Multiplication of Infectious Flacherie and Densonucleosis Viruses in the Silkworm, Bombyx mori (가잠의 전염성 연화병 및 농핵병 바이러스 증식에 관한 연구)

  • Flacherie, as one of the most prevalent silkworm diseases, causes severe economic damage to sericultural industry and its pathogens have been proved to be flacherie virus (FV) and densonucleosis virus (DNV). Multiplications of the viruses in the larvae of the silkworm, Bombyx mori, were studied by the sucrose density gradient centrifugation and electron microscopy. The quantitative and qualitative changes of nucleic acids and proteins were investigated from the midgut and hemolymph in the silkworm larvae infected separately with FV and DNV. The histopathological changes of epithelial cells of infected midgut also were examined by an electron microscope. 1. Purified fractions of FV or DNV in a sucrose density gradient centrifugation yielded one homogenous and sharp peak without a shoulder, suggesting no heterogenous materials in the preparation. Electron microscopy also revealed that FV and DNV were spherical particles, 27nm and 21nm in diameter, respectively. 2. Silkworm larvae showed a decrease in body weight on the 6th day and in midgut weight on the 3rd day after inoculation with FV or DNV. 3. DNA content was higher in the midgut when infected with FV or DNV, but the hemolymph of the infected larvae showed no difference during first 6 days after inoculation, after which DNA concentration declined rapidly. 4. RNA synthesis of silkworm larvae infected separately with FV and DNV was stimulated in the midgut, but RNA content was reduced in the hemolymph at the early stage of virus multiplication. At the late stage of virus multiplication, however, it was extremely reduced in both midgut and hemolymph. 5. The concentration of protein in the midgut and hemolymph of silkworm larvae infected separately with FV and DNV showed no difference from that of the healthy larvae at the early stage of virus multiplication, but it was significantly reduced at the late stage of virus multiplication. 6. There was no difference in the electrophoretic patterns of RNAs extracted from the midgut of healthy or virus-infected larvae. 7. The electrophoresis of proteins extracted from the midgut infected with FV or DNV, when carried out on the 1st and 5th day after virus inoculation, showed no difference from that of the healthy larvae. But, there was an additional band with medium motility in the proteins on the 8th day after virus inoculation, while a band with low mobility shown in the proteins of healthy larvae disappeared in the infected larvae. However, a band with high mobility in the healthy larvae was separated into two fractions in the infected larvae. 8. The electrophoretic pattern of hemolymph proteins of the silkworm larvae infected separately with FV and DNV was similar to that of the healthy larvae, but the concentration of hemolymph proteins in the infected larvae was lower than that of the healthy larvae at the late stage. 9. Two types of inclusion bodies were shown by the double staining of pyronin-methyl green in the columnar cell of the midgut on the 8th day after FV inoculation. 10. Electron microscopy of the infected midgut revealed that the 'cytoplasmic wall' of the goblet cell thickened on the 5th day after FV inoculation and several types of the cytopathogenic structures, such as virus$.$specific vesicles, virus particles, linear structures, tubular structures, and high electron-dense matrices were observed in the cytoplasm of the goblet cell. The virus particles were also observed in the microvilli and the structures similar to spherical virus particles were observed around the virus-specific vesicles, suggesting the virus assembly in the cytoplasm. 11. Fluorescence micrograph of the infected midgut stained with acridine orange showed that the nucleus, the site of DNV multiplication in the columnar cell, enlarged on the 5th day after virus inoculation. 12. Electron microscopic examination of DNV infected midgut revealed that the nucleolus of the columnar cell was broken into granules and those granules dispersed into apical region of the nucleus on the 5th day after virus inoculation. On the 8th day after inoculation, it was also observed that the nucleus of the columnar cell was full with the high electron-dense virogenic stroma which were similar to virus particles. These facts suggest that the virogenic stroma were the sites of virus assembly in the process of DNV multiplication.

Cross-Infection of Flacherie and Densonucleosis Virus of the Silkworm, Bombyx mori, to Mulberry Pests. (누에 무름병 및 농핵병 바이러스의 주요 뽕나무 해충에 대한 교차감염)

  • Flacherie virus (FV) and Densonucleosis virus (DNV) of the silkworm, Bombyx mori, which give the most severest damage to the silk production in korea, were fed on the mulberry wild silkworm, Bombyx mori mandarina, the mulberry pyralid, Gryphodes phyloalis, and the American fall webworm, Hypantria cunea, to investigate cross infectivity by serological and histopathological at observation. By the Ouchterlony's double difusion test the mulberry wild silkworm was infected with both FV and DNV type 1 (DNV-1) and the mulberry pyralid with DNV-1, so those were confirmed the cross infection. But the American fall webworm was not recognized the cross infection by the same method. The infection and multiplication of the FV in the mulberry wild silkworm was observed in the cytoplasm of the goblet cell with the appearance of the virus-specific vesicle. In DNV-1 infection to the mulberry wild silkworm and the mulberry pyralid, the nuclei of columnar cell in the midgut of both insects was hypertrophied and the nuclei of midgut cell of the mulberry pyralid positively stained with the feulgen stain. Multiplication of DNV-1 in the midgut cell of the mulberry wild silkworm was replicated in two different patterens as linear arrays and large masses, while that of DNV-1 in the muberry pyralid was multiplied as virus masses in several portion of the nuclei of the midgut cell.

