This study is on encapsulation of a new whitening agent, LA-PEG using solid lipid nanoparticle(SLN) method, one of nanoparticle preparation method. Classical method has high capsulation efficiency for hydrophobic compounds but has demerit of low capsulation efficiency($2{\sim}3%$) for hydrophilic compounds. Purpose of this study is preparation of SLN that has higher skin penetration effect compared with general liposome, and also has higher encapsulation efficiency of hydrophilic compounds. For SLN preparation, coconut oil, macadamia oil, and jojoba oil were used. As a result, SLN preparation using coconut oil(include LA-PEG) has the most high encapsulation efficiency and also has the smallest average particle size(270 nm). SLN prepared with macadamia oil and 1% of Tween 60 has the largest particle size. Base made with coconut oil and 2% of Tween 60 showed the fastest release and base made with macadamia oil and 2% of Tween 20 showed the latest release.

L-ascorbic acid, the representative antioxidants, has a great effect on skin whitening, collagen synthesis, and anti-aging, but has low oxidative stability during storage. Therefore, in this study, thermal and oxidation properties of L-ascorbic acid under various storage conditions (powder, aqueous phase, changes of temperature, UV-irradiation, and inflow of external air etc.) were investigated. And the storage stability of ingredient was validated in the double-spaced pouch by analysing oxidation properties under each storage conditions (powder phase and blended with essence). In oder to analyze the thermal properties, TGA, DSC, and FT-IR analysis were carried out and UV-visible spectrophotometer & redox titration were used in parallel for oxidation property analyses. From the result of experiment, L-ascorbic acid was oxidized fast when it contained lots of metallic ion, hydroxy ion in aqueous solution under high temperature, UV-irradiation & inflow external air, whereas it was not oxidized for a long time when it was stored as pure powder although it has same condition as heating up, UV-irradiation & inflow external air. Based on this result, retention period of cosmetics which is using L-ascorbic acid, less stable material in oxidation can be innovatively increased when using double-spaced pouch that is designed and produced for separating storage of active ingredients.

Ultraviolet radiation exposure is a major cause of extrinsic skin aging, which leads to skin hyperpigmentation. Loganin, a major iridoid glycoside obtained from Corni fructus, has anti-inflammatory, anti-diabetic, and neuroprotective effects. In this study, we investigated the mechanisms underlying the anti-melanogenic effects of loganin in B16F10 melanocytes treated with ${\alpha}$-melanocyte stimulating hormone (${\alpha}-MSH$) and 3-isobutyl-1-methylxanthine (IBMX). Anti-melanogenic activity was measured by treating cells with loganin at concentrations between 1 and $20{\mu}m$. Cell viability assays confirmed that doses of loganin up to $20{\mu}m$ were not cytotoxic. Loganin significantly and dose-dependently decreased intracellular melanin production. We also investigated potential molecular signaling pathways for the anti-melanogenesis effects of loganin. Western blotting showed that treatment with ${\alpha}-MSH$ and IBMX increased the phosphorylation of cAMP response element-binding protein (CREB) and the gene expressions of microphthalmia-associated transcription factor (MITF) and tyrosinase. Addition of loganin suppressed these increases, while promoting the phosphorylation of extracellular signal regulated kinase (ERK) and the anti-melanogenesis response. Our data therefore indicated that loganin could attenuate the increased melanin synthesis induced by ${\alpha}-MSH$ and IBMX treatment of B16F10 melanocytes. This attenuation appears to occur by downregulation of CREB phosphorylation and MITF and tyrosinase gene expression and upregulation of ERK phosphorylation. These finding suggests that loganin could be a valuable candidate for treatment of skin diseases related to hyperpigmentation.

