Enzyme Inhibition Activities of Ethanol Extracts from Germinating Rough Rice (Oryza sativar L.) (발아기간에 따른 벼(Oryza sativa L.)의 부위별 효소저해활성)

  • This study investigated the enzyme inhibitory effects of ethanol extracts from the different parts of rough rice (Oryza sativar L.) from its germination period. Rough rice was germinated at $37^{\circ}C$ for 6 days, then separated into hull+sprout and brown rice. ${\alpha}$-Glucosidase inhibitory activity had the highest value (39.38%) in hull+sprout extracts after 5 days of germination. ${\alpha}$-Amylase and DPP-4 inhibitory activity had the highest values (75.32% and 47.77%, respectively) in hull+sprout extracts after germination for 5 days. ACE inhibitory activity of hull+ sprout extracts increased from 43.16% at the beginning to 58.60% at 5 days, while brown rice extracts increased this activity from 0.88% at the beginning to 14.50% at 4 days. The xanthine oxidase inhibitory activity of hull+ sprout extracts increased from 62.02% at the beginning to 64.49% at one day, and then decreased. Lipase inhibitory activity had its highest value (55.17%) in hull+sprout extracts after germination for 5 days. These results indicate that the optimal germination period for increasing enzyme inhibitory activities may be 5 days, and that hull+sprout extracts have a higher enzyme inhibition activity than brown rice.

The Enzyme Inhibitory Activity of Ethanol Extracts Derived from Germinated Rough Rice (Oryza sativar L.) Treated by High Pressure (발아와 고압처리에 따른 벼(Oryza sativar L.) 추출물의 효소저해활성)

  • We evaluated the enzyme inhibitory activity of germinated rough rice (Oryza sativar L.) treated by high pressure (30MPa) for 24 h (HP24) and 48 h (HP48). In rice germinated for 1 day, the ${\alpha}$-glucosidase inhibitory activity reached its highest level, 68.32%, at HP48. The ${\alpha}$-amylase inhibitory activity increased from 32.66-57.00% at HP0, to 43.67-74.82% at HP48. On the other hand, the inhibitory activity of angiotensin-converting enzyme increased from 27.98% to 49.42% over the course of the second day of HP48. The inhibitory activity of xanthine oxidase peaked of 67.51% at HP48 and subsequently decreased. Lipase inhibitory activity increased from 24.04-47.91% at HP0, to 29.62-64.63% at HP48. These results provide useful information for the use of germinated rough rice as a functional food material and demonstrate that high-pressure treatment during the germination process efficiently increase enzyme inhibitory activity.

Allelopathic Inhibition by Extracts and Volatiles from Leaf and Seed of Sicklepod (Cassia tora L.) (결명자(決明子)(Cassia tora L.) 엽(葉)과 종자(種子)의 추출물(抽出物)과 휘발성분(揮發成分)의 타감성(他感性) 생육저해작용(生育沮害作用))

  • The growth of weeds and some other plants has been considered to be inhibited by sicklepod(Cassia tora L.) sharing the habitat. The study was conducted, for the first time, to propose that this phenomenon is not only due to competition for physical and nutritional conditions but also due to allelopathy. In addition, autotoxicity of sicklepod was examined. The results are summarized as follows. 1. Rice sheath length reduced progressively from 18 % to 36 % with increasing the concentration of treating aqueous extracts of sickle pod seeds, but rice germination was not affected. In contrast, radish hypocotyl length was not reduced by the aqueous extract treatment but radish germination was significantly reduced by 66 % at 1 : 10 and 1 : 5 treatment. 2. Total chlorophyll contents in rice seedling decreased from 50 % to 65 % by treatment of seed aqueous extracts diluted from 1 : 50 to 1 : 5 ratio. 3. Aqueous extracts of sicklepod leaves significantly reduced hypocotyl length and fresh weight in radish and germination in rice, but mung bean was slightly affected by aqueous extracts only in fresh weight. 4. Volatiles from fresh, immature seeds with husk reduced the radish germination and seedling growth and radish root growth appeared to be more sensitive to the exposure to volatiles from fresh immature seeds than both germination and hypocotyl growth. 5. Volatiles from sickepod leaves inhibited germination and growth of radish, rice and mung bean, and seedling growth was more sensitive to volatiles from leaves than germination. 6. Volatiles from sicklepod leaves reduced germination and radicle length of sicklepod itself. 7. Collectively, it is concluded that there are water-soluble and volatile substances responsible for allelopathy in sicklepod.

