Changes in the Chemical Composition and Flavor of Yeast Extracts during the Autolysis of Baker's Yeast (자기소화 시간에 따른 호모 Extract의 성분과 풍미의 변화에 관한 연구)

  • The changes in the chemical composition of yeast extracts during autolysis and their effect to the sensory quality were studied with baker's yeast, Saccharomyces cerevisiae. The amounts of extracted solids, proteins, amio-N. amino acids, especially glutamic acid, alanine and lysine, increased by the autolysis time up to 48 hrs. The results of sensory evaluation made by the multiple paired comparision test and Duncan's test indicated a significant difference is taste by the time of autolysis. In the profile test, the flavor character notes expressed by the panel were 17 different characters, 11 in aroma and 6 in taste. The character notes and the intensity of flavor changed with the time of autolysis. The sharp and beany flavor of the extracts which was autolyzed for 4 hours turned into meaty and worty flavor by 48 hours of autolysis. A proper arrangement of the flavor characters in the quantitative descriptive chart could provide a weighted value of the flavor grade. The aroma grade index and the taste grade index correlated to the amplitudes of the profile test.

Effect of fermented soybean curd residue (FSCR; SCR-meju) by aspergillus oryzae on the anti-obesity and lipids improvement (Aspergillus Oryzae로 발효시킨 비지메주의 항비만 및 지질개선 효과)

  • In this study, we designed to confirm the dietary effect of anti-obesity of fermented soybean curd residue (FSCR; SCR-Meju; Biji-meju) by A. oryzae, which is well known as a Korean traditional meju microbe. We observed that body weight gain, serum and hepatic lipid profile, as well as the activity of ROS generating enzyme and ROS scavenging enzyme in high-fat diet induced obese mice fed experimental diet (SCR and SCR-meju). Body weight gain and epididymal fat weight of HC (high-fat diet control) was markedly higher than that of NC (Normal control). Conversely, body weight gain and epididymal fat weight of the SCR (Biji) and SCR-meju (Biji-meju) group was significantly lower than that of HC; these of the SCR-meju group was lower than that of the SCR group. Furthermore, serum TG and total-cholesterol and LDL-cholesterol contents of SCR and SCR-meju groups were lower than that of HC, and HDL-cholesterol level of the SCR-meju group was significantly higher than that of HC. In conclusion, although precise mechanisms of the antiobese effects of SCR-meju in this study are unknown, the present study provides an experimental evidence that SCR-meju may prevent obesity and obesity related metabolic syndromes, such as hyperlipidemia, hypertension and diabetes, and liver disease by high-fat diet. Nevertheless, further study in this filed will be needed.

Effects of Planting Density on Growth and Yield of Vegetable Soybean Varieties (파종밀도가 풋콩 품종의 생육 및 수량에 미치는 영향)

  • The objective of experiment was to investigate the effects of planting density on growth and yield of vegetable soybean, and to clarify the optimum planting density of vegetable soybean in the middle west region of Korea. The field experiment with 4 levels of planting density was carried out at Yesan area in $2005{\sim}2006$. The days from seeding to flowering and the days from seeding to harvesting and lodging were not significantly different among planting distance. The stem length was increased as planting distance was shortened but the number of node, branch, pod per branch, pod per individual, weight of stem and pod, one hundred pod weight and rate of 2+3 seed per pod were decreased as planting density was increased. The size of vegetable soybeans was not significantly different among planting distance, but the harvest index of vegetable soybean was decreased as planting distance was shortened. Yield of vegetable soybean was increased as planting distance was decreased. However, the approriate densities for stem and pod weight per a plant, number of pod per a branch and the vegetable soybean yield of 2+3 seed per pod were different from that density. The optimal planting distance of varieties was $60{\sim}25\;cm$ in Sunheukkong and Ilpumgeomjeongkong and was $60{\sim}35\;cm$ in Galmikong.

Characteristics of Kanjang Made with Barley Bran (보리등겨로 제조한 간장의 품질특성)

  • This study was conducted to identify characteristics of kanjang made with barley bran. Characteristics of kanjang made with barley bran were compared to those of kanjang made with soybean. Total nitrogen was maintained more than 0.7% at 90 days after fermentation. In case of free sugars, the contents of arabinose, xylose, fructose, glucose and maltose were more abundant in kanjang made with barley bran than kanjang made with soybean. The contents of propionic acid and butyric acid were rich among volatile organic acids. Nonvolatile organic acid of lactic acid which was known as abundant component in kanjang was not detected in kanjang made with barley bran. In case of free amino acid, glutamic acid was the most abundant in kanjang made with barley bran, followed by proline and phenylalanine. The ratio of glutamic acid content to total amino acid content was $24.8{\sim}28.0%$. Essential amino acid was $26.7{\sim}29.9%$. Total contents of amino acids were $0.7{\sim}1.4%$. Result of sensory evaluation showed that taste of kanjang made with barley bran was good for fermented after 45 days.

