Inhibitory Melanogenesis of Bambusae caulis in Taeniam and Profiling of Related Proteins (죽여의 멜라닌 생성 억제 효과 및 관련 단백질 동향 분석)

  • Inhibitory melanogenesis by Bambusae caulis in Teaniam (Phyllostaachys nigra var. henonis Stapf) was studied. Tyrosinase inhibition activities were evaluated with six different extracts. Among them the extract with methanol showed the highest tyrosinase activity inhibition. MTT assay with B16 melanoma showed that the extract was not toxic up to the concentration of 50 ppm. The melanogenesis was clearly inhibited by the extract when it was examined by the melanin content assay in the cell. When the extract was dosed as 10 ppm, the melanogenesis was reduced to 68% in culture medium and 74% in the cell. By the proteome analysis with 2-D electrophoresis, 171 protein spots were found in the control gel and 282 spots were detected in the sample gel. Among 120 spot proteins matched, 12 spots were identified as proteins involved in the melanogenesis mechanism.

Commelina communis Ledeb Inhibits Melanin Synthesis in Alpha-MSH-stimulated B16F10 Cells (압척초추출물의 Alpha-MSH 유도성 멜라닌합성 억제 효과)

  • Commelina communis Ledeb is a widely used medication for the treatment of antidiabetic, antioxidant and hypoglycemic agent in Korea. Alpha-melanocyte stimulating hormone (${\alpha}$-MSH) is a major factor to stimulate melanin synthesis in the skin. The purposes of this study was to investigate the inhibitory effects of extract from Commelina communis Ledeb (ECC) on ${\alpha}$-MSH-stimulated melanogenesis in B16F10 cells. ECC suppressed melanin synthesis and intracellular tyrosinase activity in B16F10 cells or ${\alpha}$-MSH-induced B16F10 cells in a dose dependent manner. In study on the melanogenic protein expressions, it had especially influence on expressions of tyrosinase and tyrosinase-related protein (TRP-1). Tyrosinase and TRP-1 expressions were gradually decreased in a dose-dependent. Additionally, the extract also decreased the ${\alpha}$-MSH-induced over-expression of tyrosinase and TRP-1. This results show that the anti-melanogenic activity of ECC is correlated with the suppression of tyrosinase and TRP-1 protein expressions in B16F10 cells.

Physiological activities of Agrimonia pilosa extract (짚신나물(Agrimonia pilosa) 추출물의 생리활성)

  • In this study, we investigated the applicability of functional materials by examining various physiological activities with an extract from the Agrimonia pilosa root. The A. pilosa extract showed low cytotoxicity against murine melanoma B16F10 cells. With little or no cytotoxicity at various concentrations, the A. pilosa extract showed high levels of DPPH radical scavenging activity ($ID_{50}$, 20.70 mg/L) and anti-microbial activity against Bacillus subtilis, Escherichia coli, and Candida albicans. In particular, it had a high level of anti-microbial activities against Gram-positive bacteria. These results suggest that the A. pilosa extract can be used as a natural preservative. It also showed inhibition of tyrosinase activity ($ID_{50}$, 90.18 mg/L), as does kojic acid ($ID_{50}$, 89.13 mg/L), and especially, a higher decrease in melanin content ($ID_{50}$, 62.5 mg/L) than the arbutin level ($ID_{50}$, 100.7 mg/L) as a positive control. These findings suggest that the A. pilosa extract inhibits melanin synthesis by suppressing the intracellular tyrosinase expression. These results indicate that the A. pilosa extract may be an effective material for functional cosmetics, such as skin whitening materials.

Inhibitory effects of Flowers of Lespedeza bicolor on Tyrosinase Activity and Melanin Synthesis (싸리꽃 추출물의 tyrosinase 활성 및 멜라닌합성 억제효과)

  • In this study, we investigated the effects of the flowers of Lespedeza bicolor on melanogenesis in a mouse melanocyte cell line, B16/F10 cells. Our results show that the flowers of Lespedeza bicolor significantly inhibits melanin synthesis in concentration-dependent manner. In addition, it was also found to inhibit the activity of tyrosinase, the rate-limiting melanogenic enzyme. While the flowers of Lespedeza bicolor have no direct inhibitory effect on tyrosinase activity in cell free assay system using mushroom tyrosinase. Moreover, the flowers of Lespedeza bicolor effectively suppressed the ${\alpha}-MSH-stimulated$ melanin formation, tyrosinase activity and dendrite outgrowth. These results suggest that the flowers of Lespedeza bicolor is a potent depigmetation agent.