Comparison of Physiological Activities of Radish Bud (Raphanus sativus L.) according to Extraction Solvent and Sprouting Period (추출용매 및 발아시기에 따른 무순 추출물의 생리활성 비교)

  • This study extracted radish bud (Raphanus sativus L.) and investigated its nitrite scavenging activity, superoxide dismutases (SOD)-like activity, tyrosinase inhibition activity, xanthine oxidase inhibition activity, and angiotensin-converting-enzyme (ACE) inhibition activity according to extraction solvent and sprouting period. For nitrite scavenging activity, each extract recorded its highest level of 81.44~89.71% at pH 1.2. Radish bud extracts on sprouting days 4 and 8 showed greater scavenging activities than those on sprouting day 12 at pH 1.2 and pH 4.0. There were differences in scavenging activity according to extraction solvent based on water extract exhibiting improved scavenging activity. Ethanol extract recorded scavenging activity of 16.12% at pH 6.0, which was similar to those of ethanol and methanol radish bud extracts on sprouting day 12. SOD-like activity of radish bud extracts was in the range of 4.57~27.05%. For comparison purposes, SOD-like activity of L-ascorbic acid was 52.15%, which was higher than that of radish bud extracts. Acetone and methanol extracts showed high SOD-like activities on sprouting day 8. SOD-like activity of radish bud extracts on sprouting day 12 significantly decreased to 4.57~15.59%. Radish bud extracts recorded good tyrosinase inhibitory activities on sprouting 8 and 12, whereas methanol extracts recorded the greatest tyrosinase inhibitory activity at 62.65~84.89%. Radish bud extracts recorded xanthine oxidase inhibition activity of 21.26~29.52% on sprouting day 4, and acetone extracts showed the highest level of xanthine oxidase inhibition activity. Xanthine oxidase inhibitory activity tended to decrease with sprouting period compared early on. ACE inhibitory activity was in the range of 12.48~51.78% according to sprouting period and extraction solvent. Ethanol extracts on sprouting day 8 showed the highest ACE inhibitory activity of 51.78%. These results will hopefully contribute to research into the identification of materials and development of products for natural functional foods.

The Factors Influencing the Systemic Action of Dimethoate (O.O-dimethyl-S-(N-methylcarbamoylmethyl) phosphorodithioate) to the Rice Seeds and Phytotoxic Effects (수도종자에의 Dimethoate 침투력 및 발아저해에 관여하는 요인에 관한 연구)

  • These experiments were conducted to investigate the :actors influencing the systemic action of Dimethoate (O,O-dimethyl-S-(N-methylcarhamoylmethyl) photphorodithioate) to rice seeds and the phytotoxic effects on the seed germination. Dimethoate $(Roxion^{(R)})$ $40\%$ emulsion was used. The varieties tested were Jinheung. Nongkwang,Suwon #82, Norm #6, Paltal, Shirogane, Suseong, Pungkwang, Shin #2, Fujisaka #5, Kwanok, and Jaekeun. The permeated Dimethoate was extracted from the treated seeds by chloroform and quantities were determined by Spectrophotometer. The phytotoxicity was evaluated from the effects on the germination of the treated seeds which were kept in an incubator. The oxygen consumption was measured by Warburg Manometer at $30^{\circ}C$ for 60 minutes. Indices of KOH disintegration of seeds and chemical composition of the seeds were also determined. The results obtained were as followings; 1) The amount of permeated Dimethoate in the seeds showed remarkable differences with varieties. The amount of Dimethoate per 100 grains was greater as in the ascending order of Suseong, Kwanok, Nongkwang, Jinheung, Paltal, Fujisaka #5, Suwon #82, Norm #6, Shirogane, Shin #2, Pungkwang and Jaekeun. 2) It was observed that the total amount of Dimethoate in the seeds(mg./100 grains) were greater among the varieties with large grain than those with small grains, while reverse cases were true in the amount of Dimethoate in a gramme of seeds, probably because of the greater surface areas In a small grains for a gramme weight. 3) There was no significant correlation between the permeated amount of Dimethoate and amount of absorbed water by the seeds when the seeds were treated with $0.1\%$ Dimethoate for 24 and 48 hours. 4) The permeability of Dimethoate to seeds significantly increased in the prolonged soaking periods, higher concentration, and higher temperature. 5) When the seeds were treated with $0.1\%$ Dimethoate for 24 and 48 hours at $15^{\circ},\;20^{\circ},\; 20^{\circ},\; and \;30^{\circ}C$, the permeated amount of Dimethoate were increased at higher temperature. It seems to be that the more active penetration of Dimethoate was involved at the higher temperature. 6) The phytotoxic effects of Dinethoate on the seed germination varied with the varieties. An descending order of varietal tolerance of seeds was as followings: Jinheung, Fujisaka #5, Suwon #82, Paltal, Nongkwang, Jaekeun, Shin #2, Kwanok, Shirogane, Pungkwang, Suseong, and Norm #6. 7) There was a positive correlation between the amount of Dimethoate permeated into the seeds (mg./gram. of seeds) and phytotoxicity of seeds. 8) The Phytotoxic effects of Dimethoate showed close correlation with the degree of KOH disintegration of seeds, average germination periods, and oxygen respiration of seeds. 9) It was observed that higher protein contents of the seeds decreased the phytotoxic effects of Dimethoate. 10) Relatively high negative correlation between the degree of KOH disintegration of seeds and crude protein content of the seeds was observed. 11) The average germination period was delayed for about 2 days when the seeds were treated with $0.2\%$ Dimethoate for 24 hours at $30^{\circ}C$. 12) The oxygen consumption of the seeds treated with $0.2\%$ Dimethoate for 24 hours at $30^{\circ}C$ was greatly decreased when compared with that of the normal seeds. 13) The amount of oxygen consumption of the seeds (in 24 hours after 24 hours water soaking) was negatively correlated with the average germination periods of the seeds.