Microbiological Studies and Biochemical Changes in Fermenting Soybean Curd Residue during Fermentation (비지의 발효과정중 발효미생물 및 성분변화)

  • This study was attempted to identify microorganisms in fermenting soybean curd residues (SCR). The changes in contents of free amino acids, nucleosides, reducing sugars, and oligosaccharides were also studied. The fermentation of SCR which was by change inoculation was carried out at $55^{\circ}C$ for 48 hrs. pH increased gradually during fermentation and isolated microorganisms were identified as Bacillus subtilis and Bacillus licheniformis. Moisture content decreased from 80.8% to 58.4% at 48 hrs of fermentation and other proximate composition did not show any changes during fermentation. The content of total free amino acids increased rapidly and the number and quantities of each amino acid analyzed also increased during the course of fermentation. Glutamic acid, phenylalaine, lysine and aspartic acid were rapidly liberated during fermentation. As to the changes of nucleotides, 5'-AMP little changed during the first 36 hrs but subsequently decreased to approximately 1/6 after 48 hrs of fermentation. On the other hand, 5'-IMP plus 5'-GMP did not show almost any change during the first 36 hrs but increased about 3.5 times at 48 hrs of fermentation. However, 5'-XMP was not detected. The reducing sugar level showed rapid and steady increase throughout the fermentation and that of stachyose plus raffinose decreased slightly. From these results, a possible way of utilization of fermented SCR was proposed as a substitute for soybean in meju preparation.

Antimutagenicity and Cytotoxic Effect of Ethanol Extract from Korean Traditional Mackjang Added Sea Tangle (다시마 분말을 첨가한 막장 에탄을 추출물의 항돌연변이원성 및 세포독성 효과)

  • This study was carried out to investigate antimutagenic and cytotoxic effects of Korean traditional Mackjang added with sea tangle powder. The content percentage of most of minerals among general composition increased by adding sea tangle powder. By the Ames test, the antimutagenic effect of ethanol extract of Korean traditional Mackjang added with 5% sea tangle powder was strongest exceeding the control, 10%, and 15% sea tangle additions. The ethanol extract (400$\mu$g/plate) of Mackjang added with 5% sea tang1e powder in the S. typhimurium TA 100 strain showed inhibition rate of 95.0% against the mutagenesis induced by MNNG. The inhibition rate of ethanol extract (400$\mu$g/plate) of Mackjang added with 5% sea tang1e powder in the S. typhimurium TA98 and TA100 strains was 81.4% and 88.8%, respectively, against the mutagenesis induced by 4NQO. Under the same conditions, the suppression against B($\alpha$)P and Trp-P-1 in the TA98 and TA100 strains were 85.3% and 91.0%, and 96.5% and 96.5%, respectively. For the anticancer effects, an investigation was done on the cytotoxicity of Mackjang added with 5% sea tangle powder on the cell lines with human lung carcinoma (A549), human hepatocellular carcinoma (HepG2), and human gastric carcinoma (KATOIII). The cytotoxicities were inhibited with increasing the extract concentration. The treatment of 1.0 mg/mL Mackjang added with 5% sea tang1e powder showed relatively strong cytotoxicity of 61.2%, 61.8% and 66.8% against A549, KATOIII, and HepG2, respectively.

Establishment of optimal soybean Koji manufacturing conditions (콩 코오지의 최적 제조조건 설정)

  • To establish the optimal manufacturing conditions of soybean koji, soybean Koji prepared with Aspergillus oryzae 6-M-1 and Bacillus subtilis 3-B-1 isolated from traditional Korean meju. During 7 days of making Koji, the amount of amino-type nitrogen was getting more increase. The amount of amino-type nitrogen of Koji prepared with A. oryzae 6-M-1 was 686.16 mg% (w/w), that of Koji with B. subtilis 3-B-1 was 643.46 mg% (w/w) at seventh day of making Koji. The ${\alpha}$-amylase activity of Koji prepared with A. oryzae 6-M-1 was 1472.54 unit/g, that of Koji with B. subtilis 3-B-1 was 791.00 units/g on the seventh day of the making. The acidic protease activity of Koji prepared with A. oryzae 6-M-1 was 309.00 unit/g, that of Koji with B. subtilis 3-B-1 was 135.88 unit/g at 7th day of making. The amount of amino-type nitrogen and enzyme activities of soybean Koji prepared with A. oryzae 6-M-1 and B. subtilis 3-B-1 were produced more than those of wheat flour Koji made in factory. Sensory evaluation on a commercial doenjang and doenjangs prepared with A. oryzae 6-M-1 and B. subtilis 3-B-1 was not significantly different at p<0.05.

