Effect of Liquid Fertilizer Contained Fermentation of Lactobacillus confusa and Pichia anomala on Growth of Creeping Bentgrass (A. palustris Huds. CV. Pennlixs) (유산군(Lactobacillus confusa)과 효모균(Pichia anomala) 발효 액체비료의 시용이 크리핑벤트그래스(A. palustris Huds. CV. Pennlixs)의 생육에 미치는 영향)

  • This study was conducted to evaluate effect of liquid fertilizer contained medium of Lactobacillus confusa and Pichia anoala on growth of creeping bentgrass(Agrostis palustris Huds. CV. Pennlixs). By application method of liquid fertilizer, the experiment plot was designed as following; NF was non-fertilized treatment, control was applied with compound fertilizer and treatment 1(T-1), treatment 2(T-2) and treatment 3(T-3) were applied with compound fertilizer and liquid fertilizer solution diluted 500, 300 and 100 folds, respectively. The every treatments was arranged a randomized complete block with three replications. Compared with NF, leaf color index of control, T-1, T-2 and T-3 in creeping bentgrass was increased 7.4%, 7.5%, 77% and 7.2%, respectively and chlorophyll content of T-1, T-2 and T-3 in creeping bentgrass was increased 45.7%, 45.6%, 52.1% and 49.6%, respectively, and T-2 and T-3 was increased 4.4% and 2.7% more than control. Dry weight of T-1, T-2 and T-3 was increased 2.7%, 13.8% and 13.9% more than control, when compared to control. These results were found that turfgrass qualities and growth was improved in treatment applied to compound fertilizer and solution of liquid fertilizer diluted 300 folds.

Comparative Study on Ethanol Production with Pentose and/or Hexose by Saccharomyces cerevisiae and/or Pichia stipitis (Saccharomyces cerevisiae와 Pichia stipitis를 이용한 오탄당과 육탄당으로부터 에탄올 생산에 관한 비교연구)

  • Glucose and xylose are the most abundant materials in nature which can be used to produce ethanol by yeast fermentation. Three combinations of cultivation with glucose and xylose were carried out; separated, co-culture, and sequential fermentation with Saccharomyces cerevisiae and Pichia stipitis. In the separated fermentation, S. cerevisiae fermented glucose to produce 14.5 g/l ethanol from 29.4 g/l glucose but hardly used xylose. However, P. stipitis utilized not only glucose but also xylose to produce ethanol 11.9 g/l and 11.6 g/l from 29.4 g/l glucose and 29.0 g/l xylose, respectively. In the mixture of glucose and xylose, P. stipitis fermented both sugars, producing 21.1 g/l ethanol while S. cerevisiae fermented only glucose, producing 13.4 g/l ethanol. In the co-culture and sequential fermentation, the co-culture showed more efficient ethanol productivity with 18.6 g/l ethanol than the sequential fermentation with 12.4 g/l ethanol. To investigate the effect of nutrients in the growth of microorganisms and ethanol production, yeast nitrogen base (YNB) was used in the sequential fermentation with S. cerevisiae and P. stipitis. YNB supplemented some nutrients which S. cerevisiae used up in the broth and the culture showed increased growth rate, increased consumption of xylose, and increased ethanol productivity producing 22.5 g/l ethanol from 54.6 g/l sugar with a yield of 0.41 g/g.

Study of Tannin Reducing Effect of Aronia by Yeast Isolated from Jeotgal (젓갈에서 분리된 효모를 이용한 아로니아의 탄닌 성분 저감화 효과에 관한 연구)

