머위로부터 Bakkenolide B의 순수분리, HPLC분석 방법 및 채취 시기별 함량 분석 (Isolation, Quality Evaluation, and Seasonal Changes of Bakkenolide B in Petasites japonicus by HPLC)

  • 머위(Peatasites japonicus)의 잎으로부터 생리활성물질을 탐색하여 항천식 효과가 있는 물질로 알려진 bakenolide B를 순수분리하였으며, 1D-NMR, 2D-NMR 및 GC-MS spectrum 데이터를 이용하여 구조를 동정하였다. 머위의 잎, 엽병 및 근경 등의 채취시기별 bakkenolide B의 함량을 평가하기 위하여 HPLC 분석 방법을 확립하였다. Bakkenolide B의 민감도는 210 nm와 215 nm의 파장에서 254 nm, 235 nm 및 265 nm보다 높았으며, 분석을 위한 회귀 직선식은 y=7.841-6.262(파장 215 nm)로서 상관값($r^2$)이 0.999 이상으로서 유의성이 매우 높았다. 검출한계(LOD)는 1.05, 정량한계(LOQ)는 3.38이었다. 회수율은 3종류의 농도값에 대하여 98.6에서 103.1%로서 매우 높았다. 이러한 결과들로 미루어 볼때 머위에서 bakkenolide B의 함량은 HPLC 분석방법으로서 가능한 것으로 증명되었다. 따라서 식물체의 부위 즉 잎, 엽병 및 근경에서의 bakkenolide B의 함량을 측정한 결과, 잎에서 가장 높았고, 다음은 엽병이었으며, 근경에서 함량이 가장 낮았다. 3월 초부터 8월 초까지 1개월 간격으로 식물체의 부위별 bakkenolide B의 함량을 조사한 결과, 3월부터 8월까지 채취시기가 지연될수록 감소하는 경향이었다. 따라서 HPLC를 이용한 Petasites species의 bakkenolides 분석은 단순하고, 급속하게 분석할 수 있는 방법으로 확인되었다.

  • 머위(Petasites japonicus) 잎에서 알레르기성 천식 억제 효능이 있는 bakkenolide B의 최적 추출공정을 반응표면 분석법을 이용하여 결정하였다. Box-Behnken design에 따라 설정한 추출온도(80, 100, $120^{\circ}C$), 추출시간(30, 45, 60 min) 및 추출용매의 pH (5, 7, 9)를 독립변수로 하고, 추출물의 특성 즉, bakkenolide B의 함량과 항알러지의 측정지표인 비만세포의 탈과립반응 억제효과를 종속변수로 하여 분석을 실시하였다. Bakkenolide B의 추출함량은 추출온도, 시간, pH의 유의적인 영향을 받았으며, 추출온도 $127.1^{\circ}C$, 시간 46.6분, 추출용매의 pH 7.7에서 최대치 121.6 ${\mu}g/g$이 예측되었다. 비만세포의 탈과립반응 억제효과의 회귀분석 결과는 유의성이 인정되지 않았고 낮은 상관계수를 보였다.

산채밭의 잡초발생(雜草發生) 양상(樣相) 및 방제(防除)에 관(關)한 연구(硏究) (Studies on Occurrence and Control of Weeds in Edible Wild Greens Field)

  • 산채류(山菜類)중 달래, 머위, 참취밭에 발생하는 일년생(一年生) 잡초(雜草)를 방제(防除)하기 위하여 napropamide 21.8% 유제(乳劑) nitralin 50% 수화제(水和劑) 그리고 pendimethalin 31.7% 유제(乳劑)를 약량별(藥量別)로 처리(處理)하였을 때 잡초방제(雜草防除) 효과(效果)와 산채류(山菜類)에 나타나는 반응(反應)을 조사(調査)하였다. 1. 시험포장(試驗圃場)에서는 피, 바랭이, 여뀌, 명아주, 털진득찰, 깨풀, 속속이풀, 닭의장풀 등 9종(種)의 잡초(雜草)가 발생(發生)하였으며, 그중 피, 바랭이, 여뀌, 명아주, 털진득찰 등이 우점(優占)하였다. 2. 잡초발생(雜草發生)의 다양화(多樣化) 정도(程度)를 나타내는 Simpson 지수(指數)는 0.26~0.30로 낮아 잡초발생(雜草發生) 초종(草種)이 다양(多樣)하였다. 3. 공시약제(供試藥劑)의 잡초방제(雜草防除) 효과(效果)는 napropamide유제(乳劑)(21.8%) 872g(ai/ha) 처리구(處理區)는 81.4%~85.6%이고, nitralin수화제(水和劑)(50%) 1,000g(ai/ha) 처리구(處理區)는 79.4%~82.8%, pendimethalin유제(乳劑)(31.7%) 634g(ai/ha) 처리구(處理區)에서는 86.8%~92.2%로서 작물별로 큰 차이가 없었다. 4. 공시약제(供試藥劑)에 대한 약제(藥劑)의 반응(反應)은 napropamide유제(乳劑)에서는 발아(發芽) 혹은 출아지연(出芽遲延)으로 인하여 생장(生長)이 억제(抑制)되는 1~3정도(程度)의 약해(藥害)가 나타났으며, nitralin수화제(水和劑)에서도 그 약해 증상과 정도가 napropamide유제(乳劑)와 유사(類似)하였으며 pendimethalin유제(乳劑)처리구의 달래와 머위는 생육지연(生育遭遲延)에 의한 1~2의 약해가 발현(發現)된 반면에 참취는 잎의 기형발현(畸形發現)으로 인한 2~3의 약해가 유발(誘發)되었다.

