Distribution and Identification of Halophilic Bacteria in Solar Salts Produced during Entire Manufacturing Process (천일염 생산공정별 미생물 분포 조사 및 호염미생물 동정)

  • In this study, we determined the changes in microbial numbers in solar salts according to the manufacturing process and storage duration. The salt samples were harvested from salt farms in Shinan (area 2) and Yeonggwang (area 1). They were serially diluted ten-fold and then placed on 4 kinds of cultivable media (mannitol salt agar, eosin methylene blue, plate count agar, and trypticase soy agar). After incubation, we obtained 62 halophilic isolates from the salt samples. Coliform and general bacteria were not detected in all salt samples. By 16S rRNA sequencing analysis, we found 12 kinds of halophilic bacteria belonging to the genera Halobacillus, Halomonas, Bacillus, Idiomarina, Marinobacter, Pseudoalteromonas, Vibrio, Salinivibrio, Virgibacillus, Alteromonas, Staphylococcus and some un-known stains. In our study, we discovered two novel species that have a 16S rDNA sequence similarity below 97%.

Comparison of Bacterial Diversity in the Water Columns of Goseong Deep Seawaters (고성 심해에서 수심에 따른 해양미생물의 다양성 비교)

  • Microbial diversities in the 300 m and 500 m deep seawaters near Goseong, Gangwon Province (South Korea), were investigated. Pyrosequencing of 16S rRNA genes of marine microbes resulted in 19,474 reads from the 300 m deep seawaters, which consisted of Alphaproteobacteria (57.41%) and Gammaproteobacteria (38.85%), and 82,806 reads from the 500 m deep seawaters, which consisted of Gammaproteobacteria (99.64%) mostly. Rhodobacterales (57.31%) were dominant in the 300 m deep seawaters, but Alteromonadales (45.65%) and Oceanospirillales (34.61%) were dominant in the 500 m deep seawaters. On the bases of operational taxonomic units and diversity indexes (Shannon and Simpson), biodiversity of marine bacteria in the 500 m deep seawaters was shown to be higher than that in the 300 m deep seawaters.

  • We isolated lactobacillus phage hosting L. casei strains, and examined distribution of the phages and resistance to the sanitary reagent treatment in drainages fo Korea plant. 1) There were Type 1, 11, 111, and 1v of the phage J$_1$in plantdrainages and the order of distribution was Type 1v>Type 11>Type 1>$^1$Type 111. 2) All the phages in plantdrainages were completely removed by spreading the invert soap. 3) Sodium hypochloride, invert soap and dresol were most effective sanitary reagents, and isopropyl alcohol and ethyl alcohol were sanitary reagents.

  • To investigate the variations of physicochemical factors and microbial populations in seven stations at water region of Gangjin bay, nutritive salts, water temperature, transparency, suspended solid, salinity, COD, DO, pH, heterotrophic bacteria, fungi and fecal coliform were analysed four times from February to October, 1999. Total fecal coliform and heterotrophic bacterial population during investigation periods ranged 16.1~166.0 CFU/ml and $5.0{\times}10^3$~$13.1{\times}10^3$CFU/ml, respectively. Stational mean values of these bacterial population showed higher densities at the inlet of fresh water inflow than those of other stations. When the aspects of abiotic and biotic parameters measured were analyzed, ecosystem of Gangjin bay was regulated by factors such as inflow of fresh water, nutrient salts, salinity and variation of water temperature during four seasons.

  • File fish fillets collected from markets were analyzed for their microbial load and microflora, resulting in the level of aerobic mesophiles ranged from $3.5{\times}10^3$ to $1.1{\times}10^8CFU/g$, total coliforms from less than 2.3 to $4.6{\times}10^5MPN/g$, fecal coliforms from 1ess than 2.3 to $1.1{\times}10^5MPN/g$, Enterobacteriaceae from $3{\times}10$ to $4.4{\times}10^5CFU/g$ and fecal Streptococci from $1.6{\times}10^2$ to $7.6{\times}10^6CFU/g$ having Staphylococcus hominis, Staphylococcus hemolyticus, Micrococcus varians, Micrococcus luteus and Streptococcus cremoris as constituent microorganisms of aerobic mesophiles, Escherichia coli and Proteus rettgeri of Family Enterobacteriaceae, and Streptococcus faecium and Streptococcus avium of fecal Streptococci.

  • The localization of carbon monoxide dehydrogenases (CO-DHs) in Pseudomonas carvoxydovorans and Acinetobacter sp.1 was examined by comparison of the distribution of CO-oxidizing activity between soluble and particulate fractions obtained after disruption of CO-grown cells by sonic oscillation and of spheroplasts by osmotic shock. When the cells were broken by sonic oscillation, most of the CO-DH activity was recovered from soluble fractions. However, disryption by osmotic lysis of spheroplasts revealed that the enzyme activity is present in the cell membrane. The results indicated the CO-DHs in these cells are loosely attached to the cytoplasmic membrane.
