This work was investigated the effects of the elevated $CO_2$ and Pb contamination on the growth of Pinus densiflora. Two-years pine trees were planted in Pb-contaminated soils (500 mg/kg-soil) and uncontaminated soils, and cultivated for 3 months in the growth chamber where $CO_2$ concentration was controlled at 380 or 760 ppmv. The growth of P. densiflora were comparatively analyzed in 4 kinds of soil samples (CA : $CO_2$ 380 ppmv + Pb 0 mg/kg, CB : $CO_2$ 380 ppmv + Pb 500 mg/kg, EA : $CO_2$ 760 PPmv + Pb 0 mg/kg, EB : $CO_2$ 760 ppmv + Pb 500 mg/kg). It was measured the growth changes of the p. densiflora caused by $CO_2$ concentration and Pb contamination. The growth of P. densiflora was remarkably inhibited in the Pb-contaminated soil, although the biomass and the root elongations were not significantly affected by the elevated $CO_2$. These results suggested that the growth of p. densiflora was sensitively influenced by Pb contamination rather than $CO_2$ concentration. Compared to the initial soil, total Pb concentration in the soil samples was decreased at 760 ppmv $CO_2$ as well as at 380 ppmv $CO_2$ after 3 months. The accumulation of Pb in the roots at 760 ppmv $CO_2$ was two-fold of that at 380 ppmv $CO_2$, indicating that Pb bioavailability in the root of p. densiflora might be affected by the elevated $CO_2$.
The stf (Salmonella typhimurium fimbriae) operon consisting of stfA(CDEFG assumes to encode putative fimbriae. The complete stf operon is existed in S. typhimurium and S. choleraesuis, whereas it is absent in S. typhi. Analyses of the amino acid residues between major subunit StfA of the Stf fimbriae and those of known other fimbriaes suggested that Stf belongs to class I type fimbriae. Through comparison of StfD chaperone with the other fimbrial chaperones, and of C-terminus in subunits of Stf fimbriae, it belongs to FGS (with a short Fl-G1 loop) subfamily. In order to investigate the expression of stf operon, we have constructed a Salmonella strain containing a chromosomal stfA::lacZYA transcriptional fusion, resulting in S. typhimurium $_X8532$. The strain $_X8532$ lacked the expression of \beta-galactosidase$ under normal culture conditions. However, with longer incubation time of the S. typhimurium $_X8532$, we have isolated 21 individual strains exhibiting $Lac^+$ phenotype. $Lac^+$ phenotype was appeared as approximately 0.03 frequency per generation. All isolates expressed lacZ constitutively in the various environmental conditions. Various global regulatory proteins including RpoS, OmpR, and CpxR were not involved in the regulation of the stf operon. A S. typhimurium $_X8661$ mutant lacking stfAC function attenuated 6.7 folds more than that of wild type $_X3761$ in the mouse virulence test, suggesting in the somehow involved in the Salmonella pathogenesis.
To exploit plant growth promoting bacteria in the roots of Miscanthus sacchariflorus, a biomass energy crop, total 64 bacteria were isolated. For the investigation of plant growth promoting effects from the isolated bacteria, production of indole acetic acid (IAA) and 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activities were tested and other cultural conditions were examined. As results, 8 isolates showed plant growth promoting effects on the M. sacchariflorus and an isolate designated Agrobacterium sp. BE516 has the highest activity by enhancing the shoot elongation over 2-fold than the control. Agrobacterium sp. BE516 produced 64 ${\mu}g$ IAA per mL and showed ACC deaminase activity which is involved in the resistance to environmental stress such as high salt and drought. It could grow at low temperature in the range from 4 to $15^{\circ}C$, at pH 4.0 and at 4% NaCl. These results indicate that the Agrobacterium sp. BE516 can be useful as a bio-fertilizer for M. sacchariflorus under the stressed conditions.
