Studies on Bioresponses of Sophorae Radix (I) (고삼(苦蔘)(Sophorae Radix)의 생체 반응에 대한 연구(I))

  • 청열조습(淸熱燥濕), 거풍살충(祛風殺蟲)의 효능이 있는 고삼(苦蔘)의 체내 면역활성에 미치는 영향에 대해 실험한 결과 아래와 같은 지견을 얻었다. ICR 생쥐에 고삼(苦蔘) 수침액을 투여한 결과 백혈구수 및 면역장기 변화는 없었다. in vitro실험에서 IgM PFCs에 유의성 있는 영향을 주지 못했고, BSA를 주사한 생쥐에서의 Arthus 반응에도 영향이 없었다. 그러나 지연형 과민반응을 억제하였으며, 비특이적 면역에 관여하는 macrophage의 탐식능을 약간 저하시켰다. 이상의 결과 고삼(苦蔘) 수침액(水浸液)은 세포성 면역을 억제하는 것으로 사료된다.

  • 장관은 우리 몸에서 가장 넓은 표면적을 가지고 있으며, 외부로부터 침입하는 이물에 대항하여 면역세포의 배치가 필수적이다. 항원과 만난 적이 없는 naive lymphocyte는 임파절 등 2차 임파기관의 수지세포(dendritic cell)가 임파구에 항원을 제시할 때 레티노인산(retinoic acid)을 생산하여 제공함으로써, 소장 조직에 특이적으로 임파구가 호밍(homing)하는 능력을 부여한다. 한편, 장에 있어서는 식품항원에 대한 면역반응을 억제할 필요가 있는데, RA는 T세포의 기능분화를 억제하여 면역관용의 성립에도 관여함이 밝혀졌다.

  • 인공 면역 시스템을 사용한 정보통신시스템의 기억구조의 바이러스 침투경로를 확인하고 이를 대처하는 면역체계의 임파구를 T임파구와 B임파구라 할때 바이러스가 경유된 기억 구조를 기존의 정보 공유체제에서 전송 경유체계로 구성하여 바이러스의 침투 경유사항을 스스로 느낄 수 있게끔 항원체계를 구축한다. 항원 시스템은 항체 구조를 사용하여 항체 구조 내에 존재하는 임파구와 생체면역 체계를 형성하는 항원 시스템간의 혈청 시스템 구조를 형성하는데 혈액 구조내부의 항원 임파구와 항체 임파구를 조직하여 생체 내부에서 생체바이러스를 퇴치하는 방법으로 정보공유 및 전송시스템의 기억구조의 바이러스를 퇴치하게 된다.

Depression of Immune Response by Newcastle Disease Virus Infection (Newcastle병(病) 바이러스감염(感染)에 의(依)한 면역반응억제(免疫反應抑制))

  • The immunosuppressive activity of newcastle disease virus(NDV) and some possible role of interferon(C-IF) in viral suppression of immune response were evaluated by SRBC-induced delayed-type hypersensitivity(DTH), rosette formation in spleen cells, number of lymphocytes in peripheral blood, hemagglutinin and hemolysin response to SRBC in ICR mice sensitized with SRBC. When NDV was inoculated before or after sensitization of mouse with SRBC, virus caused a marked inhibition of DTH, and its depressive effect was dependent on the time of virus inoculation in relation to SRBC sensitization or challenge. Rosette formation of spleen cells was significantly reduced by NDV infection. The degree of the depression of rosette formation was more prominent in mice inoculated before sensitization than after sensitization and could be related to the amount of serum interferon induced by the virus. Humoral response to SRBC of virus infected mouse was significantly depressed when NDV was inoculated 24 or 48 hours before sensitization. However, there was no difference in the response when the virus was inoculated 9 hour before and at the same time of sensitization or even after that. Lymphocytes in peripheral blood of mice were markedly diminished in numbers when NDV was inoculated 48 and 24 hour before sensitization with SRBC, but they were slightly augmented when the virus was inoculated 9 hour before and at the same time of sensitization. When UV-inactivated or heat-inactivated NDV was injected to the mouse at the same time of sensitization with SRBC, DTH and rosette formation of spleen cells were slightly depressed. DTH and rosette formation in mice treated with crude-IF were generally depressed as com pared with those of control mice. These studies suggest that the NDV causes a significant depression of cell-mediated immunity, whereas the humoral immune response is not inhibited markedly, and that the depression of immune response by NDV infection may be caused by interferon produced by NDV and direct viral activity.

