Porcine reproductive and respiratory syndrome (PRRS) is characterized by reproductive failures in sows and respiratory problems in piglets. The nucleocapsid(N) protein, encoded by the open reading frame 7 (ORF7) gene, is known to be the most abundant and antigenic protein in PRRS virus. Therefore, it was suggested that the N protein could be a suitable candidate for the detection of PRRS virus-specific antibodies and diagnosis of PRRS. In the present study, the ORF7 gene encoding the N protein was cloned and expressed as a fusion protein with the glutathione S-transferase (GST) in Escherichia coli. The resulting GST-N recombinant protein was used as an antigen for an indirect sandwich enzyme-linked immunosorbent assay (i-ELISA). Expressed GST-N recombinant protein was migrated at 41 kDa and reacted with ORF7-specific monoclonal antibody by Western blotting. In order to increase the specificity of the ELISA for the detection of PRRS virus-specific antibodes, an i-ELISA was developed using an anti-GST antibody as a capture antibody. The sensitivity and specificity of developed i-ELISA were 92% and 96%, respectively. Based on these results, it was suggested that the i-ELISA is a simple and rapid test for screening a large number of swine sera for the anti-PRRS virus antibodies.
Bokbunja (Rubus coreanus Miquel) is a wild berry belonging to Rosaceae genus of which application areas have been expanded due to many health effects. Bokbunja berry contains carbohydrates, protein, fats and dietary fibers as major components and various flavonoids such as tannins, volatile components including organic acids, alcohols and hydrocarbons as minor ones. Those active compounds attribute to the following health effects: increase in immune function and antioxidant activity, rise in hormone secretion, suppression of the growth of type B hepatitis virus, and control of weight gain. To utilize the above activities, many products have been developed by using Bokbunja berry extracts in the area of foods, liquors, and cosmetics. Recently, pure compounds have been isolated from the extracts for the development of medicines. In this mini review, the state-of-the-art of Bokbunja researches and applications including papers and patents is summarized.
Innate immunity is the first line of host defense consisting of various molecules against infectious challenges. Mannose-binding lectin (MBL) belongs to the collectin protein family which takes part of innate immunity and is able to recognize specific carbohydrates on the surface of a variety of infectious agents acting as a pattern recognition molecule. In this way, MBL differentiates self from non-self and interacts with other molecules of the immune system. MBL genotype shows various MBL2 polymorphisms which are responsible for MBL deficiency in a substantial portion of the entire human population and for susceptibility to infectious disease. Therefore, it has been highlighted in the relationship between genetic variants and clinical significance. Here we focus on presenting anoverview of our understanding of MBL structure and functions.
In order to transform pokeweed antiviral protein cDNA to tobacco plant, total RNA was extracted from Phytolacca americana. PAP-II cDNA was synthesized from purified total RNA via RT-PCR and subcloned to recombinant vector pBluescript II SK-. 10 deletion mutant PAP-II cDNA fragments which were sequentially deleted from N-terminal by 90bp were synthesized from PAP-II cDNA except leading frame by PCR with primers designed in our laboratory. To select non-cytotoxic clone, pAc55M was constructed with yeast expression vector pAc55 and multicloning site(MCS). Sequentially deleted mutant PAP-II cDNAs were cloned on downstream of gall promoter of pAc55M. 6 non-cytotoxic deletion mutant PAP-II cDNA were selected. Selected cDNAs were cloned into plant expression vector pKGT101BH for transformation of these clones to plant through Agrobacterium tumefacience. After cloning, recombinant pKGT101BH carrying deleted mutant PAP-IIcDNA were transformed to Nicotiana tabacum cv. NC567. Transformed tobacco plants cultured on shooting and rooting media were transfered to green-house. About four weeks later, these plants were infected with physically infection using carborandum with PVY-VN strain. After 4 weeks, plants resistant to virus were selected , and seeds of these plants were gathered. Southern blot hybridization showed deleted fragments by 220bp and 420bp, so resistant ability of these plants is due to mutant PAP-II cDNA.
