  • The differences in microbial community structures between planktonic microorganism and biofilm in rivers were investigated using respiratory quinone profiles. The compositions of microbial quinone for 4 tributaries of the Kyongan Stream located in/flowing through Yongin City, Gyeonggi-Do were analyzed. Ubiquinone(UQ)-8, UQ-9, menaquinone(MK)-6 and Plastoquinone(PQ)-9 were observed in all samples of planktonic microorganism and biofilm for the sites investigated, Most planktonic microorganism and biofilm had UQ-8(15 to 30%) and PQ-9(over 30%) as the dominant quinone type. These results indicated that oxygenic phototrophic microbes(cyanobacteria and/or eukaryotic phytoplankton) and UQ-8 containing proteobacteria constituted major microbial populations in the river. The quinone concentration in the river waters tested, which reflects the concentration of planktonic microorganisms, increases with increasing DOC. Further research into this is required. The microbial diversities of planktonic microorganism and biofilm calculated based on the composition of all quinones were in the range from 4.2 to 7.5, which was lower than those for activated sludge(ranging from 11 to 14.8) and soils(ranging from 13.4 to 16.8). The use of quinone profile appears to be a useful tool for the analysis of microbial community structure in river.

  • The objective of this study was investigated for the microbial community structure and treatment performance of domestic wastewater in lab-scale submerged membrane bioreactor operated with anoxic-oxic cycles. Respiratory quinone profiles were applied as tools for identifying different bacterial populations. The cycle time program of bioreactor was control under anoxic/oxic of 60/90 minutes with an hydraulic retention time of 8.4 hrs. The average $COD_{Cr}$ removal efficiency of domestic wastewater was as high as 93%. The results showed complete nitrification of $NH_4^+$-N generated during oxic period and up to 50% of the total nitrogen could be denitrified. The dominant quinone types of suspended microorganisms in bioreactor were ubiquinone (UQ)-8, -10, followed by menaquinone (MK)-6, and MK-7 for anoxic period, but those for oxic period were UQ-8, MK-6, followed by UQ-10 and MK-7. The microbial diversities of bioreactor at anoxic and oxic periods, calculated based on the composition of all quinones were 10.4 and 12.2-11.8, respectively. The experimental results showed that the microbial community structure in the submerged membrane bioreactor treating domestic wastewater was slightly affected by intermittent aeration.

  • A Streptomyces strain No. 182-27, which produced amino acid antimetabolite, was isolated from soil. During the course of screening for new amino acid antimetabolites from the culture broths of Actinomycetes, we found that the strain produced a substance active against Gram-positive bacteria and its activity was reversed by L-Ieucine on the synthetic minimal agar medium in the culture broth. The morphological and cultural characteristics serve to identify the producing organism strain 182-27 as the Streptomyces, although the species of this strain should be resolved in further studies. Fermentation was carried out in the synthetic medium at $28^{\circ}C$ for 78 hours. The fermentation yield reached about 2 mg per liter of the broth. Purification was done by ion exchange resin, active carbon, silica gel column chromatography and obtained 20 mg of pure active substance from the 20 $\ell$ culture broth. The 182-27 substance was obtained as white powder, mp 18SoC. From the physicochemical characteristics of the substance, it was amino acid like substance but unknown about its chemical structure. It is active against some Gram-positive bacteria and reversed by L-Ieucine.

  • Changes of biochemical and genetic diversity of microbial communities in the soil damaged by a forest fire were analyzed. Soil samples were collected from Gangnung area where a forest fire was broken out in 2000. Two soil samples were from the burnt area, one from the naturally restoring soil (NS) and the other from the artificially restoring soil (AS). A normal, unaffected soil sample (US) was also included as a control. For the biochemical diversity, each sample was directly applied to the BIOLOG system, and the cluster analysis through a statistic process (SPSS) were performed. Genetic diversity was analyzed through DGGE using 16S-rDNA amplified from soil DNA. Among the samples tested, top soils of US and NS, and sub soil of NS revealed more than 70% of the similarity value in biochemical diversity. In case of genetic diversity, however, the similarity value was found to be in the range of 53% to 68% in all samples. This result indicates that the biochemical diversity is not always correlated with the genetic diversity in the analysis of microbial communities.

