Biological Co2 Fixation to Antioxidant Carotenoids by Photosynthesis Using the Green Microalga Haematococcus pluvialis (광합성 녹색 미세조류 Haematococcus pluvialis를 이용한 이산화탄소 고정화 및 항산화성 카로티노이드 생산)

  • As one of the $CO_2$ reduction strategies, a biological method was proposed to convert $CO_2$ to useful biomass with antioxidant carotenoids by photosynthetic microorganisms. One of the photoautotrophs, Haematococcus pluvialis is a freshwater green microalga and accumulates the secondary carotenoid astaxanthin during induction of green vegetative cells to red cyst cells. In this study, $CO_2$ fixation and astaxanthin production using H. pluvialis was conducted by photoautotrophic culture in the $CO_2$ supplemented photo-incubator. Maximum growth rate of H. pluvialis was obtained at a 5% $CO_2$ environment on basic N and P conditions of NIES-C medium. The photoautotrophic induction consisted of 5% $CO_2$ supply and high light illumination promoted astaxanthin synthesis in H. pluvialis, yielding an astaxanthin productivity of $9.6mg/L{\cdot}day$ and a $CO_2$ conversion rate of $27.8mg/L{\cdot}day$ to astaxanthin. From the results the sequential photoautotrophic culture and induction process using H. pluvialis is expecting an alternative $CO_2$ reduction technology with a function of valuable biosubstance production.

Development of Biosensor for Simultaneous Determination of Glucose, Lactic Acid and Ethanol (포도당, 젖산 및 에탄올의 동시 측정용 바이오센서의 개발)

  • The purpose of this study is to develop biosensor for determination of glucose, lactate, and ethanol in foods and food-stuffs simultaneously. The multiple cathode system was prepared with an oxygen electrode having one anode and hexagonal cathode. Glucose oxidase, mutarotase, lactate oxidase, alcohol oxidase and catalase were used for immobilization to determine glucose, lactate, and ethanol. These components including ethanol were simultaneously determined by the immobilized enzymes in the multiple cathode system. The determination of the components by enzyme sensor was based on the maximum slope of oxygen consumption from enzyme reaction of each sensor part. The response time for analysis was 1 min. The optimum condition for glucose, lactate and ethanol sensor was found to be 0.1 M potassium phosphate buffer, pH 7.0 at $40^{\circ}C$. Interferences of various sugars and organic acids were investigated. Less than 10% of error was found in determination of the components except organic acids. This difference was compensated by the modified equation. This system was confirmed by conventional methods. It was concluded that the multiple cathode system of this study is for an effective method to determine sugar, organic acid, ethanol simultaneously in foods.

  • At present, global warming and depletion of fossil fuels have been one of the big issues which should be solved for sustainable development in the future. CCS (carbon capture and sequestration) technology as the post $CO_2$ reduction technology has been considered as a promising solution for global warming due to increased carbon emission. However, the environmental and ecological effects of CCS have drawn concerns. There are needs for noble post reduction technology. More recently, CCU (carbon capture and utilization) Technology, which emphasizes transforming carbon dioxide into value-added chemicals rather than storing it, has been attracted attentions in terms of preventing global warming and recycling the renewable carbon source. In this paper, various technologies developed for carbon dioxide conversion both in gas and liquid phase have been reviewed. For the thermochemical catalysis in gas phase, the development of the catalytic system which can be performed at mild condition and the separation and purification technology with low energy supply is required. For the photochemical conversion in liquid phase, efficient photosensitizers and photocatalysts should be developed, and the photoelectrochemical systems which can utilize solar and electric energy simultaneously are also in development for more efficient carbon dioxide conversion. The energy needed in CCU must be renewable or unutilized one. CCU will be a key connection technology between renewable energy and bio industry development.

Distribution of Magnetic Field Depending on the Current in the μ-turn Coil to Capture Red Blood Cells (적혈구 포획용 미크론 크기 코일에 흐르는 전류의 크기에 따른 자기장 분포 특성)

