The Effect of Renewal Topworking on Early Y Shape Tree Formation and Yields in Peach Trees (고접갱신이 복숭아 Y자 수형 조기 구성 및 생산성에 미치는 영향)

  • The effects of the early tree shapes with renewal by top-working on Y-shaped peach tree, and the influence on the maintaining fruit quantity with gradual renewal of interstock cultivar were determined. In the comparison of the places of top-working tree, top working tree on the inside of the main branch of interstock cultivar had higher graft union rate and branch growth than those of top working tree on the outside. Tree width, basal diameter of shoot, and number of bearing shoots were smaller in top working tree than in replanted tree. Although labor time was not different to control top working tree and replanted tree, labor time was much required to manage top working tree with interstock cultivar. Accumulated fruit production was 2,384 kg/10a in top working tree and 2,025 kg/10a in replanted tree for three years. However, top working tree had no loss of fruits because interstock cultivar of top-working tree had 3,727 kg/10a of fruits. No variation on fruit quality was observed between top working tree and replanting tree. In terms of economic value of top-working tree, labor to manage interstock cultivar, fertilizer price, fruit bagging, and grading and packing price increased. However, fruit production increased, and price of seedling, rental equipment, pulling-out trees, and repairing supporting system decreased. Therefore, gradual renewal of topworking tree has effects on the maintaining fruit quantity, supplementation on fruit loss, and renewal cultivar.

  • Typhula incarnata grew over a temperature range of -5 to $20^{\circ}C$ with maximum growth at 10 to $15^{\circ}C$. Sclerotial production for T. incarnata was greatest at the higher temperature. Maximum mycelial growth of this pathogen occurred from pH 5.4 to 6.2. When carbon sources were added to a basal salt medium (Czapek's dox agar) at 5 g carbon sources/l, inulin, soluble starch, galactose, glucose, mannose, manitol, sucrose, maltose, cellobirose, trehalose, raffinose, and dextrin supported growth better than other carbon sources did. Of the twenty-three nitrogen sources tested, glycine, serine, ammonium sulfate, asparagine, asparatic acid, and ${\beta}-alanine$ were the most favorable for mycelial growth of T. incarnata. Cystine and cysteine were poor nitrogen sources. Ammonium salt of nitrogen sources supported growth better than nitrate salt of nitrogen sources. Potato dextrose agar, oat meal agar, and V-8 juice agar were the most favorable for mycelial growth and sclerotial formation. Appropriate addition of pepton to PDA decreased mycelial dry weight, but sucrose supported good growth of T. incarnata. Percent viable sclerotia of T. incarnate buried in bentgrass soil decreased from 2 months after treatment remarkably. Trichoderma riride and bacteria were isolated from non-germinated sclerotia. Live orchard grass leaf pieces within the soil were colonized by T. incarnata better than sterile and unsterile dead leaf pieces at $0^{\circ}C$. Saprophytic ability of T. incarnate on sterile leaf sheath occurred better at $0^{\circ}C$ than at $10^{\circ}C$. Saprophytic microflora consisting of Cladosporium sp., Fusarium sp., Mucor sp., Pythium sp., and unidentified fungi were the competitors for the sterilized and unsterilized substrate, but their colonization was not find on live leaf sheath buried in the soil at $0^{\circ}C$. In the effect of fungicides to Typhula snow mold disease of creeping bentgrass, mixture of polyoxin and thiram was the most effective, followed by iprodione, mixture of iprodione and oxine copper, thiophanate-methyl, myclobutanil, and tolclofos-methyl.

Germinability and Physiological Properties of Maize Seeds Affected by Matriconditioning (Matriconditioning에 의한 옥수수 종자 활력증진과 생리적 특성변화)

