Restoration and Stability of the Glass Sarira Bottle (Treasure No. 1925) from the Sarira Reliquaries Commissioned by Yi Seonggye, Excavated from Geumgangsan Mountain (보물 제1925호 금강산 출토 이성계 발원 사리장 엄구 내 유리제 사리병의 복원 및 안정성 연구)

  • 3D printing technology has been actively applied for the restoration of cultural properties. However, its application to the restoration of glass cultural properties has not yet been reported and thus requires further study. In this study, 3D printing technology was used to restore a defective part of a glass sarira bottle that forms an element of a series of sarira reliquaries commissioned by Yi Seonggye (known as King Taejo after founding the Joseon Dynasty) that was excavated from Geumgangsan Mountain (designated as Treasure No. 1925) and is currently housed at the National Museum of Korea. The defective area was reproduced using 3D printing and the printed reproduction was reproduced again using an epoxy resin. This latter piece was used as the restoration component rather than the 3D printed element. After the completion of the conservation treatment, the materials used for the 3D printing were compared with transparent materials used to restore ceramics to evaluate their usability and stability. A total of five specimens were produced, including from photocurable resin made by a stereo lithography apparatus (SLA), epoxy resin, acrylic resin, and more. They were exposed to UV for 96 hours to test for yellowing. Of the two specimens made of photocurable resins and exposed to UV, one was sprayed with a UV blocking agent but the other was exposed as-is. The UV exposure test showed that the specimen made by the SLA and sprayed with a UV blocking agent and the specimen made of epoxy resin were stable in terms of yellowing with a change in the b-value was less than 1. They are thus considered to be suitable materials for the restoration of glass cultural properties. Such glass cultural properties are often diverse in shape and their restoration can be difficult as they generally consist of a range of complex parts that hamper restoration. In this regard, diverse materials should be considered when selecting materials for the restoration of glass cultural properties.

Study on Characteristic of CO2 Hydrate Formation Using Micro-sized Ice (미세직경 얼음을 이용한 CO2 하이드레이트 제조특성 연구)

  • Gas hydrate is an inclusion compound consisting of water and low molecular weight gases, which are incorporated into the lattice structure of water. Owing to its promising aspect to application technologies, gas hydrate has been widely studied recently, especially $CO_2$ hydrate for the CCS (Carbon Capture and Storage) issue. The key point of $CO_2$ hydrate technology for the CCS is how to produce gas hydrate in an efficient and economic way. In this study, we have tried to study the characteristic of gas hydrate formation using micro-sized ice through an ultrasonic nozzle which generate 2.4 MHz frequency wave. $CO_2$ as a carrier gas brings micro-sized mist into low-temperature reactor, where the mist and carrier gas forms $CO_2$ hydrate under $-55^{\circ}C$ and atmospheric pressure condition and some part of the mist also remains unreacted micro-sized ice. Formed gas hydrate was average 10.7 of diameter at average. The starting ice particle was set to constant pressure to form $CO_2$ hydrate and the consumed amount of $CO_2$ gas was simultaneously measured to calculate the conversion of ice into gas hydrate. Results showed that the gas hydrate formation was highly suitable because of its extremely high gas-solid contact area, and the formation rate was also very high. Self-preservation effect of $CO_2$ hydrate was confirmed by the measurement of $CO_2$ hydrate powder at normal and at pressed state, which resulted that this kind of gas storage and transport could be feasible using $CO_2$ hydrate formation.

Evaluations of Corrosion Resistance of Coated Steel Using Polymer Cement Slurry (폴리머 시멘트 슬러리로 코팅한 도장철근의 내식성 평가)

  • Reinforced concrete structures under sever conditions such as marine structures, bridges and structures constructed with aggregates(dredged from sea), can be deteriorated from corrosion of the reinforcing bars. The purpose of this study is to evaluate the anti-corrosive performance of coated steel using polymer cement slurry. Polymer cement slurry with various polymer dispersions and corrosion inhibiting agent were coated to the surface of bars, and tested for accelerated corrosion tests. Tests include immersion in NaCl 10% solution, chloride ion spray, autoclave cure, autoclave cure after carbonation, penetration of NaCl 10 % solution, carbonation after penetration of NaCl 10% solution. Test results, show that the anti-corrosive performace is considerably improved by using polymer cement slurry at surface of steel. And this trend is marked by adding of corrosion inhibiting agent. This difference of the anti-corrosive properties is hardly recognized according to types of polymer dispersions. The coated steel using polymer cement slurry will be improved to a great extent compared to those of plain steel when increasing content of chloride ion in cement concrete.

