Studies on the Isolation and Identification of Bacillus sp. for the Composting of Swine Manure and the Removal of Malodorous Gases from its Liquid Compost (돈분의 퇴비화를 위한 Bacillus sp.의 분리 동정 및 그 액체 비료의 악취 제거 연구)

  • Bacillus species were isolated from swine manure to develope the microbial additive suitable for the rapid com-posting. The 3 of 4 isolated strains were identified as Bacillus cereus KD-2, B. pumilus KD-3, and B. licheni-formis KD-4. Bacillus sp. KD-1 was, however, not highly identical with any Bacillus sp. The isolated strains were analyzed their growth rates, enzyme activities, and antibacterial activities. The maximum growth tem-peratures of KD-1, KD-2, KD-3 and KD-4 were $45^{\circ}C$, $50^{\circ}C$, $53^{\circ}C$, and $55^{\circ}C$, respectively. The activities of pro-tease or amylase in mixed culture of 4 strains were similar in the range of $37^{\circ}C$ to $53^{\circ}C$ and activities of lipase in the range of $37^{\circ}C$ to $42^{\circ}C$ were twice higher than those of lipase in the range of $47^{\circ}C$ to $53^{\circ}C$. The antibacterial activity of KD-l, KD-2, or KD-3 against each other was not detected. That of KD-4 against KD-1, KD-2, or KD-3 was, however, detected. The organic compound and C/N ratio of compost fermented by the mixed culture were determined as 61.9% and 22.4%, respectively. The concentration of the ammonia gas was 12.35 mg/l in the liquid compost.

Use of Activated Soil to Bioaugment Degradation of Atrazine in Soils (토양 내의 Atrazine의 생물학적 분해 촉진을 위한 활성토의 이용)

  • Effectiveness of activated soil containing directly enriched atrazine-degrading soil microorganisms as an inoculant to bioaugment degradation of atrazine in soils was investigated. A Wooster silt loam (Typic Fragiudalf) was spiked with atrazine at a rate of 4 mg/kg soil three successive times to create activated soil. Atrazine degradation was significantly enhanced (p < 0.05) after the first treatment. After the second treatment, there was an increase in the number, based on MPN, of microorganisms utilizing atrazine as a C- and N-source by 3 logs and 1 log of magnitude, respectively. Inoculation of typical agricultural soils collected from Ohio with activated soil at a rate as low as 0.5% reduced the extractable atrazine remaining in soils to the level below 2% of that initially recovered (initially added at a rate of 4 mg/kg soil) after 4 days. Inoculation at a higher rate was required to achieve the same result in soils with non-typical properties (pH of 4.5 or organic matter of 43% w/w). Activated soil was stable, in terms of atrazine degradation activity, at least up to 6 months when it was kept at low temperature (< $10^{\circ}C$) and moistened (water content above 15%). The results of this study indicate that microorganisms capable of degrading atrazine are relatively easily enriched in soil to create activated soil. Use of activated soil can be a practical option for bioremediation of contaminated soils.

Influences of Invasive Plant on Extracellular Enzyme Activities in Riparian Ecosystems (하변토양의 미생물체외효소활성에 미치는 칩입성 식물의 영향)

  • We have measured soil enzyme activities, which represent the rates of organic matter decomposition, in four riparian ecosystems in Korea. ${\beta}$-glucosidase, N-acetylglucosaminidase, phosphatase and arylsulfatase activities were determined in five occasions over a year period in soils of control plots and plots with invasive plants, namely Sicyos angulatus and Humulus japonicus. Significantly higher enzyme activities were found in soils with invasive plant in barren land, but the difference was season and enzyme-specific. Although it was not universal changes, the invasive plants appeared to accelerate organic matter decomposition in some disturbed riparian ecosystems.

  • A bacterium producing the antifungal compound KRF-001 isolated from soil was selected and identified as Bacillus subtilis. The antibiotic KRF-001 was active against various fungi. Effecacy of KRF-001 at various concentration for controlling leaf blast of rice in the paddy field was evaluated and compared with recommended rates of kasugamycin, blasticidin- s and tricyclazole. KRF-001 caused no irritation on the skin of rabbits and LD50 for mice was deduced more than 5000 mg/kg which indicates the possibility of low toxicity or no toxicity.

  • In this study, we have tried to find the maximum microbial efficiency at the various dissolved oxygen (DO) concentration conditions in the treatment of pharmaceutical wastewater. Experiments of activated sludge process were performed to examine the relation between microbial activity and DO concentration in a continuous bioreactor. The reduction rate of COD (chemical oxygen demand) was low at the DO concentration between 0 ppm and 1.0 ppm, but it went higher and showed maximum between 1.5 ppm and 3.0 ppm. Then the COD reduction rate was quickly decreased above the DO concentration of 3.0 ppm.

