Cell Viability and Fatty Acids Composition of Zymomonas mobilis grown at different Concentrations of Ethanol (Zymomonas mobilis 균체의 지방산 분포와 균의 생존성에 미치는 ethanol 농도의 영향)

  • The aim of the present studies was to analyze the physiological background of ethanol inhibition in Zumomonas mobilis. The experiments were carried out with a number of continuous culture to give steady state concentration of ethanol. The domposition of fatty acids in the cells obtained from various conditions was analyzed and cell viability was also estimated. As results, it was found that vaccenic acid was the mafor fatty acid in the cell of Z. mobilis and the concentration was changed apparently to increase as increasing the concentration of ethanol produced from substrate utilization. Finally it was observed also that cell viability was decreased remarkably at the elevated ethanol concentration. Those changes might play important roles in the ethanol fermentation to give more complex phenomena observed at high concentration of ethanol.

A Survey for Distribution of Airborne Microorganisms in Storage of Movable Cultural Properties (동산문화재 다량 보관처의 공기 중 부유 미생물 분포 조사)

  • The temple and family or private owner have managed the storage space of movable cultural properties. Thus they lack the ability to manage professionally and systematically, movable cultural properties are in a poor environment and have been damaged by abundant dust and airborne fungi in the storage. In this study, we investigated microbes distribution in 10 storage or exhibition hall housing the movable cultural properties. As a results, concentration of collected microorganisms exhibited a large difference according to a storage and the D Relic Museum in Yeongam is the most contaminant storage, in which detected $2,000m^3$ or more. More than $166m^3$ of the fungi were detected in most storages of the other. We identified so many varieties of fungi such as Aspergillus sp., Penicillium sp., Alternaria sp. and Cladosporium sp. existing commonly in 10 storages including wood rot fungi such as Ceriporia lacerata, Ganoderma carnosum, Myrothecium gramineum and Bjerkandera sp.. This airborne fungi may damage cultural heritages. The Guideline on a concentration of airborne fungi should be estimated and management system to the preservation environment must be provided.

Microbial Diversity and Community Analysis in Lettuce or Cucumber Cultivated Greenhouse Soil in Korea (상추 및 오이 시설재배 토양의 미생물 다양성 분석)

  • The soil chemical properties, microbial community structures and biochemical properties of lettuce or cucumber-cultivated greenhouse soil samples were analyzed to assess soil health and characterize microbial distribution in 8 locations in Korea. Although most of chemical properties were within the soil management guidelines, the available phosphate, and the contents of exchangeable potassium and calcium were higher than those of recommended levels. In the culture-dependent analysis, 841 bacterial strains were isolated from the greenhouse soils and were identified at the genus level by 16S rRNA gene sequences analysis. The dominant bacterial genera were Bacillus (35.7%), Microbacterium (9.3%), Arthrobacter (5.7%) and Lysobacter (5.1%). The abundance of pseudomonads was highly variable depending on the soil samples. In the culture-independent analysis, soil microbial community was investigated by using phospholipid fatty acid (PLFA) method. Principal component analysis (PCA) showed that a specific grouping for microbial community structure in the greenhouse soils was not observed based on cultivated crops and investigated sites. The results revealed that the greenhouses soils examined are relatively sound managed in terms of soil chemical contents and microbial properties.

  • We isolated microorganisms from soil arounding the stump and surface of stump in Bubjusa temple in Boeun, Heungguksa temple in Yeosu and Leeyongwoo house in Boseong. As a result, a distribution percentage of bacteria was high and there was a region-specific difference in bacteria colony. There was greater microbial diversity and population in the Leeyongwoo house in Boseong.

Microbial Distribution at Sediments of Lake Daechung (대청호 퇴적층의 미생물 분포 밀도)

  • In order to characterize the microbial distribution at sediments of Lake Daechung, soil samples were collected at two depths of 0.5~2 cm and 19~21 cm of Chudong(static) and Hoenam(streaming) site on May 18th(before rainy season) and on August 24th in 1998(after rainy season), and then the density(CFU/g soil) of microorganisms including bacteria, actinomycetes, and fungi was investigated by the viable cell counting method. Microbial density at streaming site was on the whole 3.9-fold higher than that at static site. Bacterial densities examined before and after rainy season was revealed to be similar, whereas actinomycetes and fungi exhibited higher distribution after and before rainy season, respectively. The microbial distribution was not generally reduced with the increase of depth and was rather higher at some deep sites. On comparing with the microbial densities of grass land around the lake, bacteria, actinomycetes, and fungi at lake sediments on the average showed the distribution of 52.9%, 35%, and 7%, respectively. However, their distribution except for fungi which exhibited 71.2% was mostly found to be somewhat higher than at the shore of lake.