IgY(Immunoglobulin Yolk) is a specific antibody in egg yolk, and it protects human body from virus and antigen. There are a lot of egg yolk components such as lipoprotein and protein. To separate IgY, HPLC(High Performance Liquid Chromatography) and precipitation were used in a batch mode and SMB(Simulated Moving Bed) was adopted for continuous purification of yolk proteins. IgY and other proteins in yolk were separated by using three-zone SMB chromatography. Before performing SMB experiments, batch chromatography and PIM(pulse input method) were performed to find operation parameters and adsorption isotherms. The results of batch chromatography were compared with simulated results using Aspen chromatography. To find the most suitable separation condition in SMB chromatography, simulations in $m_2$-$m_3$ plane on the triangle theory were carried out. $m_2$ = 0.18, $m_3$ = 1.0 and ${\Delta}$t = 419 s are the best conditions for the highest purity of IgY. With this operating parameters(flow rate in three zone and switching time), the purity of raffinate results in 98.39% from Aspen chromatography simulation. Most of the simulation reached steadystate within second recycle.
Crude sap, which was extracted from six Amaranthaceae plants, inhibited local lesion formation on Nicotiana glutinosa by tobacco mosaic virus(TMV) infection. Especially the remark. able inhibitory effect to TMV infection was shown on leaves of N. glutinosa precoated with the sap from Amaranthus mangostanus. The inhibitory activity of the sap from A. mangostanus was stable to storage in vitro for I day and to dilution 1/4 of the sap with distilled water. However, its activity was lost when the sap was heated at $70^{\circ}C\;to\;100^{\circ}C$ for 10 minutes. When the leaves of N. glutinosa precoated with the sap were sprayed with water, the inhibitory effect to TMV infection was maintained for 2 days. The A. mangostanus sap readjusted pH 3, pH 5, or pH 9 with 1 N HCl or 1 N NaOH did not decline the inhibitory action but the sap absorbed with $5\%\;to\;15\%$ charcoal completely lost their action. The protein components purified from A. mangostanus sap revealed three major bands by $5\%\;to\;15\%$ polyacrylamide gel electrophoresis and the top component of which showed the inhibitory action to TMV infection.
An isolate of Cucumber mosaic virus(CMV), called as Can-CMV, was originally isolated from Canna generalis showing typical streak mosaic foliar symptoms, and its properties were investigated in this study. Whereas all known isolates of CMV could induce symptoms on their systemic hosts(four kinds of Nicotiana spp and a zucchini squash), Can-CMV induced no symptoms on its systemic hosts tested. Replication and movement of the virus on upper leaves as well as inoculated leaves-were confirmed by RT-PCR suggesting that Can-CMV could only infect systemically on N. benthamiana and N. glutinosa. Size of local lesions on the Can-CMV-inoculated leaves of Chenopodium amaranticolor was much smaller than that of Fny-CMV. Whereas Fny-CMV and LS-CMV could induce distinct necrotic local lesions on Vigna unguiculata 2 to 3 days postinoculation(dpi), chlorotic spots symptom was expressed by Can-CMV 4 to 5 dpi. Virus-specific 4 kinds of dsRNAs were isolated from leaves of N. benthamiana infected with Can-CMV, and these dsRNAs corresponded to the viral genomic RNAs and subgenomic RNAs and their patterns were indistinguishable to those of Fny-CMV and LS-CMV. By restriction mapping analysis of 950 bp of RT-PCR amplified products of coat protein gene of the virus as well as by serological analysis of gel diffusion test, Can-CMV belongs to a typical member of CMV subgroup IA. These results suggest that the Can-CMV isolated from C. generalis possesses unique pathological properties to understand further insight into the various interactions between virus and host.

Thrips infesting hot peppers were monitored in greenhouses using yellow sticky traps. In addition, the hot peppers infected with tomato spotted wilt virus (TSWV) were observed during the monitoring period. The flower thrips (Frankliniella intonsa) were initially trapped at a low density just after transplanting seedlings of hot peppers at late March. The western flower thrips (Frankliniella occidentalis) were trapped after mid April. These two thrips represented more than 98% of the total thrips attracted to the traps after May, in which F. intonsa showed higher occurrence frequency than F. occidentalis. The total number of thrips had two peaks at mid May with a small and short-term peak and at June-July with a large and long-term peak. The trapped thrips exhibited inconsistent sex ratios, suggesting a seasonal parthenogenesis. Different geographical populations were varied in cytochrome oxidase I sequences, in which local populations in Andong shared a high sequence similarity. TSWV-infected hot peppers, which might be mediated by these two thrips species, were observed and confirmed by an immunoassay kit and a molecular diagnosis using RT-PCR. In addition, the TSWV was detected in F. occidentalis collected from the infected hot peppers. Three open reading frames (NSS, N, and NSM) of the isolated TSWV genomes were sequenced and showed multiple point mutations containing missense mutations among geographical variants. When the isolated TSWV was fed to nonvirulent thrips of F. occidentalis, the virus was detected in both larvae and adults. However, the viral replication occurred in larvae, but not in adults.