해조류 첨가사료가 돌돔의 성장과 면역기능에 미치는 영향

  • 양식 생물의 질병 제어는 예방과 치료를 통해 이루어진다. 그러나 항생제의 오ㆍ남용으로 인하여 약제 내성균의 증가와 치료 효과가 감소하고, 나아가 식품안전성 문제로 양식생물의 소비를 위축시켜왔다. 따라서 질병이 발생한 후에 시행하는 화학약품 치료법은 이미 한계에 달하였다. 이에 따라 사전에 질병을 차단하는 예방법인 vaccine이나 질병에 대한 저항능력을 키우는 비특이적 면역 증강물질에 대한 관심이 꾸준히 높아지고 있다. 해조류에는 다량의 lectin이 포함되어 있는데, Lectin은 당에 결합하는 단백질의 총체적인 이름으로, defence molecule로 작용하여 어류가 섭취할 경우 외래 항원으로 인식하여 지속적으로 면역 기능을 활성화시키는 것으로 알려져 있다. 따라서 본 연구는 돌돔사료에 kelf meal을 첨가하여 공급한 후 이들의 성장과 비특이적 면역기구에 미치는 효과를 조사하였다. 실험사료는 상업용 kelf meal을 0%, 2%, 5% 첨가하고, 2반복으로 500L FRP 탱크에서 8주간 사육하였다. 실험어류는 40g 내외의 돌돔을 대상으로 하였고, 사료공급 4주 및 8주에 각 실험구의 돌돔을 대상으로 비특이적 면역기구에 미치는 효과를 조사하였다. 체액성 면역 조사로는 혈청 및 점액 lysozyme 활성과 Escherichia coli에 대한 혈청 내 보체의 살균 능력을 조사하였고, 세포성 면역 조사는 전신 식세포의 NBT 환원 실험과 말초혈액의 식세포 식균능을 측정하였다. 식균능 시험은 Escherichia coli FKC (formalin killed cell)에 대한 식균율 및 식균지수로 나타내었다. 나아가 돌돔 말초혈액의 혈구조성에 미치는 영향을 조사하기 위하여 적혈구 5,000 세포당 임파구, 전구, 호중구의 수를 계수하였다. 8주간의 사육결과 사료계수는 각각 1.20, 1.25, 1.42로 대조구와 2% 첨가구는 큰 차이가 없었으나, 5% 첨가구는 나빠지는 경향을 보였다. 사료섭취량은 kelf meal의 첨가량이 늘어나면서 감소하는 경향이었다. 비특이적인 면역 기구에 대한 자극 효과는 체액성 면역의 지표로 측정한 lysozyme의 활성도는 혈청과 점액 모두에서 대조구보다 실험구에서 활성이 높았고, 혈청 lysozyme 활성은 5% 첨가구에서 가장 높았다. 혈청 내 보체의 살균능은 실험구간 차이가 없었다. 세포성 면역의 지표로 측정한 전신 식세포의 $O_2$ 생성능(NBT 환원능)은 5% 첨가구에서 가장 높았다. 말초 혈액의 식균능은 식균율과 식균지수 모두 실험구에서 높았고, 특히 5% 첨가구에서 가장 높았다. 말초 혈액 혈구조성의 변화는 임파구와 전구의 경우 실험구에서 높았으나 호중구에서는 차이가 없었다. 이상의 결과, 양식 돌돔에 kelf meal을 2% 첨가하는 것이 사료계수에 큰 영향을 미치지 않고, 돌돔의 체액성 및 세포성 면역을 적절히 자극하는 것으로 나타나, 돌돔의 기능성 사료 첨가물로서 이용 효과가 있을 것으로 기대된다.