Compositions of Nitrogen Compound and Amino Acid in Soybean Paste(Doenjang) Prepared with Different Microbial Sources (미생물 급원을 달리한 숙성 된장의 질소성분과 아미노산 조성)

  • Fermented traditional soybean paste(doenjang), koji soybean paste by Aspergillus oryzae, natto soybean paste by Bacillus natto and koji & natto soybean paste by Aspergillus oryzae and Bacillus natto were analyzed for compositions of amino acids and contents of nitrogens. Amino type nitrogen was the highest in fermenting for 90 days as $271{\sim}868\;mg/100g$, and its contents were in the order of koji soybean paste>traditional soybean paste>koji & natto soybean paste>natto soybean paste in all samples tested. In compositions of total amino acids, glutamic and aspartic acids were rich in koji soybean paste but big differences were not observed in all samples. But some differences were observed in free amino acid compositions in all samples, that is, glutamic acid, tyrosine, lysine and aspartic acid were detected more abundantly. Sum of free amino acids for 90 days were in order of koji soybean paste>traditional soybean paste>koji & natto soybean paste>natto soybean paste. The ratios of free to total amino acids were $3.28{\sim}19.81%$ for 45 days, but increased to $10.88{\sim}25.10%$ for 90 days, and in order of traditional doenjang>koji doenjang>koji & natto doenjang>natto doenjang. Methionine and histidine showed higher ratios of free to total amino acid but lower in glutamic acid and aspartic acid. These results suggest that koji and traditional soybean paste of having high ratios of free amino acids to free and total amino acids may be more favorable in soybean paste fermentation.

Chemical Changes of Kanjang Made with Barley Bran (보리등겨로 제조한 간장의 각종 성분 변화)

  • For the development of functional jang-products, kanjang was prepared using barley bran. Optical density of barley bran kanjang was significantly high at 15 days after fermentation, and the amount of extract was $2{\sim}3$ fold higher than that of soybean kanjang. Among the flavor components identified in barley bran kanjang, the content of 2-furancarboxaldehyde was the highest, followed by 4-vinyl-2-methoxy-phenol, benzene-acetaldehyde, palmitic acid, and methyl-9,12-octadecadienoate. In barley bran and soybean mixtare (1 : 1) kanjang, the content of 2-furancarboxaldehyde was the highest, followed by benzeneacetaldehyde, diethyl phtalate, palmitic acid, and 2-chloroethyl linoleate. Flavor components detected in both barley bran kanjang and barey bran and soybean mixture kanjang were 2-furancarboxaldehyde, benzaldehyde, benzeneacetaldthyde, 4-vinyl-2-mehtoxy-phenol, 1-furfuryl-2-formy pyrrole, dimethyl-1,2-benzenedicarboxylate, diethyl phtalate, palmiticacid, dibutyl-1,2-benzenedicarboxylate, and di-(2-ethylhexyl)phthalate.

Antimutagenic and Anticancer Effects of Ethanol Extract from Korean Traditional Doenjang Added Sea Tangle (다시마 분말을 첨가한 전통된장 에탄을 추출물의 항돌연변이성 및 항암효과)

  • This study was carried out to investigate antimutagenic and anticancer effects of ethanolic extract of Korean traditional doenjang added sea tangle. Most of the mineral content of doeniang was increased by addition of sea tangle. In the Ames test, the antimutagenic effect of ethanol extract of Korean traditional doenjang added 5% sea tangle was higher than that of control (no additive), 10%, and 15% sea tangle additions. The inhibition rate of ethanol extract (200$\mu\textrm{g}$/plate) of doenjang added 5% sea tangle in the S. typhimurium TA100 strain showed 97.0% inhibition against the mutagenesis induced by MNNG. In addition, the suppression of ethanol extract (200$\mu\textrm{g}$/plate) of doenjang added 5% sea tangle in the S. typhimurium TA98 and TA100 strains showed 60.2% and 69.1% inhibition respectively, against the mutagenesis induced by 4NQO. The suppressions under the same condition against B($\alpha$ )P and Trp-P-1 in the TA98 and TA100 strains were 71.7% and 87.3%, and 66.6% and 80.8%, respectively. In the anticancer effects, the cytotoxicity of doenjang added 5% sea tangle on the cell lines with human lung carcinoma (A549), human hepatocellular carcinoma (HepG2), and human gastric carcinoma (KATOIII) were inhibited with increasing the extract concentration. The treatment of 1.0 mg/mL Doenjang added 5% sea tangle showed strong cytotoxicity of 56.4%, 87.67%, and 89.5% against A549, HepG2, and KATOIII, respectively.