  • This study was carried out with sardine to develope a new type of fish protein concentrate. Chopped sardine meat was thermally treated in two different ways, autoclaved at $121^{\circ}C$ for 1 min and boiled at $95^{\circ}C$ for 5 min. The heat treated meat was pressed, controlled to PH 7.8 with $3\%$ (w/v) of $NaHCO_3$ and hot-air dried(at $40^{\circ}C$). The dried meat was powdered (50mesh), air and vacuum packed in laminated film bag(PET/AL. foil/CCP) and stored at room temperature for 60 days. The results of product quality analysis are as follows : 1. Proximate contents of moisture, crude lipid and protein of the autoclaved and boiled product were in the range of $10.0{\~}10.2\%,\;9.0{\~}9.1\%$ and $73.8{\~}74.4\%$, respectively. Yields of the both products were $40\%$ and $32.5\%$. 2. Values of emulsion activity, emulsion stability and foam expansion of the autoclaved product were $48.7\%$, $44.1\%\;and\;44.0\%$, respectively. These values were higher than those of boiled product. 3. Water holding capacity and digestibility of the both products were in the range of $5.0{\~}5.3\%$ and $78.0{\~}78.2\%$, respectively.

Buffer Zones for Non-Target Organisms by Aerial Pesticide Application Around Rice Paddy (벼 재배지의 항공 방제시 비표적 생물에 대한 안전거리)

  • The study was carried out to establish buffer zone for the protection of the non-target organisms by aerial pesticide application. The two pesticide combination of 3-way tank-mixing of three pesticides for aerial application, ferimzone tricyclazole SC + BPMC EC + validamycin-A SL and hexaconzole EC + isoprothiolane EC+phenthoate EC were selected for the simultaneous control of key pests on paddy rice as blast, sheath blight, brown planthopper and moth. Aquatic organisms including killifish and loach in the paddy field and nearby water reservoirs were not affected by aerial application of the pesticides. However, all the water flea were killed, when they were exposed 10 m from the aerially sprayed site, while the water flea exposed in 30 m away from the site were not affected. Honeybees within 50 m in the wind direction and 20 m in the opposite wind direction showed a mortality of $7{\sim}100%$. Residues concentration of the pesticides in paddy water were not detectable level after six days from aerial application. Drifting distance of aerially sprayed droplet from the target area was within 30 m in the wind direction and 20 m in the opposite direction. Consequently, it was the buffer zones in the aerial pesticides application for the protection of the non-target organisms should be at least 50 m for aquatic organisms and 100 m for honeybees.

Effect of Cultural Practices on the Occurrence of Pod and Stem Blight and Purple Blotch, and on Soybean Growth (콩 재배방법이 미이라병과 자주무늬병의 발생 및 식물생육에 미치는 영향)

  • Deterioration of pod and seed quality by pod and stem blight and purple blotch is a serious problem in the production of vegetable soybean. Major inoculum sources for the causal pathogens, Phomopsis spp. and Cercospora kikuchii, are soybean straw and debris. Phomopsis spp. have been known to be either limited for hyphal growth or latent in immature soybean tissues. In this experiment, cultural practices using these ecological traits of the pathogens were applied as a control measure. In plastic mulching, seed infection was remark ably reduced by drip irrigation as compared to overhead sprinkling, but not reduced in no mulching cultivation. Control value of plastic mulching was 28.0% for pod and stem blight and 29.3% for purple blotch, which was lower than that of benomyl application. By the first week in June, 78% of overwintered soybean straw examined contained matured alpha spores in pycnidia, acting as primary inoculum. Secondary inoculum of phomopsix spp. was observed abundantly throughout from mid June to September on fallen cotyledons from current crops and subsequently was found on petiole debris. Consequently, both plastic mulching and drip irrigation were effective to some extent for controlling pod and stem blight and purple blotch without significant reduction in soybean yield, and the cultural practices in combination with field sanitation resulted in removing more the secondary inoculum.

Free Radical Scavenging Activity and Ascorbate Content in Various Plant Cell Lines (다양한 식물배양세포주에서 자유라디칼 포착활성과 Ascorbate 함량)

  • We investigated the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in methanol extracts of 64 cultured cell lines, which were derived from various plant species, and the ascorbate content in cell lines, which showed a high radical scavenging activity. Thirteen cell lines revealed the antioxidative activity ($IC_{50}$) by methanol extracts of less than 50 mg in cell fresh wt. Of them, six cell lines showed the same Rf value as ascorbate on the DPPH sprayed silica gel TLC. The ascorbate content in cell lines of Rosa multiflora, Scutellaria baicalensis, and Achyranthes japonica showed 48.5, 30.3, and $16.8\;\mu\textrm{g}$ per g cell fresh wt by HPLC analysis, respectively. In callus cultures of S. baicalensis, the concentration of ascorbate reached a maximun ($39{\pm}3.4\;\mu\textrm{g}/g$ cell fresh wt) on 30 days after subculture, which corresponded to the stationary growth phase, and subsequently decreased by successive culturing.