  • In this study, we investigated the biological activity of antioxidant and antibacterial activity of citrus by-product ferment. Hot water extracts and 80% methanol extracts from citrus byproduct of ferment were screened for antibacterial activity pathogenic bacteria by paper disc method. Among the various hot water extracts and 80% methanol extracts the Bacillus subtilis showed relatively strong antibacterial activities in the order. The reducing activity on the 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and ${\cdot}OH$ radical scavenging potential were sequentially screened, in search for antioxidant activities of citrus by-product ferment.

  • 자연수계 생태계는 해양, 하천, 호수, 지하수 등과 같이 그 특성에 따라 많은 종류로 구분할 수 있는데, 그중에서 하구는 물리, 화학적 성질은 물론 미생물을 포함한 많은 생물이 끊임없이 변천(transition)하는 생태계의 하나의 꼽을 수 있다. 환경의 변화는 수온, 염분, pH, Eh, 투명도, 부유물질 등의 변화를 포함하는데, 하구 감조수역에서 가장 심한 변화를 일으키는 것은 일반적으로 영양염류의 양과 종류, 수온, 염분 등이다(Morita et al., 1973: Rublee et al., 1984). 그러므로 본 연구에서는 1985년 7월부터 1986년 12월까지 낙동강에 위치한 3개 정점에서 물리, 화학적 환경, 영양염류, BOD, 세균 및 heterotrophic activity의 28개 변수의 변화를 살펴보았다. 이렇게 측정된 결과를 중회귀 및 요인 분석으로 생태계에서 세균의 분포와 활성에 미치는 영향을 분석하고자 한다.

Characterization of Anti-dementia, Cadiovascular and Antioxidant Functionalities in Korean Traditional Alcoholic Beverages (전통주의 항치매 활성과 심혈관질환 활성 및 항산화 활성 탐색)

  • Some anti-dementia, cardiovascular and antioxidant functionalities of Korean traditional alcoholic beverages were characterized. Anti-dementia acetylcholinesterase inhibitory activities were generally not detected or low except BHS-ju (19.5%) and buthyrylcholinesterase inhibitory activities were also show below 1.0%. However, $\beta$-Secretase inhibitory activities were high in PMR-ju (42.5%), WJY-ju (41.6%) and SSJ-ju (42.9%). Antihypertensive angiotensin I-converting enzyme inhibitory activities was the highest in YON-ju (85.6%), however fibrinolytic activities were not detected in all traditional alcohol beverages. Furthermore, antioxidant activities were very high in SBB-ju (72.2%) and GMB-ju (67.9%), however SOD like activities generally were show below 20%.

Characteristics of the Bacteriocin from Lactobacillus sp. Oh-B3. (Lactobacillus sp. Oh-B3로부터 생산되는 박테리오신의 특성)

  • A bacteriocin producing microorganism, which inhibits the growth of Lactobacillus sake, was screened and isolated from Kimchi. This microorganism was identified and named as Lactobacillus sp. Oh-B3, The maximum amount of bacteriocin was produced when the isolated microorganism was cultured in MRS media(pH 8.0) for 24 hours at 25℃. The bacteriocin from the isolated microorganism was purified through ammonium sulfate precipitation, dialysis and ultrafiltration. The bacteriocin was stable on the wide pH range of 2.0-9.0, and showed antimicrobial activity on some of gram positive bacteria, not on gram negative. The antimicrobial activity of bacteriocin was mostly removed by treatment of proteolytic enzymes. But, the bacteriocin was very stable on the heat treatment, and more than 50% of activity was remained at autoclaving. The action mode of the bacteriocin showed bacteriocidal pattern, being same as that of general bacteriocins.

Chemical Characteristics and Biological Activities of Herbimycin A and Dihydroherbimycin A Produced by a Soil Isolate Streptomyces sp. AO-0511 (Streptomyces sp. AO-0511이 생산하는 Herbimycin A 및 Dihydroherbimycin A의 이화학적 특성 및 생물 활성)

  • A streptomycete strain was isolated from the soil samples from Korea. The chemotaxonomy and 16S rDNA sequencing confirmed that the strain belonged to the genus Streptomyces and we named it Streptomyces sp. AO-0511. Two antibiotics, herbimycin A and dihydroherbimycin A produced by this strain were tested for their physico-chemical and biological characteristics. Both compounds were stable under acidic pH. Dihydroherbimycin A was more heat-stable and polar compared with herbimycin A. Only weak antibacterial activities were detected against Bacillus subtilus ATCC 6633 and Micrococcus luteus ATCC 9341. However, herbimycin A and dihydroherbimycin A showed strong inhibitory activities on lung cancer cells (A549 cells) and leukemia cells (HL-60). The cytotoxicity was determined using L5178Y and P388 cell lines. The results showed that herbimycin A and dihydroherbimycin A had lower toxic effects on the cells compared with the standard compounds, comptothecin and cyclosporin A. Therefore, both compounds could be good candidates for the development of new anticancer drugs.
