Tyrosinase catalyzes melanin synthesis in skin melanocytes. The effects of Paeonia sufficinalis water extract (MDP) on antioxidant and tyrosinase activities have been studied in a cell-free system and mouse melanoma B16 cell. Radical scavenging activity of MDP was tested by DPPH assay and it showed high DPPH radical scavenging activity. The cellular tyrosinase activity was measured in mouse melanoma B16 cell by RT-PCR and enzyme activity. Treatment with MDP for 24 hr resulted in decreased tyrosinase mRNA level. Tyrosinase activity was decreased, compared with control, in cells exposed to MDP. Thus, Paeonia sufficinalis water extracts may be a candidate for cosmetic use.

Many people of all ages wish to have lighter skin for cosmetic reasons, and natural products attract more attention than chemically synthesized compounds. Uncaria rhynchophylla is widely used in Asia as a traditional herbal medicine. In order to find novel skin whitening agents, the present study evaluated the antioxidant activity and potential tyrosinase-inhibiting properties of U. rhynchophylla. Specifically, this study analyzed the antioxidant capacity of a 70% ethanolic extract of U. rhynchophylla as well as its effects on tyrosinase activity and melanin synthesis. Total mRNA levels were examined using reverse transcription polymerase chain reaction. The results revealed that U. rhynchophylla extracts exhibit great antioxidant capacity and significant levels of polyphenol and flavonoid compounds. U. rhynchophylla extracts can also powerfully inhibit tyrosinase activity. This same capacity was observed in melanoma B16F10 cells; that is, U. rhynchophylla extracts suppressed intracellular tyrosinase activity and reduced the amount of melanin in treated cells. In addition, a 1 mg/ml concentration of U. rhynchophylla extract significantly reduced the mRNA expression levels of tyrosinase. U. rhynchophylla extracts decrease tyrosinase and inhibit melanogenesis in B16F10 cells. This finding suggests that U. rhynchophylla has great potential as a natural whitening agent in skincare products.

This study was performed to examine in vitro antioxidative activities such as DPPH radical scavenging activity, reducing power and tyrosinase inhibitory effect of various solvent fractions from Chionanthus retusa leaves. Ethyl acetate fraction showed potent antioxidative activity and tyrosinase inhibitory effect. The active compound was isolated from the butanol fraction by silica gel column chromatography and MPLC. The isolated compound was luteolin-4'-O-glucoside determined by $^1H$, $^{13}C$-NMR and 2D NMR. Compared with several antioxidant compounds, luteolin-4'-O-glucoside exhibited effective DPPH radical scavenging activity and reducing power in a concentration dependent manner. Bioassay with pure luteolin-4'-O-glucoside showed a dose-independent inhibitory effect on L-DOPA oxidation by mushroom tyrosinase and its $IC_{50}$ values were established as 23.2 ${\mu}g/mL$. Therefore, we may suggest that luteolin-4'-O-glucoside can be used as a food additive possessing the potent antioxidative activity and skin-whitening cosmetic material.
Comparison of four phenolic compounds among three different parts of Panax ginseng C.A. Meyer was performed in this study. Contents of esculetin, p-coumaric acd, quercetin, maltol in the ethyl acetate fraction were determinated by HPLC analysis. The main root has a highest concentration of p-coumaric acid and the leave has a highest concentration of quercetin and esculetin. This ethyl acetate fraction of the leaves exhibited higher depigmenting activity and lower cell toxicity than the other parts of ginseng. In addition, it showed that quercetin and esculetin among phenolic compounds highly inhibited melanin biosynthesis in ginseng. It implies that leaves of ginseng may be used as skin whitening herb. Quercetin and esculetin proved to be active compounds in the leaves of ginseng.
Rodgersia podophylla was a native medicinal plant cultivated in Korea. During the search for a new whitening natural herb, we found that underground part of Rodgersia podophylla showed inhibitory activity against melanin biosynthesis in melan-a cells by brocking tyrosinase activity. In addition, this plant exhibited protective effect on UV-B region. These results suggest that underground part of Rodgersia podophylla might be used as whitening agent for the skin.