Qunatitative analysis of liquiritin and glycyrrhizin in glycyrrhizae radix by HPLC-MS/MS (HPLC-MS/MS에 의한 감초의 liquiritin과 glycyrrhizin의 분석)

  • Licorice, Glycyrrhizae Radix is widely used as a herbal medicines and a dietary supplements in East Asia. We employed high performance liquid chromatography electrospray ionization tandem mass spectrometry to determine liquiritin and glycyrrhizin in the Glycyrrhizae Radix. Liquiritin and glycyrrhizin in Glycyrrhizae Radix were ionized by positive ion pneumatically assisted electrospray and detected by HPLC-MS/MS in the multiple-reaction monitoring (MRM) mode using precursor ${\rightarrow}$ product ion combinations at m/z $436.2{\rightarrow}257.0$ and $823.4{\rightarrow}453.4$, respectively. The assay had a calibration range from 10 to 3,000 ng/mL. The limits of detection (LOD) of the liquiritin and glycyrrhizin were 0.4 ng/mL and 0.01 ng/mL, respectively. The reproducibility and repeatability (relative standard deviation) at different analyte concentrations varied from 103 to 113 % and 0.95 to 1.8 %, respectively. According to the above results, HPLC-MS/MS method permits assignment of tentative structures such as liquiritin and glycyrrhizin in the Glycyrrhizae Radix.

Survival and Growth Characteristics of Foodborne Pathogen in Romaine Lettuce (로메인 상추에서 병원성미생물의 생존 및 증식 특성)

  • The purpose of this study was to provide safety management information by analyzing the survival and growth-related properties of foodborne pathogens from Romaine lettuce. After cultivating E. coli O157:H7 for 72 h on Romain lettuce via spray inoculation, the bacteria population increased by 2.0 log CFU/g from the initial population, confirming the possibility of survival and multiplication of the pathogen thereon. The study also revealed that there is no significant difference in the cultivation of E.coli O157:H7 after 72 h from inoculation on damaged and undamaged lettuce leaves. As a result of investigating distribution of E.coli O157:H7 on damaged lettuce leaves, it was found that the bacteria is unlikely to adhere on the smooth surface of undamaged leaves and, thus, results in a low population density, whereas the bacteria cluster on the rough surface of damaged leaves and easily enter through the damaged tissues. Furthermore, after 24 h of cultivation of the pathogenic microbe in the extract with concentrations of 10-100%, utilization of the lettuce extract by the pathogen was found to be 8.9 log CFU/mL E. coli O157:H7, 8.6 log CFU/mL L. monocytogenes, and 8.8 log CFU/mL P. carotovorum. The increase in the population of both the pathogenic microbe and foodborne pathogen reached over 4 log CFU/mL, implying the microbe can utilize the lettuce extract as a source of nutrition. Compared to the initial inoculation concentration in 0.1% lettuce extract, the final concentration has increased up to 2.7 log CFU/mL E. coli O157:H7, 1.3 log CFU/mL L. monocytogenes, and 2.9 log CFU/mL P. carotovorum. Accordingly, the study confirms that the minimal growth concentration of the pathogenic microbe is lower than 0.1% and that the pathogen possibly survive and multiply inside the lettuce leaves given the lettuce extract with concentration of 0.1% is consistently supplied through the damaged tissues.

Deterioration Diagnosis and Source Area of Rock Properties at the West Stone Pagoda, Gameunsaji Temple Site, Korea (감은사지 서탑의 풍화훼손도 진단 및 석재의 산지추정)

  • The rock properties of the West pagoda in the Gameunsaji temple site are composed mainly of dark grey porphyritic granodiorite with medium grained equigranular texture and developed with small numerous dioritic xenoliths. These xenoliths occurred with small holes due to different weathering processes. As a weathering results, the rock properties of this pagoda occur wholly softened to physical hardness because of a complex result of petrological, meteorological and biological causes. Southeastern part of the pagoda deteriorated seriously that the surface of rock blocks showed partially exfoliations, fractures, open cavities in course of granular decomposition of minerals, sea water spray and crystallization of salt from the eastern coast. The Joint between blocks has small or large fracture cross each other, contaminated and corrupted for inserting with concrete, cement mortar, rock fragments and iron plates, and partially accelerated coloration and fractures. There are serious contamination materials of algae, fungus, lichen and bryophytes on the margin and the surface on the roof stone of the pagoda, so it'll require conservation treatment biochemically for releasing vegetation inhabiting on the surface and the discontinuous plane of the blocks because of adding the weathering activity of stones and growing weeds naturally by soil processing on the fissure zone. Consisting rock for the conservation and restoration of the pagoda would be careful choice of new rock properties and epoxy to reinforce for the deterioration surfaces. For the attenuation of secondary contamination and surface humidity, the possible conservation treatments are needed.