  • To observe the effects of seed matriconditioning on the emergence, plumule growth, and seed physiological properties of normal and aged seeds of three corn(Zea mays L.) genotypes (dent, sugary, and shrunken-2), normal and artificially aged seeds were matriconditioned at 50, 75, 100, and 125% moisture contents of vermiculite at $25^{\circ}C$ for 24 hours and the germinability was examined at $25^{\circ}C$ and in cold test (7 days at 1$0^{\circ}C$followed by $25^{\circ}C$). Emergence rate, emergence speed, and plumule growth were smaller in the order of dent>sugary>shrunken-2. Matriconditioning did not affect the emergence rate of normal dent and sugary seeds, while it enhanced emergence speed and plumule growth. In contrast, the emergence rate, emergence speed, and plumule growth of normal shrunken-2 and aged seeds of all genotypes were enhanced as the moisture content of vermiculite increased up to 125% both at $25^{\circ}C$ and in cold test. Matriconditioning promoted $\alpha$-amylase activity and DNA and soluble protein contents, while it reduced the leakage of total sugars and electrolytes from the seeds when soaked in water.

Amended Soil with Biopolymer Positively Affects the Growth of Camelina sativa L. Under Drought Stress (가뭄 조건 하에서 바이오폴리머 혼합 토양이 Camelina sativa L.의 생장에 미치는 긍정적 영향)

  • The biopolymer (BP) used in this study is mainly composed of xanthan gum and ${\beta}$-glucan derived from microorganism and has been introduced as a novel material for soil stabilization. However, the broad applicability of BP has been suggested in the field of geotechnical engineering while little information is available about the effects of BP on the vegetation. The goal of this study is to find the BP effects on the growth of Camelina sativa L. (Camelina) under drought condition. For more thorough evaluation of BP effects on the plant growth, we examined not only morphological but also physiological traits and gene expression patterns. After 25 days of drought treatment from germination in the soil amended with 0, 0.25, 0.5, and 1% BP, we observed that the BP concentration was strongly correlated the growth of Camelina. When plants were grown under drought stress, Camelina in 0.5% BP mixture showed better physiological parameters of the leaf stomatal conductance, electrolyte leakage and relative water content compared to those in control soil without BP. Plant recovery rate after re-watering was higher and the development of lateral root was lower in BP amended soil. RNA expression of Camelina leaf treated with/without drought for 7 and 10 days showed that aquaporin genes transporting solutes at bio-membrane, CsPIP1;4, 2;1, 2;6 and TIP1;2, 2;1, were induced more in the plants with BP amendment and drought treatment. These results suggest that the soil amended with BP has a positive effect on the transport of nutrients and waters into Camelina by improving water retention in soil under drought condition.

Development of salt-tolerant transgenic chrysanthemum (Dendranthema grandiflorum) lines and bio-assay with a change of cell specificity (내염성 국화 형질전환 계통 육성 및 저항성 검정과 세포특성 변화)

  • Recently the increasing of vinyl and green houses and development of reclaimed land including Saemangeum induced the need for breeding salt-tolerant crops which can survive and grow in high salinity soil. So we try to develop salt-tolerant transgenic chrysanthemum (Dendranthema grandiflorum.) lines by using anti-porter gene TANHX and HVNHX. Through marker selection and plant regeneration step, we could get 284 putative transgenic chrysanthemum lines. On selected putative transgenic plants, 40 candidates were used for genetic analysis and 30 lines could be made up of target size band on PCR, so about 75% of marker selected lines were decided as real transgenic lines. Selected 284 transgenic lines were also used for salt-tolerance test as a range of NaCl 0.2 ~ 1.2% (300 mM). As a result of salt-tolerance test, 15 selected transgenic lines could live and grow on the continuous supply of 0.8% (200 mM) NaCl solution and another 7 lines were could survive under 1.2% (300 mM) NaCl solution. This salt-tolerant transgenic lines under salt stress also lead a cell alternation especially a guard cell. A stressed guard cell be swelled and grow larger in proportion to NaCl concentration. TTC test for cell viability on transgenic chrysanthemum lines pointed out that more strong salt-tolerant lines can be live more than another under same salt stress. The numerical value of strong salt-tolerant 7 transgenic lines were 0.206 ~ 0.331 under 1.2% NaCl stress, and then it's value is more larger than middle salinity lines' 0.114 ~ 0.193 and non-transgenic's 0.046. And the proline contents as indicated stress compound also pointed out that HVNHX introduced salt-tolerant transgenic lines were less stressed than other under same salt stress. The contents of strong salt-tolerant transgenic lines were 2.255 ~ 2.638 mg/kg and it is much higher than that of middle salinity lines' 1.496 ~ 2.125.