Eruca sativa, called arugula, is a perennial plant in the Brassicaceae family, an edible plant commonly used in Italian cuisine. To study as a cosmetic material application E. sativa was extracted with 70% ethanol (ES). Then ES was fractionated using n-hexane, chloroform, ethyl acetate, n-butyl alcohol and water (EHex, EEA, ECHCl3, EBuOH and EDW). EEA showed mushroom tyrosinase inhibitory activity. ES, EEA and EBuOH showed inhibition of tyrosinase activity. As a result, ES is expected to have skin whitening efficacy. ES was applied to 0.05, 0.1% the toner and emulsion formulation to test the stability. The anti-microbial activity of eight bacteria and fungi including Staphylococcus aureus and Propionibacterium acnes which cause dermatitis and acne was evaluated. EEA showed effects in all of microorganisms. The toner and emulsion containing ES with 0.05, 0.1% were passed in the challenge test. At -20, 4, 25, 55 ℃ and daylight, there was no significant change on pH, viscosity for 4 months. However, emulsions had phase separation phenomenon at 55 ℃, so the base formulation needs improvement. In addition, through the skin penetration test, EEA penetrated 0.058% in 6 hr, predicting the clinical efficacy. This means that E. sativa can contribute whitening agent and the synergistic effect of preservatives.
Melanin is a natural pigment found in most plants and animals, and it is involved in determining the color of the skin and hair. Melanogenesis is a reactive occurrence in melanocytes aiming to protect the skin from external stimuli, such as ultraviolet rays. Tyrosine is involved in the biosynthesis of the substrate tyrosine into melanin. However, melanin overproduction can lead to skin diseases, such as melasma, blotching, hyperpigmentation, and skin cancer. Although many studies have been conducted on whitening substances, such as kojic acid and arbutin, some countries have banned or refrained from using them due to their side effects. Therefore, this study assessed the potential of horseradish (HR) as a new whitening agent in cosmetic products. For efficient extraction, subcritical water extraction was conducted. The results showed that the horseradish subcritical water 200℃ (SW 200) extract showed high DPPH radical scavenging ability, total phenolic contents (TPC), inhibiting tyrosinase activity and inhibiting melanin production of B16-F10 melanoma cell lines. To investigate its cytotoxicity to the B16-F10 melanoma cell lines, MTT reduction assay and morphological changes were observed. No cytotoxicity was found in horseradish methanol extract and SW 200. In conclusion, this research suggests the possibility of horseradish subcritical water may be useful as a natural whitening ingredient to be used in cosmetic products.
The solvent extracts of Nelumbo nucifera G. were investigated for antioxidant activities, whitening and anti-wrinkle effects to apply as a functional ingredient in cosmetic products. For their industrial application, the cosmetic products were also prepared with advanced formulation techniques such as W/O/W multiple emulsion. Total phenolic and flavonoids contents increased in Nelumbo nucifera G.-Leaf (NN-L). The electron donating ability of Nelumbo nucifera G.-Flower (NN-F) or Nelumbo nucifera G.-Leaf (NN-L) extracts were above 85% at a concentration of 500 ppm. The superoxide dismutase (SOD)-like activity of Nelumbo nucifera G. (NN-L) extracts was about 60% at a concentration of 1,000 ppm. The xanthine oxidase inhibitory effect of NN-L extract was higher than that of NN-F and NN-S extracts. The tyrosinase inhibitory effect, which is related to skin-whitening, was 36% in NN-F at 1,000 ppm. For anti-wrinkle effect, the elastase inhibition activity of NN-L was about 30% at 1,000 ppm. The results of stability test showed that W/O/W multiple emulsion (ME) containing Nelumbo nucifera G. extracts. The electron donating ability of the ME containing NN-F and NN-L were about 60% at a concentration of 100 ppm. The superoxide dismutase (SOD)-like activity of the ME containing NN-L was 30% at 1,000 ppm. The tyrosinase inhibitory effect, which is related to skin-whitening, was 34% in the ME containing NN-F at 1,000 ppm. In anti-wrinkle effect, the elastase inhibition activity of the ME containing NN-L was about 55% at 1,000 ppm.

In order to discover physiologically active substance, we investigated a powder obtained by processing of Opuntia ficus-indica var. saboten Makino trunk. The powder was treated by sulfuric acid and then extracted by several solvents such as methanol, methylene chloride, ethanol etc. Among them, the best antimicrobial activity was showed by methylene chloride extract. To identify materials exhibiting physiological activation, the acid hydrolyzed extract was separated by 7 fractions through preparative silica gel TLC. The effective fraction exhibiting the best broad antimicrobial activity was identified, named as MBT-01108. From structural analysis of the products extracted to acid hydrolysis, a compound exhibiting the antimicrobial activities is identified to levulinic acid. Levulinic acid isolated from Opuntia ficus-indica var. saboten Makino may be applicable as a natural preservative of food or cosmetic and for prevention of bacterial diseases, an ingredient of acne, ageing and whitening cosmetics and an antimicrobial agent.
Quantitative difference in five phenolic acids between white and red ginsengs was measured in this study. As the results, white ginseng has higher contents of cinnamic acid, quercetin and p-coumaric acid than red ginseng. Maltol was mainly included in red ginseng. These five compounds were recently reported to have tyrosinase inhibitory effects. These reports led us to investigate the de-pigmenting effect of ginseng products. In our examination of effect on tyrosinase activity, UV-protection and melanin production in melan-a cells, ethyl acetate traction of white ginseng extract and cinnamic acid showed potent de-pigmenting properties. The results indicated that white ginseng might be useful as skin whitening material and cinnamic acid proved to be one of active ingredient.
