Identification of host plant species of Balanophora fungosa var. indica from Phnom Bokor National Park of Cambodia using DNA barcoding technique (캄보디아 프놈보콜국립공원의 Balanophora fungosa var. indica의 숙주식물에 대한 DNA barcoding 기법을 통한 동정)

  • During the floristic survey on Phnom Bokor National Park, Kampot, Cambodia, we encountered Balanophora fungosa var. indica, which is a tropical holoparasitic plant. To identify its host species, we collected host roots and trees nearby and tried to identify them using DNA barcoding approach. We applied plastid rbcL and matK gene regions as DNA barcode markers, and successfully amplified and sequenced the markers from 15 host roots and seven tree samples. Obtained host root sequences were identified as Primulaceae, Celastraceae, Myrtaceae, and Oleaceae, while trees nearby are Oleaceae, Myrtaceae, Sapindaceae, Rosaceae, Clusiaceae, Ericaceae, and Lauraceae. At genus level, host species are identified as Myrsine, Euonymus, Syzygium, and Olea, but failed in species discrimination. Myrsine (Primulaceae) and Olea (Oleaceae) are reported here as host species of B. fungosa var. indica for the first time. Further sampling and comparative work, and DNA barcoding will help recognize the biodiversity of the area and host species of Balanophora, together with their evolution.

Jasminum attenuatum Roxb. ex G. Don (Oleaceae): a new record to the flora of Vietnam (Jasminum attenuatum Roxb. ex G. Don: 베트남 미기록종)

  • Jasminum attenuatum Roxb. ex G. Don, a species of Oleaceae, was recently found in Kon Ka Kinh National Park of Vietnam that represents a new record to the flora of the country. The plant can be differentiated from its allied species J. latipetalum C.B. Clarke and J. simplicifolium subsp. funale (Decne) Kiew by the shape of its leaf blade, in having 5-20 flowered inflorescence, linear bracts, and obconic and glabrous calyx with shorter lobes. The present study provides the species description, illustrations, micrographs by scanning electron microscope of the pollen grains, and important taxonomic notes. Furthermore, a comparison of the diagnostic characters between this species and the above mentioned related species is made.

  • This paper explores the types (species of trees) of 25 pieces of waterlogged wood excavated from the area between Gwangju and Jangseong during road expansion by the Gwangju National Museum. These 25 pieces of wood include nine pieces of Quercus (Lepidobalanus Cerris)sp., six pieces of Quercus (Lepidobalanus Prinus)sp., three pieces of Castanea sp., two pieces of Salix sp., one piece of Alnus sp., one piece of Prunus sp., one piece of Morus sp., one piece of Chionanthus sp., and one piece of Acanthopanax sp.

이팝나무의 항산화 및 항균활성

  • 이팝나무(Chionanthus retusus)는 물푸레나무과에 속하는 낙엽교목으로 육도목, 유소수라고도하며 중국이나 일본에서는 잎을 차대용으로 사용한다고 하여 다엽수라고도 부른다. 지사, 건위, 중풍 등의 치료에 효과적이고, 강장제, 해열제, 이뇨제 등으로 사용되었다고 알려져 있는 이팝나무 잎의 생리활성을 검색하여 유용자원으로 활용하기 위한 기초 연구로 메탄을 추출물을 각종 용매를 이용하여 분획한 후 DPPH를 이용한 항산화 활성, 아질산염 소거 및 항균 활성 등의 생리활성에 대한 실험 결과는 다음과 같다. DPPH에 의한 항산화 활성을 측정한 결과 에틸 아세테이트 분획물과 부탄올 분획물에서 가장 높은 활성을 나타내었으며, reducing power의 경우 에틸 아세테이트 분획물에서 그리고 농도가 증가함에 따라 환원력이 증가하는 것으로 나타났다. 아질산염 소거능은 pH가 낮을수록, 농도 의존적으로 그 활성이 높게 나타났다. 항균활성은 에틸 아세테이트 분획물에 대하여 특히, Escherichia coli와 Bacillus cereus 균주에 대해 각각 16 및 18mm 로 높은 항균력을 나타내었다.