Microbial Colonization of the Aquatic Duckweed, Spirodela polyrhiza, during Development (수생식물 개구리밥 (Spirodela polyrhiza)과 미생물)

  • Fresh specimens of the aquatic macrophyte, Spirodela polyrhiza, have been examined employing scanning and transmission electron microscopy. Observations revealed the occurrence of microbial colonization during development. Submerged parts of the small, free-floating S. polyrhiza body exhibited a variety of microorganisms such as bacteria, cyanobacteria, and diatoms throughout their development. However, immature and/or young plants normally demonstrated much less microbial colonization compared to mature plants. During the study, heavy colonization by the microorganisms was routinely encountered at maturity, especially in the fully developed abaxial fronds and root caps. The mucilaginous layer was shown along the root caps, and the microorganisms appeared to be either clustered or attached to this layer. In contrast, only moderate degrees of colonization were observed in the root, and little to no colonization was observable in the adaxial frond surface. Transmission electron microscopy clearly demonstrated the microbial colonization to be external in the S. polyrhiza specimen examined in the current study. The association between the microorganisms and S. polyrhiza has been considered non-harmful, as no frond senescence and almost no mechanical penetration of the plant by the microorganisms were noticed during the study.

  • Protective effects on subcellular localization of Photobacterium leiognathi CuZnSOD(PSOD) were examined in Escherichia coli SOD mutant cells on the treatment of paraquat, heat shock $(37^{\circ}C{\to}42^{\circ}C{\to})$, hydrogen peroxide and copper sulfatem respectively. The physiological characteristics of the periplasmic and cytoplasmic PSOD localized differently are dependent on the conditions in this experiment. Cells expressing SOD periplasmically in the treatments of paraquat and $H_2O_2$ respectively were somewhat better protective effects cells expressiong SOD cytoplasmically at comparable level and SOD expression level showed, the most consistently important variable. However, this was reversed in the treatments of heat shock and $CuSO_4$, respectively.

Phylogenetic diversity of marine bacteria dependent on the port environment around the Ulleng Island (울릉도 항구의 해양환경에 따른 해양미생물의 분포 변화)

  • Pyrosequencing of 16S rDNA tags was used to obtain the bacterial diversity and community structure in the uncultured seawaters as well as in the cultured seawaters, which were collected from the 7 ports (Cheonbu, Hyunpo, Taeha, Namyang, Sadong, Dodong, and Jeodong) and 1 seashore (Guam) around the Ulleng island, Korea. Alphaproteobacteria were the most abundant group in the clean seawaters such as seawaters of Taeha and Sadong ports. Gammaproteobacteria proportion increased depending upon the wastewater amounts mixed with the seawaters such as seawaters of Namyang, Dodong, and Jeodong ports. The genuses of Alteromonas (from samples of Cheonbu, Taeha, Guam, Namyang, Sadong), Shewanella (from sample of Jeodong), and Vibrio (from samples of Hyunpo and Dodong) were dominant group in each of the cultured seawaters incubated in marine broth (Difoco). The results suggest that the incoming wastewaters to the port seawaters contribute to the dynamic change of the marine bacterial community around the Ulleng island.

Microbial Distribution as an Environmental Factor for the Conservation of Cultural Properties in the War Memorial in Korea (전쟁기념관내 문화재 보존을 위한 환경요인으로서의 미생물의 분포)

  • Two isolation methods using the open plate and air sampler were applied to examine microbial distribution as an environmental factor for conservation of cultural properties in the War Memorial in Korea. The numbers of fungi were the abundant in the air of entrance and inside of the exhibition room where visitors were crowded, compared with inside of repository rooms. Fungal and bacterial distributions in the exhibition room during exhibition period were higher than during nonexhibition period, These results are due to the inflow of air current carrying microoganisms to the exibition rooms from outside with visitors. Fungi isolated from the War Memorial were identified as 19 species and one thermophilic fungus, Neurospora crassa. The majority of the isolated fungi are seemed to cause damage to cellulose materials in the Memorial.

  • 일반적으로 쌀막걸리 중에는 풍미에 관여한다고 생각되는 핵산관련물질이 핵산분해효소인 PDase (phosphodiesterase) 및 PMase (phospomonoesterase) 등의 작용을 받아 대부분이 분해되는 것으로 알려져 있다. 따라서 저자등은 쌀막걸리 제조공정을 통하여 핵산관련물질의 분포와 그와 관련된 핵산 분해효소에 대한 실험을 하였다. 본보에서는 제국중 핵산관련물질과 핵산관련물질을 분해하는 효소의 경시적 변화를 조사하였으며 이들 분해효소의 효소학적 성질을 검토하여 몇가지 결과를 얻었으므로 보고하는 바이다.
