Anatomical Characteristics of Three Korean Bamboo Species (국내산 대나무 3종의 해부학적 특성)

  • Bamboo is one of the major biomass resources in the world. To obtain valuable information for effective use of bamboo resources in Korea, the anatomical characteristics of the commercial Korean bamboo species (Phyllostachys pubescens, Phyllostachys nigra, and Phyllostachys bambusoides) were analyzed. The structures in bamboo culm were observed by optical and scanning electron microscopy. Also the crystalline properties as relative crystallinity and crystallite width were measured by an X-ray diffraction method. The three Korean bamboo species had the vascular bundle type I with tylosoid in intercellular space. In the outer part of culm, vascular bundles showed denser spacing than inner part. The fiber length in outer part samples of the three bamboo species showed longer than inner part samples. Furthermore, the fiber length showed a significant difference between inner part and outer part in three bamboo species, showing the longest fiber length in Phyllostachys bambusoides. Phyllostachys pubescens showed the greatest diameter in vessel and parenchyma on cross section. Parenchyma cells in Phyllostachys pubescens and Phyllostachys bambusoides showed similar length and width in both radial and tangential sections. The relative crystallinity and crystallite width in outer part samples of the three bamboo species showed higher values than those in inner part samples, with the greatest values from Phyllostachys bambusoides.

Growth and Biomass Production of Fast Growing Tree Species Treated with Slurry Composting and Biofiltration Liquid Fertilizer (SCB액비가 속성수의 생장 및 biomass 생산에 미치는 영향)

  • Fifteen clones of poplars, 2 clones of willows, and yellow poplar were used to evaluate the effects of 5 treatments such as SCBLF (slurry composting and biofiltration liquid fertilizer), general slurry liquid fertilizer, chemical fertilizer, groundwater, and control (no treatment) on vitality, growth performance, and biomass production. Five cuttings for each tree species were planted in 3 replications. After planting cuttings, a coppice was induced by cutting off stems at 10cm above the ground. Data were collected for first growing season and trees were harvested at the end of October. Maximum mortality rate i.e. 96% was recorded in the cuttings treated with groundwater and minimum 92% with control (no treatment). In all tree species, sprouting of stump was not differ significantly among the treatments. Total nitrogen concentrations of leaves and stump sprouts were higher in the treatment of SCBLF than the control, 26.6% and 22.9%, respectively. Biomass production was highest in the stumps treated with chemical fertilizer, $1.98Mg\;ha^{-1}\;year^{-1}$, and lowest in control ($1.34Mg\;ha^{-1}\;year^{-1}$).

Characteristics of Volatile Fatty Acids Release During the Hydrolysis of Rice Straw (볏짚의 가수분해과정중 유기산 생성 특성)

  • Objective of this study was to investigate the effects of pre-treatment of rice straw by sizes(3cm, milled), temperatures($35^{\circ}C\;and\;55^{\circ}C$), with/without NaOH treatment, and RPM on the characteristics of volatile fatty acids production. The concentration of total volatile fatty acids (TVFAs) was increased with the hydrolyzed time. Concentration of VFAs in milled rice straw was higher than that in 3 cm cut. With the alkali treatment, the concentration of TVFAs were sharply increased, which showed 3 times higher than non-treatment. Concentration of VFAs was high at 150 rpm at $35^{\circ}C$, and at 80rpm and 200 rpm at $55^{\circ}C$. Among them acetic acid was dominant, which showed the similar increase with TVFAs. It was also observed that in the case of fibrous material, the contents of cellulose and hemi-cellulose were reduced a little, but no change in lignin.

