Collection, Conservation, and Utilization of Plant Genetic Resources -emphasis on weeds- (유용식물 유전자원의 수집 보존 및 이용에 관한 연구 -잡초를 대상으로-)

  • Germinating ability of 54 weed species collected throughout Korea in 1987 and 1988 was determined in incubator at $25^{\circ}C$ for 15 days. The seeds of Echinochloa sp, collected from 70 different places throughout Korea, were classified by Yabuno's seed morphology method. Plant characters of two identified Echinochloa species, one having two subspecies were evaluated. SDS-PAGE(sodium dodecyl sulfate polyacrylamide gel electrophoresis) of seed protein assay was used to identify locally collected Echinochloa species classified by Yabuno's seed morphology method. 1. Out of 54 weed species collected, 31 species showed germination at 15 days after incubation, and 14 species showed over 50% and no germination was observed in 24 species, indicating that approximately 40 species had varied degree of germinating ability. 2. It was confirmed that two annual species of Echinochloa such as E. oryzicola and E. crus-galli occurred in Korea and one of which particularly E. crus-galli has two subspecies such as E. c. var. praticola and E. c. var. crus-galli. 3. E. oryzicola was distinguished from E. c. var crus-galli and E. c. var praticola by presence or absence of one major protein band in B zone. However, it was rather difficult to distinguish E. crus-galli species because they showed similar protein pattern.

Taxonomic Identity of Echinochloa crus-galli (L.) Beauv. var. crus-galli in Dokdo (독도 돌피의 분류학적 실체)

  • Molecular study were conducted to evaluate taxonomic identities of Echinochloa crus-galli (L.) Beauv. and Echinochloa crus-galli var. echinata (Willd.) Honda in Dokdo. Echinochloa crus-galli complex of two species 26 individuals analyse based on nuclear ribosomal DNA (ITS region) and cpDNA (trnH-psbA, trnL-F). At a result, two species were same sequence. Characters the length of the lemma and the length of the awn traits were identity of the species was unclear. According to, Taxonomy treatments that is based on existent morphological characters should thinks again. On the other hand, in the case of ITS, Echinochloa crus-galli (L.) Beauv. and Echinochloa crus-galli var. echinata (Willd.) Honda at the Dokdo forms from other clades with individuals that is collected at land area and Ulleungdo. These result is showing that is flowing independent evolution trends.

Hormonal Effect on the Callus Induction from Perennial Weeds (다년생잡초(多年生雜草)로부터 Callus 유도(誘導)와 생장조절제(生長調節劑)의 영향(影響))

  • This experiment was conducted to evaluate effect of various hormones on callus induction, and on plantlet formation on various media, and to detect of Londax [Methyl 2-[[[[[(4,6-dimethoxy pyrimidin-2-yl) amino] carbonyl] amono] sulfonyl) methyl] benzoate] and Basta[Ammonium-(3-amino-3-carboxy-propyl)-methyl phosphinate] on callus growth and reaction of succinate dehydrogenase in callus against TTC, using various species such as Eleocharis kuroguwai, Cyperus serotinus, Oryza sativa (samgangbyeo) and Echinochloa crusgalli P. Beauv. var. caudata Kitagawa. The optimal levels of 2,4-D in MS medium seems to be different among species tested, 2.0 ppm for rice and E. crusgalli, 1.0 ppm for Eleocharis kuroguwai, and 4.0 ppm for C. serotinus derived callus from shoot-tip. In case of combination of 2,4-D with BA, 1.0 plus 0.3 ppm appeared the most appropriate level to induce callus from rice and E. kuroguwai, and I.0 plus 0.1 ppm for C. serotinus and E. crusgalli. When 2,4-D treated with TIBA, 1.0 plus 0.5 ppm appeared the most appropriate rates to induce callus derived from seeds of rice, E. crusgalli, seeds of C. serotinus and E. kuroguwai, 1.0 plus 0.3 ppm for shoot-tip of C. serotinus. Positive reaction of succinate dehydrogenase against TTC was observed regardless of calli and herbicides tested, indicating that they all are alive, and these herbicides were not able to kill the calli tested within the short period of time 20 hrs treatment. Regardless of plant species used, the rate of plantlet formation from callus was very low. However, some plantlet formed from E. crusgalli at 0.8 ppm of 2,4-D plus 8.0 ppm of kinetin, and from E. kuroguwai at 1.6 ppm of 2,4-D plus 16.0 ppm of kinetin, showing effectiveness of 2,4-D with kinetin mixture treatment. No callus was induced from C. serotinus treated with Basta from $10^{-6}M$ to $10^{-3}M$. In general, rice was the least susceptible to Basta among plant species tested, followed by E. crusgalli, and E. kuroguwai. In Londax treatment, rice showed the least inhibition rate in callus growth. Callus was induced from rice even at $10^{-3}M$ of Londax. However, $10^{-3}M$ of Londax completely inhibited callus induction from the test species. Rice showed most tolerant to both herbicides, indicating the existence of different responses among plant species.

