in this study is critical to fishery management for this species.

Predicting the Goshawk's habitat area using Species Distribution Modeling: Case Study area Chungcheongbuk-do, South Korea (종분포모형을 이용한 참매의 서식지 예측 -충청북도를 대상으로-)

  • This research aims at identifying the goshawk's possible and replaceable breeding ground by using the MaxEnt prediction model which has so far been insufficiently used in Korea, and providing evidence to expand possible protection areas for the goshawk's breeding for the future. The field research identified 10 goshawk's nests, and 23 appearance points confirmed during the 3rd round of environmental research were used for analysis. 4 geomorphic, 3 environmental, 7 distance, and 9 weather factors were used as model variables. The final environmental variables were selected through non-parametric verification between appearance and non-appearance coordinates identified by random sampling. The final predictive model (MaxEnt) was structured using 10 factors related to breeding ground and 7 factors related to appearance area selected by statistics verification. According to the results of the study, the factor that affected breeding point structure model the most was temperature seasonality, followed by distance from mixforest, density-class on the forest map and relief energy. The nature of the goshawk's breeding environment and habit to breed inside forests were reflected in this modeling that targets breeding points. The northern central area which is about $189.5 km^2$(2.55 %) is expected to be suitable breeding ground. Large cities such as Cheongju and Chungju are located in the southern part of Chungcheongbuk-do whereas the northern part of Chungcheongbuk-do has evenly distributed forests and farmlands, which helps goshawks have a scope of influence and food source to breed. Appearance point modeling predicted an area of $3,071 km^2$(41.38 %) showing a wider ranging habitat than that of the breeding point modeling due to some limitations such as limited moving observation and non-consideration of seasonal changes. When targeting the breeding points, a specific predictive area can be deduced but it is difficult to check the points of nests and it is impossible to reflect the goshawk's behavioral area. On the other hand, when targeting appearance points, a wider ranging area can be covered but it is less accurate compared to predictive breeding point since simple movements and constant use status are not reflected. However, with these results, the goshawk's habitat can be predicted with reasonable accuracy. In particular, it is necessary to apply precise predictive breeding area data based on habitat modeling results when enforcing an environmental evaluation or establishing a development plan.

A Study on the Emergence Period and Geographic Distribution of Cicadinae (Hemiptera: Cicadidae) in Korea Using Bioacoustic Detection Technique (생물음향 탐지기법을 이용한 한국 매미아과의 출현 시기 및 서식지 분포 특성 연구)

  • The purpose of this study is to observe the period of mating calls of cicadas in South Korea to identify the emergence period and geographic distribution for each cicada species. The study sites were 19 protection areas nationwide. The mating calls of cicadas were collected over the 12 months of 2019. A bioacoustics measuring device was installed to record the mating calls of cicadas in WAV, 44,100Hz format for 1 minute every hour. The temperature was recorded once or twice every hour using a micro-meteorological measuring device. Nine species of Korean cicadinae were studied. The start and end periods of mating calls were recorded for each cicada species for the subsequent analysis. The analysis results showed that nine cicada species appeared in the 19 protection areas. The chronological order of mating call periods for each species was as follows: Cryptotympana atrata (7/12 - 9/30), Meimuna opalifera (7/27 - 10/20), Hyalessa fuscata (7/25 - 10/9), Graptopsaltria nigrofuscata (7/28 - 9/5), Platypleura kaempferi (7/3 - 9/29), Suisha coreana (9/14 - 10/30), Leptosemia takanonis (6/26 - 8/2), Auritibicen intermedius (7/27 - 9/28), and Meimuna mongolica (8/8 - 9/11). The mating call period was between 35 (Meimuna mongolica) and 89 (Platypleura kaempferi) days, with the average being 62 days. The elevation above sea level for the habitats of each species was as follows: 5 - 386 m for Cryptotympana atrata, 7 - 759 m for Meimuna opalifera, 7 - 967 m for Hyalessa fuscata, 42 - 700m for Graptopsaltria nigrofuscata, 7 - 700 m for Platypleura kaempferi, 5 - 759 m for Suisha coreana, 7 - 759 m for Leptosemia takanonis, 397 - 967 m for Auritibicen intermedius, and 7 - 42 m for Meimuna mongolica. The average temperature of the habitats of each species was as follows: 23.9℃ for Cryptotympana atrata, 21.8℃ for Meimuna opalifera, 22℃ for Hyalessa fuscata, 23℃ for Graptopsaltria nigrofuscata, 22.9℃ for Platypleura kaempferi, 14.6℃ for Suisha coreana, 20.6℃ for Leptosemia takanonis, 19.3℃ for Auritibicen intermedius, and 24.4℃ for Meimuna mongolica. In terms of the habitat distribution of species, Meimuna opalifera, Hyalessa fuscata, and Platypleura kaempferi were distributed in more than 15 protection sites. Cryptotympana atrata was distributed in the lowlands in the southwest. Graptopsaltria nigrofuscata was distributed in the western area of the Korean Peninsula. Suisha coreana was distributed in areas excluding high mountain areas and parts of the southeast area. Leptosemia takanonis was distributed in areas near the mountains. Auritibicen intermedius was distributed locally in the high mountain areas. Meimuna mongolica was distributed locally in flat wetlands.

