Variation of Seed and Germination Characteristics of Natural Populations of Abies koreana Wilson, a Korean Endemic Species (한국특산(韓國特産) 구상나무 천연집단(天然集團)의 종자(種子).발아특성(發芽特性) 변이(變異))

  • This study was conducted to investigate the variation of seed and germination characteristics among populations and among individuals within populations of Abies koreana Wilson distributed in Korea. Cones was collected from 4 natural populations and their seven seed qualities as well as three germination behaviors was measured and tested. Statistical analysis showed that there were significant differences among populations as well as among individuals within populations in 8 traits except for percentage of sound seeds and seed vigor. Coefficients of variation in percentage of sound seeds, germination percentage and germination rate is relatively high (43.8~57.2%) compared to other traits (11.0~28.3%). The germination of seed started on the seventeen day after sowing and it was completed within seventy-nine days. The final seed germination capacity was found high except for Mt. Duckyu (8.5%) and varied from 39.7% to 47.7%. Mean germination time ranged between 38.1 and 43.6 days and germination rate varied from 1.98 to 1.39 ea./days. As a result of simple correlation analysis between seed characteristics and germination characteristics, the percentage of sound seeds showed a significant positive correlation with the germination percentage and the germination rate but showed a negative correlation with the mean germination time.

  • For the study of method for promoting the germination of Korean sod seeds harvseted newly some physical and chemical treatments were tested in this experiment. The summarized results of these tests are as follows: 1. Storage of wet seed in low temperature of 0~$2^{\circ}C$ during 2~30 days was seemed the most effective treatment for promoting the germination of sod seed. 2. The treatment that Soaking and mixing up the seeds in cone sulphuric acid about one and half minutes and then washing off in the water promoted fairly the germination of sod seeds. 3. Better germination of sod seeds was found in the light than in the dark or soil. 4. Cutting the top of seed and crushing of seed coat with sand seemed to promote slightly the germination of sod seeds. 5. No promoting effect for germination of sod seeds was found with treatments of Gibberellin, $NH_4$ $NO_3$ and NAA.

Effects of Germination in Brown Rice by Addition Chitosan/Glutamic acid (Chitosan/Glutamic acid 처리가 현미의 발아에 미치는 영향)

  • To improve the levels of ${\gamma}$-aminobutyric acid (GABA) in germinated brown rice, chitosan and glutamic acid were treated during the brown rice germination. The GABA contents in germinated brown rices were 425.7 nmole/g and 637.0 nmole/g at germination temperature of $25^{\circ}C$ and germination time of 72 hrs. Response surface methodology(RSM) was used to monitor characteristics of germination from brown rices. As glutamic acid and chitosan concentration were increased, the GABA content was also increased. The ranges of optimum conditions were $105{\sim}160\;ppm$ in chitosan concentration and $200{\sim}290\;ppm$ in glutamic acid concentration. Predicted values at the optimized conditions were acceptable in comparison with experimental values.

  • This research was carried out to promote the seed germination of Styrax japonica, by several pretreatments and to examine its seedling growth and seed component variations according to time and pretreatment. The results are as follows ; 1. Styrax japonica seeds are usually germinated after more than one year cold stratification. However, when the seeds were stratificated for 5 months after dipped in the solution of 70% $H_2SO_4$ for 30 minutes and also treated with $GA_3$(3000ppm), they showed 55% germination. This demonstrated that the seeds could be germinated in one year when pretreated. 2. The growth of one-year-old seedlings after pretreatment was the same as that of two-year-old seedlings when compared 160 days after seeding. 3. Before seeding, one-year-old, pretreated seeds of Styrax japonica showed increased moisture content but decreased crude fat, carbohydrate and exude ash, which were similar to the components of two-year-old seeds.