산초나무(Zanthoxylum schinifolium Siebold & Zucc.)는 운향과(Rutaceae)에 속하는 낙엽활엽관목으로 주로 열매, 잎, 줄기는 식·약용으로 이용한다. 최근 산초나무 부위별 추출물에서 항산화활성, 항염증효과, 미백효과 등에 관한 연구가 이루어지면서 산초나무의 가치가 더욱 증가하고 있다. 본 연구는 산초나무의 효율적인 유·무성 증식법을 구명하기 위해 종자 처리에 따른 발아특성, 생장조절물질 처리에 따른 가지삽목과 뿌리삽목의 발근특성을 조사하였다. 산초나무 종자는 3개월간 저온습사저장(종자:모래=1:1, -4℃)을 실시할 경우 약 64.0%가 발아하였다. 습사저장된 종자를 파종 직전에 GA3를 이용하여 0, 500, 1,000 및 2,000ppm 농도에서 처리한 결과 각 처리구별 발아율은 유의한 차이를 나타내지 않았다(F=1.556, p=0.260). 다만, 500ppm 농도에서 72.0%가 발아하여 가장 높은 발아율을 보였으며 1,000ppm 이상 처리구에서는 발아율이 다소 감소하는 경향을 나타냈다. 무성증식 방법은 2월 중순에 산초나무 가지삽수와 뿌리삽수를 채취한 후 IBA와 NAA를 각각 0, 500, 1,000 및 2,000ppm 농도로 처리하였다. 가지삽목을 실시한 대부분의 삽수에서 신초가 발생했으나, 발근율은 1.3~6.3%로 현저히 낮았다. 반면, 뿌리삽목은 3개월 후 부정아 발생율이 처리에 따라 58.3~83.3%로 조사되었으며, 발근율은 0~66.7%를 나타나 처리구별 발근율에 유의한 차이가 있었다(F=3.744, p=0.020). IBA 처리가 근삽수의 발근에는 효과적이었으며, 특히 IBA 2,000ppm 처리에서 66.7%의 발근율을 보여 가장 효과적임을 알 수 있었다. 결론적으로, 산초나무의 효율적인 유성증식을 위해서는 종자를 3개월간 저온습사저장하는 것이 효과적이며, 무성증식 방법으로는 가지삽목보다 뿌리삽목을 이용하는 것이 더 효과적인 것으로 판단된다.

Paeony has pharmacological activities such as anti-inflammatory, anti-allergic, anti-bacterial, central inhibitory, gastric secretion inhibition, and antispasmodic activities. In addition, its antioxidant activity and whitening effect being reported, thus it is being explored as raw materials for cosmetics. We compared the changes in the contents of paeoniflorin and paeonol in Peony extracts, depending on the changes of extracting solvents, temperature and time. The HPLC method was set up for simultaneous analysis, the system suitabilities were confirmed by using the calibration curves and the QC samples for each assay batch. Paeonol was detected only in roots, and paeoniflorin was higher in leaf and flower than root. Higher concentrations of both ingredients were extracted when the root was used after grinding to a suitable size, and when 30% 1,3-butylene glycol was used as the extraction solvent. Also the concentrations tended to increase at higher temperature and longer time, but the increase was gradual at over $75^{\circ}C$ and 4 hours. The ratio of root, leaf and flower was determined to be 2+2+1g/0.5kg of batch, reaching the contents criteria of paeoniflorin and paeonol. Finally, we selected as the best extraction condition when the raw materials are mixed with 2+2+1g/0.5kg and extracted with 30% 1,3-butylene glycol as an extraction solvent at $75^{\circ}C$ for 4 hours, considering both the concentrations of two components and the cost of raw materials and manufacturing process, The extraction units were scaled up to 10 kg under this condition.

This study was performed in order to investigate the functional components of 5 kinds of marine algae. We have collected 5 samples of marine algae, such as the sea mustard (Undaria pinnatifida), sea tangle (Laminaria iaponice), sea weed fusiforme (Hizikia fusiforme), green laver (Entetomotpha), laver (Phophyratenera), which have been harvested in Jeollanam-do. In order to examine the functional effects, 5 kinds of marine algae were extracted with hot water ($80^{\circ}C$, 4 hr), ethanol and methanol (R.T., 4 hr), and subcritical water extract (SWE, 3 MPa, $90^{\circ}C$, $150^{\circ}C$, $210^{\circ}C$). A higher yield of extract was obtained through SWE method (3 MPa, $210^{\circ}C$) in all of the samples obtained. The highest total sugar content was 427.4 mg/g in green laver extracted with SWE (3 MPa, $210^{\circ}C$). The content of the SWE total phenolic compounds was higher than that of the water and solvent (methanol, ethanol) extracts. The anti-oxidative activities of the extracts from 5 kinds of marine algae were examined through the DPPH radical scavenging activity test. The SWE (3 MPa, $150^{\circ}C$ and $210^{\circ}C$) of the marine algae was the highest among all of the extracts. As per the results, the SWE of the marine algae contained more functional components and it had a higher antioxidant activity than those of the other extracts. The $IC_{50}$ value of tyrosinase in seaweed fusiforme and laver were higher than those of the other samples. These results strongly support the possible use of marine algae as functional materials.
This study was carried out to investigate the antioxidant effects of extracts from 9 species of forest plants in Korea. DPPH, ABTS, $NaNO_2$, hydrogen peroxide radical scavenging activity and reducing power activity were evaluated to measure the antioxidant activities of plant extracts. As a result, Geranium thunbergii has been identified as the most effective antioxidant resource. Also, total phenolic content was highest in Geranium thunbergii ($303.94{\pm}0.63mg\;GAE/g$) among 9 species extracts. Total flavonoid content was highest in Rosa multiflora ($24.32{\pm}0.22mg\;QE/g$) and proanthocyanidin content was highest in Vitis ficifolia ($279.00{\pm}4.58mg\;CE/g$) among 9 species extracts. In addition, the protective effect of plant extracts in $H_2O_2-induced$ human dermal fibroblast (HDF) cell systems were also assessed. Significant protective effects in $H_2O_2-induced$ human dermal fibroblast (HDF) cell systems were found in all plant extracts, especially in Geranium thunbergii. These results suggest that Geranium thunbergii could be a potential natural resource for antioxidant activity.