Ecological Characteristics of Local Collections of Sagittaria pygmaea Miq. and Sagittaria trifolia L. and Their Geographical Differentiation (올미와 벗풀 지방수집종(地方蒐集種)들의 생태적(生態的) 특성(特性) 및 그들의 지리적(地理的) 분화(分化)에 관한 연구(硏究))

  • To study ecological characteristics of Sagitiaria pygmaea and Sagittaria trifolia occurring in Korea their propagules were collected from 3 locations (Sagittaria pygmaea: Chuncheon, Suweon, Milyang; Sagittaria trifolia: Suweon, Iri, Jeonju) in 1981, cultured and replanted 4 times (May 20, June 5, June 20, July 5) in 1982. Sagitraria pygmaea from Suweon flowered earlier than those from Chuncheon and Milyang in the plants planted on May 20, but this was reversed in another planting dates. Three storied inflorescence was observed newly in Sagittaria pygmaea. Sagittaria pygmaea from Iri and Jeonju had more number of tillers, but less number of tubers per tiller than those from Suweon. Sagittaria rrifolia from Chuncheon flowered earlier than those from Suweon and Milyang. Sagirtaria trifolia from Milyang was narrower in the upper leaf width and less in the number of tubers per plant than those from Chuncheon and Suweon. Each of local collections may be regarded as different ecotype based on the above differences.

Ecology of the Forest Birds in Mt. Chiri, Korea (한국산(韓國產) 산림조류(山林鳥類)의 서식생태(棲息生態) - 지리산지역(智異山地域)을 중심(中心)으로 -)

  • For the purpose of contributing to wild birds protection and propagation, distribution characteristics and ecological breeding habitats were surveyed in Mt. Chiri from September, 1986 to December, 1989 by the use of the nestboxes installed in four study sites by the altitude. The results were as follows ; 1. During the study period, observed birds were 2,403 individuals belong to 59 species, especially, at the 1,400m above altitude the rare species, Falco tinnunculus and the protecting bird species such as Ninox scutulata, Strix aluco and Prunella collaris were observed. 2. In the inhabitation structure of birds by the altitude, Fringilla montfringilla was the highest dominance (41.33%) among the inhabitated birds in the surveyed area. 3. Phasianus colchicus and Streptopelia orientalis which were mainly inhabited in hilly area were observed until the altitude of 900m. 4. The 92.5% of nest boxes was mainly used by the small birds, Parus spp. and Sitta europaea. These species preferred the nest boxes installed at the edge or along the mountaineering path, because they could get food and water easily from those places. 5. The utilization rate of the nest boxes was showed 86% at 500m, 96% at 900m, 98% at 1,400m and 90% at 1,700m above the altitude, respectively. 6. The breeding rate of birds in the nest boxes was showed from 6% to 46% by altitude. Parus varius was the highest breeding species in the surveyed area. These birds inhabited above 500m laid eggs from 15th, April to 22th, May(n=22), but those above 1,700m laid eggs from 28th, April to 27th, May(n=13), that 13 days were delayed at high altitude. 7. Food items of the chicks of the 5 species collected by the collar method were animal matters ; 91.3% of the food was forest insects such as Phalera asiimilis and Pleuroptya batteata except 8.7% of spiders.