The Effects of Experimental Warming on Seed Germination and Growth of Two Oak Species (Quercus mongolica and Q. serrata) (온난화 처리가 신갈나무(Quercus mongolica)와 졸참나무(Q. serrate)의 종자발아와 생장에 미치는 영향)

  • Population growth and the increase of energy consumption due to civilization caused global warming. Temperature on the Earth rose about $0.7^{\circ}C$ for the last 100 years, the rate is accelerated since 2000. Temperature is a factor, which determines physiological action, growth and development, survival, etc. of the plant together with light intensity and precipitation. Therefore, it is expected that global warming would affect broadly geographic distribution of the plant as well as structure and function ecosystem. In order to understand the effect of global warming on the ecosystem, a study about the effect of temperature rise on germination and growth in the plant is required necessarily. This study was carried out to investigate the effects of experimental warming on the germination and growth of two oak species(Quercus mongolica and Q. serrata) in temperature gradient chamber(TGC). This study was conducted in control, medium warming treatment($+1.7^{\circ}C$; Tm), and high warming treatment ($+3.2^{\circ}C$; Th) conditions. The final germination percentage, mean germination time and germination rate of two oak species increased by the warming treatment, and the increase in Q. serrata was higher than that in Q. mongolica. Root collar diameter, seedling height, leaf dry weight, stem dry weight, root dry weight, and total biomass were the highest in Tm treatment. Butthey were not significantly different in the Th treatment. In the Th treatment, Q. serrata had significantly higher H/D ratio, S/R ratio, and low root mass ratio (RMR) compared with control plot. Q. mongolica had lower RMR and higher S/R ratio in the Tm and Th treatments compared with control plot. Therefore, growth of Q. mongolica are expected to be more vulnerable to warming than that of Q. serrata. The main findings of this study, species-specific responses to experimental warming, could be applied to predict ecosystem changes from global warming. From the result of this study, we could deduce that temperature rise would increase germination of Q. serrata and Q. mongolica and consequently contribute to increase establishment rate in the early growth stage of the plants. But we have to consider diverse variables to understand properly the effects that global warming influences germination in natural condition. Treatment of global warming in the medium level increased the growth and the biomass of both Q. serrata and Q. mongolica. But the result of treatment in the high level showed different aspects. In particular, Q. mongolica, which grows in cooler zones of higher elevation on mountains or northward in latitude, responded more sensitively. Synthesized the results mentioned above, continuous global warming would function in stable establishment of both plants unfavorably. Compared the responses of both sample plants on temperature rise, Q. serrata increased germination rate more than Q. mongolica and Q. mongolica responded more sensitively than Q. serrata in biomass allocation with the increase of temperature. It was estimated that these results would due to a difference of microclimate originated from the spatial distribution of both plants.

  • 부처꽃 하면 초여름에 곧게 뻗은 빨간 꽃차례도 아름답지만 자생화 중 가장 수명이 긴 화종이라는게 생각난다. 몇 년 전 우리 자생화들의 종자수명을 알아보고자 냉장고에서 보관 중이던 약 50종의 종자를 당년산부터 최장 12년산까지 뿌려 발아력을 조사한 적이 있는데, 당시 부처꽃만 100% 가까이 발아되어 정말 수명이 긴 종류구나 하는 것을 느꼈던 기억이 난다. (중략)

The Effects of ultrasonic irradition on germination of mung bean (녹두 발아에 미치는 초음파 조사의 영향)

  • The experiment was carried out to examine on effect of ultrasonication on the growith of a mungbean while a mungbean is germinated and to measure amino acid content of a mungbean during its germination. Mungbean seeds soaked are divided into three groups; the first group was control group, untreated and naturally germinated. the second was ultrasonic treated group(UTG I), irradiated by ultrasonication for 8, 16 and 24min respectively immediately after soaking. the third was ultrasonic treated group(UTG II), irradiated by ultrasonication after 48hr germinating. The results were as foolows; Fresh weight, moisture, length of mungbean increased as its germination proceeds. while dry weight decreased. Ultrasonication tends to inhibit its germination of mungbean and results in the decrease in weight, length and moisture. Specially ultrasonication had a marked effect on UTG II. The longer the time of ultrasonication was, the more was the growth inhibited. In the control groups, the content of amino acid increased until 72 hours, but after that, it decreased. of the amino acid determined, glutamic acid decreased marekedly in the cotyledons, While the opposite was the case with aspartic acid in the hypocotyls. Comparing with the control groups, UTG I, the content of amino acid decreased. As the ultrasonic treaed time was increased, the concentration of amino acid in hypocotyl cotyledon tended to decreased. UTG II, the content of amino acid was lower than control groups, but higher than UTG I, and the effects of ultrasonication were similar to UTG I.