  • Aronia (Black chokeberry, Aronia melanocarpa) belonging to the Rosaceae family, is native to eastern North America. Aronia contain high levels of flavonoids, mostly anthocyanins and proanthocyanidins, which are known as condensed tannins. The dominant proanthocyanidins in aronia are (-)-epicatechin and (+)-catechin. The concentration of proanthocyanidins in aronia is higher than in other berries, however due to the astringent taste it is not desirable for consumption. Therefore, the purpose of this study is to evaluate the effect of aronia on the reduction in tannins by yeast isolated from regional Jeotgal. We isolated strains of yeast with high ${\beta}$-glucosidase activity from Jeotgal, with the MTY2 strains exhibiting a reduction in final tannin concentration according to thin layer chromatography (TLC) analysis. MTY2 was confirmed as Kazachstania servazzii using an 18S rDNA sequence and named as K. servazzii MTY2. K. servazzii MTY2 showed most significant growth when K. servazzii MTY2 was cultured in a solution of 10% (w/v) glucose, 3% (w/v) tryptone and 0.1% (w/v) sodium chloride. According to the high performance liquid chromatography (HPLC) analysis, the (+) - catechin peak is present, but (-) - epicatechin peak was reduced at culture condition added with 10% glucose in medium.

Changes in Microorganisms, Enzymes and Texture of Dongchimi by High Hydrostatic Pressure Treatment (초고압처리에 의한 동치미의 미생물, 효소 및 조직감의 변화)

  • Effects of high hydrostatic pressure on microorganisms, pectin enzymes and texture of dongchimi (pickled radish roots) were investigated. Dongchimi was pressurized at $200{\sim}686\;MPa$ for 5 min when its pH reached to 4.0. Total aerobes, initial concentration of $4.05{\times}10^8\;CFU/mL$, were completely inactivated by high hydrostatic pressure at 600 MPa. Lactic acid bacteria, yeasts and molds, initial concentration of $3.25{\times}10^8\;CFU/mL\;and\;3.55{\times}10^4\;CFU/mL$, respectively, were completely inactivated by high hydrostatic pressure at 400 MPa. Leuconostoc mesenteroides appeared to be the most barotolerant lactic acid bacteria because it was the sole bacteria survived at 380 MPa. Pectinesterase (PE) and polygalacturonase (PG) activities increased after high hydrostatic pressure treatment. Residual PE activity was 193 after pressurization at 500 MPa, and residual PG activity was 191 after pressurization at 686 MPa when the initial enzyme activity of control was set to 100. The hardness of pressurized dongchimi was higher than control.

Analysis of Volatile Components in Korean Rice Wine by the Addition of Yulmoo (탁주 제조 시 율무 첨가에 따른 휘발성 성분의 분석)

  • In order to use nutraceutical components in Yulmoo (Coix lachryma-jobi L. var. ma-yuen Stapf), the acceptability of Yulmoo for Takju (Korean rice wine) was examined in terms of the production of volatile components. When Yulmoo was added to the Takju made by commercial koji as a substitute of starch at 0 (rice 100%), 15, 30, 60, and 100%, the ethanol content slightly increased to 13.6, 15.2, 15.2, 14.1, and 13.8%, the Production of isobutanol and iso-imyl alcohol increased as the amount of Yulmoo increased. While the use of Yulmoo to the Takju as the nuruk (Korean traditional koji), inoculated with Aspergillus kawachii (AK) and Rhizopu japonicus (RJ), decreased the contents of n-propanol, iso-butanol, and iso-amyl alcohol as well as the contents of ethanol from 14.3% to 11.2 [AK], 7.5 [RJ], and 10.4% [AK + RJ]. The Yulmoo nuruk in Takju increased the production of acetaldehyde and ethyl acetate indicating the negative effect in the organoleptic evaluation. It was determined that the addition of Yulmoo as a starch source or as nuruk has a critical effect on the production of fusel oils and other volatile compounds in Takju fermentation.

  • In this study, the characteristics of Jeung-pyun hatter were investigated by wine yeast. The processing conditions were optimized by physicochemical characterization including pH, volume, reduced sugar. The effect of yeast concentration, moisture content on the fermentation time and temperature were investigated in view of improving productivity. It was found that the volume was increased at maximum state when the fermentation was carried out at 35 $^{\circ}C$ with 0.1% yeast concentration 60% of moisture. The quality of Jeung-Pyun was most preferable in the condition of 0.1% wine yeast(Pasteur Red) for 8 hrs at 35$^{\circ}C$.

