  • Interferon alpha is an immunomodulator that is used as an antiviral agent to treat chronic active viral hepatitis C. However, interferon can induce or exacerbate sarcoidosis. We report a case of 42-year-old man with an exacerbation of pulmonary sarcoidosis after the cessation of interferon and ribavirin therapy for chronic hepatitis C. The patient's sarcoidosis improved spontaneously and he continues to be monitored regularly without steroid therapy.

Regulation of RANTES and MCP Expression in Human Nasal Mucosal Fibroblasts (비점막(鼻粘膜) 섬유모세포에서의 RANTES와 MCP의 발현 조절)

  • Background: Fibroblast functions both as a structural element and as a vital immunoregulatory cell. Fibroblasts regulate inflammation through governing of chemokine expression. In order to elucidate the mechanisms by which the expressions of chemokines were regulated, the co-stimulatory effects of Th1 and proinflammatory cytokines were compared using nasal mucosal fibroblasts. Methods: Human nasal mucosa was obtained from surgery for septal deviation and the growth of fibroblasts was established. Fibroblasts from 4th to 6th passage were stimulated with various combinations of cytokines. To inhibit selected signaling pathways, fibroblasts were pretreated with cyclosporin A, wortmannin, staurosporine, and dexamethasone prior to the stimulation with cytokines. The supernatants were collected and chemokines were detected with a sandwich enzyme-linked immunosorbent assay. Results: $TNF-{\alpha}/IFN-{\gamma}$-induced production of RANTES was inhibited by all inhibitors used. MCP-1 was produced constitutively and $TNF-{\alpha}$-induced or $TNF-{\alpha}/IFN-{\gamma}$-induced production of MCP-1 was not inhibited by cyclosporin A or wortmannin, but by stauroporine or dexamethasone. All inhibitors used in this experiment inhibited $TNF-{\alpha}/IFN-{\gamma}$-induced or $IL-1{\beta}/IFN-{\gamma}$-induced production of MCP-2 in nasal mucosal fibroblasts. Although staurosporine or dexamethasone showed strong inhibitory effects, cyclosporin A or wortmannin did not inhibit the production of MCP-3 by $IL-1{\beta}/IFN-{\gamma}$ treatment. Conclusion: Chemokines were strongly induced by stimulation of cytokines in combination and showed different pattern of inhibition by the inhibitors. Therefore, it was assumed that cytokines acted on multiple pathways or on unknown pathways which converged to gene-specific transcription factors.

새로운 nucleoside계 항암제, ara-CDP-DL-PCA.Na(BR-28702-2)의 약효연구 및 급성독성 시험.

  • 항암 및 면역조절작용을 가지고 있으며 그 자체가 서방성 prodrug으로서 약효를 나타낼것으로 기대되는 ara-C와 etherphospholipid의 conjugate인 ara-CDP-DL-PCA.2Na, ara-CDP-DL-PBA.2Na, 및 ara-CDP-DL-PMA.2Na 3종의 BR-8702-2의 micellar soultion을 투여시료로 하여 제암력 평가를 실시하였다. DBA/2J 마우스(평균 체중 25g, 수컷)에 L$_{1210}$임파성 백혈병 세포를 이식한 후, 24시간 후 약물을 복강내에 투여하는 실험계 에서 400mg/kg/day, 단회투여 및 80 혹은 100mg/kg/day, 1~5일간 투여로 ILS%값이 229~543으로 우수한 제암력을 보였다. 또한 BDF$_1$ mice(15~20g)의 axillary region에 3㎣의 Lewis Lung Tumor를 피하로 이식한후 약물투여를 통한 제암효과를 관찰하였다. 100, 200, 300mg/kg/day의 단회 투여계 에서는 수명연장 효과가 없었다. 한편, 20, 40, 60mg/kg/day, 1~5일간의 투여계 에서는 ara-CDP-DL-PCA.2Na만이 효과가 있었는데 농도에 역순하여 저농도인 20mg/kg/day, 1~5일간의 투여계에서 가장 효과가 있었으며 그때의 ILS%는 32.3%였고 투여기간중의 체중변화는 거의 보이지 않았다. 한편 NICOM 370 Dynamic Light Scattering을 이용하여 투여시료로한 micellar solution의 입자도를 분석한 결과 ara-CDP-DL-PCA.2Na는 4.2nm size의 것이 99.48%를 차지하고 있었다. ara-CDP-DL-PCA.2Na의 ICR 마우스를 이용한 급성독성 시험에 있어서 경구투여에서의 LD$_{50}$값은 암,수컷 모두 5000mg/kg이상 이었고, 정맥내 투여 에서는 432mg/kg이었다. 실험과정중 생존동물의 일반적 이상소견등은 없었으나 정맥내 투여의 경우에서 체중증가 억제현상이 있었다.

