SCIENCE - Overview of the Immune System (학술 4 - 면역기구(免疫機構)의 재음미(再吟味))

  • 2011년도 Nobel 생리(生理) 의학상(醫學賞) : 자연(自然)(선천)(先天) 면역(免疫)(innate immunity)의 활성화에 관한 연구업적으로 B. A. Beutler와 J. A. Hoffmann, 그리고 수지상세포(樹枝狀細胞)(dendritic cell; DC)발견과 적응(適應)(획득)(獲得)면역(免疫)(adaptive immunity)에 있어서의 그들 세포의 역할을 밝혀낸 R. M. Steinman의 공동수상으로 금년도 Nobel 생리 의학상 수상자가 결정되었다는 보도가 지난 10월 3일 있었다(1-3). 그들의 업적을 요약하면 대략 다음과 같다. (Steinman교수는 Nobel수상자 발표 3일전인 9월 30일 암으로 사망함). 그들은 면역기구(immune system)의 활성화의 관건(key)이 되는 원리를 밝혀냄으로써, 면역기구에 관한 우리들의 이해를 혁신하였던 것이다. 과학자들은 오랫동안 세균(bacteria)이나 기타 미생물병원체들에 의한 공격에 대비하여 그들 자신을 방어하는 사람이나 기타 동물체에서의 면역응답(免疫應答)(immune response)의 문지기들을 탐색해 왔다. Beutler와 Hoffmann은 그와 같은 병원미생물을 인식하여 생체의 면역응답의 첫 단계인 자연면역을 활성화 할 수 있는 수용체 단백질(toll-like receptor protein)을 규명한 것이다(4,5). 한편 Steinmann은 면역계의 수지상세포(DC)와 병원미생물이 생체로부터 배제되는 면역응답의 후기단계인 적응면역을 활성화하고 조절하는 그들의 독특한 재능을 규명해 낸 것이다(6-8). 그들 3명의 발명은, 면역응답의 자연 및 적응 양상(樣相)이 어떻게 활성화되는 가를 밝혀냄으로써 질병의 기전에 관한 참신한 식견(識見)을 제공한 것이다. 그들의 연구는 감염병(感染病)(infectious disease), 암(癌)(cancer) 그리고 염증성질환(炎症性疾患)(inflammatory disease)에 대응하는 예방과 치료의 개발을 위한 새로운 방법을 개척한 것이다.

Efficacy Study of Activation on Macrophage in Germanium-fortified Yeast (게르마늄 강화 효모의 대식 세포 활성화 효과에 관한 연구)

  • The aim of this study was to evaluate an efficacy about activation on macrophage, using model that measured cell viability, nitric oxide (NO), iNOS (inducible nitric oxide synthase) expression and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) on Raw 264.7 cells following treatment of Germanium-fortified Yeast in 0, 5, 10, 25, 50, 100, $200\;{\mu}g/ml$ and the same concentration of dried yeast without germanium. Cell viability (%) and NO produced in activated-macrophage were dose-dependant, a significant increase of the cell viability (132.5%) and NO in $10\;{\mu}g/ml$ (p < 0.05). Increase in iNOS level was in $10\;{\mu}g/ml$. $TNF-{\alpha}$ was produced dose-dependant, e.g. in activated-macrophage with a significant increase of the $TNF-{\alpha}$ in 5 and $10\;{\mu}g/ml$ (p < 0.05). Therefore, Germanium-fortified Yeast had an efficacy of NO mediated iNOS and $TNF-{\alpha}$ production by activated macrophage. This result showed that Germanium-fortified Yeast induced activation of cellular immunity, returned to normalcy on injured immune system and procured anticancer system by activation of macrophage, which was important in immune and anticancer function.

Induction of Macrophage Activation of Paeonia lactiflora according to Extraction Conditions (추출조건에 따른 작약의 대식세포 활성화 유도)

  • 작약은 염증성 질환을 치료하기 위해 사용되어 온 전통 약용식물이다. 최근 작약은 대식세포에서 면역조절인자의 분비를 증가시키고 포식작용을 증가시킨다고 보고되었다. 그리하여 본 연구에서 추출조건별 작약의 대식세포 활성화 유도를 비교하였다. 온도조건 별 작약추출물은 4℃에서 60℃까지는 면역조절인자의 분비를 증가시켰지만, 80℃에서는 면역조절인자의 분비가 다소 감소하였다. 60℃에서 시간별 추출조건에서는 1시간에 24시간까지 면역조절인자의 분비가 유사하였다. 따라서 본 연구결과를 종합해 볼 때, 작약은 60℃에서 1시간 추출하는 것이 대식세포 활성화를 위한 최적 조건이라고 판단된다.