Bioethanol Production from Popping Pretreated Switchgrass (팝핑전처리한 스위치그라스로부터 바이오에탄올 생산)

  • Switchgrass was selected as a promising biomass resource for bioethanol production through popping pretreatment, enzymatic saccharification and fermentation using commercial cellulase and xylanase, and fermenting yeast. The reducing sugar yields of popping pretreated switchgrass after enzymatic saccharification were above 95% and the glucose in thesaccharificaiton solution to ethanol conversion rate after fermentation with $Saccharomyces$ $cerevisiae$ was reached to 89.6%. Chemical compositions after popping pretreatment developed in our laboratory were 40.8% glucose and 20.3% xylose, with much of glucose remaining and only xylose decreased to 4.75%. This means that the hemicelluloses area broke off during popping pretreatment. FE-SEMexamination of substrate particles after popping pretreatment was showed fiber separation, and tearing and presence of numerous micro pores. These changes help explain, enhanced enzymatic penetration resulting in improved hydrolysis of switchgrass particles after popping pretreatment.

Enhancement of Ethanol Productivity by Air Supplement in Immobilized Cell Reactor System (균체고정화 생물반응기에서 산소공급에 의한 에탄올 생산성 향상)

  • To achieve higher ethanol productivity in the fermentation system, a continuous ethanol production has been investigated with the air-supplement in a packed-bed immobilized cell reactor system. Yeast cells were immobilized using sodium alginate gel. The results showed that, when the feed medium was saturated with oxygen through aeration into the medium reservoir, the maximum ethanol productivity of the reactor was enhanced from 35 g/$\ell$-gel-hr to 55 g/$\ell$-gel-hr at the residence time of 10-20 min. and the residence time for the 90% conversion of substrate to ethanol was reduced from 40 min. to 25 min. In case of 18% glucose medium, the maximum productivity was increased from 35 g/$\ell$-gel-hr to 45 g/$\ell$-gel-hr and time required for 90% conversion was from 90 min to 70 min. This behavior of air-supplemented reactor system might be due to the fact that both growth and viable fraction of yeast within the Eel were increased during reactor operation.

  • Temperature phase anaerobic co-digestion process was conducted with the one to one mixture of food wastewater with livestock wastewater, and the presence and the dynamics of various pathogenic microorganisms was analyzed. The mixture contained various enteric and pathogenic bacteria, such as Escherichia coli. Enterobacteriaceae, Coliform bacteria. Staphylococcus aureus, Salmonella, Shigella, Listeria, and Yeast. Anaerobic digestion has become stabilized around 21 days after the reaction started, showing about 80% to 90% of remarkable reduction rates of microorganisms until this period in acidogenic reactor (AR) and methanogenic reactor (MR), respect ively. After stabilization, the average reduction rate of organic matter was recorded as around 60% in MR. Most microorganisms in the effluent were not detected at around the last period of the reaction, except Listeria and S. aureus, which showed the growth even at the last day of the reaction.

Retardation of Kimchi Fermentation by the Extracts of Allium tuberosum and Growth Inhibition of Related Microorganisms (부추추출물의 김치발효 지연 및 관련 미생물 증식억제)

  • The effect of retarding the fermentation of Kimchi by the extract of leek(Allium tuberosum) were tested by measuring the changes in pH, acidity and total cell number as well as number of microorganisms involved in Kimchi fermentation such as Lactobacillus, Leuconostoc, Pediococcus, Streptococcus and yeasts. The changes of pH and acidity of Kimchi stored at $25^{\circ}C$ indicated that the shelf-life of Kimchi with leek extract was retarded by 1.5 days compared with Kimchi without leek extract. Growth of Lactobacillus, Leuconostoc, Pediococcus and yeasts in Kimchi were remarkably inhibited by adding the leek extract at the initial and the 1st day of fermentation. This result suggested the methanol extract of leek can be sucessfully used for the extension of shelf-life of Kimchi.