  • The ${\mu}$-turn coil having a width of ${\mu}m$ on the GMR-SV (giant magnetoresistance-spin valve) device based on the antiferromagnetic IrMn layer was fabricated by using the optical lithography process. In the case of GMR-SV film and GMR-SV device, the magnetoresistance ratios and the magnetic sensitivities are 4.4%, 2.0%/Oe and 1.6 %, 0.1%/Oe, respectively. In the y-z plane the distribution of magnetic field of GMR-SV device and $10{\mu}$-turns coil which put under the several magnetic bead(MB)s with a diameter of $1{\mu}m$ attached to RBC (red blood cell) was analyzed by the computer simulation using the finite element method. When the AC currents of 20 kHz from 0.1 mA to 10.0 mA flow to the 10 turns ${\mu}$-coil, the magnetic field at the position of $z=0{\mu}m$ at the center of coil was calculated from $30.1{\mu}T$ to $3060{\mu}T$ in proportion to the current. The magnetic field at the position of $z=10{\mu}m$ was decreased to one-sixth of that of $z=0{\mu}m$. It was confirmed that the $10{\mu}$-turn coil having enough magnitude of magnetic field for the capture of RBC is possible to use as a biosensor for the detection of magnetic beads attached to RBC.

The Research on the Phytosociological Characteristics of Abies nephrolepis Maxim. Community in Mt. Seorak, Korea (설악산 분비나무군락의 식물사회학적 특성)

  • This study carried out the plant sociological survey of Abies nephrolepis forest in Mt. Seorak, which is in danger of deterioration due to the accelerated climate change. We examined seventy quadrats obtained from the survey and used the TWINSPAN technique to classify communities. We then performed the DCA method for the sequence analysis and analyzed the characteristics of each community. A. nephrolepis forest of Mt. Seorak is composed of four communities (A. nephrolepis-Lonicera caerulea var. edulis community, A. nephrolepis-Acer komarovii community, A. nephrolepis-Ac. pseudosieboldianum community, and A. nephrolepis-Betula costata community). Each community showed a different distribution according to location because different microenvironments are formed depending on location such as altitude and slope direction, resulting in different species composition. Each community showed differences in environmental characteristics such as altitude, rock ratio, soil characteristics, and litter layer thickness. As a result, there were significant differences between communities in the number of species and individuals, coverage, tree size, and species diversity, as well as differences in species composition. The A. nephrolepis-L. caerulea var. edulis and A. nephrolepis-Ac. komarovii communities were located in high altitude with high rock ratios and had little development of tree layer. On the other hand, the A. nephrolepis-Ac. pseudosieboldianum and A. nephrolepis-B. costata communities were relatively in low altitude with high soil ratio and had the development of tree layer with high species diversity.

  • The aging and declining agricultural population in the modern society requires improvement of the agricultural environment and is one of the representative problems. And since most of the work systems always require a transport work, the ratio of labor consumed in the transport work is very high. Accordingly, many types of transport vehicles are being developed and sold, and in the early days, most of them are powered transport vehicles using fossil fuels. However, it is paying attention to next-generation eco-friendly energy such as hydrogen, fuel cells, solar power, and bio due to the strengthening of international environmental regulations such as global warming and the Convention on Climate Change and the depletion of fossil fuels. Therefore, in this study, the ultimate goal is to develop an eco-friendly, easy-to-operate, safe agricultural electric vehicle that replaces fossil fuels. It was designed with a focus on controlling a wide range of vehicle speeds and securing stability of electric agricultural vehicles. Considering the performance and design, it is composed of a frame, a driving part, a steering part, and a controller system, and we are going to review and manufacture each part. It is believed that the manufactured electric vehicle for agriculture can be easily and conveniently operated in an agricultural society where young manpower is scarce, and can be helpful to the agricultural society through high efficiency.

  • Here, we introduce a laser-induced graphene synthesis technology and its applications for the electric/electronic device manufacturing process. Recently, the micro/nanopatterning technique of graphene has received great attention for the utilization of these new graphene structures, which shows progress developments at present with a variety of uses in electronic devices. Some examples of practical applications suggested a great potential for the tunable graphene synthetic manners through the control of the laser set-up, such as a selection of the wavelength, power adjustment, and optical techniques. This emerging technology has expandability to electric/electronic devices combined together with existed micro-packaging technology and can be integrated with the new processing steps to be applied for the operation in the fields of biosensors, supercapacitors, electrochemical sensors, etc. We believe that the laser-induced graphene technology introduced in this paper can be easily applied to portable small electronic devices and wearable electronics in the near future.

