Atractylodis Rhizoma Alba is commonly used herbal medicine and it has been known that has immuno-regualtory effects and anti-cancer effects. The inhibition of osteoclastogenesis is essential for the prevention and treatment of osteoporosis. The aim of this study was to evaluate the effects of Atractylodis Rhizoma Alba on osteoclast differentiation in vitro and on resorbing activity of osteoclast. Osteoclast formation was evaluated in bone marrow cells (BMC) in the presence or absence of Atractylodis Rhizoma Alba. The expression of c-fos, tartrate-resistant acid phosphatase (TRAP), OSCAR, DC-STAMP, cathepsin K, MafB and NFATc1 mRNA in osteoclast precursor were assessed by RT-PCR. The levels of TNF receptor-associated factor-6 (TRAF-6), c-fos and NFATc1 protein were assessed by Western blot analysis. Also the correlation with MAPKs and NF-${\kappa}B$ pathways were measured by using Western blot analysis. With bone resorption study, I tried to evaluate the inhibitory effects of Atractylodis Rhizoma Alba on mature osteoclast function. Atractylodis Rhizoma Alba inhibited the RANKL induced osteoclastic differentiation from bone marrow macrophage in a dose dependant manner without cellular toxicity. Gene expression of c-fos and NFATc1 was significantly down regulated with Atractylodis Rhizoma Alba treatment. Atractylodis Rhizoma Alba markedly inhibited the RANKL-induced osteoclastogenesis through suppression of nuclear factor kappa b (NF-${\kappa}B$) pathway, down stream pathway of p38, ERK and JNK pathway. Taken together, I concluded that Atractylodis Rhizoma Alba have beneficial effect on osteoporosis by inhibition of osteoclast differentiation and by inhibition of functioning osteoclast. Thus I expect that Atractylodis Rhizoma Alba could be a treatment option for osteoporosis.
Objectives : This study was aimed to evaluate the anti-starvation stress effect of Atractylodis Macrocephalae Rhizoma on mice. Methods : First, we divided the mice into 6 groups: Normal Group (group with no starvation), Control Group (administered normal saline 6 times before starting 36 hours starvation), Sample A Group (administered Atractylodis Macrocephalae Rhizoma 0.25g/kg 6 times before starting 36 hours starvation), Sample B Group (administered Atractylodis Mocrocephalae Rhizoma 0.5g/kg 6 times before starting 36 hours starvation), Sample C Group (administered Atractylodis Macrocephalae Rhizoma 1.0g/kg 6 times before starting 36 hours starvation), and Sample D Group (administered Atractylodis Macrocephalae Rhizoma 3.0g/kg 6 times before starting 36 hours starvation). Blood was collected from the retro-orbital plexus and then we measured the plasma corticosterone level from the blood. Rectal temperature was measured right after the blood collection. Results : 1. The plasma corticosterone level in Sample A. B, C, and D Groups decreased compared with the Control Group. Sample A and Sample B Groups showed significant differences (p<0.05, p<0.01) compared with the Control Group. 2. The rectal temperature in Sample A, B, C, and D Groups increased compared with the Control Group. Sample A, B. C, and D Groups showed significant differences (p<0.001, p<0.001, p<0.01, p<0.001) compared with the Control Group. Conclusions : Based on the above results, it might be recognized that Atractylodis Macrocephalae Rhizoma has anti-starvation stress effect, and that further study is needed from various viewpoints.
Traditional herbal drugs of Korea, namely, Atractylodes Rhizome(蒼朮) and Atractylodes Rhizome White(白朮) which are derived from and old-grown rhizome and a newly-grown rhizome of Atractylodes japonica Koidz.(Compositae), respectively, were investigated their potential liver-protective activities against hepatotoxicity induced by $CCl_4$ intoxication in mice. Each herbal drug was extracted separately with chloroform, methanol and butanol. No extract of Atractylodes Rhizome showed a significant reduction of the duration of hypnosis produced by hexobarital after $CCl_4$ intoxication in mice. In addition, the the treatments of Atractylodes Rhizome extracts produced no decrease of aspartate aminotransferase(EC 2.6.1.1) activity in serum. However, the methanol extract of Atractylodes Rhizome White exhibited a marked protection from hepatotoxicity induced by $CCl_4$ intoxication in mice. It produced significant reductions of the duration of hypnosis and serum enzyme activity, but no other extract showed liver-protective activity against $CCl_4-induced$ hepatotoxicity.