Fixation and Histochemistry of Biological Tissues Using the Microwave Fixator Equipped with Infrared-Temperature Sensor (적외선 온도감응기를 장착한 마이크로파 고정기에 의한 생체조직 고정효과와 조직화학적 특성)

  • The present study was carried out to investigate the effect of microwave fixation in comparison with that of chemical fixation in preparing the microscopic samples. The microwave fixator was equipped with infrared-temperature sensor, and that was designed to compensate air temperature in the microwave fixator. In the microwave fixation, rat tongue was well preserved in terms of muscular fasciculus and pancreas stained by Feulgen reagents showed clear reaction products in the nucleus. Reaction products by PAS method in duodenal villi appeared specifically at the goblet cells. In electron microscopy, pancreatic cellular components such as secretory granules and collagen bundles were well preserved in both fixations. In aspect of histochemical reaction and electron microscopy, high quality was due to the protein content of microwave fixed specimen. The microwave fixation method saved total duration engaging microscopic preparation.

  • The digestive tract of the black sea bream, Acanthopagrus schlegeli is composed of esophagus, stomach, intestine, anus and four or five pyloric caeca. Pyloric caecum is a blind sac in shape and originated from pyloric portion of the stomach. Relative length of But (RLG), that is length of digestive tract to standard length, is 1.04 (n=10). Histological layer of the digestive tract is composed of serous membrane, muscular layer, undeveloped submucosal layer and mucosal layer. The mucosal folds of the esophagus are regular branched form, Esophageal muscularis mucosae is well-developed. Mucosal epithelial layer is composed of cuboidal or columnar epithelium and mucous secretory cell. Microvilli are absent in the free surface of mucosal epithelium. The mucosal folds of the stomach are regular unbranched form. The stomach has a well-developed muscular layer and muscularis mucosae. Microvilli are present in the free surface of mucosal surface epithelium. The fundic portion of the stomach have a well-developed gastric gland and more numerous secretory granules than the other parts. The mucosal folds of the pyloric caeca and the intestine are irregular branched form, Intestine is divided into the anterior, mid and posterior intestines with length of mucosal folds and histological features, Posterior intestine has a more developed striated border and goblet cells than the other parts. Mid intestine has a more abundant absorptive cells than the other parts in the intestine and pyloric caeca.

Effects of CheongGeumGangHwa-Tang(CGGH), GwaRuJiSil-Tang(GRJS) on mucin secretion from airway goblet cells (청금강화탕(淸金降火湯 ) 및 과루지실탕(瓜蔞枳實湯)이 호흡기(呼吸器) 배상세포(杯狀細胞)로부터의 뮤신 분비(分泌)에 미치는 영향)

  • Objective : This study is intended to investigate whether the two oriental medical prescriptions, CheongGeumGangHwa-tang(CGGH) and GwaRuJiSil-tang(GRJS), significantly affect mucin release from cultured hamster tracheal surface epithelial(HTSE) cells. Materials and Methods : Confluent HTSE cells were metabolically radio labeled with 3H-glucosamine for 24 hrs and chased for 30 min in the presence of CGGH or GRJS to assess the effect of each agent on 3H-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase(LDH) release. Also, the effects of CGGH and GRJS on contractility of isolated tracheal smooth muscle were investigated. Results : (1) CGGH and GRJS significantly increased mucin release from cultured HTSE cells, without cytotoxicity : (2) CGGH and GRJS did not affect contractility of isolated tracheal smooth muscle. Conclusions : These results suggest that the effects of CGGH and GRJS should be further investigated, and that it would be gainful to invesigate, from among oriental medical prescriptions, what novel agents have these mild expectorant effects on mucin secretion from airway goblet cells.

Surgical Treatment of Esophageal Adenocarcinoma in Barrett's Esophagus - A case report - (바렛 식도에 발생한 식도 선암종의 수술적 치료 - 1예 보고 -)

  • Barrett's esophagus is precancerous lesion of esophageal adenocarcinoma, but this has been rarely reported in Korea. A 81-year-old man with esophageal adenocarcinoma was admitted to our hospital, and we performed a distal esophagectomy and end-to-end esophagogastrostomy. The microscopic examination of the resected tissue revealed the intestinal metaplasia with goblet cells around the esophageal adenocarcinoma, which indicates this was a Barrett's esophagus. We report here on this case along with a review of the relevant literature.

