Chemical Characteristics in Fermented Cordycepin-enriched Cordyceps militaris (발효 동충하초의 이화학적 특성)

  • The comparative effects of fibrinolytic, antioxidative activity and electrophoretical protein patterns with Cordyceps militaris powder (CM) and cordycepin-enriched Cordyceps militaris JLM0636 powder (CCM) and fermented cordycepin-enriched Cordyceps militaris JLM0636 powder by several microscopic organisms were investigated. In addition, nutritional materials such as mineral, protein, and fatty acids were also measured. The protein concentration was higher in CCM than that in CM. The protein concentration in fermented CCM was the highest in CCM treated by Aspergillus kawachii among the various samples. When total protein patterns of CM, CCM, and both fermented CCMs were analyzed by native- and SDS-PAGE, there were slightly varietal differences in electrophoretical protein patterns. Major minerals were K, Ca, Mg, and Zn. Major fatty acids were palmitic acid, stearic acid, oleic acid, linoleic acid, and linolenic acid. Fibrinolytic activity was the highest in the fermented CCM by Bs treatment among the various samples. The ${\alpha},{\alpha}^{\prime}$-diphenyl-${\beta}$-picrylhydrazyl (DPPH) radical scavenging activity was slightly stronger in the CCM treated with Aspergillus kawachii among the various samples; however, these samples all exhibited relatively low levels of activity compared with the butylated hydroxytoluene (BHT). These results may provide the basic data necessary to understand the biological activities and chemical characteristics of Cordyceps militaris JLM0636 powder fermented by several microscopic organisms to develop functional foods.

  • Immunoblotting and competitive indirect enzyme-liked immunosorbent assay(Ci-ELISA) was used for detection of ${\beta}$-lactoglobulin(BLG) in dairy products, such as milk, dried milk and fermented milk. In immunoblotting, human IgE weakly recognized proteins of fermented milk, but still responded to dried milk even though become weak. Rabbit polyclonal antibody to BLG, used as a model of antigen, and milk allergic patients' IgE was used in the ELISA. Reactivities of Abs were the highest in market milk. BLG in fermented milk was detected in a low content. This result indicates the fermented milk have the lowest BLG content and could be used as hypo-allergenic food for milk-allergic individual.

Effects of Fermented Milk Intake on Hepatic Antioxidative Systems in Alcohol treated Rats (알코올 투여한 쥐에서 발효유의 섭취가 간조직 내 항산화 체계에 미치는 영향)

  • Effects of fermented milk, $Kupffer's^{\circledR}$, intake on hepatic antioxidative systems were investigated in rats fed ethanol (3 g/kg B.W.) for 2 weeks. Serum AST and ALT were $88.7{\pm}6.5\;and\;41.2{\pm}4.1IU/L$ in control group, $114.6{\pm}7.1\;and\;64.7{\pm}3.8IU/L$ in alcohol group, and $94.0{\pm}5.5\;and\;44.7{\pm}5.3IU/L$ in fermented milk (FM) group, respectively. Fermented milk intake decreased hepatic glutathione peroxidase and superoxide dismutase activities of FM group to level of control group (p<0.05). Glutathione S-transferase activity of fermented milk group increased by 122% compared to control group. These results suggest antioxidative activities of lactic acid bacteria and ingredients in $Kupffer's^{\circledR}$ improve antioxidative system in alcohol-treated rats.

  • 대두유중에서 L, acidophilus와 K. fragilis를 단독발효시키는 것 보다 혼합배양 함으로써 젖산의 생성이 촉진되었는데 균간의 어떠한 상호작용에 의하여 산의 생산이 증가되었는가를 연구하였다. 대두유와 Soywhey를 사용하였을 때의 젖산 생성속도는 L. acidophilus 단독보다는 K. fragilis 와 혼합배양하는 것이 빨랐다. 이들 균을 단독 또는 혼합배양하여 경시적으로 발효 대두유중의 당 성분을 HPLC로 분석하였다. 그 결과 L. acidophilus가 발효하기 어려운 sucrosed raffinose, stachyose를 K. fragilis가 생산하는 효소에 의하여 젖산균이 발효할 수 있는 glucose, fructose로 분해되기 때문에 혼합배양 함으로써 젖산 발효가 촉진된다는 것을 알았다.

  • 본 연구는 가시오가피의 마크로파지 활성 효과를 평가하고, 발효유에 첨가할 경우 젖산균 성장에 미치는 영향에 대하여 실시하였다. 가시오가피 열수 추출물이 NO 분비능이 높은 반면, 알콜 추출물은 $IL-1{\alpha}$$TNF-\alpha$ 분비능이 높았다. 첨가 농도 간에는 열수 추출물 및 알콜 추출물 공히 1,000 ${\mu}g/mL$ 첨가하였을 때 유의성 있게 증가하였다. 가시오가피 열수추출물이 젖산균에 미치는 영향을 조사한 결과, 가시오가피 첨가량이 증가할수록 젖산균 성장이 억제되었고, pH는 완만히 감소됨에 따라 발효유에 가시오가피 열수 추출물을 첨가할 경우 set type보다는 stirred type이나 drink type으로 해야 발효유 제조에 적합할 것으로 보인다.