Effect of Water-Extracts from Sludge Compost on Seed Germination (퇴비의 부숙 과정 중 추출물이 종자 발아력에 미치는 영향)

  • In order to evaluate the physicochemical properties of sludge compost and to identify the effects water-extracts from sludge compost in 2, 6, 8, 12 weeks of decomposing process on seed germination and root elongation in cabbage, lettuce, soybean and barley plants was investigated. The content of total nitrogen in sludge compost increased slightly in 6 weeks decomposing process, and then decreased gradually. Organic matter content decreased continuously overall decomposing process. As decomposition was processing, pH of sludge compost decreased slightly, and EC increase within 6 weeks decomposing process, and then decreased. The content of nitrogen in water-extracts from sludge compost increased within 8 weeks decomposing process and decreased in 12 weeks decomposing process. The content of ammonium nitrogen was similar with that of total nitrogen, and the ratio of ammonium and nitrate increased within 8 weeks, and then decreased. Cation content and EC decreased the late of decomposing process and pH didn't change. The water-extracts from sludge compost during decomposing process inhibited seed germination and root elongation in cabbage (Brassica campestris), lettuce (Lactuca sativa), barley (Hordeum vulgare) and soybean (Glycine max). The inhibition of root elongation in cabbage was greater than that of relative seed germination, whereas relative seed germination in lettuce was more inhibit than root elongation. Relative seed germination and root elongation in soybean were inhibited slightly, but those of in barley was inhibited strongly. In this study, we would identify the effects of water-extracts from sludge compost on seed germination and root elongation was different to the species of seed. The inhibition of seed germination and root elongation treated with the water-extracts which extracted from sludge compost in the early stage of decomposing process was greater than that of in the late stage of decomposing process.

Antibiotic Properties of an Entomopathogenic Fungus, Beauveria bassiana, on Fusarium oxysporum and Botrytis cinerea (살충성 진균 Beauveria bassiana의 Fusarium oxysporum과 Botrytis cinerea에 대한 항균활성)

  • 식물병원균 Botrytis cinerea와 Fusarium oxysporum에 대한 생물학적 방제의 기초자료를 얻기 위하여 살충성 진균 Beauveria bassiana의 식물병원균에 대한 균사성장저해, 포자발아 억제, 균사와 포자의 형태변화 등의 효과를 살펴보았다. 평판배지 상에서 두 식물병원균의 균사생장이 저해되었으며, 저해효과는 배지종류에 따라 달랐는데 B. cinerea의 경우 PDA배지에서 가장 크게 저해되었으며 F. oxysporum의 경우에는 TSA배지에서 가장 크게 저해되었다. B. bassiana는 $25^{\circ}C$에서 6일간 배양했을 때 F. oxysporum에 대한 항균력이 가장 높았으며 동시에 최대의 균체량을 생산하였다. 또한 B. bassiana의 배양여액을 식물병원균에 30% 농도로 첨가하여 배양했을 때 식물병원균의 포자 발아율은 B. cinerea, F. oxysporum에서 각각 30%(control: 88.2%), 10.0%(control: 78.6%)로 낮아졌으며 발아 개시 시간도 4~8시간 지연되었다. 현미경을 통한 미세구조관찰에서는 10%의 B. bassiana배양여액을 첨가했을 때 F. oxysporum의 포자 크기가 1/2~1/3으로 줄어들었으며 균사와 격막의 형태도 비정상적으로 변하여 균사외막과 격막-격막 사이의 구분이 불명확해졌다.