Effects of Several Pre-treatments on Seed Germination or Sophora japonica L. (회화나무 종자발아에 미치는 전처리의 효과)

  • This experiment was conducted to study the effects of sulphuric acid, cutting, cold stratification and hot water on the germination of Sophora japonica seeds, which have difficulty to germinate because they have hard and thick seed coats. The seeds were immersed in 30, 60 and 90% sulphuric acid for 30 minutes as seed scarifications, and cut 1/8, 2/8 and 3/8 of the opposite parts of radicles. As cold stratification the seeds were wrapped in wet towel, and then stored in plastic bags in a refrigerator $(4^{\circ}C)$ for 3, 5 and 10 days. The seeds were immersed in hot water $(90{\sim}95^{\circ}C)$ for 2,5 and 10 minutes. Pretreated seeds represented different germination properties, respectively. After sulphuric acid treatment, the seeds showed an increase in germination (G) and germination index (GI). Compared to the other treatments, 90% sulphuric acid showed the highest G (31.7%) and GI (6.2). The G and GI of cut seeds decreased with the increase of seed cutting lengths. And G and GI of cold stratificated seeds were not significantly different among the days treated (p=0.258). Two minutes treatment of hot water showed lower G and Gl than control, and the seeds were not germinated in 5 and 10 minutes treatments of hot water. At the result of relative growth rate and T/R ratio of seedlings from pretreated seeds, the seedlings from seeds in 90% sulphuric acid treatment represented the highest relative growth rate and T/R ratio.

In Vitro Culture of Immature Embryo Obtained by Crossing between Tetraploid Grape 'Fujiminori' and Triploid 'Summer Black' (포도 4배체 '후지미노리'와 3배체 '썸머블랙'의 교배로 얻은 미숙배의 기내배양)

  • For the germination and differentiation of immature embryos obtained by artificial crossing between tetraploid grape 'Fujiminori' (Vitis vinifera ${\times}$ V. labruscana) and triploid 'Summer Black' (V. labruscana ${\times}$ V. vinifera), were incubated in vitro using MS medium supplemented with $GA_3$ or coconut water (CW) at various concentrations. The percentage of embryo formation of 'Fujiminori' ${\times}$ 'Summer Black' was 64.3%. Embryo germination percentage was higher than 95% in all the $GA_3$ treatments at the concentrations of 0.01, 0.05, 0.25, and $1.25mg{\cdot}L^{-1}$. However, only 15.8-31.6% of the germinated embryos successfully developed into normal plantlets. At higher concentration of $GA_3$, the plantlets developed infirm hypocotyls with over elongated and less enlarged structure. Among the treatments of CW at the concentrations of 5, 10, 15, and 20% (v/v), 10% and 15% were more effective and plantlet achievement percentage were 68.4 and 66.7%, respectively. The addition of 10% CW was most effective to obtain plantlets with optimal shoot length, node and root numbers. 15% CW was suitable to obtain plantlets with longer roots. Accordingly, the embryo culture using the MS medium supplemented with 10-15% CW was observed to be more efficient for germinating and growing the immature embryos produced from artificial crossing between tetraploid grape 'Fujiminori' and triploid 'Summer Black'.

Effects of low dose $\gamma$-ray on the early growth of tomato and the resistance to subsequent high doses of radiation (저선량 $\gamma$선 조사가 토마토의 초기생육과 후속고선량 $\gamma$선 저항성에 미치는 영향)

  • Tomato (Lycopericum esculentum $M_{ILL}$ cv. Seokwang and cv. Housemomotaro) seeds were irradiated with the doses of $1{\sim}20$ Gy from $^{60}Co$ $\gamma$-ray source to investigate the effect of the low dose $\gamma$-ray radiation on the early growth and resistance to subsequent high dose of radiation. Germination rate of seeds irradiated with low dose $\gamma$-ray was enhanced in Seokwang cultivar but not in Housemomotaro cultivar. Seedling height increased in 4 Gy and 8 Gy irradiation group of both cultivars. Plant height of Seokwang cultivar was depressed in low dose irradiation group but fresh weight was increased in 2 Gy and 4 Gy irradiation group. In Housemomotaro cultivar, plant height increased in 12 Gy and 20 Gy irradiation group and fresh weight increased in 4 Gy and 20 Gy irradiation group. Growth inhibition of tomato plants by high dose radiation was noticeably reduced by pre-irradiation of low dose radiation. Resistance to subsequent high dose of radiation was enhanced in 2 Gy and 8 Gy Irradiation group of Seokwang cultivar and in 2 Gy and 12 Gy irradiation group of Housemomotaro cultivar.