Papaya (Carica papaya L.) is one of the crops widely planted in tropical and subtropical areas. The papaya fruit has low calories and are plentiful in vitamins A and C and in minerals. A major problem in papaya production is a plant disease caused by the papaya ringspot virus (PRSV). The first PRSV-resistant GM papaya expressing a PRSV coat protein gene was developed by USA scientists in 1992. The first commercial GM papaya cultivars derived from the event was approved by the US government in 1997. Development of transgenic papayas has been focused on vaccine production and limited agricultural traits, including insect and pathogen resistance, long shelf life, and aluminum and herbicide tolerance. Approximately 17 countries, including the USA and China, produced transgenic papayas and/or commercialized them, which provoked studies on biosafety assessment and development of GM-detection technologies. For the biosafety assessment of potential effects on human health, effects of long-term feeding to model animals have been studied in terms of toxicity and allergenicity. Studies on environmental safety assessment include influence on soil-microbial biodiversity and transfer to soil bacteria of GM selection markers. Many countries, such as Korea, the European Union, and Japan, that have strict regulations for GM crops have serious concerns about unintended introduction of GM cultivars and food commodities using unauthorized GM crops. Transgene- and/or GM event-specific molecular markers and technologies for genomics-based detection of unauthorized GM papaya have been developed and have resulted in the robust detection of GM papayas.

종양은 아직 그 발생 원인과 기작이 정확히 밝혀져 있지 않으므로 정확한 정의를 내리는 것은 어려우나 일반적으로 종양이란 정상 세포가 갖고 있는 세포 분열의 특이성을 상실하여 일어나는 조직의 자율적인 과잉 성장을 말한다. 이와 같은 세포의 비정상적인 증식에 의한 종양을 임상 및 병리 형태학적으로 양서 종양과 악성종양으로 분류한다. 양성 종양을 일으키는 종양 세포는 정상 세포와 비슷할 뿐 아니라 그 주변 세포들과 확실한 경계를 이루고 있으며 증식도 느리며 다른 부위로의 전이가 없다. 이에 반해 악성종양은 일반적으로 증식도 빠르고 이형의 세포로서 주변의 조직으로 확산, 전이될 뿐만 아니라 최종적으로 숙주인 개체를 사망시킨다. 악성종양은 다시 상피 조직에서 유례한 암, 비상피 조직에서 유래된 육종, 백혈구에서 유래된 백혈병 등으로 구별하지만 그의 본질은 거의 같으며 모든 악성종양은 통속적으로 암이라고 불리운다. 종양의 발생원인으로 크게 3가지로 나눌 수 있는데 화학물질, 바이러스 및 유전적 요인에 의한 것으로 알려졌다. 