Antioxidant, antibacterial, and skin penetration tests were conducted to investigate the skin absorption of Pyrus serotina var leaf extracts using polymer micelles and their applicability to cosmetic ingredients. Total polyphenol content was found to be $118.83{\pm}9.39mg/g$ in Pyrus serotina var leaf ethanol extract and $106.89{\pm}4.45mg/g$ in Pyrus serotina var leaf hydrothermal extract. The DPPH radical scavenging activity was found to be the highest radical scavenging activity of $74.39{\pm}7.48%$ of the Pyrus serotina var leaf ethanol extract at the concentration of 500 mg/L. The SOD-like activity was $91.62{\pm}0.43%$, the highest value at the concentration of 1,000 mg/L in the hydrothermal extract. After the experiment, antioxidation, wrinkle improvement and whitening activity were confirmed, and the Pyrus serotina var leaf extract was highly likely to be realized as antioxidant and antibacterial material. In the skin penetration experiment with the Pyrus serotina var leaf ethanol extract, the permeation amount of total accumulated tannic acid was found to be Formulation 2 ($55.45{\mu}g/cm^2$), Formulation 1 ($46.43{\mu}g/cm^2$), Formulation 0 ($34.36{\mu}g/cm^2$). In the liposome's skin penetration experiment containing pear leaf hydrothemal extract, the total amount of accumulated tannic acid permeation was found to be Formulation 5 ($75.01{\mu}g/cm^2$), Formulation 4 ($64.01{\mu}g/cm^2$) and Formulation 3 ($36.60{\mu}g/cm^2$). Through this study, we confirmed the possibility of antioxidant and wrinkle effects of Pyrus serotina var leaf extract. In addition, as a result of skin penetration through the production of polymer micelles and liposome containing Pyrus serotina var leaf extract, It will be more usable in cosmetic industry.

Artemisia fukudo Makino belongs to the Asteraceae family, is a halophyte that can grow in salinity soils and is known for its various physiological activities. However, few studies were comparing it according to the growth environment. Therefore, in this study, we analyzed the antioxidant activity, functional ingredients, and safety of Artemisia fukudo Makino according to the growth environment. Total polyphenol content was the highest in native mature leaves, but 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) and 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity was higher in cultivated leaves than in native leaves. The total mineral content, excluding sodium, was higher in cultivated than in native leaves. The content of potassium and calcium was higher in cultivated leaves than in native leaves. In addition, heavy metal analysis showed that cultivated leaves were generally lower than those of native leaves. Residual pesticides were not detected in all samples. In conclusion, since there is no significant difference in cultivated leaves compared to native leaves, it was judged that cultivated leaves could be used as a variety to be grown and mass-produced.

The tight junction, one of Intercellular junctions, performs a variety of biological functions by bonding adjacent cells, including the barrier function to control the movement of the electrolyte and water. Recent studies have revealed that unusual expression of tight junction-related genes have been shown to be related in cancer development and progression. Recently, there are many reports that control of tight junction proteins expression is closely related to the skin moisture. In this study, we are focusing on the regulating mechanism of tight junction-associated genes by the steviol and its derivatives. Steviol, used as a sweetner, is known to chemical compound isolated from stevia plant. The MTS (3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium, inner salt) assay was carried out in HaCaT cells (human keratinocyte cell line) in order to determine the cytotoxicity. As a result, while steviol showing cytotoxicity from $250{\mu}M$, steviol derivatives are not cytotoxic more than $250{\mu}M$ concentration. We have observed a change in the tight junction protein via quantitative real-time PCR. Claudin 8 among tight junction proteins is only significantly reduced up to 30% in the presence of steviol. In addition, cell migration was inhibited by steviol, not by stevioside and rebaudioside. Finally, we could observe that steviol, not stevioside and rebaudioside, is able to increase the skin barrier permeability through the transepithelial electric resistance (TEER) measurements. These results suggest that the steviol and its derivatives are specifically acts on the tight junction related gene expression, but steviol derivatives are more suitable as a cosmetic material.