  • We evaluated the effects of butterbur (Petasites japonicus Maxim) addition to the diet on lipid profiles and antioxidant biomarkers such as total glutathionine, TBARS value, carbonyl value, GPx, GR, SOD and paraoxonase activity in the plasma or liver of mice. The distribution of body fat deposition, total cholesterol (TC) contents, and atherogenic index in the plasma were significantly decreased in the butterbur group. The levels of GSH and the activity of GR and SOD were significantly higher in the liver of the butterbur group than in that of the control group. Lipid oxidation of the liver and kidney and protein oxidation of the liver and heart were decreased in the butterbur group. Additionally, the DNA damage, as determined using the comet assay (single cell gel assay) with alkaline electrophoresis and as quantified by measuring the tail length (TL), was decreased in the butterbur group. The results of the present study showed that a diet with added butterbur exerts degenerative disease-protective effects on oxidative DNA damage and lipid peroxidation.

머위 잎 추출액의 염색성 -매염제가 염색성에 미치는 영향- (A Study on the Dyeing Properties of Petasites Japonicus Leaf Extract)

  • This study were carried out to investigate the effects of mordants and mordanting methods under the various dyeing conditions such as temperature, time, pH, repetition of dyeing in the silk fabrics with Petasites japonicus leaf. It was the most high K/S values of the silk fabrics dyed with Petasites japonicus leaf according to co mordanting and used cupric sulfate, ferric sulfate as mordanting agents. K/S values of the silk fabrics dyed with Petasites japonicus leaf were increased gradually with dyeing temperature, dyeing time, repetition of dyeing was higher and the optimum pH was pH 7. Silk fabrics were colored yellowish orange and surface color(munsell value) was changed from 8.3YR to 2.0Y by using mordanting agents and those of the silk showed high tone when mordanting with cupric sulfate but decolored and darked when mordanting with ferric sulfate. Washing fastness of silk fabrics were good in 4 ~ 5 grade, so washing fastness of the silk fabrics was significantly improved when washed with the neutral detergent.

  • This study was performed to investigate a vasodilatory efficacy and its underlying mechanisms of the mixture of Cnidium officinale, Petasites japonicus and Coptis chinensis (CPC), CPC relaxed rat aortic vascular strips in endothelium-independant manner precontracted with phenylephrine or KCI(50mM), but the magnitude of relaxation was greater in KCI induced contraction. L-NAME, iNOS inhibitor, and methylen blue(MB), cGMP inhibitor, did not attenuate the relaxation responses of CPC. Furthermore, the contraction by increaseing $Ca^{2+}$ concentration (0.3-10.0mM) to a $Ca^{2+}$-free high $K^+$ (60mM) was significantly reduced by CPC pretreatment. These results suggest that the relaxation effect of CPC is related with the block of $Ca^{2+}$ influx via $Ca^{2+}$ channel.

머위 분말 첨가 쌀쿠키의 항산화 활성 및 품질 특성 (Antioxidant Activities and Quality Characteristics of Rice Cookie with Added Butterbur (Petasites japonicus) Powder)

  • This study evaluated the antioxidant activity and quality characteristics of rice cookie with added butterbur powder in a ratio of 0, 5, 10, 15, and 20% to confirm the possibility of butterbur as a functional food. The moisture content, spread factor, leavening rate, and hardness of rice cookies increased with an increase in the amount of butterbur powder; whereas a decrease in the pH and baking loss rate was observed. The L and b values decreased as the amount of butterbur powder increased, but the value was the lowest when 5% of butterbur powder was added. The sensory liking score showed the highest preference for 10% butterbur powder regarding appearance, flavor, taste, texture, and overall preference. In the principal component analysis (PCA), the addition of 10% butterbur powder positively affected the measure of food acceptance in terms of organoleptic properties of butterbur. Besides, as the amount of added butterbur powder increased, the antioxidant activity of rice cookies increased. Based on these results, it appears that the addition of butterbur powder to rice cookies in a 10% ratio can produce rice cookies with excellent antioxidant activity, overall quality, and high preference.

