Two New Records of Marine Hydromedusae (Cnidaria: Hvdrozoa) in Korea (한국 해산 히드라해파리 (자포동물문: 히드라충강) 2미기록종)

  • Some hydromedusae were collected from Shiwhaho (Kyonsgi-do), Korea Strait and Yeosu, Korea during years 2000-2002. They were identified into Sorsia tubulosa (M. Sars, 1835) of the order Anthomedusae, and Liriope tetraphylla (Chamisso and Eysenhardt, 1821) of the order Trachymedusae respectively. They are new to the Korean fauna. The order Trachymedusae is first recorded in Korea. The long tube-like manubrium and four marginal tentacles are characteristics of S. tubuloso. In L. tetraphylla, the four triangle-shaped gonads and their positions upon half of surface of subumbrella, the seven blind centripetal canals in a quadrant and the long fat manubrium are its distinct characteristics. Resulting from this work, the Korean hydromedusae identified so far consist of 13 species of 11 families in five orders.

Analysis of Chalcone Synthase and Flavanone 3-Hydroxylase Activity in Lilium Cultivars (Lilium품종의 Chalcone Synthase와 Flavanone 3-Hydroxylase 효소학적 분석)

  • In this work, we analyzed the activity of control enzymes of flower color biosynthesis, chalcone synthase (CHS) and flavanone 3-hydroxylase (FHT) using biochemical and enzymological methods in Lilium longiflorum and 11 Lilium cultivars. The results obtained are as follows ; Naringenin (NAR) was synthesized in all Lilium cultivars tested by the catalytic activity of CHS which used malonyl-CoA and 4-coumaryol-CoA as substrates. Substrate-specific activity of CHS was observed because eridictiol (ERI), which uses caffeoyl-CoA as a substrate, was not detected in tested cultivars. In next step, dihydroflavone product was synthesized by FHT using flavanones as a substrate. FHT synthesized dihydrokaempferol (DHK) by using NAR as substrates. A remarkable activity of FHT was observed in other 11 cultivars.

  • This experiment was carried out to establish growth characterestics and number of bulblet, bulb weight, and diameter of bulblet on scale tissue culture in Lilium brownii. Flowering date of Lilium brownii was 25th, May with 50cm hight in field condition. Vimyl house condition was flowered 28th May with 54cm hight. Number of bulblet, bulb weight, diameter of bublet was good position of outer>Middle>inner scale in Lilium brownii. Number of bulblet formation was the best with 3.5 in large scale. Concentration of BA was the best at 1.5mg/L with 3.8 bulblet with bulb weight, weight and diameter of bulblet.

Development of an Efficient In Vitro Screening Method for Selection of Resistant Lily Cultivars Against Fusarium oxysporum f. sp. lilii (백합 구근부패병 저항성 검정을 위한 기내 간편 검정법 개발)

  • The soilborne fungus Fusarium oxysporum f. sp. lilii (Fol) is a serious threat to all lily cultivars, especially infecting bulbs and flowers. It has become increasingly important to develop varieties resistant against the bulb rot disease. Genetic diversity of cultivars and reliable screening methods are required for this purpose. Here, an efficient in vitro screening system for evaluating resistance to Fol in 38 in vitro-grown lily plants was investigated. Various factors including culture conditions of Fol, inoculum density, appropriate plant materials, inoculation method and duration, and incubation period of plant materials after inoculation were combined to optimize the screening method. As a result, we suggest optimal conditions for an in vitro screening system for the selection of Fol-resistant lily cultivars as follows. Fol was grown on potato dextrose agar (PDA) medium for 6 days at $25^{\circ}C$ in darkness and used as working inoculation. Spore suspensions were prepared (inoculum density: $1.0{\times}10^4$ $spores{\cdot}mL^{-1}$), and then leaf segments $1.5{\times}2.0cm^2$ were inoculated by dipping for 22 hours at $25^{\circ}C$ in dark. Later, leaves were cultured on 0.6% agar plates at $25^{\circ}C$ and 50% humidity with a photoperiod of 16 hours light/8 hours dark (fluence rate of $40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) to examine the progress of bulb rot. After 7 days, disease levels were classified into indices 1 (no symptom) to 6 (serious bulb rot). Soil inoculation of Fol carried out with resistant or susceptible lily cultivars that had been selected through in vitro screening confirmed the reproducibility of results. Therefore, the in vitro screening method established in this study is efficient and reliable for selection of lily cultivars resistant against bulb rot disease.