타고난 면역이 활성화한 육계병아리의 혈액 항산화계 균형과 TNF-$\alpha$ 농도에 미치는 콩 추출물 함유 미역제품 사료의 영향

  • Effect of dietary 2.0 % brown seaweed(Undaria pinnatifida) with bean extract on anti-oxidant system and TNF-$\alpha$ levels were evaluated in blood of 2 week-old broiler chicks activated innate immune response. Dietary brown seaweed and activation of innate immune response decreased MnSOD activities. while activation of innate immune reponse only increased CuZnSOD activities in erythrocyte cytosol. Activation of innate immune response lowered plasma SOD activity in birds fed seaweed with bean extract, increased peroxide levels, and decreased peroxidase activity in plasma. Brown seaweed with bean extract reduced TNF-$\alpha$ levels and increased ovotransferrins concentrations in plasma. The result indicated that dietary 2.0 % brown seaweed with bean extract affect innate immune response changing anti-oxidant system and TNF-$\alpha$ levels in broiler chicks.

Phenotype Changes in Immune Cell Activation in Obesity (비만 환경 내 면역세포 활성화 표현형의 변화)

  • Immune and metabolic systems are important factors in maintaining homeostasis. Immune response and metabolic regulation are highly associated, so, when the normal metabolism is disturbed, the immune response changed followed the metabolic diseases occur. Likewise, obesity is highly related to immune response. Obesity, which is caused by an imbalance in energy metabolism, is associated with metabolic diseases, such as insulin resistance, type 2 diabetes, fatty liver diseases, atherosclerosis and hypertension. As known, obesity is characterized in chronic low-grade inflammation. In obesity, the microenvironment of immune cells became inflammatory by the unique activation phenotypes of immune cells such as macrophage, natural killer cell, T cell. Also, the immune cells interact each other in cellular or cytokine mechanisms, which intensify the obesity-induced inflammatory response. This phenomenon suggests the possibility of regulating the activation of immune cells as a pharmacological therapeutic strategy for obesity in addition to the common pharmacological treatment of obesity which is aimed at inhibiting enzymes such as pancreatic lipase and α-amylase or inhibiting differentiation of preadipocytes. In this review, we summarize the activation phenotypes of macrophage, natural killer cell and T cell, and their aspects in obesity. We also summarize the pharmacological substances that alleviates obesity by regulating the activation of immune cells.

Investigating the Role of Microglia in Maternal Immune Activation in Rodent Models (모체 면역 활성화 유도 설치류 모델에서 미세아교세포의 역할 조사)

  • Epidemiological studies suggest that maternal infection, maternal stress, and environmental risk factors during pregnancy increase the risk of brain development abnormalities associated with cognitive impairment in the offspring and increase susceptibility to schizophrenia and autism spectrum disorder. Several animal models have demonstrated that maternal immune activation (MIA) is sufficient to induce abnormal brain development and behavioral defects in the fetus. When polyinosine:polycytodylic acid (poly I:C) or lipopolysaccharide (LPS), which is commonly used in maternal immune activation animal models, was introduced into a pregnant dam, an increase in pro-inflammatory cytokines and microglial activity was observed in the offspring's brain. Microglia are brain-resident immune cells that play a mediating role in the central nervous system, and they are responsible for various functions, such as phagocytosis, synapse formation and branching, and angiogenesis. Several studies have reported that microglia are activated in MIA offspring and influence offspring behavior through interactions with various cytokines. In addition, it has been reported that they play an important role in brain circuits through interactions with neurons and astrocytes. However, there is controversy concerning whether microglia are essential to brain development or lead to behavioral defects, and the exact mechanism remains unknown. Therefore, for the potential diagnosis and treatment of brain developmental disorders, a functional study of microglia should be conducted using MIA animal models.