Development of Seed Pelleting Technology for Rice and Cabbage (벼 및 배추종자 Pelleting을 물질채색 및 기술개발)

  • Seed pelleting have successfully been used in many crops for better crop estab-lishment and for mechanizing seeding process for small crop seeds in developed countries. In this experiment various pelleting materials and binders were tested to get basic information on the shape, hardness and germination of pelleted seesds of cabbage (cv. Seoul Beachoo) and rice (cv: Ilpoom). PLL-11, paper clay, lime and coal ash were good materials to make smooth shape of the pellets with pel gel and AG-11 as binders, and PLL-ll as material and pelgel and AG-11 as binders were the best among them in consideration of shape and hardness together. The hardness of the pelleted seeds were differed with each other depending on both of the pelleting materials and binders. Pelleted cabbage seeds coated by pelgel as binder with different materials showed lower germination percentge than control in general, but the seeds pelleted by PLL-11 with different binders showed no restraint effects. When the cabbage seed pelleted by PLL-11 with pelgel as binder showed almost same germination percentage as control. The pH and electrical conductivity of the extract from bentonite and zeolite were very higher than other materials tested and germination percentage showed a little lower than control when the cab-bage seed planted on the filter paper damped with the extract. As a result, PLL-11 as pelleting material and pel gel and AG-11 as binder appeared the good materials to make pellets of cabbage seeds and rice in consideration of shape, hardness and germination.

Effects of Seed Treatment with Several Systemic Insecticides to Rice, Barley and Soybean (수도, 대맥 및 대두에 대한 침투성살충제의 종자분의처리효과)

  • An experiment was carried out to investigate possible use of several systemic insecticides as seed treatment to control some insect pests of rice, barley and soybean. The insecticides were impregnated with active carbon dust and $2\%$ water solution of methyl cellulose as skicker. The seeds were dressed with the insecticides: Fussol (Monofluoroacetamide), Dimethoate (0, 0-dimethyl-S-(N-methyl carbamoylmethyl) phosphorodithioate), Metasystox (0, 0-dimethyl 0-2 ethylthioethyl phosphorodithioate), Dimecron (0, 0-dimethyl (diethylamide-1-chlorocrotonyl) Phosphate). The seeds were treated at the rate of $4\%$ by grain weight. Effects of seed treatments with systemic insecticides on the seed germination and insecticidal effectiveness were tested. Fer rice, no seed germination was observed in Fussol treatment. Dimethoate, Metasystox and Dimecron showed a slight reduction in germination when seeds were sown 1 day after treatment, but they showed gradual reduction in germination when seeds were sown 20 and 45 days after treatment. Especially Diriethoate treatment showed significant reduction in germination after 20 and 45 days of storage. All the insecticides reduced seedling stands of barley somewhat. No plant emergence was observed in Fussol treatments after 25 and :to days of storage. In case of the 40 days of storge in barley seed treated with Dimethoate, Hetasystox and Dimecron there was no gignificant reduction in plant emergence except Fussol treatment. Dimethoate was highly toxic to the soybean seed viability. However, there was no significant reduction in the soybean germination after 1 day of storage. in Fussol, Metasystox and Dimecron treatments After 25 days of storag in Fussol and Dimethoate treatments, no plant emergence was observed, and there was $13.3\%$ emergence in Metasystox treatment, and $93.0\%$ emergence in Dimecron treatment. Dimethoate treatment in rice seed(variety Suwon # 82) showed $90\%$ mortality at the twentieth day and $33.3\%$ at the thirtieth day after sowing against the green rice leafhopper, Nephotettix cincticeps But no visible effectiveness was observed in Metasystox and Dimecron treatments. Dimethoate treatment in barley seed resulted in low population density of aphids. Rhopalosiphum spp. Final observation on the number of eggs per plant was made at the fiftieth day after sowing; 2 in Dimethoate, 4 in Fussol, 6 in Metasystox, 7 in Dimecron, and 13 in check plots. All the insecticides tested resulted in good insecticidal effectiveness against the aphids, Aulocorthum solani on soybean plant at the ninth day infestation after sowing. Dimethote, Metasystox, and Dimecron treated soybean plants was still effective in controlling the aphids to the eighteenth day infestation.