Immunomodulatory Effects of Polysaccharides Derived from Persimmon Leaves on Cyclophosphamide-induced Immunosuppressed Mice (감잎 유래 다당체가 면역억제 마우스의 면역조절에 미치는 영향)

  • We prepared 2 different crude polysaccharides from persimmon leaves, by hot water (PLW-0) and pectinase digestion (PLE-0), respectively. PLW-0 and PLE-0 showed similar sugar compositions and contained 11 different sugars, including rarely observed sugars such as 2-methyl-fucose, 2-methyl-xylose, apiose, and aceric acid. However, the uronic acid content of PLE-0 was lower than that of PLW-0, because of pectinase treatment. In normal mice, administration of PLW-0 and PLE-0 increased the spleen index and splenocyte proliferation. The effect of PLE-0 on the spleen index and splenocyte proliferation was greater than that of PLW-0. We subsequently assessed the immunomodulatory activities of PLW-0 and PLE-0 on cyclophosphamide (CY)-induced immunosuppressed mice. We revealed that mice treated with PLW- 0 or PLE-0 showed increased splenocyte proliferation, NK cell activity, and white blood cell numbers. Taken together, our results indicate that PLW-0 and PLE-0 can enhance immune function in normal mice and modulate CY-induced immune suppression.

Effect of bee pollen extract on activation of dendritic cells and induction of Th1 immune response (꿀벌 꽃가루 열수 추출물의 수지상 세포 활성화 및 Th1 반응에 미치는 효과)

  • Dendritic cells (DCs) are potent antigen-presenting cells that play a pivotal role in modulating both innate and adaptive immunity. This study examined the immunomodulatory activities of hot-water extracts of bee pollen (BPW) in bone-marrow derived DCs (BMDC) and mice splenocytes. BMDCs isolated from mice were treated with 250 and $500{\mu}g/mL$ BPW for 24 h. BPW, up to $500{\mu}g/mL$, did not display any cellular toxicity against BMDCs. In fact, it functionally induced BMDC activation via augmentation of CD80, CD86, and major histocompatibility complex (MHC) class I/II expression and pro-inflammatory cytokine (tumor necrosis factor; $TNF-{\alpha}$, interleukin; IL-6, and $IL-1{\beta}$) production. Interestingly, BPW treatment significantly increased the production of interferon $(IFN)-{\gamma}$ in splenocytes, suggesting its possible contribution to Th1 polarization in immune response. Taken together, these findings suggest that BPW may regulate innate and adaptive immunity via DC activation and Th1 polarization in immune responses.

Advance Understanding and New Treatment of Alopecia Areata (원형탈모증(alopecia areata)의 최신 이해와 치료)

  • Alopecia areata (AA) is a common and tissue-specific autoimmune disease of hair follicle resulting in the loss of hair on the scalp and elsewhere on the body. Hair follicles is a unique organ because it has its own immune system and hormonal milieu and has a different immune state at each hair cycle stage. The collapses of anagen-dependent hair follicle immune privilege arise autoimmune attack, inducing ectopic MHC class I expression in the hair follicle epithelium and autoantigen presentation to autoreactive CD8+T cells, which results in AA. Clinical and experimental studies have pointed that psychological stress may also influence the hair follicle immune/hormone systems and contribute to the induction of AA. The key pathogenesis of AA is associated with immune privilege guardians (including ACTH, ${\alpha}-MSH$, and $TGF-{\beta}$), natural killer group 2D-positive (NKG2D+) cells (including NK and CD8+T cells), and stress hormones (including CRH and substance P). Effective treatments for AA are still demanded. One of the future targets of treatment will be the modification of hair follicle immune privilege including stress. Recent studies have reported that JAK inhibitors and immunomodulators used in other autoimmune disease, such as psoriasis, atopic dermatitis, and rheumatoid arthritis, Tregs, platelet-rich plasma therapy, statins, and prostaglandin anaolgues are effective for AA. Here the article reviews the recent understanding in the pathogenesis associated with perifollicular endocrine/immunology and new treatments of AA.