Effect of Fruits from Pyrus ussuriensis var. hakunensis (Nakai) T.B. Lee on Macrophage Activation (산돌배(Pyrus ussuriensis var. hakunensis (Nakai) T.B. Lee) 열매의 대식세포 활성화 유도 활성)

  • In this study, we investigated in vitro immunostimulatory activity of fruit extracts from Pyrus ussuriensis var. hakunensis (Nakai) T.B. Lee (PUF) using mouse macrophage RAW264.7 cells. PUF increased the production of immunostimulatory factors such as NO, iNOS, IL-1β, IL-6 and TNF-α, and phagocytic activity in RAW264.7 cells. The inhibition of TLR2 and TLR4 blocked PUF-mediated production of immunostimulatory factors in RAW264.7 cells. In addition, the inhibition of MAPKs signaling pathway reduced PUF-mediated production of immunostimulatory factors. From these results, PUF may have immunostimulatory activity via TLR2/4-mediated activation of MAPKs signaling pathway. Therefore, PUF expected to be used as a potential immune-enhancing agent.

New Technology - Immunoregulatory Actions of Polysaccharides from Natural Plant Resources (해외 기술 - 천연소재 유래 다당체의 면역력 증진 기능)

  • 기능성 다당체는 자연계에 두루 존재하며 특히 많은 천연소재 유래 다당체의 경우 거의 부작용없이 광범위한 면역조절 기능을 나타내는 것으로 보고되고 있다. 이들 다당체의 면역증진 기능성은 다양한 경로와 타켓을 대상으로 나타나고 있으며 각 다당체의 기능성 차이는 그들에 구조적 차이에 기인한 것으로 판단되고 있다. BRM의 일종으로 천연소재 유래 다당체는 면역기관이나 T와 B 림프구, 대식세포, NK cell, LAK 세포 및 수지상 세포 등 면역세포의 활성화, 사이토카인 및 항체의 생성, 적혈구의 면역기능에 영향을 미쳐 생체방어 기능, 즉 면역기능을 증진시켜 주는 것으로 보고 되었다. 또한 lentinan, PSK, PSP, PUPS 등과 같이 이미 잘 알려진 다당체의 경우, 이들 다당체가 지닌 품질 안정성, 정확한 효능, 낮은 독성 및 부작용 등을 근간으로 다양한 항암치료제의 보조제로서 활용가능성도 높아지고 있다. 반면 다당체를 이용한 기능성 구명 연구에서는 몇 가지 제한이 있는 것도 사실이다. 첫째로 비교가능한 적절한 기능성 다당체의 비교 표준이 부재하다는 것이며 두 번째로는 대부분의 다당체 관련 연구가 보다 철저한 구조적, 정성적 연구결과 없이 주로 추출물을 이용한 연구로 진행되어 정확한 분자수준에서의 면역조절 기전을 구명하기 어렵다는 점이다. 마지막으로 정확한 기능 구명을 위한 신호전달경로 관련 연구, 수용체 관련연구 및 임상연구 등이 부족하다는 점이다. 따라서 향후 다당체를 이용한 면역력 증진 등 기능성 구명연구의 활성화를 위해서는 보다 정확한 기능성 다당 분획의 구조분석 및 동정 연구가 수행되어야 할 것이며 이는 분자 수준의 기능성을 이해하는데 주요한 요인이 될 것으로 판단되었다.

  • Effect of dietary brown seaweed (Undaria pinnatifida) levels on the performance, nutrients utilization, and blood anti-oxidant system was studied in broiler chicks activated innate immune response. Brown seaweed 2.0 % diet improved performance of broiler chicks and resulted in enhanced feed efficiency due to the increased NB and decreased UAN excretion significantly (P<0.05). Dietary brown seaweed reduced SOD activity in erythrocyte cytosol and enhanced peroxidase activity in plasma significantly(P<0.05). Activation of innate immune response increased SOD activity and peroxide levels in blood. The results indicated that dietary brown seaweed affected SOD and peroxidase activity and the increased performance in birds fed brown seaweed 2.0 % diet related with decreased decomposition of body protein and the change of anti-oxidant systems in blood of broiler chicks during activation of innate immune response.

  • Efficient production of antigen specific cytotoxic T cells is critical for appropriate adoptive immune response. In vitro culture and expansion of human T lymphocyte clones are very sophisticated and subtle procedure in immune cell therapy and hard to control. Therefore, many groups devoted their efforts to manipulate artificial antigen presenting cells (aAPCs) that can induce T cell activation and clonal expansion. To mimicking of natural antigen-presenting cells, aAPCs encompass basic signal molecules required for T cell activation: MHC:antigen complexes, co-stimulatory molecules and soluble immune modulating molecules. Orchestrated organization of these molecules is important for efficient T cell activation. Here, we discuss how those molecules have been incorporated in several aAPC models, but also how physical properties od aAPC are important for interaction with T cells.
