The crude polysaccharide fraction from fruit body of Auricularia auricula were obtained by using hot water extraction and ethanol precipitation. As the crude polysaccharide fraction contained the brownish dark colored compounds, the adsorption study of pigments from the crude polysaccharide using activated carbon was carried out. The pigment compounds showed an absorption characteristic with $\lambda_{max}$ of 230 nm and the absorbance at 230 nm was taken as color intensity. Adsorption capacity of pigment depended on increase of the activated carbon to sample loading ratio. The adsorption capacity increased with increase of pH and temperature in the pH range of 3.0-7.0 and temperature range of 25-40$^{\circ}C$, but decreased in the temperature range of 40-70$^{\circ}C$. The optimum capacity was obtained at addition of 16.7 mg activated carbon per mL sample solution (concentration = 3 mg/mL) at pH of 7.0 and temperature of 40$^{\circ}C$. Treatment for 10 min was sufficient to achieve the 80% decolorization and 1.25 fold purification of polysaccharide. Langmuir isotherm and pseudo second-order kinetic model provided the best fitting for adsorption of the brownish dark colored compounds onto powdered active carbon. The activation energies of adsorption from the Langmuir isotherm parameter in the ranges of 25-40$^{\circ}C$ and 40-70$^{\circ}C$ was -2.54 and 4.38 kcal/g, respectively. The results of low activation energy also indicated that the adsorption process was a physical adsorption which was controlled by diffnsion.

The optimization of submerged culture conditions for mycelial growth and exopolysaccharide (EPS) production in an edible mushroom Tremella fuciformis were studied in shake flasks and bioreactors. The temperature of $28^{\circ}C$ and pH 8 in the beginning of fermentation in agitated flasks was the most efficient condition to obtain maximum mycelial biomass and EPS. The optimal medium constituents were as follows (g l-1): glucose 20, tryptone 2, $KH_2PO_4$ 0.46, $K_2HPO_4$ 1 and $MgSO_4H_2O$ 0.5. The fungus was cultivated under various agitation and aeration conditions in a 5L stirred-tank bioreactor. The maximum cell mass and EPS production were obtained at a relatively high agitation speed of 200 rpm and at an aeration rate of 2 vvm. The flow behavior of the fermentation broth was Newtonian and the maximum apparent viscosity (35 cP) was observed at a highly aerated condition (2 vvm). The EPS productivity in an airlift reactor was higher than that in the stirred-tank reactor. The EPS was protein-bound polysaccharides consisted of mainly mannose, xylose, and fructose. The molecular weights of EPS were determined to be $1.3{\sim}1.5{\times}10^6$.
To minimize cultivation costs, prevent insect-pest infestation, and improve the production efficiency of thermophilic mushrooms, plant substrates obtained from local areas in Cambodia were used for production of both spawn and mushrooms. In this experiment, different sawdusts different organic wastes and grain ingredients and analyzed for improvement of spawn-production efficiency. Four thermophilic mushroom species, Pleurotus sajor-caju (oyster mushroom, Sambok), Ganoderma lucidum (deer horn shaped), Auricularia auricula (ear mushroom), and Lentinula edodes (shiitake), were used to identify efficient new substrates for spawn and mushroom production. Although the mycelia in the rubber tree sawdust medium showed a slightly slower growth rate (10.9 cm/15 days) than mycelia grown in grains (11.2 cm/15 days in rice seeds), rubber tree sawdust appeared to be an adequate replacement for grain spawn substrates. Th findings indicate that rubber tree sawdust, sugarcane bagasse, and acacia tree sawdust supplemented with rice bran and calcium carbonate could be new alternative the substrates for. Although sugarcane bagasse and rubber tree sawdust showed similarly high biological efficiencies (BE) of 60% and 60.8%, respectively, acacia tree sawdust exhibited relatively a low biological efficiency of 22.4%. However, it is expected that acacia sawdust has potential for the mushroom cultivation when supplemented with currently used sawdust substrates in Cambodia, because of its relatively low price. The price of the sawdust (20 kg sawdust= 6500 Riel or 1.6 USD) currently used was 6.5 times higher than the price of acacia sawdust (201000 Riel or 0.25 USD). Therefore, utilization for acacia sawdust for mushroom cultivation could become feasible as it would reduce by producing costs of mushrooms in rural areas of Cambodia.