An Immunohistochemical Study on the Endocrine Cells in the Gastrointestinal Tract of the Mandarin Fish (Siniperca scherzeri) (쏘가리의 위장관 내분비세포에 관한 면역조직화학적 연구)

  • The regional distribution and relative frequency of neurohormonal peptides-producing cells were demonstrated in the gut of the stomach teleost, the Mandarin fish, Siniperca scherzeri Steindachner, using 7 types of specific antisera raised against mammalian regulatory peptides. The gastrointestinal tract of the Mandarin fish was divided into three portions from proximal to distal, stomach, small intestine and large intestine. Cells showing immunoreactivities against regulatory peptides were situated in the epithelial lining, between epithelial cells, and gastric or intestinal gland regions with various frequencies along with gastrointestinal tract. Mast of immunoreactive cells in the epithelial lining portion were generally spherical or spindle shape having long cytoplasmic process that were reached to the lumen (open type cell) while cells showing round in shape (closed type cell) were found in the gastric gland of the stomach occasionally. Serctonin-, samatostatin-, gastrin-, cholecystokinin (CCK)-8- and human pancreatic polypeptide (HPP)-immunoreactive cells were observed in this study. However, no insulin- and glucagon-immunoreactive cells were found. Serotonin- and somatostatin-immunoreactive cells were restricted to the stomach regions with moderate and numerous frequencies, respectively. Gastrin-immunoreactive cells were demonstrated in the stomach and small intestinal portions with a few and moderate frequencies, respectively and CCK-8-immunoreactive cells were restricted to the small intestinal portions with moderate frequency. In addition, HPP-immunoreactive cells were demonstrated in the stomach and small intestine with numerous frequencies, respectively. In conclusion, the distribution and relative frequency of these immunoreactive cells in the gastrointestinal tract of the Mandarin fish shows peculiar patterns compared to those of other stomach and/or stomachless teleost.

Immunohistochemical study of pancreatic poly-peptide-immunoreactive cells in the pancreas of the Korean native goat (Capra hircus) during developmental stages (발생단계에 따른 한국재래산양 췌장에서 pancreatic polypeptide 면역반응세포에 대한 면역조직화학적 연구)

  • The regional distribution and relative frequency of the pancreatic polypeptide (PP)-immunoreactive cells in the pancreas of the Korean native goat (Capra hircus) were studied by immunohistochemical methods (PAP methods) using specific antisera against to PP during developmental stage, fetus, neonate, 1-month-old, 6-month-old and adult. The different regional distribution and relative frequency of PP-immunoreactive cells in the pancreas of Korean native goat were observed during development. In the exocrine portions, they were detected in the all ages and distributed in the interacinar regions. PP-immunoreactive cells were increased from neonate to 1-month-old but thereafter decreased with developmental stage. In the pancreatic duct, PP-immunoreactive cells were demonstrated from 1-month-old but they were decreased with developmental stages in these regions. These cells were observed in the subepithelial connective tissue of the pancreatic duct with a few frequencies in 1-and 6-month-old. However, they were located in the interepithelial cells of the pancreatic duct in the adult with rare frequency. In the endocrine portion (pancreatic islets), PP-immunoreactive cells were observed from neonate, and the regional distribution of PP-immunoreactive cells from 1-month-old was divided into two patterns : dispersed 1) in the marginal regions of the pancreatic islets with moderate or a few frequencies, and 2) in the whole pancreatic islets (in a case of 1-month-old) or in the central cores (in a case of 6-month-old and adult) with numerous frequency. In conclusion, the different regional distribution and relative frequency of PP-immunoreactive cells in the pancreas of the Korean native goat were observed during development. It is suggested that the changing of feeding habits and physiological conditions during different developmental stages may cause these differences.
