Generation Method of Spatiotemporal Image for Detecting Leukocyte Motions in a Microvessel (미소혈관내 백혈구 운동검출을 위한 시공간 영상 생성법)

  • This paper presents a method for generating spatiotemporal images to detect the leukocyte motions in a microvessel. By using the constraint that the leukocytes move along the contour line of a blood vessel wall, the method detects leukocyte motions and then generates spatiotemporal images. the translational motion by a movement in vivo is removed first by the template matching method. Next, a blood vessel region is detected by the automatic threshold selection method to binarize the temporal variance image, then a blood vessel wall's contour is expressed by B-spline function. With the detected blood vessel wall's contour as an initial curve, the plasma layer of the best accurate position is determined to be the spatial axis by snake. Finally, the spatiotemporal images are generated. The experimental results show the spatiotemporal images are generated effectively through the comparison of each step of three image sequences.

  • In this paper, we propose a detection method of the leukocyte motions in a microvessel by using spatiotemporal image analysis. The leukocyte motions that adhere to blood vessel walls can be visualized to move along the blood vessel wall's contours in a sequence of images. In this proposal method, we use the constraint that the leukocytes move along the blood vessel wall's contours and detect the leukocyte motions by using the spatiotemporal image analysis method. The generated spatiotemporal image is processed by a special-purpose orientation-selective filter and then subsequent grouping processes are done. The subsequent grouping processes select and group the leukocyte trace segments among all the segments obtained by simple thresholding and skeletonizing operations. Experimental results show that the proposed method can stably detect the leukocyte motions even when multiple leukocyte traces intersect each other.

Preparation of Magnetic Chitosan Microsphere Particles (나노 크기의 마그네타이트 입자를 이용한 자성 키토산 미소구체의 제조)

  • Magnetite nanoparticles, which have been extensively used in many fields, were encapsulated with a natural polymer, chitosan, to improve their biocompatibility. We have synthesized magnetite $(Fe_3C_4)$ nanoparticles using chemical coprecipitation technique with sodium oleate as surfactant. Nanoparticle size can be varied from 1.2 to 7.4nm by controlling the sodium oleate concentration. Magnetite phase nanoparticles could be observed from X-ray diffraction. Magnetic colloid suspensions containing particles with sodium oleate and chitosan have been prepared. High magnetic property chitosan-microsphere particles were prepared from oleate-coated magnetite suspension using spray method. The surftce, and tile morphology of the magnetic chitosan microsphere particles were characterized using optical microscope and scanning electron microscope. Magnetic hysteresis measurement were performed using a superconducting quantum interference device (SQUID) magnetometer at room temperature to investigate the magnetic properties of the chitosan microspheres including magnetite nanoparticles. The SQUID measurements revealed superparamagnetism of nanoparticles.

  • 핵의학(nuclear medicine )이 란 방사성 및 안정 핵종의 동위원소표지 화합물을 인체에 투여하여, 관심장기의 형태 및 기능을 평가하여 해부학적 또는 생리학적 상태를 진단, 치료하는 의학의 전문 분야이다. 핵의학에 이용되던 방사성 핵종은 1960년대 까지만 해도 $^{131}$ I이 주였으나 1970년대 부터는 $^{99}$Mo -$^{99m}$ Tc 발생기와 $^{99m}$ TC으로 표지된 방사성의 약품이 활발히 이용되면서 $^{131}$ I을 대신하게 되었다. 원자로-생산핵종들의 특성은 중성자가 과잉이어 붕괴시 배타입자를 방출하는 점으로, 이것이 각종 질병의 치료에 이용되고 있다. 특히 각종 표시 화합물의 성질을 이용하여 원하는 부위에 방사선을 집중시킬 수 있음이 외부조사보다 유리한 점이다. 방사성핵종을 이용한 악성종양의 치료에 가장 성공적인 것은 분화된 갑상선 암환자에서 $^{131}$ I을 사용한 것이며, 갈색세포종 등에 $^{131}$ I-MIBG도 효과적이다. 악성종양의 골전이 치료에 베타선을 방출하는$^{32}$P, $^{186}$ Re, $^{153}$ Sm 등이 이용되었다. 종양의 동맥에 주입하여 세동맥이나 모세혈관에 걸리는 기름, 교진 또는 입자에 의한 치료에 $^{131}$ I-lipiodol, ethiodol, $^{32}$P 또는$^{90}$ Y흡사 ceramic resin 미소구 $^{166}$ Ho 유산중합체 미소구 등이 이용된다. $^{166}$ Ho, $^{198}$ Au, $^{32}$P, $^{90}$ Y, $^{169}$ Er, $^{186}$ Rc, $^{131}$ I, $^{211}$ At 등 의 방사성 핵종의 교질, 미소구 또는 단세포군 항체표지 형태로 직접 종양내 또는 공동이나 체강에 투여하는 치료법이 있다. 류마치스 관절염의 슬관절에 $^{165}$ Dy colloid를 주사하는 $^{166}$ Ho-MAA도 활발히 이용되고 있다.