머위(Petasites japonicus maxim)를 급여한 rat와 mouse에 대한 병리학적 관찰 I. 육안적 및 병리조직학적 관찰 (Pathological changes on rats and mice fed with Petasites japonicus Maxim I. Macroscopical and histopathological observations)

  • In order to know the toxic effect and carcinogenic activity in rats and mice fed with juice of Korean native Petasites japonicus Maxim of its pellet(4% or 8%) which were dried, milled and mixed with basal diet, the investigations were carried out by macroscopy and histopathology. Macroscopically, although remarkable changes were not observed in the liver of mice, there were slight to moderate swelling of rat livers in the whole groups at 12 to 14 weeks after feeding and milky spots in rats fed with its juice and 8% pelleted Petasites japonicus Maxim diet and a normal diet for 1 week alternatively for 14 weeks. Moreover, moderate to severe swelling and milk spots were recognized in livers of all rats fed with its juice and 8% pellet or 8% pelleted Petasites japonicus Maxim for 16 weeks. But, in cases of rats fed with its juice and 4% pellet or 4% pelleted Petasites japonicus Maxim, only swelling of livers was recognized moderately or severely. Histopathologically, major lesions were found in livers of both rats and mice. There were congestion, hemorrhage, fatty change, focal necrosis, megalocytosis and hyperplasia of endothelial cell in livers of mice and rats, the additional lesions such as proliferation of bile duct and nodular regeneration with diffuse regenerating cells were seen in livers of rats. In addition, preneoplastic lesions, the areas of milky spots macroscopically, were observed in livers of rats fed with Petasites japonicus Maxim for 14 to 16 weeks. In a few cases, haemangioendothelial sarcoma in livers was detected in rats fed with Petasites japonicus Maxim for 16 weeks. Petasites japonicus maxim growing naturally in Korea seem to exhibit toxic effect especially in liver and it contained a causative agent of primary liver tumors.

산채류(山菜類) 가열즙(加熱汁)의 돌연변이 억제 작용에 관(關)한 연구(硏究) (Desmutagenic Activity of Heated Mountain Herb Juices)

  • 10종류(種類)의 산채류가열즙(山菜類加熱汁)에 대한 돌연변이원성(突然變異原性)을 Spore rec-assay, Ames test 그리고 DNA 절단실험(切斷實驗)으로 검토(檢討)하였으며 가열즙(加熱汁)의 조제(調製)는 $100^{\circ}C$에서 20분간 가열하여 가열즙(加熱汁)을 얻었다. Spore rec-assay 결과 10종류(種類)의 시료(試料)모두 변이원성(變異原性)은 없었으며 금속이온의 영향에서 소리쟁이가열즙은 $Pb^{2+}$, 곰취와 참나물가열즙의 경우는 $Zn^{2+}$의 존재하에서 변이원성을 나타내었다. Ames 시험과 DNA 절단시험에서도 시료모두 변이원성은 없었으며 DNA 절단(切斷)시험에서는 곰취, 머위, 잔대, 개미취가열즙의 경우는 25mM $Cu^{2+}$ 공존하에서 절단활성(切斷活性)이 촉진되었다. 그러나 두릅, 원추리, 잔대, 참나물 그리고 참취가열즙(加熱汁)은 $Fe^{2+}$의 공존시 DNA 절단활성(切斷活性)을 억제(抑制)시키는 결과(結果)를 얻었다. Benzo${\alpha}$pyrene에 대한 돌연변이(突然變異) 억제작용(抑制作用)에서는 시료(試料) 모두 농도증가(濃度增加)에 따라 강(强)한 변이원(變異原) 억제작용(抑制作用)을 나타내었다.

머위 분획물의 In Vitro 라디칼 소거능 및 신경세포의 산화적 스트레스 보호 효과 (In Vitro Radical Scavenging Effect and Neuroprotective Activity from Oxidative Stress of Petasites japonicus)

  • This study was focused on the evaluation of radical scavenging effect and the protective activity against oxidative stress of the extract and fractions from Petasites japonicus. P. japonicus was extracted with methanol and then fractionated into 4 fractions [n-butanol, ethyl acetate (EtOAc), methylene chloride, and n-hexane]. The extract and fractions showed strong 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity. Among all the fractions, particularly, the EtOAc fraction showed the strongest effect with the $IC_{50}$ value of $0.02{\mu}g/ml$. In addition, the fractions also showed strong hydroxyl radical scavenging activity and nitric oxide scavenging activity as well. Furthermore, cell viability generated by the P. japonicus extract and 4 fractions were examined under C6 glial cellular model. The C6 glial cells showed high generation of reactive oxygen species (ROS) and decrease in cell viability by the treatment generator of hydrogen peroxide. However, the production of ROS formation was decreased by the treatment of the fractions of P. japonicus and also founded that the EtOAc fraction led to significant increase in the cell viability at concentration $100{\mu}g/ml$. Results from this work indicated that P. japonicus showed protective effects against oxidative stress and its EtOAc fraction may be served as a useful natural antioxidant.