Syntaxonomy and Distribution Characteristics of the Herbaceous Vegetation on Running Waterside in the Main Stream of Geumho River, Daegu and Gyeongsanbuk-do, Korea (금호강(대구, 경북)본류에서 유수변초본식생의 군락분류와 그 분포특성)

  • The Herbal Vegetation on Running Waterside in the Main Stream of Genmho River, Daegu and Gyeongsanbuk-do, Korea was investigated by the methodology from June to August of 1994 and analyzed distribution characteristics of vegetation that was related Water Environment. The vegetation is divided into 6 communities of Persicaria sieboldi-Persicaria hydropiper community, Persicaria thunbergii community, Impatiens textori community, Zizania latifoloa community, Phragmites communis community and Echinochloa crus-galli var. oryzicola community. A BOD and COD that indicate degree of contamination in water were showed 0.9 ppm and 1.6 ppm at upstream and 15.9 ppm and 24.8 ppm at downstream, respectively. The distribution patterns of vegetation have dominated by Impatiens textoricommunity in the upstream, by Persicaria sieboldi-Persicaria hydropiper community from upstream to mid-upstream, by Persicaria thunbergii community from upstream to midstream, by Zizania latifoloa community from nidstream to mid-downstream and by Phragmites communis community and Echinochloa crus-galli var. oryzicola community from mid-downstream to downstream. Especially, I suggested that Persicaria sieboldi-Persicaria hydropiper community and Persicaria thunbergii community will use as biotic indicator in water environment.

  • This experiments were conducted to find out the physiological characteristics of lowland weed for physiological control. Temperature, moisture, light, oxygen. carbon dioxide and soil texture effect on germination and sprouting of weed seed. But this research was conducted to know the relationship between oxygen concentration and germination on 7 species weed.

  • This study was conducted to determine the activity of ALS, content of endogenous free amino acids and fatty acids affected by herbicide mixture of cyhalofop-butyl, pyribenzoxim and pyrazosulfuron-ethyl. $I_{50}$ values (concentration required for 50% inhibition of ALS activity) of pyribenzoxim herbicide on the activity of ALS in Echinochloa crus-galli and Cyperus serotinus in vitro were recorded at 4${\times}$100 nM and 5${\times}$10 nM, respectively, while $I_{50}$ values of pyrazosulfuron against E crus-galli and C. serotinus were 4.5${\times}$10 nM and 4${\times}$10 nM, respectively, and the mixture of two herbicides showed additive effect on ALS activity at the low application rate, and independent effect at the high application rates of two herbicides. The inhibition rates of the three herbicides mixture treatment on the three branch-chain amino acids such as valine, leucine and isoleucine were 74.6%, 66.6% and 57.9% in C. serotinus and 36.6% 51.1% and 48.1% in E. crus-galli, respectively. A little bit higher inhibitory effect on the three branch-chain amino acids in C. serotinus and E. crus-galli seedlings was observed in two herbicide mixture of pyribenzoxim with pyrazosulfuron than three herbicide mixture of cyhalofop with pyribenzoxim and pyrazosulfuron. The interaction among three herbicides showed non-antagonism on the amounts of endogenous free amino acids.