  • In the present study, we intended to investigate whether polycationic peptides including poly-L-lysine (PLL) and poly-L-arginine (PLA) specifically inhibit the mucin release and do not affect significantly the release of the other releasable glycoproteins with less molecular weight than mucin's from cultured airway goblet cells. Confluent primary hamster tracheal surface epithelial (HTSE) cells were metabolically radiolabeled with 3H-glucosamine for 24 hr and chased for 30 min in the presence of varying concentrations of either poly-L-arginine (PLA) or poly-L-lysine (PLL) to assess the effects on 3H-mucin release and on the total elution profile of the treated culture medium. The results were as follows : (1) PLL 78,000, PLL 9,600 and PLA 8,900 inhibited mucin release in a dose-dependent manner; (2) These polycationic peptides did not inhibit the release of the other releasable glycoproteins with less molecular weights than mucin's. We conclude that these polycationic peptides 'specifically'inhibit mucin release from airway goblet cells. This finding suggests that these polycationic peptides might be used as a specific airway mucin-regulating agent.

Study on the Visual Cells in the Retina of Macropodus ocellatus (Pisces, Osphronemidae) Freshwater Fish from Korea (한국산 담수어류 버들붕어, Macropodus ocellatus (Pisces, Osphronemidae) 망막의 시각세포에 관한 연구)

  • Using both light and scanning electron microscopies, it was investigated on the visual cells as well as the eyes of Macropodus ocellatus (Pisces, Osphronemidae). This species had a circular lens and yellowish cornea. The eyes had $3.5{\pm}0.2mm$ which is $31.1{\pm}3.0%$ in a percentage of eye diameter relative to head length. The retina ($158.2{\pm}10.6{\mu}m$) was built of several layers, including the visual cell layer which consists of three types of cells: single cons ($27.8{\pm}1.6{\mu}m$) and equal double cone ($33.9{\pm}3.7{\mu}m$), and large rods ($57.3{\pm}1.3{\mu}m$). The visual cell layer then was classified into the correct pattern. All visual cells were clearly distinguished from two parts (inner and outer segments). The elongated rod cells were extend to the bottom of the retinal pigment epithelium. In scanning electron microscopy, the outer segment links to inner segment by so-called calyceal piles. The M. ocellatus single and double cones appearance form a flower-petal arrangement, which is a regular mosaic pattern that contains quadrilateral units by four double cones surrounding a single cone.

REDIFFERENTIATION FROM TISSUE CULTURE AND ISOLATION OF VIABLE PROTOPLASTS IN PANAX GINSENG C.A. MEYER (고려인삼의 조직배양에 의한 기관형성과 원형질체배양에 관한 연구)

  • Ginseng cotyledon calli were cultured on 1/2MS media supplemented with combination of various growth regulators to induce more embryoids and plantlets in a short period. And tissues of ginseng root and calli were also incubated under various factors or conditions to establish methods for the isolation of viable protoplasts in Panax ginseng C.A. Meyer. The calli derived from cotyledon produced numerous embryoids in 1/2MS media containing 0.5mg/$\ell$ 2,4-D and 0.5mg/$\ell$ kinetin after 2 months' culture. But only shoot formation was less frequent. Further development of these embryoids occurred on 1/2MS medium supplemented with the same concentration of BA and GA. Viable protoplasts were isolated from the root tissue and callus of ginseng. The specific conditions for the isolation of viable protoplasts were required of ginseng materials, root tissue and callus, being processed. For the production of viable protoplasts from 1-year old ginseng root tissue, an enzyme mixture of $2\%$ cellulase 'Ono-zuka' and $0.5\%$ macerozyme, an enzyme solution pH of 5.2 to 5.8, a 7- to 8- hour incubation period at $28{\pm}1^{\circ}C$, and 0.9M mannitol as osmoticum in the cell enzyme mixture were optimum, while the treatments with an enzyme mixture of $2\%$ cellulase 'Onozuka', $2\%$ macerozyme and $1\%$ driselase, and 25-hour incubation period at $28{\pm}1^{\circ}C$, were more efficient for the production of viable protoplasts from ginseng callus.

Visual Cells in the Retina of Iksookimia longicorpa (Pisces; Cobitidae) of Korea (한국산 미꾸리과 어류 왕종개 Iksookimia longicorpa 망막의 시각세포)

  • The visual cells in the retina of Iksookimia longicorpa (Pisces, Cobitidae) were investigated by light and scanning electron microscopes. The retina ($216.42{\pm}13.36{\mu}m$) has several layers, and the visual cell layer consists of unequal double cones and large rods. In a double cone, two members are unequal such that one cone is longer than the other (long element $26.42{\pm}1.7{\mu}m$, short element $16.82{\pm}1.1{\mu}m$). The cones form a row mosaic pattern in which the partners of double cones are linearly oriented with a large rod. The visual cells observed have an outer segment (hematophilic), inner segment (eosinophilic). In scanning electron microscopy, the outer segment links to inner segment by so-called calyceal piles (calyceal processes) of membrane discs surrounded by double membranes.