Objectives : This study aimed to evaluate the effect of Atractylodis Rhizoma Alba water extract on streptozotocin (STZ)-induced diabetes in rats. Methods : Male Sprague-Dawley rats were divided into four groups ; normal, STZ-control and Atractylodis Rhizoma Alba (A) water extract-administrated group. Rats in which diabetic was induced by intraperitonal injection with STZ(60 mg/kg body weight). STZ-induced diabetic rats were orally administrated A extract daily for 5 weeks at doses of 200 or 500 mg/kg. Fasting blood glucose, total cholesterol, triglyceride and blood urea nitrogen were measured in sera of rats. Total volume of urine and urinary creatinine were also measured. Histopathological examination and immunohistochemical staining for the expression of insulin and ${\alpha}$-SMA in pancreas and kidney were performed, respectively. Results : There were no differences in body and kidney weights between STZ-control and A extract-administrated groups. However, serum triglyceride level was significantly decreased in A extract-administrated groups compared with those of STZ-control group. Histopathological analysis of pancreas and kidney revealed increased the number of islets and insulin-positive beta-cells in pancreas, and decreased morphological changes of glomerulus and ${\alpha}$-SMA expression in kidney after the administration of A extract. Conclusions : These results suggest that Atractylodis Rhizoma Alba has a biological action on STZ-induced diabetes in rats via decreasing the serum levels of total triglyceride, and suppressing the morphological changes of pancreas and kidney.
Objectives : The present study was designed to investigate effects and molecular mechanisms of Atractylodes macrocephala Koidzumi extracts(AMK) on the improvement of adipocyte dysfunction induced by TNF-${\alpha}$ in 3T3-L1 adipocytes. We examined whether AMK could directly influence the inflammation and insulin resistance in 3T3-L1 adipocytes. Methods : Potential roles of AMK in the lipolysis, production of inflammatory adipokines and ROS, expression and phosphorylation of ERK, JNK, and $I{\kappa}B{\alpha}$ protein, and expression of $PPAR{\gamma}$ and C/EBP${\alpha}$ were investigated in this study. Results : Our data demonstrated that TNF-${\alpha}$ significantly increased lipolysis, levels of MCP-1, IL-6, and ROS and phosphorylation of ERK, JNK, and $I{\kappa}B{\alpha}$ protein, while TNF-${\alpha}$ reduced the expression of $PPAR{\gamma}$ and C/EBP${\alpha}$ in adipocytes, suggesting that TNF-${\alpha}$ induced a condition with the occurrence of inflammation and insulin resistance. Those alterations induced by TNF-${\alpha}$ were prevented by the treatment of AMK. AMK down-regulated the phosphorylation of ERK, JNK, and $I{\kappa}B{\alpha}$ protein and up-regulated the expression of $PPAR{\gamma}$ and C/EBP${\alpha}$ on TNF-${\alpha}$-induced inflammation and insulin resistance. Conclusions : Thus, our results indicate that AMK can be used to prevent from the TNF-${\alpha}$-induced adipocyte dysfunction through MAPK, $NF{\kappa}B$ and $PPAR{\gamma}$ pathways.
Objectives: This study was to investigate the antioxiative capacity, antiobesity effects of Atractylodes Rhizoma Alba, Houttuyniae Herba, Lonicerae Flos, Scutellariae Radix, and Coptidis Rhizoma on Raw 264.7 and 3T3-L1 cell lines. Methods: Three different types of herb extracts (A. Rhizoma Alba, H. Herba, L. Flos, S. Radix, and C. Rhizoma; water 100%, ethanol 30%, ethanol 100%) were used in this study. Total polyphenol compound, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, reactive oxygen species (ROS) activity, NO production and cell proliferation were measured. Results: Total polyphenol compound measurement of L. Flos, A. Rhizogenes, and C. Rhizoma extracts were higher than A. Rhizoma Alba, H. Herba. DPPH radical scavenging activity, ROS activity and NO production of S. Radix, C. Rhizoma extracts were lower than L. Flos, A. Rhizoma, and H. Herba. Conclusions: Metformin and S. Radix, C. Rhizoma, A. Rhizoma Alba, and L. Flos extracts combination groups showed synergistic effect on adipocyte differentiation inhibition and antioxidative activity.