Physical and Chemical characteristics of Cokes Using Ash-Free Coal as binder (무회분 석탄(AFC)을 바인더로 이용한 코크스의 물리적 및 화학적 특성)

  • Coke strength was increased by adding ash-free coal (AFC) binder. In this study, the effect of the AFC binder on the physical and chemical properties of coke was experimentally investigated to understand the molecular mechanism for the improved coke strength. For reduced $CO_2$ emission in steelmaking industry, torrefied biomass fuel mixed with coal binder is also considered. The interface between the base coal and AFC was thus examined using Scanning Electron Microscope (SEM). The coke strength was commonly measured by performing the indirect tensile test and Nuclear Magnetic Resonance (NMR) spectroscopy in $^1H$ and $^{13}C$ modes. For comprehensive mechanism study of the enhanced coke strength thus obtained, ordinary coal for thermal power plant use was carbonized with AFC for subsequent SEM examination. The NMR spectroscopy results of coke samples positively revealed that the tensile strength was proportional to the average number of aromatic rings.

Response of Nutrient Solution and Photosynthetic Photon Flux Density for Growth and Accumulation of Antioxidant in Agastache rugosa under Hydroponic Culture Systems (식물공장에서 양액의 종류 및 PPFD가 배초향의 생장 및 항산화 물질에 미치는 영향)

  • Agastache rugosa, is a perennial medicinal plant commonly used in Chinese herbalism, and may have anti-atherogenic and antibacterial properties. Here in this study, we investigated the growth and variations in antioxidant contents of A. rugosa in response to nutrient solution and photosynthetic photon flux density (PPFD) with artificial lighting for a hydroponics culture. Fluorescent light at 150, and $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD with a 16/8 (light/dark) photoperiod, combined with four different nutrient solutions [developed by Horticulture experiment station in Japan (HES), University of Seoul (UOS), Europe vegetable research center (EVR), Otsuka-house 1A (OTS)], were used in a hydroponics culture system for 6 weeks. The shoot and root dry weights of A. rugosa grown with the OTS were significantly higher than those of other nutrient solutions. The amount of tilianin was the highest grown with the OTS, followed by EVR, HES, and UOS. Total acacetin content was the highest in A. rugosa grown under EVR which was statistically similar with OTS. The A. rugosa grown under $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD produced higher fresh weight and both acacetin and tilianin contents than that grown under $150{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD. The present results suggested that OTS along with $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ PPFD could be an optimum growing condition for better growth and higher accumulation of tilianin and acacetin contents in A. rugosa with hydroponic culture systems in a plant factory.

Evaluation for Wood Pellets from Pinus densiflora Wood Damaged by Forest Fire (산불피해 소나무재의 목질펠릿으로의 이용가능성 평가)

  • The properties of the wood pellets from damaged wood by forest fire and sound woods were investigated. Oven-dry densities of pellets made from sound wood and the damaged wood of Pinus densiflora were 0.93~0.94 and 0.86~0.88, respectively. Compressive strength of the pellets from the woods was ranged from 145 to $168kgf/cm^2$, and there was little difference between the sound wood and the damaged wood of P. densiflora. Ash content for the sound wood and the damaged wood of Pinus densiflora were 0.32~0.37% and 0.25~0.35%, respectively, and calorific values of each pellet were ranged from 18 to 19 MJ/kg. The result of elemental analysis in carbon. hydrogen and oxygen showed 45.8~48.8%, 6.2~6.5% and 46.5~48.0%, respectively. Consequently, there seem no significant difference in the properties between damaged and sound wood pellets.

Biological Co2 Fixation to Antioxidant Carotenoids by Photosynthesis Using the Green Microalga Haematococcus pluvialis (광합성 녹색 미세조류 Haematococcus pluvialis를 이용한 이산화탄소 고정화 및 항산화성 카로티노이드 생산)

  • As one of the $CO_2$ reduction strategies, a biological method was proposed to convert $CO_2$ to useful biomass with antioxidant carotenoids by photosynthetic microorganisms. One of the photoautotrophs, Haematococcus pluvialis is a freshwater green microalga and accumulates the secondary carotenoid astaxanthin during induction of green vegetative cells to red cyst cells. In this study, $CO_2$ fixation and astaxanthin production using H. pluvialis was conducted by photoautotrophic culture in the $CO_2$ supplemented photo-incubator. Maximum growth rate of H. pluvialis was obtained at a 5% $CO_2$ environment on basic N and P conditions of NIES-C medium. The photoautotrophic induction consisted of 5% $CO_2$ supply and high light illumination promoted astaxanthin synthesis in H. pluvialis, yielding an astaxanthin productivity of $9.6mg/L{\cdot}day$ and a $CO_2$ conversion rate of $27.8mg/L{\cdot}day$ to astaxanthin. From the results the sequential photoautotrophic culture and induction process using H. pluvialis is expecting an alternative $CO_2$ reduction technology with a function of valuable biosubstance production.