Effects of Houttuynia cordata Thunb Extract Inhibits on the Migration and Proliferation of Vascular Smooth Muscle Cell (어성초 추출물의 혈관 평활근 세포 이주 및 증식 억제 활성에 관한 연구)

  • Houttuynia cordata Thunb.[H.cordata]belonging to Saururaceae, is a wild medicinal herb of perennial plants, and grows well in a place with a lot of shade and moisture. The medical action of H.cordata is reported to have an antitumer effect, toxicity-suppressive effect, antifungal effect, diuretic effect, and antioxidative action, but its effect hasn't been reported on cardiovascular diseases, such as ateriosclerosis and hypertension yet. This study intended to confirm the effect of the water extract of H.cordata on the migration and proliferation of rat aortic smooth muscle cells. Such results show that the water extract of H.cordata suppresses the migration and proliferation of rat aortic smooth muscle cells. It is believed that a useful clue will be offered later to the prevention of cardiovascular diseases such as ateriosclerosis and hypertension, and the development of their medicines on the basis of the fact.

  • The attractiveness of the gingiva is determined by its color, shape, and the shape and location of the boundary between the teeth and the gingival tissue. The standards beauty, balance, and health of the gingiva are all different, but the general public would agree that a coral pink gingiva is more beautiful than black or brown gingiva. Hence, one would be able to smile more confidently in public if he or she receives a gingival pigmentation removal surgery that changes the color of black or brown gums to a beautiful pink color with relative simplicity. The color of one's gingiva varies from pale pink to deep bluish purple, depending on many health components. The most prominent among these include the vascular supply, epithelial thickness, the degree of keratinization, and the presence of pigment in the epithelium. Melanin, carotene, reduced hemoglobulin, and oxyhemoglobulin are the main pigments contributing to the normal color of the oral mucosa. Fine Structure and Detoxification Kinetics in Kupffer Cells after Injection of Endotoxin in Rats (내독소 투여에 의한 Kupffer 세포의 미세형태학적 해독반응)

  • The aim of this study was to clarify the role of Kupffer cells in the mechanism of endotoxin-induced liver injury. The study on fine structure of Kupffer cells was performed after the injection of endotoxin. The endotoxin(Escherichia coli lipopolysaccharide 026 : B6. 1.5mg/100 g of body weight) was intraperitoneally injected in Sprague-Dewley rats. Animals were sacrificed at 1/4, 1/2, 1, 2, 4, 8, 16, 24, 72 and 120 hours after the injection of endotoxin. Livers were extirpated and processed to be examined by light and electron microscopy. The results obtained were summerized as follows: Early changes observed in liver after endotoxin injection included the increased number and hypertrophy of Kupffer cells, infiltration of neutrophils and presence of fibrin thrombi within the sinusoids. The continuous increase of the Kupffer cells in number with hypertrophy, congestion and infiltration of inflammatory cells within the sinusoids were observed. Hepatocytes showed fatty change and occasional necrosis. At 72 hours the congestion decreased. At 120 hours the number of Kupffer cells was increased, but the morphology of Kupffer cells became similar to that of the control group. The numbers and sizes of primary and secondary lysosomes and amount of euchromatin of Kupffer cells increased. Swellings and increase in number of mitochondria, Golgi complex, smooth endoplasmic reticulum, rough endoplasmic reticulum were evident. Microthrombi were present within the sinusoids. The swelling of rough endoplasmic reticulum and mitochondria, decrease of glycogen particles, fatty change, hypoxic vacuoles, pyknotic nuclei and occasional necrosis were observed in hepatocytes. At 72 hours the number of secondary lysosomes in Kupffer cells decreased. At 120 hours the morphology of Kupffer cells became similar to that of the control group. According to these results, it was postulated that the endotoxin was initially taken up by pinocytosis into Kupffer cells and degraded in secondary lysosomes of activated Kupffer cells. Kupffer cells may play an important role in the defense mechanism of liver during endotoxemia. The dysfunction of Kupffer cells and ischemia by sinusoidal microthrombi may cause liver injury.