  • 가시오가피와 더덕 추출물을 첨가한 발효유가 마우스의 면역 기능에 미치는 영향을 알아보기 위하여 선발된 수컷 마우스를 대상으로 하여 발효유에 가시오가피와 더덕 열수 추출 건조물(가시오가피:더덕=8:2)을 혼합하여 투여한 그룹 1mg/mL(A), 3mg/mL(B), 9mg/mL(C)의 3 그룹으로 나누어 임상 적용 경로인 경구 투여를 선택하여 발효유를 각각 3mL/kg씩 위내로 직접 투여하였다. 대조군은 발효유만 먹인 그룹(D)과 식염수만 먹인 그룹(E)을 두었다. 실험 결과 모든 그룹에서 마우스의 증체량과 체중 증가는 유의성이 없었고, 7주와 10주령에 안락사 시킨 마우스의 각 장기인 간, 신장, 심장, 폐, 고환의 무게에서도 유의성이 없었다. 비장 계수는 7주령에서는 B군에서 유의성 있는 차이를 보였고 10주령에는 C군에서 유의성 있는 증가가 관찰되었다(p<0.05). 비장 계수는 식이 지속 기간이 길어질수록 증가되었고, 흉선 계수는 B군에서 유의성 있는 증가가 관찰되었다(p<0.05). 항체 생산 세포수는 가시오가피와 더덕을 급여 한 군 모두에서 대조군에 비해 증가하였으며, 특히 A, C군에서는 유의성(p<0.05)이 있었다. 모든 그룹에서 항체 생산 세포수는 10주령에서 7주령에 비하여 감소하는 경향을 보였다. 항원에 대한 항체 생성량을 알아보기 위하여 혈구 응집 반응을 실시한 결과 가시오가피와 더덕을 급여한 군에서 대조군에 비하여 증가하는 경향을 보였고 7주령 A, C 군에서는 유의성 있는 차이를 보였으나 10주령 에서는 C군에서만 유의성있는 차이가 나타났다(p<0.05).

Antibacterial activity of lactic acid bacteria isolated from traditional fermented foods and development of a starter for fermented milk (전통발효식품에서 분리한 유산균의 항균 활성 및 발효유 스타터 개발)

  • This study was conducted to investigate the antibacterial activity of lactic acid bacteria isolated from traditional fermented foods and to develop a new starter for fermented milk. The isolates were identified using 16S rDNA sequencing and named Lactobacillus plantarum A, Leuconostoc lactis B and L. acidophilus C. The activity of these strains to inhibit the growth of food-borne human pathogens (Escherichia coli NCTC 12923, Salmonella Typhimurium NCTC 12023, Listeria monocytogenes NCTC 11994) was measured using the paper disc method. All these strains showed strong antibacterial activity against Li. monocytogenes NCTC 11994. The experiment groups were the fermented milks with these strains, and the control group was the fermented milk with the commercial starter (ABT 5). The change of pH, acidity and viable cell counts were measured during their aging time. All the experiment groups showed a significant difference in their aging times compared to the control group. However, the sensory test showed that the experiment groups can be used as useful starters for fermented milk. This result suggests that L. plantarum A, Leu. lactis B and L. acidophilus C have the potential to be developed as new starters for fermented milk.

  • This study was carried out to investigate characteristics of acid and alcohol fermented milk by mixed starters made by Lactobacillus bulgaricus (KCTC 3635) and Kluyveromyces marxianus (KCTC 17212) for 36 hours when the curds were formed. Final pH and titratable acidity were about 4.5 and 0.68%, respectively. The viable cell counts of lactic acid bacteria and yeast for alcohol fermented milk were increased to $3.2{\times}10^9 CFU/mL$ and $5.3{\times}10^9 CFU/mL$, respectively. The ethanol contents increased to 0.35% during fermentation. Antitumor activities of the fermented milk against tumor cell lines, such as HEp-2, HEC-1B, SW-156 and SK-MES-1 showed to 86.6, 70.3, 60.4 and 57.14%, respectively.

  • This study was desiged to carry out an observation study on the cognitive level of health benefitis of fermented milk and subsequntly, on prevention of gastrointestinal diseases for the consumers in Korea. Data used in this study were collected from two different source ; 1) 987 university students living in Seoul, were selected randomly and interviwed induvidual from May 25 to September 30 1994, to investigate the awareness status as a consomer of fermented milk, and 2) health benefits of fermented milk with respect to prevention gastric intestinal diseases such as typhoid fever, paratyphoid fever and bacillary dysentery in Korea with the raw data from the Yearbook of Health and Social Statistics by Ministry of Health and Social Affairs (1976${\sim}$1995) and Ministry of Agriculture, Forestry & Fisheries (1976${\sim}$1995), Republic of Korea. The results of this study were as follows : The effects of femented milk on health appeared to be attributed to taking care of an environment of the gastrointestinal tract regarding diarrhea, constipation and digestion, with showing 75.1% of male and 71.1% of female of university students in Seoul answered positively, The correlation coefficients of statistics between amounts of consumption per capita of fermented milk and incidence rates of gastrointestinal diseases during the period from 1975 to 1994 in the whole country was r = -0.308 (p<0.05). those of Seoul a major comsuming area of fermented milk in the country and Kangwon province, a minor consuming area were r = -0.704 (p<0.01) and r = +0.262 (n.s.), respectively. In conclusion, the results obtained in this study suggest that fermented milk exert its effect on gastrointestinal tract. Furthermore, the results warrant a futher study in the field of preventive medicine and health science with respect to efficacy of fermented milk consumption.

  • Lactobccillus acidophilus와 Saccharomyces cerevisiae를 대두유에서 혼합 배양시켜 최적 젖산발효조건을 검토하였다. 대두유에서의 혼합배양시 20시간 이상 배양했을 때 산생성이 촉진되었고 배양온도는 34$^{\circ}C$ 부근으로 했을 때 가장 좋았다. 대두유에 sucrose와 skim milk가 젖산발효에 미치는 영향을 조사한 결과 sucrose 는 1.5% 첨가시 skim milk는 2.0% 첨가시 첨가하지 않고 배양할 때 보다 2배 정도의 산생성을 보였다.
