Denaturing Gradient Gel Electrophoresis and Culture-based Analysis of the Bacterial Community in Cheonggukjang, a Korean Traditional Fermented Soybean Food from Gangwon Province (DGGE 방법과 배양법을 이용한 강원지역 전통 발효 청국장에서 미생물의 다양성 분석)

  • Bacterial communities derived from cheonggukjang and raw rice straw collected from a Mireuksan farm and a Heungup cheonggukjang in Gangwon province were investigated using both culture-based method and denaturing gradient gel electrophoresis (DGGE) analysis. Pure cultures, which were isolated from raw rice straw and cheonggukjang and cultured on tryptic soy agar plates (53-76 colonies per plate), were identified by analysis of 16S rRNA sequences. The traditional culture-based method and analysis of PCR-amplified 16S rRNA by DGGE revealed that for samples collected from the Mireuksan farm, Pantoea agglomerans and Bacillus subtilis were the predominant species in the raw rice-straw and cheonggukjang, respectively. For samples collected from the Heungup cheonggukjang, Bacillus amyloliquefaciens was the predominant species in both raw rice straw and cheonggukjang. Other microorganisms, including members of Pantoea, Bacillus, Enterococcus, Enterobacter, Pseudomonas, Rhodococcus, and Acinetobacter, were also present in the raw rice-straw and cheonggukjang, as were bacteria that could not be cultured.

  • To understand the drift-diffusion of HAB(Harmful Algal Bloom) in this paper, we used three-dimensional hydrodynamic model POM(Pringceton Ocean Model) and Lagrangian particle track module. First, the results of residual flow that considered tide, wind, temperature, salinity, and TWC(Tsushima Warm Current) effect was tend to northeast in the coastal area and the flow in the offshore region showed results similar to TWC. To understand of HAB's movement, released each area that southern Kamak bay(Case 1), Mijo coast(Case 2), and southern Mireukdo coast(Case 3) assumption that red tide occurred. The areas where the HAB occurs frequently. As a result of HAB occurred in southern Kamak Bay(Case 1), mainly drifts to Narodo coast and Yeoja bay that located on the west side. Case 2 was mainly drifts to Yokjido coast and Saryangdo coast Especially, HAB occurred in Mireukdo coast(Case 3) relatively many particles drift to eastward as the influence of the TWC.

Folk Ideas, Daoist Images, and Daoist Texts from the Late Joseon Dynasty (구한말 민중사상과 도교이미지, 그리고 도교서 언해)

  • In the late Joseon Dynasty, ideas in folk religions were closely related to Daoist themes. There were, for instance, folk ideas centered on Prophecies of Jeong Gam (鄭鑑錄 jeonggamrok) that developed into 'raising island-armies (海島起兵說 hado gibyeongseol),' the future utopian movement known as the 'South Joseon Faith (南朝鮮信仰 namjoseon sinang),' and faith around 'Maitreya's Descensionist-Birth (彌勒下生 mireuk hasaeng).' People aimed to transform their country based on these ideas. Associated folklore tended to come from fengshui (風水) and books on prophecies and divination (圖讖 docham), and both of these drew heavily upon Daoist concepts. On the other hand, Daoist texts began being translated as national projects under King Cheoljong (哲宗), and many more were translated and published later under King Gojong (高宗). The nature of these Daoist texts mostly consisted of either morality books (善書 seonseo) or precious scrolls (寶卷 bogeon). The problem was that these ordinances and the Daoist texts of regents were among the main causes of civil war during the Qing Dynasty. In this regard, the translation of the Daoist texts conducted as a national project provided a theoretical basis for the people wishing to foment civil war or transformation. This raises the question of why King Gojong implemented a Daoist translation project in his nation. In an effort to answer this question, this article summarizes the popular ideas of the late Joseon Dynasty and explains how they were closely related to Daoism. In addition, this article summarizes the facts about how Daoism has emerged from a national crisis but developed a function of protecting the state (鎭護) in Korean history. Further described is the situation under which Daoism was summoned during the Japanese Invasion of Joseon (壬辰倭亂). Analysis is provided to show that King Gojong's intention was to translate Daoism due to Daoism's role in protecting the state. In addition, the relationship between current Daoist rites and customs in Korea and King Gojong's dissemination of Daoist oaths and vouchers is confirmed.

A Study on the Changes of the Site Layout of Beopjusa(Temple) on Sokrisan(Mt.) (속리산(俗離山) 법주사(法住寺) 가람배치(伽藍配置)의 변천(變遷)에 관한 연구(硏究))

  • This study is to find out the characteristics of the changes of the site layout in Beopjusa which was built on Maitreya faith in the Shilla.. According to the analysis of it in this study, we make conclusions as follows; 1) The reconstruction of Beopjusa means a start in Dhamalsama(法相宗) and then it was supposed to be Buddhist temple which was formed by intersecting axis of centering around a wooden pagoda(捌相殿) with a main Buddhist hall and a lecture hall. 2) After the middle of Koryo dynasty, Beopjusa was changed to building layout of intersecting with Yongwabojeon(龍華寶殿) and Daeungbojeon(大雄寶殿) because of harmony with Avatamsaka(華嚴宗) and Dhamalsama centering around Avatamsaka. 3) The buildings of Zen Buddhism was built in the early Chosun dynasty owing to a prevalence of Zen Buddhism in the late Koryo dynasty. And since 17th century, Buddhist halls were each built in their a faith system according to interpenetrated Buddhism(通佛敎). 4) The courtyard type of mountainous district was made on interpenetrated Buddhism. On the other hand, the site layout of Beopjusa is being maintained by centripetal spatial organization through the wooden pagoda as object.