Enhancement of in vivo Radiosensitization by Combination with Pentoxifylline and Nicotinamide (Pentoxifylline과 Nicotinamide의 병용에 의한 생체내 방사선 감수성 증강 효과)

  • Pentoxifylline (PENTO) has been known to improve RBC fluidity, and thus improve the flux of RBC through narrow capillaries. Additionally, PENTO also decreases the $O_2$ affinity of hemoglobin by increasing 2,3-DPG levels, thereby increasing the $O_2$ release from RBC. Nicotinamide (NA) has been reported to decrease the number of acutely hypoxic cells in tumors by temporarily increasing tumor blood flow. Therefore, the purpose of this study was to examine whether the combination of PENTO and NA (PENTO+NA) would reduce the radioresistance of the Fsall murine fibrosarcoma by oxygenating the hypoxic cells. We obsewed a significantly enhanced radiation-induced growth delay of the FSaII tumors by PENTO+NA. Thus the enhancement ratio was between 2.5 and 2.8 in growth delay assay. The $TCD_{50}$ of control tumors was about 57 Gy, but that of PENTO+NA treated tumors was about 32Gy. Thus $TCD_{50}$ was modified by a factor of 1.8. We also observed that PENTO+NA exerted no effect on the radiation-induced skin damage after the legs without bearing tumors were exposed to X-irradiation. In order to clarify radiosensitizing effects of PENTO+ NA, changes in tumor blood flow and intratumor pOf were measured using laser Doppler flowmetry and $O_2$ microelectrode methods. The tumor blood flow significantly increased at 10 min. after injection of PENTO+ NA. Furthermore, we also found that PENTO+ NA significantly increased intratumor $pO_2$ from 8 to 19 mmHg. We concluded that PENTO+MA was far more effective than NA alone or PENTO alone. The increase in the response of tumors in vivo to X-irradiation appeared to be due mainly to an increase in the tumor oxygenation. Further studies using various concentrations of PENTO alone and in combination with NA to obtain better sequencing and maximal radiosensitization are warranted.

Telomerase Activity in Non-small Cell Lung Cancer (비소세포폐암에 있어서의 Telomerase 활성도)

  • Although many reseraches have been persued to detect the molecular tumor marker to define the cancer, ideal tumor marker which speak for the characteristics of malignancy and has high sensitivity and specificity is not known. One of the characteristics of the malignant cells is indefinite proliferative potential, in other word, immortality. The expression of telomerase and stabilization of te10meres are con omitant with the attaiunent of immortality in tumor cells; thus the measurement of telomerase activity in clinically obtained tumor samples may provide important information which would be useful as a diagnostic marker to detect immortal cancer cells. Telomerase activity was analyzed in 12 non-small cell . lung cancer cell lines and 41 primary non-small cell lung cancers with the use of a PCR-based assay. All the cell lines and the majority of tumors displayed telomerase activity, but telomerase was not detectable in most of the corresponding pathologically-normal tissues. Telomere length was not correlated with telomerase activity. The present study indicate that measurement of telomerase activity may be useful as a molecular tumor marker in non-small cell lung cancer.

Antitumor Effect of Schizandrin by Inhibiting Angiogenesis (Schizandrin의 신혈관형성억제에 의한 항암효과)

  • Schizandra chinensis extract has been known to possess a variety of efficacy including antitumor. However, it remains unclear how schizandrin, which is a major biological active ingredient of Schizandra chinensis, exerts antitumor effect. This study was designed to investigate the mechanism by which schizandrin inhibits tumor growth and metastasis. In in vivo test using tumor model mice injected with B16BL6 cell line, mice treated with 10 and 100 ${\mu}g/ml$ schizandrin showed a significant inhibition by $73.79{\pm}6.43%$ and $90.46{\pm}1.72%$, respectively, compared with positive tumor controls. Schizandrin did not exert a significant toxicity for the normal cells (HUVECs) and tumor cell lines (A549, B16BL6, Du145, Huh7). Treatment with schizandrin at 10 and 100 ${\mu}g$/head significantly inhibited the tumor-induced angiogenesis by $68.04{\pm}32.21%$ and $103.8{\pm}34.99%$ compared with the positive control group, respectively. Using in vivo lung metastasis model, tumor metastasis assay revealed that 10 and 100 ${\mu}g$/head schizandrin significantly decreased the metastatic lung tumor by $37.51{\pm}8.15%$ and $75.53{\pm}4.38%$ compared with positive controls, respectively. On the other hand, schizandrin did not affect the adherence of B16BL6 cell line to extracellular matrix protein. These results demonstrate that schizandrin exerts inhibitory effect on tumor growth and metastasis by inhibiting angiogenesis. This study thus suggest that schizandrin may be a candidate molecule target for cancer drug development.