A Study on the Cadmium Tolerance of Some Native Herb Plants (수종 야초류의 카드뮴 내성에 대한 연구)

  • This research was conducted to test cadmium tolerance by five speices of native herbs for screening cadmium tolerance plant. We examined germination rate, root and shoot growth rate of seedling, fresh biomass of native herbs exposed to cadmium solution for 14 days. Especially, germination rate of Abutilon avicennae and Amaranthus retroflexus was over 90% in all concentration, but germination rate of Aeschynomene indica, Echinochloa crus-galli var. oryzicola and Echinochla crus-galli var. frumentacea began to decrease when they exposed to 15 mgCdSO₄ /L. The higher the Cd concentration, the length of root, shoot grew lower. The fresh biomass of Echinochloa crus-galli var. oryzicola, Echinochla crus-galli var. frumentacea and Amaranthus retro-flexus were not so different from each control, but those of Abutilon avicennae and Aeschynomene indica began to decrese greatly when exposed to 60 mgCdSO₄ /L. EC/sub 50/(Effective Concentration 50%) of Abutilon avicennae, Aeschynomene indica and Amaranthus retroflexus was 44.6∼77.5 mgCdSO₄ /L for root growth, 38.6∼114 mgCdSO₄ /L for shoot growth, 60∼107.5 mgCdSO₄ /L for fresh biomass. Among the native herbs, sensitivity of cadmium ordered as root length > fresh biomass > shoot length. These results indicate that Abutilon avicennae is a tolerant species for cadmium.

Growth Responses of Rice and Barnyardgrass Varieties to Flooding at Early Growing Period (벼와 피의 침관수에 따른 생장 반응)

  • Barnyard grasses are dominant weed that are not eliminated easily in the direct water seeding cultivation. So, deep water treatment can reduce their growth at the early growing stage of submerged paddy. This study was carried out to investigate the reducing growth of the barnyard grasses through flooding at seedling stages of rice plant in the green house. Under the normal condition, the plant height of rice variety, Samgwangbyeo, and 3 species of barnyard grass, E. caudata, E. pratocola and E. utilis were not showed the difference up to 10 days after seeding while the plant height of 3 barnyard grass species, especially E. utilis, was more elongated than the rice at 25 days after seeding. Plant height of the 3 barnyard grasses were not elongated largely during 3, 5 and 7 days of flooding treatment at 10 days after seeding. Interestingly, the rice seedlings was grown over 20 cm, and the flooding tolerance of rice seedling was higher than the 3 barnyard grass varieties. However, after flooding treatment for 3, 5 and 7 days, the elongation of plant height of 3 barnyard grasses, especially E. utilis was more speedy compared to rice seedlings as 6 to 9 days passes. And the protein spots from barnyard grasses were also reduced and eliminated more than the spots of rice seedling after flooding treatment.

Response of domestically collected Echinochloa species to cyhalofop-butyl and pyribenzoxim herbicides, their absorption and translocation (국내 피 수집종에 대한 cyhalofop-butyl과 pyribenzoxim의 약제반응 및 흡수이행)

  • To investigate the physiological and biochemical differences between susceptible and tolerant species of Echinochloa species to the foliar applied cyhalofop-butyl and pyribenzoxim herbicides, herbicidal response, the rate of absorption, translocation and metabolism were studied. Among 148 individuals of Echinochloa species collected from 41 sampling sites in Korea which were classified into 7 geographical regions, based on morphological characteristics of plant and seed type, 46 lines were used for further studies. From them, E. crus-galli var. praticola collected from Hwanggan and E. crus-galli var. crus-galli collected from Namyangju were selected as the most susceptible species to cyhalofop-butyl and pyribenzoxim, respectively. Meanwhile, E. oryzicola(from Cheju) and E. crus-galli var. crus-galli(from Asan) were selected as tolerant species to cyhalofop-butyl and pyribenzoxim, respectively. Application of radio-labelled herbicides on the 1st leaf to both susceptible and tolerant Echinochloa species exhibited that the applied $^{14}C$-Cyhalofop-butyl and $^{14}C$-pyribenzoxim were more easily absorbed and translocated into stem tissues than root. Absorption of $^{14}C$-Cyhalofop-butyl increased rapidly at 1 h after application and reached the maximum at 12 h after application. However, the absorption rate of $^{14}C$-pyribenzoxim was not changed. Two metabolites from cyhalofop-butyl-treated plants and one metabolite from pyribenzoxim-treated plants were separated by TLC. The amount of metabolite 1 in cyhalofop-butyl-treated tolerant species was significantly higher than that in the susceptible one, suggesting differences in detoxification ability between susceptible and tolerant species.
