The Contents of Organic Acid and Fatty Acid in Traditional Soy Sauce Prepared from Meju under Different Formations (형상이 다른 메주로 제조한 재래식 간장 중의 유기산과 지방산 조성)

  • Three kinds of soy sauce were prepared using the brick type of conventional menu(A), the brick type of meju of Aspergillus oryzae (B) and the grain type of menu Aspergillus oryzae (C). Organic acid and fatty acid were analyzed In accordance to aging time of those products Citric acid, lactic acid, acetic acid, malonic acid, butyric acid, oxalic acid, and propionic acid were dejected in all kinds of soy sauce. The content of lactic acid was shown higher than those of any other organic acids. The content of lactic acid was much higher at beginning of preparation and at 180 days in soy sauce B than any other conditions. The content of acetic acid was much higher at beginning of preparation, at 120 days in soy sauce C and at 180 days in soy sauce B than any other conditions. The content of citric acid was highest at beginning preparation in soy sauce C, and that was highest in soy sauce B except beginning preparation to 120 days. Myristic, palmitic, stearic, oleic, linoliic, linolenic, arachidonic acid were detected in all kinds of soy sauce after 180 days. The content of oleic acid were shown 32.59∼53.79% in soy sauce B and in soy sauce C. The content of stearic acid was shown 49.7oA In soy sauce A. Linolinec acid and arachidonic acid were detected in only soy sauce C.

  • Enzyme activities, production of killer toxin and some functionality of forty seven yeasts isolated from traditional Meju were investigated in culture broth and cell free extracts. Activities of $\alpha$-galactosidase, invertase and inulinase were detected in cell free extracts of 38 strains, 43 strains and 45 strains, respectively and acidic and neutral protease activities also were detected in culture broth of all the strains, $\beta$-Galactosidase activity was detected in cell free extracts of OE-20 and S-14 strains. Killer toxins were produced by OE-12, S-8 (Candida spp.), OE-19 (Zygosaccharomyces spp.) and S-3 (Saccharomyces spp.). Culture broth of OE-23 and S-9 showed 61.3% and 59.2% of antioxidant activity to $\alpha$, $\alpha$-diphenyl-$\beta$-picrylhydrazyl(DPPH), but nitrite-scavenging ability as well as inhibition of tyrosinase and polyphenol oxidase were not appeared in all the strains.

  • As a result of survey of one hundred grain weight and been husk/whole bean ratio of 25 soybean varieties, the one was ranged 12.1∼27.1g and Saeal was highest as 27.1g, and Kwangan was lowest as 12.1g, while the other was revealed that Sinpladal 2 was highest as 9.40%, and Jangyebo was lowest, as 6.13%. Contents of crude protein were ranged 38∼48.5% which Tanback was highest as 48.5%, and Jangyeob was lowest as 39%. Crude lipid contained 17∼22%, and Muhan was highest as 48.5% and Tanback was lowest as 39%. Range of water uptake ratio and hardness after soaking were 235.9∼202.8% and 0.890∼2.593kg/3.14$\textrm{mm}^2$ and Jinpum was highest as 2.593kg/3.14$\textrm{mm}^2$, while Tanwon was lowest as 0.066kg/3.14$\textrm{mm}^2$.

Changes in Microflora and Enzyme Activities of Traditional Meju during Fermentation at Sunchang Area (순창지역 메주 발효 중 미생물과 효소역가의 변화)

  • Meju for doenjang and kochujang was prepared as a model at Sunchang areaand monitored with major changes. Kochujang meju was prepared on September 12 and doenjang meju on November 12, 1995. Kochjang meju was found to be naturally fermented at 80~90% RH, 15~2$0^{\circ}C$ and doenjang meju was at 80~90% RH and 0~5$^{\circ}C$. The shapes of kochujang meju and doenjang meju were doughnut-type and rectangular, respectively. Weight losses during fermentation were 48% and 28%, respectively. The pH drop and acip production of kochujang meju were negligible. However, pH of doenjang meju decreased from 6.29 to 5.88 and acidity increased from 0.08 to 0.23% as lactic acid. Protein in meju was found to be rapidly solubilized during the early stage of fermentation. Soluble protein cotents of kochujang meju after 7 days and 60 days were 8.23%, respectively. The doenjang mejus were 2.15% after 20 days and 5.72% after 60 days. Soluble suger content increased with the fermentation time. The soluble sugar content was higher in kochjang meju. Acidic protease was highly produced during meju fermentation. $\alpha$-Amylase and $\beta$-amylase were detected in the kochujang meju, of which glutinous rice consisted, but negligible in doenjang meju. Lipase was detected in kochujang meju, but was, negligible in doenjang meju. Microbial population increased drastically after 7 days of fermentation in kochujang meju and 20 days of fermentation in doenjang meju.

Studies on the Fungal Isolates of Mucorales Collected from Korean Home Made Mejus and Nuluks (한국전통 식품의 원료인 메주와 누룩에서 분리된 접합균에 대한 연구)

  • The fungal isolates of Mucorales, directly collected from Korean traditional raw materials of Nuruk (raw material for Korean rice wine) and Meju (raw material for Korean soysauces), were compared with those of Rhizopus oryzae purchased. The fungal isolates of Rhizopus, Mucor, and Absidia mostly identified as based on the morphological observations, were evaluated with the PCR-polymorphic bands. The PCR-polymorphic bands of the genomic DNA reacted with the primers of OPD series tenmer were various, but showed averaged 4 to 6 in the agarose-electrophoresis. The dissimilarity coefficient (DC) between two isolates were compared by the cluster analyses, dendrogams and polar ordinations. The isolates of R. oryzae known. showed several groupings within the lower value of DC and were divided to two groups of amylo-process and other fungi with other purposes. The isolates unidentified were identified by the DC made of this results. Taxonomy of these isolates made by the morphological observations were consistent with those resulted above in most case but not in all aspects. More works were needed with the isolates known for detail informations of Mucorales.