Comparative anatomy of petiole in Forsythieae, Fontanesieae and Myxopyrum (Oleaceae) and its systematic implication (개나리족, 향선나무족, Myxopyrum속(물푸레나무과) 엽병의 해부학적 형질 및 분류학적 유용성)

  • A comparative study of the petiole anatomy in the tribes Forsythieae (Abeliophyllum, Forsythia) and Fontanesieae (Fontanesia), including one related genus Myxopyrum belonging to Myxopyreae, was carried out using light microscopy. The anatomical characteristics of the distal, medial and proximal parts of the petiole were studied to document any differences. We are described in detail based on their quantitative and qualitative characteristics. Three crystal types (acicular, druse, and prismatic crystals) were found in both Fontanesia and Myxopyrum within all of the studied taxa. Uni-cellular non-glandular trichomes were found in Abeliophyllum and three Forsythia taxa (F. europaea, F. giraldiana, and F. japonica). All features were compared and the vascular patterns of the petiole were distinguished in two types: Type 1A: Trace continuous arc - without crystals (Abeliophyllum, Forsythia), 1B: with crystals (Myxopyrum), Type 2: Trace invaginating at ends with crystals (Fontanesia). A detailed anatomical description of the studied taxa is provided, and its systematic importance is also briefly discussed. In conclusion, some petiole anatomical characteristics (e.g., the main vascular patterns, the presence/absence of crystals) can be useful for diagnostic features as well as partly for supporting the recently proposed molecular phylogeny.

Genetic diversity of Forsythia ovata Nakai (Oleaceae) based on inter-simple sequence repeats (ISSR) (ISSR 자료에 기초한 만리화(물푸레나무과)의 유전적 다양성)

  • We investigated the genetic diversity of an endemic rare species, Forsythia ovata Nakai by examining 93 ISSR amplicons in 84 individuals distributed among five populations. The overall percentage of polymorphic ISSR amplicons was 54.8% and mean number of amplicons per ISSR primer was 6.6. The amount of genetic diversity was relatively lower than other shrub species. The Mt. Seokbyeong and Mt. Seorak B populations had the highest level of genetic diversity. Although the Seokgae-jae population had the lowest level of genetic diversity, the population was genetically the most distinctive from the other populations. About 30.6% of the total variation was allocated between five populations, which was slightly higher than other shrub species. Such a pattern of genetic variation may have resulted from the limited distribution and small population sizes of F. ovata. The UPGMA dendrogram based on Nei's genetic distance showed some decisive geographic patterns. These results suggest that, in addition to the preservation of the natural stands, the conservation of larger number of populations with small number of individuals per population is more effective for the dynamic ex situ conservation and for maintaining the genetic diversity of F. ovata than smaller number of populations with large number of individuals.

The systematic consideration of leaf epidermal microstructure in the tribe Forsythieae and its related genera (Oleaceae) (개나리족 및 근연분류군(물푸레나무과)의 잎 표피 미세구조에 대한 분류학적 검토)

  • A comparative study of leaf epidermal microstructure in tribes Forsythieae (Abeliophyllum: 1 species, Forsythia: 12 taxa - 10 species, 2 varieties), Fontanesieae (Fontanesia: 2 species) including one related genus Myxopyrum belonging to Myxopyreae (Myxopyrum: 5 taxa - 4 species, 1 subspecies) was carried out using scanning electron microscopy (SEM) in order to evaluate their significance in the taxonomy. The leaves of investigated taxa are either hypostomatic or amphistomatic, but former is more frequent. The size range of the guard cells is 17.14-47.58 ${\times}$ 11.59- 44.25 ${\mu}m$: the smallest one was found in Forsythia giraldiana (17.48-22.96 ${\times}$ 11.64-12.88 ${\mu}m$), while the largest one was measured to Myxopyrum pierrei (31.50-41.75 ${\times}$ 32.53-44.25 ${\mu}m$). Anomocytic stomatal complex are most frequent type (rarely paracytic), usually both anomocytic and anisocytic occur in one leaf. In surface view both adaxial and abaxial anticlinal walls of the subsidiary cells are variable (e.g., straight/curved, undulate, sinuate, undulate/sinuate). Three types (simple unicellular and multicellular non-glandular, subsessile glandular) of trichomes are found in leaves. Finally, the systematic significance of the leaf epidermal micromorphological characters in idenfitication and elucidation of these tribe, between or within the genera including among the species is also briefly discussed.

Jasminum albicalyx Kobuski (Oleaceae): a new record from Vietnam (Jasminum albicalyx Kobuski: 베트남 미기록종)

  • We report a new record of Jasminum albicalyx Kobuski in Vietnam. The plant differs from J. pedunculatum Gagnep., a closely related taxon, by foliar and floral characters such as less number of lateral veins, presence of domatia at vein axils on the abaxial surface of leaf, smaller bracts, white calyx having more number of lobes and less number of lobes in corolla. Morphological description, line drawing and color photographs are provided in addition to the species key of allied species of Jasminum albicalyx in Vietnam.