Effect of Medium, Growth Regulators and Activated Charcoal on Plant Regeneration from in vitro Culture of Dioscorea japonica TUNBERG (산약(山藥)의 기내배양(器內培養)에 미치는 배지(培地) 및 생장조절물질(生長調節物質)과 활성탄(活性炭)의 영향(影響))

  • This study was carried out to investigate the optimal propagation condition through stem node tissue culture of yam supplemented with concentration levels of growth regulators and activated charcoal in 1 /8MS white media. For shoot induction and plant regeneration from yam stem node tissue wre more effectivein combination of auxin, kinetin and activated charcoal than combination of auxin and kinetin. Shoot induction and plant regeneration were more effective in 1/8 medium suplemented with 1AA $2mg\;/\;\ell,\;kinetin\;2mg\;/\;\ell$ and activated charcoal $1g\;/\;\ell$ than in white medium, but in case of white medium, Shoot induction and plant regeneration wre effective in culture medium supplemented with NAAa $4mg\;/\;\ell,\;kinetin\;2mg\;/\;\ell$ and activated charcoal $1g\;/\;\ell$. Growth status in optimal propagation condition from yam stem node tissue brought the results of multiple plant, elongated plant height and increase in number of leaves and roots, but tissue culture effect by kinds of media in number of multiple plant were more effective in 1 /8MS medium than in white medium.

Plant regeneration and soil acclimatization through photoautotrophic culture from leaf explant of a rare species in Sedum tosaense Makino (희귀수종인 주걱비름(Sedum tosaense Makino)의 잎절편으로부터 기내 식물체 재분화 및 광독립배양을 통한 토양순화)

  • The aim of this study was to establish plant regeneration from leaf explants of Sedum tosaense Makino, which is globally rare and endangered species. The leaf explants of S. tosaense were cultured on the MS medium supplemented with different concentration of BA and NAA for callus induction. Callus induction was showed the highest (100%) on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA. The highest number of shoots were regenerated when callus were cultured on MS medium containing $2.0mg{\cdot}L^{-1}$ BA and $1.0mg{\cdot}L^{-1}$ NAA for 5 weeks. The axillary bud were cultured on the MS media supplemented with combination of BA and NAA for in vitro propagation. The highest number of adventitious shoot (7.9 per explants) formed at $1.0mg{\cdot}L^{-1}$ NAA and $2.0mg{\cdot}L^{-1}$ BA. For rooting, MS medium supplemented with or without $2.0g{\cdot}L^{-1}$ activated charcoal was tested. The optimal results were observed using MS medium supplemented with $2.0g{\cdot}L^{-1}$ activated charcoal, on which 85.7 (No. of root), 4.6 cm (length of root). 1,200 ppm $CO_2$ and 350 ppm $CO_2$ were supplied for make certain the effects of $CO_2$ on pre-acclimatization by photoautotrophic culture. 1,200 ppm $CO_2$ treatment was established higher than 350 ppm $CO_2$ treatment. Soil acclimatization of in vitro plantlets was the best in mixture soil consisted of peat moss and perlite with 100% survival rate and they showed the maximum growth.

Growth and Flowering of Campanula Species as Affected by Duration, Temperature, and Light Condition during Chilling Treatment (저온처리 기간, 온도 및 광 조건이 자생초롱꽃의 생육과 개화에 미치는 영향)