Microscopic Observation of Pellets Fabricated with Torrefied Larch and Tulip Tree Chips and Effect of Binders on the Durability of the Pellets (반탄화 낙엽송 및 백합나무 칩으로 제조한 펠릿의 현미경 관찰과 펠릿의 내구성에 대한 바인더의 영향)

  • This study was conducted to investigate the effects of several variables on the durability of wood pellets fabricated with torrefied larch (LAR) and tulip tree (TUT) chips. Microscopic observation by scanning electron microscope-energy dispersive X-ray spectrometer was also performed to identify the surface of the wood pellets visually. In addition, torrefied-LAR and TUT pellets were fabricated with the addition of moisture, lignin, starch and protein as binders, and durabilities of the pellets were analyzed statistically. Durabilities of torrefied-LAR and TUT pellets were lower than one of non-torrefied-LAR and TUT pellets. Durabilities of both pellets fabricated with the wood chips, which were torrefied with $230^{\circ}C$ and 30 min, were the highest among all torrefaction conditions used in this study. From the microscopic observations, lignin was distributed broadly on the surface of non-torrefied wood pellets, whereas congregated partially on the surface of torrefied wood pellets. Durability of LAR pellets increased with the addition of moisture, but that of TUT pellets was reduced. Addition of binders contributed to increase the durability of LAR and TUT pellets. As a binder, lignin and protein were more effective than starch for improving the durability. In conclusion, mild torrefaction treatment, such as $230^{\circ}C$ and 30 min, might be an optimal condition to minimize the durability reduction of the LAR and TUT pellets. In addition, when torrefied woody materials with high and low specific gravities are used as a raw material for the production of durable wood pellets, it might be required to adjust moisture content and torrefaction conditions of woody materials, respectively.

Effect of End-taping Treatments on Prevention of Drying Defects during Radio-Frequency/Vacuum Drying of Domestic Yellow Poplar Log Cross Section (엔드테이핑 처리들이 백합나무 원반의 고주파진공건조 중 건조결함 예방에 미치는 영향)

  • This study was carried out to investigate the effects of the Inside end-taping (Inside-ET) and Outside end-taping (Outside-ET) treatments on prevention of drying defects during radiofrequency/vacuum (RF/V) drying domestic yellow poplar log cross section with size of 20~75 mm in thickness and 120~470 mm in diameter. The treatments of Inside-ET and Outside-ET were quite effective for producing sound log cross sections with thinner than 33 mm and smaller than 270 mm in diameter. Even the 60 mm- and 75 mm-thick log cross sections, if the diameter was smaller than 30 cm, it might expect preventing formation of border checks by appropriate end-taping treatments. The positions of the observed border checks differed in diameter ranging at a relative radius about 0.50~0.63 diameter, and it was estimated that it was necessary to select the border position of end-taping treatment properly according to the diameter of log cross sections. It was observed that the radius of 0.3 diameter was the critical location for pretreatment for preventing formation of border checks in the log cross section with middle diameter. There were severe border checks and V-shaped cracks in the log cross section with large diameter. This was because of the severe variation of moisture content along radial direction of natural characteristic of domestic yellow poplar.

Effects of Fumigation Methods on Insect Disinfestation and Quality of Cut Rose and Lily (훈증방법이 살충력 및 절화장미·백합의 품질에 미치는 영향)

  • Effects of traditional fumigation (methyl bromide; MB) and combined fumigation (methyl bromide/phosphide; $MB/PH_3$) on the disinfestation, and phytotoxicity and longevity of cut flowers (lily and rose) were investigated. Combined fumigation with low concentrations of $7/3g/m^3\;(MB/PH_3$) at $10^{\circ}C$ immediately after transportation from harvested place was found to be much more effective in disinfestation of aphid, mite, and thrips than fumigation with $MB\;48g/m^3$ only at $20^{\circ}C$ after simulated transportation for 2 days. Quality and longevity of cut flowers measured after fumigation were better in flowers treated by combined fumigation ($7/3g/m^3,\;MB/PH_3$) at $10^{\circ}C$ for 2.5-3 hrs, irrespective of pretreatment with pulsing solution immediately after transporation, even though phytotoxicity varied with cut flowers fumigated. On the other hand, fumigation with $MB\;48g/m^3$ only at $20^{\circ}C$ after simulated transportation for 2 days showed tendency of enhanced insect disinfestation, but caused phytotoxicity to both lily and rose, regardless of pretreatment and moreover, reduced the longevity of the cut flowers.

