Bacillus cereus에서 유래한 Lactate Dehydrogenase 동질효소 유전자의 대장균 내 발현 및 효소특성 규명 (Enzymatic Characterization of Bacillus cereus Lactate Dehydrogenase Isozymes Expressed in Escherichia coli)

  • Lactate dehydrogenases (LDHs)는 세포 내의 생화학적 대사경로에서 중요한 역할을 담당하는 효소로서 오랜 동안 많은 관심을 받았다. 본 연구에서는 다양한 미생물 genome database의 탐색을 통해 Bacillus cereus ATCC 14579 genome으로부터 LDH로 추정되는 3종의 유전자를 발견하고, 대장균에서 클로닝 및 대량 발현하였다. 모든 BcLDH 동질효소들의 상동부위 아미노산 잔기 대부분이 기존의 $NAD^+$-의존형 LDH와 높은 상동성을 보였다. 한편 314개의 아미노산으로 이루어진 BcLDH1과 2는 86%의 서열 상동성을 보였으나, BcLDH3와는 49%의 상동성을 나타냈다. 흥미롭게도 BcLDH1만이 $NAD^+$ 조효소 존재 하에서 L-lactate와 pyruvate 간의 상호전환 활성을 나타냈으며, 그 외의 동질효소들은 거의 활성을 보이지 않았다. 결론적으로 BcLDH1은 전형적인 $NAD^+$-의존형이며, L-lactate에 특이적인 dehydrogenase 효소임을 확인하였다.

  • A method of optimum design of an axial rotary sensible heat exchanger for the heat recovery of exhaust gas from the air conditioning space was developed in consideration of economics of investment cost and profit according to the installation of heat exchangers. Leakage rate of exhaust gas was calculated and the correlation for the pressure drop due to leakage of exhaust gas was proposed. Heat transfer between the matrix and exhaust and intake gas was analysed to calculate the effectiveness of heat exchanger, which was used for the optimum design of rotary heat exchanger. The results show that optimum rotational speed increases as the length of rotor increases and there exists optimum NTU which maximizes the gain of total cost according to the installation of rotary heat exchanger.

  • To evaluate the building energy performance, many building simulation programs are used and its capabilities are developed. Despite of its increased capabilities the weather data used In the Building Energy performance evaluation, are still using the same limited set of data. This often forces users to find or calculate weather data such as illuminance, solar radiation, and ground temperature from other sources to calculate it. Also, proper selection of a right weather data set has been considered as one of important factors for a successful building energy simulation. In this paper, we describe TMY2 data, a generalized weather data format developed for use, and applied to Seoul region and examine the differences comparing to existing weather data. A set of 23 years raw weather data base has been developed to provide the weather data file for building energy analysis in Seoul.

  • Method of optimum design of a compact sensible plate-fin heat exchanger for the heat recovery of exhaust gas from the air conditioning space was developed in consideration of the econamics of investment cost and profit according to the installation of heat exchanges. In the counterflow heat exchanger, the frontal area was fixed and the length of heat exchanger was optimized in order to maximize the net gain according to the setting of the heat exchanger. In the cross flow heat exchanger, the size of the exchanger was also optimized to maximize the net gain.

공기식 집열창 시스템이 설치된 건물의 동적부하 해석 (Time Dependent Thermal Load Analysis of the Building with an Airflow Window System)

  • It has been known that the application of an airflow window system reduces the energy consumption compared with conventional double pane window in a building. But how to analyze thermal load in a building with an airflow window system has not been well known. so two kinds of method (Mode 1 and Mode 2) to analyze time dependent thermal load of the building with an airflow window system are presented in this study. The results of load analysis about the model building(total area : $4521m^2$, 3 floors) by Mode 2 show that the maximum cooling and heating load in a building with an airflow window system are decreased about 12-17% and about 19.5% than with double pane glass window, and yearly energy consumption with an airflow window system is saved about about 20% than with double pane glass window.

뇌졸중후 우울증의 정도와 운동기능의 회복과의 상관관계에 대한 연구 (The correlation between post-stroke depression and the recovery rate of motor functions)

  • Objectives : The degree of post-stroke depression was observed and then correlated to the recovery rate of the motor functions of the above treated stroke patients. Methods : The BDI SCALE(Beck Depression Inventory Scale) and motor grades of 50 diagnosed stroke patients who were hospitalized in Dong-Seo Oriental Hospital between the period of May 2002 to September 2002 were measured. After a 1 month recovery period the BDI SCALE and motor grade of the above mentioned patients were again measured and a correlation was observed. Results : A lower BDI SCALE was observed in patients with a higher motor grade recovery rate. Conclusion : The treatment of post-stroke depression is imperative for positive effects on the motor functions of stroke patients.

  • This study has been designed and performed to identify the effect on insomnia patients according to th injection of herbal medicine induced from Sanjoincho.The result of the injection shows as follows.1. Total duration of sleeping time showed 1.1500${\pm}$1.5433 hours of increase. (p〈0.01, respectively)2. Delaying time before the onset of sleep showed 0.8158${\pm}$1.3251 hours of increase in sleep onset insomnia(p〈0.05, respectively)3. The wake times showed 1.375${\pm}$1.4079 times of decrease in sleep maintenance isomnia. (p〈0.05, respectively)

  • Purpose: To investigate whether or not acupuncture of GB34 produces a significant response of the modulation of somatomotor areas by functional magnetic resonance imaging (fMRI) study. Materials and methods: The acupoint, GB34, located in the back of the knee, is known to be effective in recovering motor function after stroke. Using 3T MRI scanner, functional MR imaging of the whole brain was peformed in 12 normal healthy subjects during two stimulation paradigms; acupuncture manipulation on GB34 and sham points. This study investigates the activation of the motor cortex elicited by a soft and an intensified stimulation of GB 34.Three different paradigms were carried out to detect any possible modulation of the Blood Oxygenation Level Dependent (BOLD) response in the somatomortor area to motor stimulation through acupuncture.

  • This is an experimental study to identify the performance of air-to-air rotary heat exchanger with polyurethane foam matrix. The experimental apparatus including heating AHU(Air Handling Unit), cooling AHU, sensor chamber, and heat exchanger testing unit was designed and manufactured in this study. The performance of heat exchanger with porous polyurethane foam matrix was tested with variations of the density and the thickness of matrix, regulating the wind velocity and the rotational speed of matrix. The actual heat recovery effectiveness, air leakage rate, and pressure drop of heat exchanger were measured and analyzed.