Design and Operation of Self-Powered Arduino System for Solar Energy Harvesting (태양에너지 하베스팅을 위한 자가발전 아두이노 시스템의 설계 및 동작)

  • In this paper, we design a self-powered Arduino system for solar energy harvesting and explain its operation. To perform the operation, the Arduino system senses the amount of solar energy that changes every moment and adjusts the ratio of the active mode and sleep mode operation time according to a given solar light intensity. If the intensity of sunlight is strong enough, the Arduino system can be continuously driven in active mode and receive sufficient power from sunlight. If not, the system can run in sleep mode to minimize power consumption. As a result, it can be seen that energy consumption can be minimized by reducing power consumption by up to 81.7% when using sleep mode compared to continuously driving active mode. Also, when the light intensity is at an intermediate level, the ratio between the active mode and the sleep mode is appropriately adjusted according to the light intensity to operate. The method of self-control of the operating time ratio of active mode and sleep mode, proposed in this paper, is thought to be helpful in energy-efficient operation of the self-powered systems for wearables and bio-health applications.

The Study on the Height Characteristics of Abies Nephrolepis Community in South Korea - In the Case of Seorak·Odae·Taebaek National Park - (우리나라 분비나무의 수고 특성 연구 - 설악·오대·태백산국립공원을 대상으로 -)

  • This study investigated whether population dynamic analysis based on the height characteristics of Abies nephrolepis was feasible. It was necessary because existing population dynamic analyses based on age and diameter at breast height (DBH) made it difficult to reflect the slow growth characteristics of Abies nephrolepis in harsh environments of high altitudes. The limitations of population dynamics analysis based on the age and DBH distribution of Abies nephrolepis in Seoraksan, Odaesan, and Taebaeksan National Parks, where Abies nephrolepis populations are representative, were verified, and the characteristics of height growth were investigated to comprehensively analyze whether a vertical structure based on height could reveal the population dynamics. The result of this study showed some limitations in understanding the population dynamics of Abies nephrolepis based on age distribution due to practical difficulties in sampling all trees and variations in age distribution within the same community depending on factors such as light conditions. Moreover, it was challenging to differentiate the distribution of DBH classes at fine levels, making it difficult to reflect the rapid growth characteristics of Abies nephrolepis when light conditions become suitable after prolonged stays in smaller DBH classes under shade conditions. However, a comprehensive analysis of the height characteristics of Abies nephrolepis revealed that the density corresponding to the population dynamic characteristics of Abies was high and adequately reflected the predominant tree death at similar height stages, as well as the U-shaped population dynamics at the lower stratum. Moreover, it was possible to identify a transition point in height values under shaded conditions, where the annual growth of Abies nephrolepis individuals in the lower stratum increases significantly, indicating that Abies nephrolepis individuals can escape from competition with other shrubs and undergo vigorous growth only at this height level. Therefore, this study confirmed that a vertical structure based on height can be utilized to understand the population dynamics of Abies nephrolepis in high altitudes, and it is expected that future studies on height characteristics can intuitively reveal the maintenance status of Abies nephrolepis populations in the field.

Establishing Optimal Conditions for LED-Based Speed Breeding System in Soybean [Glycine max (L.) Merr.] (LED 기반 콩[Glycine max (L.) Merr.] 세대단축 시스템 구축을 위한 조건 설정)

  • Plant breeding is a time-consuming process, mainly due to the limited annual generational advancement. A speed breeding system, using LED light sources, has been applied to accelerate generational progression in various crops. However, detailed protocols applicable to soybeans are still insufficient. In this study, we report the optimized protocols for a speed breeding system comprising 12 soybean varieties with various maturity ecotypes. We investigated the effects of two light qualities (RGB ratio), three levels of light intensity (PPFD), and two soil conditions on the flowering time and development of soybeans. Our results showed that an increase in the red wavelength of the light spectrum led to a delay in flowering time. Furthermore, as light intensity increased, flowering time, average internode length, and plant height decreased, while the number of nodes, branches, and pods increased. When compared to agronomic soil, horticultural soil resulted in an increase of more than 50% in the number of nodes, branches, and pods. Consequently, the optimal conditions were determined as follows: a 10-hour short-day photoperiod, an equal RGB ratio (1:1:1), light intensity exceeding 1,300 PPFD, and the use of horticultural soil. Under these conditions, the average flowering time was found to be 27.3±2.48 days, with an average seed yield of 7.9±2.67. Thus, the speed breeding systems reduced the flowering time by more than 40 days, compared to the average flowering time of Korean soybean resources (approximately 70 days). By using a controlled growth chamber that is unaffected by external environmental conditions, up to 6 generations can be achieved per year. The use of LED illumination and streamlined facilities further contributes to cost savings. This study highlights the substantial potential of integrating modern crop breeding techniques, such as digital breeding and genetic editing, with generational shortening systems to accelerate crop improvement.