The Comparison of Protein Patterns of Several Species in Bivalvia by SDS Polyacrylamide Gel Electrophoresis (SDS Polyacrylamide Gel 電氣泳動에 依한 斧足綱數種의 蛋白質패턴의 比較)

  • SDS-polyacrylamide gel electrophoresis for the proteins obtained from the plasma, Scapharca subcrenata and suapharca broughtonii, and for the proteins of muscles of several species in Bivalvia was performed. The protein patterns of plasma showed little difference between S. subcrenata and S. broughtonii in lower molecular weight weight proteins. However, the protein patterns of muscles of other species, which wre used in this study, were more shown in the lower molecular weights than the higher molecular weights in difference. Thus it is thought be an interesting fact. The protein band of blood corpuscles, 17,800 dalton, was not appeared in S. broughtonii, but this band was appeared in S. subcrenata. Henceforth this is the significantly important difference in these two species. But the protein patterns obtained from muscles of the species did not show a difference in a range of molecular weights between 10,000 and 100,000 daltons. Meanwhile, several protein bands obtained from Meretrix lusoria were similar to those of Mercenaria stimpsona. Hence, in this study, 6 protein bands which exist all species in Bivalvia and 4 characteristic protein bands in S. subcrenata and S. broughtoniionly, were investigated. And in four species of Eulamellibranchia, two protein bands in comon and the characteristic band of 23,000 dalton which is belong to Meretrix lusoria and Mercenaria stimdsona, were found. The molecular weights of the characteristic protein patterns, which are contained in each species, were measured and compared.

Effect of Diluted H2SO4 and NaOH Treatment on Chemical Composition of Larch and Yellow Poplar (황산 및 수산화나트륨처리가 낙엽송과 백합나무의 주요 화학조성에 미치는 영향)

  • In this study, both mild acid and alkali treatments with 1.0 wt% and 2.0 wt% of $H_2SO_4$ and NaOH solution were applied to evaluate the effects on chemical compositions of wood biomass. Yellow poplar (Liriodendron tulipifera L.) and larch (Larix kaempferi C.) were chosen due to major species planted in Korea. Chemical treatments of biomass were carried out by being soaked in either acid or alkali solution with 1:20 ratio for 72 hours at ambient temperature. Afterward, lignin, 5 major reduced sugars, ash contents and elemental composition were determined. To statistically understand the relationship between samples and chemical treatments, the Tukey test, simple linear regression model and ANOVA analysis were introduced using a statistical software R. As results from both wet chemistry and statistical analysis, yellow poplar was more affected on the lignin and xylose contents by acid treatments under these experimental conditions. Meanwhile, larch was more affected on the composition of galactose and lignin by alkali treatments. A series of results in this study would show that equivalent chemical treatment makes a change the chemical composition of each species.

Occurrence and Changes of Botrytis elliptica resistant to fungicides (살균제 저항성 백합 잎마름병균(Botrytis elliptica)의 발생과 변화)

  • Five hundred sixteen isolates of Botrytis elliptica were isolated from infected leaves of Lilium longiflorum from Kangwon alpine areas in Korea during tile seasons from 1998 to 2000 and resistance of these isolates against some fungicides were examined. The isolation frequency of phenotypes resistant to benomyl, procymidone, and diethofencarb were 90.1, 32.4, and 40.9%, respectively. The isolates were divided into six phenotypic groups; RSS, RRS, SSR, SRR, RSR and RRR, representing sensitive (S) or resistant (R) to benzimidazole, dicarboximide, and N-phenylcarbamate fungicides in order. The percentage of six phenotypes were 40.7, 8.5, 7.2, 2.7, 19.8, and 21.1%, respectively. The RSS phenotype was the most frequently isolated, and tile SRR consisted of the extremely minor populations. In comparison studies on tile overwintering ability of each phenotype in relation to the others, the most frequently isolated RSS and SSR had the higher fitness ability than the less frequently isolated RSR, SRR, and RRR. Recently, population increase of tile RSR and RRR phenotypes may have resulted from the increased applications of the mixture of carbendazim and diethofencarb to control benzimidazole-resistant B. elliptica since 1998. The results of this study indicate that careful application of the fungicides is necessary to achieve effective control of leaf blight on lily in Korea.