A total of 167,955 microorganisms were isolated from 366,661 clinical specimens. Among them, 6,517 strains of the Candida spp. were isolated from the department of laboratory medicine in "C" hospital from Jan. 1, 2005 to Dec. 31, 2009. All clinical specimens were reviewed by the medical records of patients with Candida by the method of retrospectiveness. From this, we got the some isolated pure cultured yeasts. We identified these yeast by the identification kit system of VITEKII and VITEKII-ID-YST card. The isolation frequencies of Candida spp. were as follows. 56.4%,of C. albicans, 17.7% C tropicalis, 10.7% C glabrata and 9.5% C parapsilosis. The isolated frequency of Candida spp. in 2009 was 1.9 times higher than that in 2005. The clinical materials showing over 10.0% isolation rate were in sputum (30.1%), random urine (25.0%), 15.8% blood (15.8%) and catheterized urine (13.5%) in Candida spp.. The clinical department of showing over 7.0% isolation rate were in pulmonary medicine (20.5%), renal medicine (11.0%), infection disease medicine (10.4%), critical care medicine (10.0%), hematooncology (9.6%), general surgery (7.5%) and gastrointestinal medicine (7.4%) in Candida spp.. In monthly analysis, Candida spp. were most friquency isolated in July (10.6%), but lowest one in February (6.1%). Candida spp. were most frequently isolated in patient of over 50 years old (16.7-40.1%) than those isolated from the patients under the age of 0-49 (1.3-7.5%).

Nuclease was secreted to the environmental media from the Escherichia coli JM107 tranformant harboring the extracellular nuclease gene of Serratia marcescens in the plasmid of pNUC4. Under the growth conditions, the amount of secreted enzyme was increased in parallel with bacterial growth conditions, the amount of secreted enzyme was increased in parallel with bacterial growth. The enzyme was purified using chromatofraphic procedures of Matrex green gel and heparin agarose affinity gel, resulted in 50-fold purification with 15% recovery of the enzyme. The apparent molecular weight of the enzyme was estimated to be 29Kda by sodium dodecylsulfate denaturing gel electrophoresis. Using the purified enzyme, polyclonal antibody was obtained from the rabbit. The specificity of the antibody was confirmed by immunoblotting and immunoprecipitaion. For the investigation of cellular distribution of the enzyme, cells were fractionated into three fractions; cytoplasm, periplasm and extracellular fluid. While more than 80% of the enzymatic activity was detected in the extracellular fluid and periplasm, a little was found in the cytoplasm, indicating that the enzyme was likely to be immediately exported to the membrane for excretion after biosynthesis. These results were confirmed again by immunocytochemistry technique using the antibody.
We studied the effect of fermented Rhus verniciflua stem bark (FRVSB) extract (used in herbal med-icine by Koreans) on the microbial growth and enzyme activity of 12 soybean-fermenting microorganisms, including Bacillus spp., lactic acid bacteria, yeast, and other harmful bacteria. The ethanol and methanol extracts of FRVSB inhibited the growth of Bacillus subtilis, Bacillus licheniformis, Bacillus cereus, and Zygosaccharomyces rouxii, and in the disk diffusion assay, their inhibition zone diameters were 11.06-12.23, 12.32-18.38, 11.47-11.84, and 13.59-14.21 mm, respectively. The water extract did not show any inhibitory effect. In fact, the water extract addition enhanced the growth of B. subtilis and B. licheniformis by 1.3-4.5 fold and that of B. cereus by 1.2-1.4 fold. However, the water extract did not affect the growth of Lactobacillus plantarum, Lactobacillus mesenteroides, Saccharomyces cer-evisiae, and Escherichia coli. The addition of water extract increased the amylase and protease activity of B. subtilis and B. licheniformis.
In this study we tried to transform an antimicrobial peptide gene (Shiva) under the promoter of tomato phenylalanine ammonia-lyase (tPAL5) into tobacco and potato plants. Antimicrobial peptide gene was isolated originally from giant silk moth (Hyalophora cecropia) and modified ie nucleotide sequence to increase antimicrobial activity. Transgenic tobacco plants were regenerated and their seeds were tested on the media containing kanamycin (500 mg/L). The results of PCR amplification and genomic Southern blot hybridization confirmed the integration of construct (tPAL5 promoter-Shiva-NOS-GUS-NOS) into chromosome. We observed that one of the transgenic tobacco plants showed chromosome rearrangement when integrated. In case of potato transformation, the efficiency of regeneration was maximized at the medium containing Zeatin 2mg/L, NAA 0.01mg/L, GA$_3$ 0.1mg/L. We also observed the high expression of GUS (${\beta}$-glucuronidase) enzyme which was located next to the terminator sequence of nopaline synthase gene (NOS) in the vascular tissue of stem, leaves of transgenic potatoes. This result suggested that a short sequence of Shiva gene (120 bp) and NOS terminator sequence might be served as a leader sequence of transcript when translated.