Immunological Detection of Cytosolic Immature and Plastidial Mature EPSP-synthase after Glyphosate Treatment in Tomato(Lycopersicon esculentum) Apical Meristem (Glyphosate 처리후 토마토 정단부 세포질과 원형 엽록체에서 immature 및 mature EPSP-synthase의 면역학적 검정)

  • Glyphosate had no effect on 5-enolpyruvylshikimate-3-phosphate synthase(EPSP-synthase) biosynthesis per se. But it inhibited clealy the activity of EPSP-synthase. EPSP-synthase seemed to be synthesized as a higher molecular weight(54 kDa) presusor protein and to be transported into plastid. The apparent molecular weight of mature EPSP-synthase in plastid is 45 kDa. Thus, the molecular size of transit peptide appeared to be about 9 kDa. The etiolation for 48 h after glyphosate application did not exhibit the inhibition of translocating level of EPSP-synthase across chloroplast envelope in actively growing meristematic leaves. But even when the plants were etiolated 2 hr after glyphosate treatment, a complete inhibition did not occur at least within 12 hr, i.e. 2 hr after beginning light period, suggesting that EPSP-synthase biosynthesis appeared to be not completely light dependent and the level of EPSP-synthase translocation to chloroplast could be controlled by an unknown regulatory mechanism of light dependent herbicidal effect of glyphosate.

It is the first Research Institute Company, based on the 'Law for Daedeok Innopolis Development'. this company made a product of HemoHim which is the supportive functional food for the cancer treatment. This technology is very unique and get the Triad Patent Families. This paper deal with the new technology based innovation and the product is very sensitive because of the consumer oriented. We study the successful technology commercialization process of HemoHim and suggest a implication for the company and decision makers.

Radioiodine Labeling of Insulin Using Dimethylsulfoxide as a Labelling-Aid

  • Using dimethylsulfoxide (DMSO) as a labelling aid, insulin--$^{126}$ I of radioimmunoassay use has been effectively prepared. A small amount of DMSO was added to usual labelling mixture ana the reaction time was controled. The labelled insulin obtained in such a way showed improved bindabilities to the antibody and thus expressed larger dose-gradients in the plots of standard dose-response curves even though the labelling rate was decreased to some extent. However, by extending the reaction time to about 1 min, average labelling yield of 30% could be obtained. The average increase of bindability (B/F) in definite antiserum dilution was 2.5 comparing with 1.5 obtained in the absence of DMSO. Thus, the net bindability increase was 70% of those obtained in tile absence of DMSO. By means of a NMR spectrometry, it has been confirmed that the DMSO in the labelling mixtutre is converted to dimethylsulfone by chloramine-T. The results, generally agreed with the Stags's postulation, were discussed in view of a competitive oxidation of DMSO with disulfide linkages of the insulin molecule by the chloramine-T.

Enhancement of Growth and Bioactivity of Pleurotus eryngii Mycelia by Spent Coffee Ground (커피박 첨가에 의한 새송이버섯 균사체의 생육 및 생리활성 증가)

  • Pleurotus eryngii. one of the most popular edible mushrooms, has been well known for its biological activities such as antioxidation, antitumor and immune modulation. Spent coffee ground(SCG) that is a waste product from the coffee industry has been continuously investigated for its reutilization. In this study, SCG was added to the fungal cultuvation medium and analyzed for its effect on the growth and physiological activity of P. eryngii mycelia. It was clearly demonstrated that SCG could accelarate mycelia growth. 1% SCG culture was very notable by showing 2.5-fold higher dry cell weight comapred to the control culture, which suggested SCG as an excellent activator for the growth of P. eryngii mycelia. By the addition of SCG, polyphenol content was increased by two fold but there was no change in polysaccharide content. In the analysis of DPPH scavenging activity, SCG was determined as a valuable source in order to significantly increase the antioxidative activity of the mycelium.