  • Genomic DNA of Bacillus stearothemzophilus, which expressed alkalophilic and thermophilic xylanases, was partially digested with HindIII, cloned into pBR322, and subsequently transferred into the Escherichra coli HB101 cells. Three among 5, 000 transformants screened formed clear zones around their colonies. From the functional clones, three recombinant plasmids (pMG11, pMG12 and pMG13) had been isolated, and they were identified to carry the same 4 kb HindIII fragment originated from B. stearothemzophilus which was responsible for the xylanase activity. pMGl3, however, had the foreign DNA of opposite orientation compared to the other two recombinant plasmids. This recombinant plasmid gave much lower xylanase activity. B. stearothermophilus was observed to produce at least three xylanase activities as evidenced by the PAGE-xylan zymogram. The xylanase from E. coli HB101/pMG12 was judged to correspond to the largest among the three B. stearothermophilus xylanases observed in the zymogrom. The enzyme hydrolyzed xylooligosaccharides larger than xylotriose and degraded xylan to produce xylobiose and xylotriose as major products. The xylanase was considered to have trans-xylosidase activity, too.

  • In soil and sediment environment, microorganisms play major roles in biochemical cycles of ecological significant elements. Because of its ecological significance, microbial diversity and community structure information are useful as indexes for assessing the quality of subsurface ecological environment and bioremediation. To achieve more accurate assessment, it is requested to gain sufficient yield and purity of DNA extracted from various soil and sediment samples. Although there have been a large number of basic researches regarding soil and sediment DNA extraction methods, little guideline information is given in literature when choosing optimal DNA extraction methods for various purposes such as environmental ecology impact assessment and bioremediation capability evaluation. In this study, we performed a thorough literature review to compare the characteristics of the current DNA extraction methods from soil and sediment samples, and discussed about considerations when selecting and applying DNA extraction methods for environmental impact assessment and bioremediation capability evaluation. This review suggested that one approach is not enough to gain the suitable quantity and yield of DNA for assessing microbial diversity, community structure and population dynamics, and that a careful attention has to be paid for selecting an optimal method for individual environmental purpose.

  • Microbial diversity of soil samples from ancient stone-lined tombs was investigated. The tombs, discovered at Eoryung Ocheon-Ri site, Korea, were estimated to be belonged to middle class people from an ancient country, Gaya, which existed till AD 559 at the southern part of Korea. Nine fungal stains and 70 bacterial strains were isolated from the twelve soil samples, which were collected from the tomb Nos. 5 and 6. Ribosomal DNA sequence analysis discovered 5 fungal and 22 bacterial strains belonged to 10 genus groups from the tomb No. 5 while 1 fungal and 28 bacterial strains belonged to 6 genus from the tomb No. 6. The higher microbial diversity suggests that the tomb No. 5 was constructed warmer season than the tomb No. 6. Moreover, the discovery of Staphylococcus warneri, which is found as part of the skin flora on human and animals, and Bacillus aquimaris, which is a marine bacterium and can be discovered from tidal flat, from the surface of large dagger suggests that the ancient people may use meat and seafood at the burial ceremony.

  • To investigate the substrate variety of a putative non-metal dependent isomerase, the tagatose-6-phosphate isomerase (E.C. 5.3.1.26) structural genes (lacB; 510bp and lacA; 430bp) of Staphylococcus aureus were subcloned and co-expressed. Based on the substrate configuration, various aldoses were surveyed for substrate of ketose isomerization. Among the 10 aldoses tested, D-ribose and D-allose were isomerized by the enzyme. The subunit A and B showed more than $95\%$ activity for D-ribose and $75\%$ for D-allose in the presence of 1mM EDTA compared with non-EDTA conditions, which implying tagatose-6-phosphate isomerase is a non-metal dependent isomerase. Each of subunit A or subunit B alone showed no activity for any of the substrates tested. The affinity constant ($K_m$) of tagatose-6-phosphate isomerase against D-ribose and D-allose were 26 mM and 142 mM, respectively.