최초의 발암물질로 알려진 benzopyrene에 의한 발암 등 연구에 의해 화학적 발암원들은 직접 발암 물질로 작용하는 것이 아니라, 일단 체내에서 대사된 후 이들 대사 산물일 DNA 등에 작용함으로써 발암이 유도 되는 것으로 밝혀졌다. 이와 같이 화학적 변화를 거친 후에야 DNA에 영향을 미치는 것외에 다른 화학물질들은 또 다른 기작을 통해 암을 유발하는데 쥐의 피부에 benzopyrene을 한 번 처리하면 암을 유발하지 않으나 여기에 phorbol ester를 처리하면 높은 빈도로 암이 형성된다. 여기서 benzopyrene과 같이 세포의 DNA에 돌연변이를 일으키는 작용을 하는 발암물질을 발암개시제라 하고 phorbol ester처럼 그 자체로는 발암능이 없으나 발암개시제에 노출된 세포에 영향을 미쳐 발암능을 크게 강화시켜 주는 것을 발암촉진제라고 한다. 암은 세포증식 제어계에 DNA가 이상을 일으킨 현상을 말하는 데 이와 같은 DNA의 변형된 유전정보에 의해 암과 관련된 단백질을 합성하므로 이 DNA를 암유전자라 부르며 바이러스에서 유래된 것을 V-one 그리고 세포에서 유래된 것을 c-one이라 한다. 암유전자는 본래 암을 형성하기 위한 것이 아니라 증식제어 유전자로서 변이나 비정상으로 활성화 됨으로써 암을 유발시키므로 proto-oncogene이라 부른다. 또한 고등동물의 유전자 중에는 세포성장을 억제하는 유전자들이 있으며 이들은 세포의 성장 생존 분화를 조절하는 것으로 생각되고 있다. 따라서 이들 유전자는 세포의 암변형을 억제하는 기능을 가지고 있다. 이들 유전자의 이상으로 세포성장 억제기능이 상실되면 세포의 과잉 성장이 초래되면 결과적으로 암으로 유발하는 것으로 추측되고 있다. 최근의 연구에 의하면 암세포에서 암억제 유전자의 이상과 암유전자의 활성화가 함께 발견 되면서 정상세포가 암으로 변형되는 과정에는 암억제 유전자의 이상과 암유전자의 활성화가 동시에 관여한 가능성이 높은 것으로 알려져 있다. 정상 세포가 암을 유발하기 위해서는 발암의 다단계설에서와 같이 여러 단계의 변과가 필요한데 여러 가지 요인에 의해 정상세포의 염색체가 변화되어 정상세포들이 가지고 있는 세포분열의 특이성을 상실하고 증식이 빠르고 저항력이 강한 세포가 선택 되어지고 비정상 서ㅔ포으 과잉 분여러에 의해 종양이 형성되며 이어서 혈관의 신생을 촉진하는 맥관형성, 전이 등이 과정을 거쳐 신체의 다른 부위로 전이된다. 20세기 초까지는 암은 노화와 함께 자연발생적으로 일어나는 피할 수 없는 질병으로 여겨졌으며 그 치료도 조기에 발견된 암환자에게 외과적인 치료를 하는 것이 최선의 방법 이었다. 그러나 현재에는 암환자의 80% 이상이 환경적 요인에 의해 암이 발생 된다고 믿어지고 있다. 과거 치료에 중점을 둔 것에서 점차 예방의 가능성과 그 방법의 모색에 관심을 갖게 되었으며 치료적인 면에서도 외과적 수술 이외에 방사선 치료, 항암제의 투여 등 약물요법, 면역요법의 이용 이외에 더 나아가 gene theraph 및 tumor vaccines 개발에 대한 관심도 증가되고 있다. 국제암연구협회에서는 인간에게 발암이 가능한 물질의 종류를 정기적으로 발표하고 있는데 지금까지 발암 가능성이 높다고 널리 알려진 위험요인을 크게 나누어 보면 다음과 같다. 흡연, 음주, 식이요인, 호르몬 및 기타 요인으로 약물, 자외선 등을 들 수 있는데 현재까지 이들 요인에 의한 발암 기작이 완전히 규명된 것은 아니지만 이들에 의한 발암의 확률이 높다는 것은 사실이므로 이 위험요인에 노출되는 것을 방지함으로써 암발생을 예방할 수 있을 것이다. 암발생의 예방법으로는 암발생 자체를 예방하는 것과 이미 발생한 암환자를 조기 발견하고 치료하는 방법이 있을 수 있다. 현재까지의 여러 연구 결과들을 보면 유전적인 요인을 제외한 대부분의 발암 위험인자들은 개개인의 생활습관과 밀접히 관련되어 있음을 알 수 있다. 이를 바탕으로 1992년 대한 암협회에서는 '암 예방 14개 권장 사항'을 발표하여 국민 홍보활동을 하고 있는데 그 내용의 반 이상이 식생활 습관과 관련되어 있을 정도이므로 암예방에 있어서의 식품의 역할이 매우 크다 할 수 있다. 따라서 건전한 생활습관과 더불어 적절한 식품의 섭취는 암예방을 위한 기본이 될 것이다.

제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