For the investigation of the effects of mushrooms on prevention of hypercholesterolemia, dietary hyperc-holesterolemic rats were fed for 2 weeks with basal diet containing 5% G. lucidum, 5% L. edodes, 5% A. judae and 10% G. lucidum, 10% L. ededos or 10% A. judae mushroom. Concentrations of total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, phospholipid, free-cholesterol, cholesteryl ester, VLDL and chylomicron in serum were analyzed. The result obtained are as follows : Concentrations of total cholesterol, LDL, LDL-cholesterol in serum were lowest in the group 6 (10% L. edodes mushroom) and HDL-cholesterol concentration in serum was significantly higher than the concentrations of the control group. Concentrations of triglyceride, phospholipid, free-cholesterol, cholesteryl ester in serum were lower than the concentration of the control group, and the concentrations of triglyceride, cholesteryl ester in group 6 were lower than those of the other groups. Concentrations of phospholipid and free-cholesterol in group 3 were lower than those of the other groups.
To investigate antitumor components of Korean higher fungi, the carpophores of Schizophyllum commune and Auricularia auricula-judae collected in Kyeong Buk Province were extracted with hot water or 0.1N-NaOH solution. The concentrated extracts were precipitated by addition of ethanol, and the precipitates were purified by dialyzing through visking tube and polysaccharide fractions were obtained. They were found to show antitumor activity against sarcoma 180 implanted in mice. Especially, the inhibition ratio of the extract of Auricularia adicula-judae was 90.8% in the doses of 100mg/kg/day for the period of ten days. The tumor in five of the eight mice was completely regressed. The components of these aqueous extracts were found to be polysaccharide and protein. The hydrolysis of the respective polysaccharide yielded four monosaccharides. After hydrolysis of the protein fraction, 15 amino acids were identified in the respective fraction of S. commune and A. auricula-judae.
Effects of cultural conditions on the production of xylanase by Tremella fuciformis, symbiotic fungi and mixed fungi were investigated. The optimum carbon source for high production of xylanase by T. fuciformis, symbiotic fungi and mixed fungi was xylose. The optimum nitrogen source for both T. fuciformis and symbiotic fungi was $KNO_3$, whereas mixed fungi was $(NH_4)_2SO_4$. The optimum culture period for high production of xylanase was 5 days for both T. fuciformis and mixed fungi, and 6 days for symbiotic fungi, respectively. The optimum temperature for T. fuciformis and symbiotic fungi was $40^{\circ}C$, and the corresponding value for mixed fungi was $45^{\circ}C$. Xylanase activity was high at pH 6 for T. fuciformis and symbiotic fungi, and pH 7 for mixed fungi. Except $Hg^{2+}$ and $Pb^{2+}$, metal ions in T. fuciformis inhibited the activity of xylanase, and, thermal stability of xylanase in T. fuciformis, symbiotic fungi and mixed fungi maintained 80% of activity until $50^{\circ}C$. The Michaelis constant (Km) of xylan was $6.25{\times}10^{-5}\;M$ in T. fuciformis, $5.6{\times}10^{-2}\;M$ in symbiotic fungi, $5.2{\times}10^{-2}\;M$ in mixed fungi.

'Yong-A' was bred by Jeollanamdo Agricultural Research and Extension Services in 2015. It was primarily selected through monospore crossing between JNM21008 and JNM21013 in 2009. The strain is named as JNM-Mi-194 and 'Yong-A', showing major characteristics with good production yield. The cultivation characteristics, yield potential, and fruiting body characteristics of 'Yong-A' are as follows; The wrinkle of the fruiting body was of branching type its shape was wave type. MCM, YM, Malt, and PDA media were suitable for growth of this cultivar. The number of effective stipes was 13 ea/0.9 kg. The minor axis of pileus was 5 cm and the major axis was 9 cm. The yield was 291 g per plastic bag (0.9 kg). Yong-A required 60 days for mycelial running at $20^{\circ}C$. The growth and primordial period required 24 days, which was shorter than that of JNM-Mi-21002 (the conventional cultivar). Somatic incompatibility was observed between parental lines and Yong-A. Analysis of genetic diversity in the new variety 'Yong-A' showed a different profile compared to that of the parent strain when Random Amplified Polymorphic DNA primers were used.