Production of Fermentable Sugar from Lipid Extracted Algae using Hot Water Pretreatment (열수전처리를 이용한 탈지미세조류로부터 발효당 생산 공정 개발)

  • The microalgae have cellulose as a main structural component of their cell wall and the lignin content in microalgae is much lower than other lignocellulosic biomass. Therefore, fermentable sugar production from microalgae (Tetraselmis KCTC 12236BP) can be carried out under pretreatment without high temperature and high pressure. It was investigated that the effect of hot-water pretreatment using sulfuric acid for lipid extracted algae which is expected to be a next generation biomass. The effects of three major variables including extraction temperature, acid concentration and time on the enzymatic hydrolysis were investigated. Among the tested variables, temperature and acid concentration showed significant effects and optimum pretreatment conditions for the economic operation criteria were obtained as follows: reaction temperature of $120^{\circ}C$, sulfuric acid concentration of 2 mol and pretreatment time of 40 min. Under the optimum conditions of acidic hot water pretreatment, experimentally obtained hydrolysis yield were 95.9% which showed about 2.1 fold higher compared with enzymatic hydrolysis process. Therefore, acid pretreatment under mild condition was proven to be an effective method for fermentable sugar production from lipid extracted microalgae.

Effects of Stand Age Classes on Biomass Expansion Factors and Stem Densities in Chamaecyparis obtusa Plantations (편백 조림지에서 영급이 바이오매스 확장계수와 줄기밀도에 미치는 영향)

  • Biomass expansion factors and stem density values were commonly used in converting stand volumes into total carbon stocks for the purpose of national inventories of greenhouse gas emissions and carbon sequestration. The objective of this study was to examine the influence of stand age classes on aboveground and total biomass expansion factors, and stem density values in Chamaecyparis obtusa species. A total of 25 representative sample trees based on the three different stand age classes were destructively sampled to measure green weights and dry weights of the major four(root, stem, branch and foliage) portions of C. obtusa species grown in Jangseung-gun of southern Korea. According to the results of this study, as stand age classes increase, total biomass expansion factors tended to be decreased with the ranges from 3.64 to 1.44, while the stem density values tended to be slightly increased with the ranges from $0.35(g/cm^3)$ to $0.44(g/cm^3)$. There were statistically significant differences in biomass expansion factors and stem density values between stand age classes, but became nearly constant after 30 years old for C. obtusa species. This information could be very useful to improve a national-scaled inventory of greenhouse gas emissions and carbon sequestration for the C. obtusa species by applying different biomass expansion factors and stem density values.

Change of Solubilization Characteristics of Rice Straw by Different Pre-treatments (전처리 방법에 따른 볏짚의 가용화 특성 변화)

  • In order to find a feasibility of applying an agricultural biomass to the anaerobic digestion system, the effects of pre-treatment of rice straw was investigated by different sizes,temperatures, ultrasonic frequencies, and with/without NaOH treatment on the change of soluble organic matter, which is generated most dominantly in Korea. It was observed that SCOD(soluble chemical oxygen demand) as the index for the decomposition of rice straw increased with the smaller particle size, higher reaction temperature and alkali treatment. With treatment of 5% NaOH, it was shown that the highest concentrations of SCOD were observed at 9,000 mg $L^{-1}$ and 6,000mg $L^{-1}$ at $35^{\circ}C$ and $55^{\circ}C$, respectively. Agitating with ultrasonic irradiation could be enhanced SCOD of rice straw less than 3 cm with 40 kHz of ultrasound. Optimal RPM in this study was at 150 rpm regardless of reaction temperatures.