The presence of biological response modifiers (BRMI-like effect was confirmed in peritoneal exudate (PE) of ToxopLnsmo gondii-infected ICR mice which inhibited Concanavalin A (Con A)-induced peritoneal Iymphocyte (PL) proliferation. During 5 days of PL incubation with $10{\;}\mu\textrm{g}/ml$ Con A with or without PE, 3H-thymidine uptake was measured for the last 24 hrs. Compared to uninduced control, PL proliferated by 7.3-fold with Con A induction_ When PE of infected mice was added, PL proliferation was inhibited by $74.0{\;}{\pm}{\;}11.9%$ whereas inhibition by PE of normal mice was $16.4{\;}{\pm}8.3%$. Inhibitory effect of PE increased exponentially from 3 days up to 4-5 days of survival after the infection. Inhibitory activity of PE was decreased concentration-dependently. Also the inhibition was diminished when the PE was treated with heat of $95^{\circ}C$ for 10 min orprecipitated with 10% trichloroacetic acid (TCA). In SDS-PAGE of PE, many minor bands appeared newly. Heat-labile protein molecule in PE exerted inhibitory activity to Con A- induced Iymphocyte proliferation.
Twenty four Holstein steers (average body weight $714{\pm}13.60kg$) were used in this experiment to determine the effect of supplementing of microbial culture and coated vitamin-C on growth performances and carcass characteristics in finishing Holstein steers. Holstein steers were randomly assigned to feeding groups of control group (Con, 12 kg of basal diet/head/day), microbial culture group (MC, 12 kg of basal diet + 30 g of microbial culture/head/day) and coated vitamin-C group (CVC, 12 kg of basal diet + 10 g of coated vitamin-C/head/day). MC and CVC groups were higher in ADG compared to control (P<0.05). FCR was also lower in MC and CVC groups than control group (P<0.05). Back fat thickness, rib-eye area, marbling score, meat color and yield index were not changed by supplementing microbial culture and coated vitamin-C. MC group was higher for maturity compared to control and CVC group (P<0.05). CVC group was higher for fat color compared to control and MC group (P<0.05). Based on the results obtained from the current study, supplementation of microbial culture and coated vitamin-C as an alternative to antibiotic might increase growth performances and enhance carcass characteristics in finishing Holstein steers. However, more studies are needed to find out the optimum supplementing period of microbial culture or coated vitamin-C for high quality meat production from Holstein steers.

A sediment exposure experiment was conducted to investigate the influence of acid volatile sulfides (AVS) and simultaneously extracted metals (SEM) in sediments on the bioavailability and toxicity of Cd, Ni and Zn to a marine polychaetes Neanthes arenaceodentata. The test animals were exposed to contaminated sediments spiked by metal mixtures of Cd, Ni, Zn (0.5~15 $\mu$mol/g of total SEM) in low (~1 $\mu$mol/g), medium (~5 $\mu$mol/g) and high AVS series (~10 $\mu$mol/g) to determine bioaccumulation, mortality and individual growth rate in each treatment after 20 days. Cd and Zn bioaccumulation in test animals increased with increasing of overlying water (OW) concentration controlled by AVS. In contrast, Ni bioaccumulation increased with increase of SEM concentration. Mortalities and growth inhibitions of N. arenaceodentata observed in only treatments with [SEM-AVS]>0, due to a high level of OW-Zn. With regard to the mortality, the 20-d LC5O value fur OW-Zn was 9.3(8.0$\pm$11.0) $\mu$M. The LOEC (Lowest Observed Effect Concentration) for Tissue-Zn was 7.8 $\mu$mol/g and the NOEC (No Observed Effect Concentration) was 6.2 $\mu$mol/g. Regarding the inhibition of individual growth rate, the LOEC fer Tissue-Zn was 5.9 $\mu$mol/g, and NOEC was 5.1 $\mu$mol/g. In this study, the toxicity of dissolved metals, especially for Zn, was overemphasized due to the reduced distribution coefficients (K$\_$d/s) of metals in the experimental sediments.
