Ascorbic acid and Lipid Contends of the Silkworm eggs(Bombyx mori)during its development of Embryo. (가잠난 배자발육 과정에서 Ascorbic acid와 Lipid의 변동에 관하여)

  • As a result of investigating the change of Ascorbic acid and Lipid which have a relation with metabolism of a silkworm egg in the process of the growth of embryo is silkworm eggs. The following facts have been found 1) Ascorbic acid has gradually increased before the period of the Byong B embryo and it has decreased after period of Byong B embryo. 2) Triglyceride and Total cholesterol has gradually increased before the period of the Byong B embryo and it has decreased after period of the Byong B embryo. 3) Phospholipid has gradually decreased before the period of the Byong A embryo and it increased during the Byong B embryo and decreased again at same stage. It has increased from the head pigment of embryo to hachting. 4) Free Fatty acid decreased during the Byong A embryo stage and increased from the Byong B embryo stage to the Ki A embryo stage and decreased again and increased shortly before the hachting.

  • 조류는 발생학적 특성상 수정과 초기 배발생을 제외한 거의 대부분의 개체 발생 과정이 난각 속에서 진행된다. 또한 배자가 발생하는 동안 배자의 뼈를 구성하는 석회성분의 거의 대부분은 난각에서 유래한다. 난각 석회성분은 배자유래의 융모성 뇨막(chorioallantoic membrane; CAM)의 모세혈관을 통하여 배자로 이동하다. 배자 발생후기로 가면 융모성 뇨막은 전체 난각과 거의 대부분 밀착하여 배자 발생 후기에 필요한 많은 양의 석회성분을 난각에서 배자로 공급하게 된다. 그러므로 한 개의 난각 속에서 두 개의 배자가 정상적으로 발생되기란 거의 불가능하다. Change in Soluble and Insoluble Proteins during Embryonic Development of the Silkworm, Bombyx mori (누에알의 배자발육에 따른 가용성 및 불용성 단백질의 변화)

  • PBS soluble- and insoluble-extracts form silkworm eggs were analysed by native-PAGE and SDS-PAGE. During the embroyonic development, soluble proteins which are mainly composed of ESP, vitellin and 30K proteins showed similar degradation pattern in both electrophoretic analyses. Several peptides which seemed to be intermediated forms of yolk proteins were detected in latter part of embryogenesis, while a protein band newly appeared in one day elapsed after acid treatment. When insoluble extracts from silkworm eggs were analysed with SDS-PAGE, several peptides were detected at the later stages of the embryonic development, and newly hatched larvae. These peptides are considered to be structure proteins for embryonic morphogenesis.

  • We investigated effects of a topical treatment of methoprene(0.5-5.0$\mul$/egg), a juvenLle horm mane analogue, on embryo development in the gypsy moth, Lymantri$\alpha$ dispar. Methoprene lowered egg hatching rate, and also reduced the mean wet weights of hatched 1st instar larvae w with the most effect shown at the highest concentration. The differences in protein(p < 0.01) and carbohydrate(p < 0.05) contents between control and methoprene(5$\mul$/ egg) treatment g groups were observed during embryo development.

  • This study was aimed for a development of a useful gene promoter which has a transcript expressional specificity in the early embryonic period of the silkworm, Bombyx mori. To select a useful gene expressed in the early embryonic stage, we constructed and analyzed a PCR-base subtraction cDNA library. In subtractive hybridization analysis, we confirmed four clones as differently expressed genes(BmNanos-like, BmNanos-P, BmNanos-O, BmVasa mRNAs). Northern hybridization and real time PCR results reveled that the BmNanos-like gene promoter is suitable for the silkworm transgenic vector system. Further defined studies on molecular functions and biological roles of their promoters will give us well-fined information and its application.

Preservation Experiment of Hibernating Silkworm Eggs by 2 Year Cold Refrigeration (월년잠종의 2년간 냉장에 의한 보존 시험)

  • Long term preservation experiment through refrigeration was conducted for 2 year on 300 lines of silkworm races preserved, as one method for the development of long term safe preservation technique. Experiment with 6 treatments was conducted for 680 days from July 1 st 2000 to May 1st 2002. Embryonic development was conducted to each treatment. There are no differences among treatments and races in 400 days preservation, the stage of whole embryonic development was Eul B and condition of eggs was good. In 650 days preservation experiment, differences were reveled among races and treatment, the level of whole embryonic development was Byeong A and condition of eggs was good. The order of embryonic development is European races >Tropical, Korean races >Japanese, Chinese races, thus European races showed fast embryonic development. Control(treatment A) and treatment C showed faster development than other treatments. And treatment D and F showed stable individual stage among all treatments. The test of shape characteristics and embryo which were conducted in hatching period showed 61% of high line succession possibility in average of 6 treatments. But treatment A and B showed no hatching, 3 lines of treatment C, 48 lines of treatment D, 1 line of treatment E, and 29 lines of treatment F showed slow development. And treatments D and F which showed stable embryo condition had highest possibility. The two treatments D and F showed good result among six treatments, and the preservation period of treatment D and F are 235 days and 310 days, and exposure period in $-2.5^{\circ}C$...was longer than other treatments. Numbers of hatched lines of treatment D and F are 48 and 29, and occupied 15.6% and 9.4% of all tested lines, respectively. Average hatching ratio of treatment D and F were 54.% and 71.6%, and average dead egg ratio were 12.6% and 2.4%, respectively. These results show that average ratio of hatching dead eggs in treatment D and F are higher than general line. Thus reconsideration of hatching condition on treatment D and F is needed.

  • Antheraea yamamai vitellin was purified from matured eggs by polyacryamide gel electrophoresis, also stage dependent appearance, immunological comparison and relative content of the protein were investigated. 1. Vitellogenin, the precursor of vitellin, was first detected in the larval hemolymph at the late spinning stage by polyacrylamide gel electrophoresis and immunoelectrophoresis. 2. The electrophoretic mobility of the vitellin was identical with that of Bombyx mori and of Bombyx mandarina. However, the specific antiserum against A. uamamai vitellin did not react with either that of Bombyx mori or Bombyx mandarina in immumo-diffusion test. 3. Relative content of A. yamamai vitellin to the total soluble egg protein was 46.0 percent and did not change till eight days after oviposition. But the content started to decline from ten days after oviposition and was negligible in the five or serventeen month old eggs.

  • To identify the structural basis of mutations that affect synaptic transmission we have begun quantitative ultrastructural descriptions of synapses in cultured Drosophila embryonic neurons. In wild-type cultures, synapses are distinguished by the parallel arrangement of a thickened pre- and post synaptic membrane separated by a synaptic cleft. The presynaptic active zones and postsynaptic densities are defined by electron dense material close to the membrane. Presynaptic regions are also characterized by the presence of one or more electron dense regions, presynaptic densities, around which a variable number of small, clear core synaptic vesicles (mean $35.1{\pm}1.44$ nm in diameter) are clustered. Subsets of these vesicles are in direct contact with either the presynaptic density or the membrane and are considered morphologically docked. A small number of larger, dense core vesicles are also observed in most presynaptic profiles.