The fungal isolates of Scopulariopsis collected from Korean home-made Mejus (메주에서 분리된 Scopulariopsis 속의 분리균)

  • From five years' previous work, the fungal isolates of Scopulariopsis were reported to be important flora at the late stage of meju fermentation. Mainly, the white or pale brown powders of spore mass of these fungi were observed on the surfaces of rectangular mejus, and to be an important sign for well-done Korean traditional home - made mejus. Out of the five isolates previously collected and stored, two kinds of Scopulariopsis isolates were identified as S. brevicauli and S. fusca. The microscopic differences between two were found to be branching patterns of annellophore and ornamentations of spore wall (warty and smooth). However, the intermediate form between two ornamentations of spore wall were also observed in our isolates. This observation was consistent with other result made from the protein electrophoresis. The isolates of Scopulariopsis were considered to be similar or superior to those of Aspergillus species, as compared with production of protease and amylase related enzymes. Thus, these isolates were speculated to be important fungi in Korean traditional home - made meju fermentation and also in production of protease and amylase.

Isolation of Aflatoxin Producing Fungi from Korean Rice and Soybean Paste (한국산 쌀과 메주에서의 Aflatoxin생성 진균분리)

  • In view of the warmth and hunidity in Korea, there is little possibility to avoid the fungal contamination of rice soybean paste which are major foodstuffs in this country. In order to isolate aflatoxin producing fungi from these foodstuffs, 6 samples of rice were collected from the farmhouses at Cbangsung and 40 samples of soybean paste in Seoul areas. The peptone glucose chloramphenicol, Czapek-Dox agar and malt agar were used for the isolation and identification of fungi, and Adye and Meteles medium and/or Sahouraud medium for the production of toxins. The cultures for the toxin production were done at $25^{\circ}C$ for $5{\sim}7$ days. Eleven Penicillium species were isolated. from 6 samples of rice, and 37 Aspergillus flavus, 48 Aspergillus species, 1 Penicillium species and 105 others were isolated from 40 samples of soybean paste. Among these, one Penicillium species isolated from rice was found to be aflatoxin producing strain, and the toxin was quantitatively measured. Aflatoxin appeared to be more favorably produced in the glucose Sabouraud medium than in the Adye and Meteles medium.

  • Aflatoxin $B_1$, $B_2$, $G_1$, and $G_2$were quantitatively detected by the high pressure liquid chromatography on a Micropak-CN column, with Hexane-THF-IPA-water, using a Lichrosorbpacked flowceil in the fluorometric detector. Under those conditions, the minimum detectable amount of aflatoxin $B_1$ was 0.2 ng. HPLC was used in determining amount of aflatoxins in the commercially manufactured soybean food and home-made Meju. Aflatoxin producing abilities of strains used in the industrially fermented soybean food were also studied with the HPLC technique. Although aflatoxin-like substances were detected in a few samples on TLC, they were not identified with the HPLC retention times of standard aflatoxins. The commercial fungal strains used in Korea had no aflatoxin producing abilities.

Physicochemical and Sensory Characteristics of Traditional Doenjang Prepared using a Meju Containing Components of Acanthopanax senticosus, Angelica gigas, and Corni fructus (가시오가피, 당귀와 산수유 성분이 함유된 메주로 제조한 전통 된장의 이화학적 특성 및 관능적 특성)

  • Changes in the quality characteristics of doenjang prepared with a meju containing components of Acanthopanax senticosus, Angelica gigas and Corni fructus during storage were evaluated. Three types of doenjang produced using each of these components showed lower Hunter's L and a values than the values of the control during the initial stage of storage: however, these values were similar to those of the control after 60 days of storage. Doenjang containing A. gigas showed a similar degree of browning to that of the control during the early stage of storage, while the degree of browning observed in the other samples was much higher than that og the control during this period. Despite these initial differences, the browning in all samples became similar after extended storage. The level of acidity increased gradually for up to 60 days, after which it decreased slightly. The pH of all treatments decreased with storage time. The amino-N contents of all types of doenjang increased gradually, reaching peak values after 60 days. Doenjang with A. senticosus and A. gigas had a strong characteristic flavor that reduced the native flavor of doenjang. Doenjang with C. fructus showed a similar overall palatability to that of the control.

  • This study was designed to investigate the changes in microflora and enzyme activities of traditional kochujang meju during fermentation for 60 days. The pH of meju decreased continously up to 40 days of aging and then increased slightly thereafter, while the change in titratable acidity showed the opposite trend to that of pH. The viable cell count of aerobic bacteria increared gradually for up to 40 days of fermentation and then decreased slightly thereafter, while that of molds and yeasts showed a rapid increase up to 40 days of fermentation and then leveled off. ${\alpha}-amylase$ activity increased slightly for up to 40 days of meju fermentation and then stabilized. On the other hand, ${\beta}-amylase$ and glucoamylase activities did not show a significant change for up to 20 days of fermentation and then increased rapidly at 40th day of fermentation. Acidic, neutral and alkaline protease activities increased sharply up to 40 days of aging and then decreased significently at 60th day of fermentation. These results suggest that meju fermented for 40 days had the highest quality in terms of the number of microflora and enzyme activity.