Metallurgical Study of Bronze Artifacts Excavated from Miruksa Temple (미륵사지 출토 청동유물의 금속학적 연구)

  • Metallurgical studies of the bronze artifacts excavated from Miruksa Temple were performed by chemical analysis and metallographic observation. Alloy systems of the bronze artifacts were classified into two groups of Cu-Sn and Cu-Sn-Pb, according to the items. The contents of impurities such as Sb, As, Ni and Fe in bronze artifacts are within the limiting range of the mod ern standard bronze castings. Chemical compositions of the kitchen utensils such as bronze vessels and dishes in the Unified Silla dynasty, are in the follow ing range, Cu : 74.8-79.4% and Sn : 18.6-21.1%. Chemical composition of the Buddha-image in Koryo dynasty are 820Cu-7.0Sn-10.3Pb, showing increased Pb content and decreased Sn content. The results of chemical analysis suggest that the chemical compositions were good controlled. Any casting defects such as voids and shrinkage holes are not found microscopically, indicating high casting skill. Zinc atoms are not contained in the all bronze artifacts of Miruksa Temple site. This is the common facts founded in the east asian bronze artifacts of Korea, China and Japan. It is comparable with the European bronze of Cu-Sn-Pb-Zn system, after the Middle Age.

  • History is bound to change according to the interpretation of modern people, and based on this, we must find the will to practice for our future. In order to examine the ideology of a country, we can know the cultural aspect of that country through its religious ideology and its cultural heritage. This paper focuses on the stone pagoda of the Baekje period. In order to collect data on the 5-story stone pagoda of Jeongnimsaji, I would like to study the formative features of the 5-story pagoda of Jeongnimsaji by visiting the Jeongnimsaji Museum and familiarizing myself with domestic and foreign professional books, preceding papers, and references based on prior research.

Flexural Behavior of Granite Reinforced with Titanium Metal Rods (티타늄 금속봉으로 보강된 화강암의 휨거동)

  • In this study, flexural tests of granite reinforced with titanium metal rods were carried out to repair and restore the damaged stone-made cultural heritage, the Stone Pagoda of the Mireuksa Temple Site. A total of twelve specimens were tested. The primary test parameters are the reinforcement ratio and the location of the reinforcement. For restoration, epoxy resin was used for joining the separated stones, and titanium metal rods were used for structural reinforcement. Test results showed that the flexural failure took place in specimens with a reinforcement ratio of 0.8% or less, and shear failure occurred when the reinforcement ratio was 1.68% or more. The peak load of the reinforced stone was found to be highly related to the reinforcement ratio. The peak load increased with increasing the reinforcement ratio. Also, the flexural behaviors of the reinforced stones were affected by the location of the reinforcement. Based on the test results, this study recommends the reinforcement ratio of the reinforced granite to induce ductile behavior.

  • 아시아 국가들은 근대사에 들어 서양 제국주의와 식민지주의로 인해 정치·사회적 과도기를 경험하였다. 지역종교들도 이러한 영향에서 예외는 아니었는데 19-20세기에 나타난 신종교들이 그 좋은 사례들이다. 한국도 이러한 지구적인 현상에 포함되었다. 주변국들의 문화적 계몽주의와 조선왕조의 쇄국정책 사이의 정치적 혼란은 방향 감각 없는 국가적 위기를 초례했고, 정치·사회적 부패와 국가적 불안정은 중·하류층 시민들로 하여금 고통스러운 삶을 극복하기 위해 새로운 진리나 믿음을 찾게 동요하였다. 근대사에 근원을 둔 대순진리회는 한국에서 가장 영향력 있고 성공적인 신종교일 뿐 아니라 현대사회의 기존 종교들에게도 많은 도전을 주고 있다. 그렇다면 이 신종교운동의 근원은 어디에 있을까? 그들의 교리나 가르침은 어떤 것일까? 다른 신종교적 현상들과 어떻게 구별될까? 이 논문은 창시자인 강증산의 역사적인 출현과 업적을 "성취관념"에서 분석해 볼 뿐 아니라 대순진리회가 기존의 유교, 불교, 도교의 가르침을 단순히 혼합한 것이 아니라 음양합덕, 신인조화, 해원상생, 도통진경의 특유한 가르침들은 미륵사상, 천지공사, 인존사상과 함께 그들의 최고신인 구천상제가 후천세계를 위해 성취한 완전한 미션을 반영하는 창조적인 사상임을 논증할 것이다.

Distribution of Organic Matter and Trace Metals in Surface Sediments and Ecological Risk Assessment in the Tongyeong Coast (통영연안 표층퇴적물에서의 유기물과 미량금속 분포 특성 및 생태위해성 평가)

  • We investigated the distribution of organic matter and trace metals in surface sediment from Tonyeong coast. To determine the status of trace metal pollution, we also conducted an ecological risk assessment. Relatively high concentration of TN (total nitrogen), TOC (total organic carbon), and AVS (acid volatile sulfide) was found in surface sediment located in the narrow channel (site 35-38). Spatial distribution of Cd, Cr, Ni, Co, Hg, and Zn in surface sediment was similar and high Cu concentrations were found in narrow channel. The assessment of heavy metal pollution was derived using the Enrichment factors (EF). The enrichment factor indicated that Cd was no enrichment (EF<1), Pb, Cr, Ni, Co, Zn, and Hg were minor enrichment (1