  • Multisystem inflammatory syndrome in children (MIS-C) is a rare complication of coronavirus disease 2019 (COVID-19), causing multi-organ damage affecting the heart, lungs, kidneys, digestive tract, and nervous system. As the cases of MIS-C have been increasing following the COVID-19 pandemic, the importance of appropriate management for MIS-C is becoming increasingly apparent. Immunomodulating agents such as anakinra, infliximab, and steroids are regarded as supplementary therapy to the first-line treatment with intravenous immunoglobulin. However, these immunomodulating therapies can potentially precipitate opportunistic infections, including those caused by cytomegalovirus (CMV), Epstein-Barr virus, and tuberculosis, or increase the risk of co-infections. Herein, we report a case of a 3-year-old boy who was treated with immunoglobulin, infliximab, and high-dose methylprednisolone for MIS-C, and subsequently developed a persistent fever lasting 32 days caused by a CMV infection.

Effects of Salt Concentration on Motility and Expression of Flagellin Genes in the Fish Pathogen Edwardsiella tarda (염 농도가 어류 병원체 Edwardsiella tarda의 운동성과 편모발현에 미치는 영향)

  • E. tarda, a fish pathogen, can survive in seawater under relatively high salt conditions as well as in fish under physiological salt conditions. Bacterial growth under different salt concentrations may influence the expression of genes involved in bacterial structure and physiology. The growth rate of E. tarda culture in high salt (3.5% NaCl) was similar to that in low salt (1.0% NaCl, physiological salt concentration). Interestingly, the strain moved much faster in low salt conditions than in high salt conditions. Electron microscopic observation demonstrated that the bacterial cells grown in high salt had less or no flagellation. Obvious flagellation was observed in the parental strain E. tarda CK41 grown in low-salt condition. Two putative genes coding flagellin were identified in the E. tarda genome sequences. The amino acid sequence comparison of each gene revealed 93% identities. A flagellin gene was PCR amplified and cloned into a cloning vector. Using an E. coli protein expression system, a part of flagellin protein was overexpressed. Using the purified protein, an anti-flagellin antibody was raised in the rabbit. Immunoblot analyses with flagellin specific antibody demonstrated that E. tarda CK41 expressed falgellin in low salt conditions, which is consistent with the results seen in motility assay and microscopic observation. This is the first report of salt regulated flagella expression in E. tarda.

Antioxidant and immunoregulatory effects of Korean Rhamnaceae (국내 자생 갈매나무과 식물의 항산화 및 면역반응조절 효과 탐색)

  • We studied the antioxidative and immunoregulatory properties of 70% ethanol extracts from the leaves, branches, and fruit of 13 species of the Rhamnaceae collected during the bearing season (Berchemia berchemiifolia, Berchemia floribunda, Hovenia dulcis, Paliurus ramosissimus, Rhamnella franguloides, Rhamnus crenata, Rhamnus davurica, Rhamnus koraiensis, Rhamnus parvifolia, Rhamnus ussuriensis, Rhamnus yoshinoi, Sageretia thea, and Ziziphus ujube). Total phenolic and flavonoid content values were determined using calibration curves of gallic acid and rutin, respectively. ABTS and DPPH radical scavenging assays were performed to determine antioxidative potential. Anti-inflammatory effects were studied using the nitric oxide (NO) inhibition assay in LPS-induced RAW264.7 cells. Extracts of Berchemia berchemiifolia, Sageretia thea, and Ziziphus jujuba had the highest total phenol and flavonoid content values. Extracts of Berchemia berchemiifolia, Paliurus ramosissimus, Rhamnella franguloides, and Sageretia thea had the highest ABTS radical scavenging activities. Berchemia berchemiifolia and Sageretia thea had the highest DPPH radical scavenging activities. Furthermore, extracts of Berchemia berchemiifolia, Berchemia floribunda, Paliurus ramosissimus, Sageretia thea, and Ziziphus jujuba showed strong anti-inflammatory activity through inhibition of NO production. The results of this study suggest that Rhamnaceae extracts can be used as natural antioxidants and immunomodulators.