Atractylodes macrocephala has been used for renal anorexia, gastroenteritis, cold, dyspepsia in Korean folk medicine. Specially aerial parts has been eaten as edible mountain herbs. In order to investigate the efficacy of antioxidant activity the activity guided fractionation and isolation of physiologically active substance were peformed. For the investigation of the active components from Atractylodes macrocephala MeOH extracts of aerial parts of Atractylodes macrocephala Koidzumi L. were suspended with H$_2$O, partitioned by CHCl$_3$. In order to investigate the efficacy of antioxidative activity the activity guided fractionation and isolation of physiologically active substance were peformed. CHCl$_3$, H$_2$O, 30% MeOH, 60% MeOH, MeOH fractions were examined antioxidative activity by DPPH method. It was revealed that 30% MeOH and 60% MeOH fractions have significantly antioxidant activity. From 30% MeOH and 60% MeOH fraction, six flavonoids (7-methoxy-pinocembrin-7-O-${\beta}$-D-glucopyranoside, apige nin-8-C-${\beta}$-D-glucopyranoside, 4'-caffeoyl-luteolin-6-glucopyranoside, luteloin-6-C-${\beta}$-D-glucopyranoside, apigenin-6-C-${\beta}$-D-glucopyranoside, luteolin) and four phenylpropanoids (3-feruloylquinic acid, 4,5-di-O-caffeoylquinic acid, feruloyl acid, 3,5-di-O-caffeoylquinic acid) were isolated. To investigate the antioxidant activities of each compounds, we measured radical scavening activity with DPPH method and anti-lipid peroxidative efficacy on low density lipoprotein (LDL) with TBARS assay. Six compounds (III, IV, V, VI, IX, X) which have antioxidant factor showed significant activities.
The rhizome extract of Atractylodes japonica Koidzumi(Compositae) exhibited a particular inhibition on the proliferation of cultured human tumor cell lines, in vitro. Thus, the intensive phytichemical investigation of the MeOH extract of Atractylodes japonica have been conducted by the way of activity-guided purification. The repeated column chromatographic separation of the n-hexane soluble part of extract resulted in the isolation of four sesquiterpenes (1-4) and a polyacetylene component (5). Chemical structures of them were identified as atractylon (1), atractylenolide Ⅰ(2), atractylenolide Ⅲ(3), eudesma-4(15),7(11)-dien-8-one (4) and 1,3-diacetyl-atractylodiol (5) by spectroscopic means. Among the isolates, compound 2-4 were shown to give moderate inhibitory effect in a dose dependent manner on the proliferation of cultured human tumor cell lines such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system) and HCT 15(colon), respectively.
Objectives : When herbal medicines are extracted together, they may interact with each other, leading to change of chemical characteristics. This study aimed to evaluate the influence of Atractylodes rhizomes (Atractylodes japonica, A. macrocephala, and A. chinensis) on the chemical features of the roots and rhizomes of Glycyrrhiza uralensis, which is are commonly combined with herbal medicines in many herbal formulae, when they are co-decocted. Methods : Liquiritin apioside, liquiritin, ononin, and glycyrrhizin levels of G. uralensis in hot-water extracts prepared by the combination of Atractylodes rhizomes with various weight ratios (G. uralensis : Atractylodes rhizomes = 10:0, 10:5, 10:10, and 10:20) and extraction times (60, 90, and 120 min) were quantified using a HPLC-diode array detector and compared by statistical analysis. Results : The concentrations of liquiritin apioside, liquiritin, ononin, and glycyrrhizin from G. uralensis roots and rhizomes mostly reduced when co-extracted with Atractylodes rhizomes, and the addition of A. chinensis most reduced their contents between Atractylodes combination groups. A. japonica and A. macrocephala rhizomes also showed differences of liquiritin and glycyrrhizin levels at 10 g and 20 g groups of Atractylodes rhizomes. Extraction times also affected the concentrations of liquiritin, ononin, and glycyrrhizin mostly during 60 and 90 min. Conclusions : Atractylodes rhizomes might alter the chemical characteristics of G. uralensis when these herbs are co-decocted. This study provides the understanding of the chemical interactions of herbal medicines during the extraction in hot water.

To determine whether metabolite fingerprinting for whole cell extracts based on Fourier transform infrared (FT-IR) spectroscopy can be used to discriminate and compare metabolic equivalence, standard medicinal parts from four medicinal plants (Cynanchum wilfordii Hemsley, Atractylodes japonica Koidz, Polygonum multiflorum Thunberg and Astragalus membranaceus Bunge) and their in vitro-produced adventitious roots were analyzed by FT-IR spectroscopy. The principal component analysis (PCA) and partial least square discriminant analysis (PLS-DA) from the FT-IR spectral data showed that the whole metabolic pattern from Cynanchum wilfordii was highly similar to Astragalus membranaceus. However, Atractylodes japonica and Polygonum multiflorum showed significantly different metabolic patterns. Furthermore, adventitious roots from Cynanchum wilfordii and Astragalus membranaceus also showed similar metabolic patterns compared to their standard medicinal parts. These results clearly show that mass proliferation of adventitious roots may be applied to aquire novel supply of standard medicinal parts from medicinal plants. However, the whole metabolic pattern from adventitious roots of Atractylodes japonica and Polygonum multiflorum were not similar to their standard medicinal parts. Furthermore, FT-IR spectroscopy combined with multivariate analyses established in this study may be applied as an alternative tool to discriminate the whole metabolic equivalence from several standard medicinal parts. Thus, we suggest that these metabolic discrimination systems may be applied for metabolic standardization of herbal medicinal resources.