Inhibitory Effect of Fructus Ligustri Lucidi Hexane Extract on Melanin Biosynthesis (여정자 헥산추출물의 멜라닌합성 억제효과)

  • The purpose of this study was to investigate the effects of Fructus Ligustri Lucidi (FLL) on the process of melanogenesis. Cell viability of B16F10 cells was measured by MTT assay and melanin content was assessed using the method of Hosoi with some modifications. Methanol extract of Fructus Ligustri Lucidi (MFLL) significantly inhibited melanin synthesis in a concentration-dependent manner, and it reduced the activity of tyrosinase, the rate-limiting melanogenic enzyme. Additionally, $\alpha$-melanocyte stimulation hormone ($\alpha$-MSH)-induced hyperpigmentation was down-regulated by MFLL. MFLL was fractionated by organic solvent. In the present study, Hexane extract of Fructus Ligustri Lucidi (HFLL) inhibited tyrosinase activity and melanin production of B16F10 cells in the absence or presence of $\alpha$-MSH. Ethyl acetate, butanol and water layers did not affect tyrosinase activity and melanin production. Meanwhile ethyl acetate, butanol and water layers showed DPPH free radical scavenging activity. These results suggest that extract of FLL could be used as functional biomaterial in developing a skin whitening agent having the antioxidant activity.

Ability of Lipophilic Extract Obtained from Plants to Inhibit Tyrosinase Activity in Reverse Micelles (역미셀계를 이용한 지용성 식물체 추출물의 tyrosinase 저해효과 분석)

  • The abilities of petroleum ether-extracts prepared from 75 plants to inhibit tyrosinase activity were evaluated in reverse micelles composed of isooctane/AOT(100 mM)/phosphate buffer(20 mM, pH 8.0) containing tyrosinase(105.3 units/mL) and 3,4-dihydroxyphenylalanine(0.18 mM). Compared with control which has no plant extracts, garlic could completely inhibit in vitro melanogenesis by tyrosinase, and Chinese quince, sweet potato, onion, radish bud and apple did more than 60%. Lipophilic extracts of medicinal plants and herbs such as rosemary, coriander, cinnamomi ramulus, crataegii fructus, ramulus biotae folium, mume fructus, menthae herba, eucommiae cortex and clove also inhibited tyrosinase activity more than 60%. When the extraction yield of lipophilic materials was considered together with their inhibition effect on tyrosinase, it was possible to select plants of which tyrosinase inhibitors could be produced in high quantity from unit weight. Using reverse micelles, the analysis of the capacity of lipophilic materials to inhibit tyrosinase activity which was difficult up to present could be possible.

Anti-photoaging Effects of Fermented Soybean (Bio-Peptone®) (대두 발효물(Bio-Peptone®)의 광노화 억제 효과)

  • Soybean (Glycine max), as one of the foods with high plant proteins, contains a large amount of bioactive compounds and known to be effective in cardiovascular disease and obesity as well as in improving skin condition. The purpose of this study was to investigate the anti-photoaging effects of soybean fermented with Lactobacillus Rhamnosus ($Bio-Peptone^{(R)}$) by assessment of cytotoxicity against UVB, collagen synthesis after UVB-irradiation, tyrosinase activity, and melanin synthesis. Results showed that $Bio-Peptone^{(R)}$ protected skin fibroblasts against UVB-induced cytotoxicity and increased type I collagen synthesis. Furthermore, $Bio-Peptone^{(R)}$ significantly inhibited tyrosinase activity and reduced melanin contents. This study suggests that $Bio-Peptone^{(R)}$ has protective effects against UVB-induced skin damage. Thus, it is concluded that $Bio-Peptone^{(R)}$ is able to prevent skin damage against UVB and thus acts as anti-photoaging materials by protecting skin forming wrinkles and skin pigments.

The effects of Hominis Placenta extract on Melanin synthesis of B16 melanoma cells (자하거(紫何車) 약침액(藥針液)이 B16 melanoma 세포주의 멜라닌 합성에 미치는 영향)

  • Objective : This research was carried out for the development of medicine for vitiligo treatment and focused on the effect of Hominis Placenta extract on Melanin synthesis of B16 melanoma cells. Methods : Acitivity of tyrosinase playing a vital role in synthesis of Melanin and the quantity of Melanin, which is the final product in cultured B16 melanoma cells, effects of Hominis Placenta extract were measured. Results: The results indicated that Hominis Placenta extract increased both the amount of Melanin and the activity of tyrosinase according to the concentration, and they also supported by western blot analysis. Conclusion : The results suggests that Hominis Placenta extract has an advantageous effect on the promotion of Melanin synthesis and will contribute to the development of vitiligo treatment through further related studies.

  • Objective : This research was carried out to compare the effect of Gorynju and a Soju extract Psoraleae fructus on melanin synthesis of B16 melanoma cells. Methods : To investigate melanin synthesis of B16 melanoma cells, this research was measured cell survival, tyrosinase activity, melanin synthesis, western blot. Results : Both Gorynju and Soju extract Psoraleae fructus, cell toxicity depended on the density. Tyrosinase activity depended on the density of Gorynju extract Psoraleae fructus and statistic was showed significant(0.5, 1, 2, 3 ${\mu}g/ml$), in a Soju extract Psoraleae fructus, 1 ${\mu}g/ml$ were showed significant. Melanin synthesis was showed significant in a Soju extract Psoraleae fructus(3, 4 ${\mu}g/ml$). Western blot was showed to depend on the density of Gorynju and a Soju extract Psoraleae fructus. Conclusions : In a tyrosinase activity and a melanin synthesis, the intermediate alcohol of Gorynju and a Soju may be suitable to use.