  • The experiment investigated effect of duration, temperature, and light condition during chilling treatment on growth and flowering of four Campanula species in a factorial experiment. Two parent species, Campanula punctata Lam. var. rubriflora Mak. and C. Punctata Lam., and their two $F_1$ hybrids, C. punctata Lam. ${\times}$ C. punctata Lam. var. rubriflora Mak. ('Jiknyeo') and C. punctata Lam. var. rubriflora Mak. ${\times}$ C. punctata Lam. ('Gyeonu'), were used. Plants were cultured in vitro for five weeks at $25^{\circ}C$ under about $75{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD before being chilled at 4 or $25^{\circ}C$ for 3, 6, or 9 weeks under a darkened or lighted (about $10{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD) condition. After chilling treatment, plants were transplanted to 10 cm pots filled with a commercial growing medium and were transferred to environment-controlled growth chambers and subsequently to a greenhouse to observe their reproductive growth. Growth of all species and flowering of a $F_1$ hybrid 'Jiknyeo' were affected by duration, temperature, and light condition during chilling treatment. The greatest growth and survival percentage were observed in C. punctata Lam. var. rubriflora Mak. The survival percentage was greater when plants were chilled in a lighted than darkened condition, whereas it decreased when plants were chilled more than six weeks in vitro. Among the four species tested, flowering was observed only in a $F_1$ hybrid 'Jiknyeo' with 62.5% flowering plants when it was chilled at $25^{\circ}C$ for three weeks under a lighted condition. Percent flowering plant was affected by duration, temperature, and light condition during chilling treatment. Three-week chilling at $4^{\circ}C$ under a darkened condition significantly reduced days to flowering. These results suggest that the low temperature requirement for flowering is not qualitative but quantitative in Campanula species. Further experiment with more number of plants is necessary to ascertain this conclusion.

Productivity Evaluation of Rosemary Shoots using Artificial Light Sources in Multi-layer Cultivation (다단재배에서 인공광원을 이용한 로즈마리 어린순의 생산성 평가)

  • This study was aimed to investigate the effects of layer-by-floor environmental conditions and lower shelf supplemental lighting on the productivity of fresh shoots when growing rosemary in multi-layer cultivation. The 10-cm cuttings from stock plants of common rosemary (Rosemarinus officinalis) were planted in a 128-hole tray, rooted, and then transplanted into pots of 750, 1,300, and 2,000 mL. Afterwards, they were placed on multi-layer shelves (width × length × height: 149 × 60 × 57 cm, 3-layer) in a two-linked greenhouse and cultivated using the sub-irrigation. The productivity of young shoots by layer of the multi-layer shelf was the highest on the third floor (top floor), but productivity decreased sharply after September due to stem lignification caused by excessive light during the summer. Conversely, the lower two layers exhibited faster growth rate of young shoots until the late cultivation period, but the quality decreased due to stem softening and leaf epinasty. To address the excessive light problem on the third floor during the summer, shading was implemented at 30% opacity in July and August, resulting in a 210% increase in rosemary young shoots count and a 162% increase in fresh weight per unit area compared to the unshaded control. To improve the lighting deficiency on the lower layer, supplemental lighting with LED at 30 W increased rosemary young shoot harvest by 168% from June to September compared to no supplemental lighting, but it decreased productivity after September. Therefore, when growing rosemary in multi-layer, it is judged that intensive production of young shoots is possible if the third floor (top layer) is shaded with 30% of light from July to August to prevent stem lignification, and the lower layer is temporarily supplemented with LED 30 W from June to September to increase young shoot growth.

Characteristics of Vegetation Structure of Burned Area in Mt. Geombong, Samcheok-si, Kangwon-do (강원도 삼척 검봉산 일대 산불 피해복원지 식생 구조 특성)

  • In 2000, a total of 23,794ha of forest was lost due to the East Coast forest fire, and about 70% of the damaged area was concentrated in Samcheok. In 2001, artificial restoration and natural restoration were implemented in the damaged area. This study was conducted to understand the current vegetation structure 21 years after the restoration of forest fire damage in the Samcheok, Gumbong Mountain area. As a result of classifying the vegetation community, it was divided into three communities: Quercus variabilis-Pinus densiflora community, Pinus densiflora-Quercus mongolica community, and Pinus thunbergii community. Quercus variabilis, Pinus densiflora, and Pinus thunbergii planted in the artificial restoration site were found to continue to grow as dominant species in the local vegetation after restoration. As for the species diversity index of the community, the Quercus variabilis-Pinus densiflora community dominated by deciduous broad-leaf trees showed the highest, and the coniferous forest Pinus thunbergii community showed the lowest. Vegetation in areas affected by forest fires is greatly affected by reforestation tree species, and 21 years later, it has shown a tendency to recover to the forest type before forest fire. In order to establish DataBase for effective restoration and to prepare monitoring data, it is necessary to construct data through continuous vegetation survey on the areas affected by forest fires.
