Species Composition and Biomass of Marine Algal Community on the Mid-Western Coast of Korea (서해 중부 연안 해조군집의 종조성과 생물량)

  • The species composition and biomass of intertidal benthic algae at 12 widely dispersed stations representative of mid-western coast of Korea were studied. A total of 151 species of marine algae including 19 Cyanophyta, 20 Chlorophyta, 32 Phaeophyta and 80 Rhodophyta is listed. Three species were found at all 12 stations: Sargassum thunbergii, Stylonema alsidii and Gracilaria verrucosa. Biomass per unit area exhibited a wide range of variation, ranging from a low of 41 g dry $wt{\cdot}m^{-2}$ at both Paeksajang and Taechon to a high of 549 g dry $wt{\cdot}m^{-2}$ at Uihangri, the northernmost station of the study area. The latter value is higher than those reported from any other stations in western coast of Korea. Biomass was composed largely of Phaeophyta at nearly every station. Sargassum thunbergii was dominant over the study area. The distribution of Corallina spp., however, is disjunct, since those occur as dominant species in biomass at the northern stations and to a lesser extent at the southern stations, but are nearly absent from the middle part of the study area.

Endophytic Fungal Diversity Isolated from the Root of Halophytes in Taean Peninsula (태안반도에 자생하는 염생식물의 뿌리로부터 분리한 내생 진균의 다양성)

  • Halophytes of seven species, Carex scabrifolia Steud., Limonium tetragonum Bullock, Salicornia europaea L., Suaeda glauca Bunge, Suaeda japonica Makino, Suaeda maritima Dumort., and Triglochin maritimum L. were collected from the Taean Peninsula. Thirty-seven endophytic fungi were isolated from the root of halophytes, and analyzed using the DNA sequences of internal transcribed spacers (ITS). The diversity of all endophytic fungi was analyzed using diversity indices. The endophytic fungi associated with the halophytes belonged to seven orders; Eurotiales (78%), Capnodiales (5%), Hypocreales (5%), Agaricales (3%), Corticiales (3%), Glomerellales (3%), and Pleosporales (3%). At the phylum level, the endophytic fungi were composed of Ascomycota and Basidiomycota. At the genus level, Alternaria, Aspergillus, Cladosporium, Paecilomyces, Penicillium, Phanerochaete, Schizophyllum, Talaromyces, and Verticillium were confirmed. Among them, Penicillium was the most abundant in the roots of the halophytes. This study analyzed the distribution and diversity of endophytic fungi on halophytes in the Taean Peninsula.

Anti-obesity effect of Korean Hamcho (Salicornia herbacea L.) powder on high-fat diet-induced obese rats (고지방식이로 유도한 비만 흰쥐에서 함초 (Salicornia herbacea L.) 분말의 항비만 효과 탐색)

  • Purpose: The objective of this study was to examine whether Hamcho freeze-dried powder and Hamcho freeze-dried nano powder have an anti-obese effect on high fat diet-induced obese rats. Methods: Sprague Dawley rats were divided into 6 different groups, each of which was bred for 8 weeks with a different experimental diet: normal diet group (NC), high fat diet group (HFC), high fat diet + 2% Salicornia herbacea L. freeze-dried powder group (2% HS), high fat diet + 5% Salicornia herbacea L. freeze-dried powder group (5%HS), high fat diet + 1% Salicornia herbacea L. freeze-dried nano powder group (1% HSN), high fat diet + 2% Salicornia herbacea L. freeze-dried nano powder group (2% HSN). Results: The 5% HS group showed a weight loss effect in body weight, liver and adipose tissue by reducing the amount of dietary intake and food efficiency ratio. In addition, the 5% HS group showed a significant reduction of serum leptin concentration, while having a beneficial effect on the improvement of lipid metabolism such as increase the serum concentration of adiponectin. However, Hamcho freeze-dried nano powder did not show a weight loss effect. Conclusion: Overall data indicated that Hamcho seemed to have anti-obesity effects. In particular, consumption of 5% Hamcho freeze-dried powder might have beneficial effects on body weight, serum adiponectin level, and lipid profiles since it led to reduced body weight and growth of adipose tissues by suppressing dietary intakes. However conduct of further research studies is needed for analysis of active components and biochemical action mechanism of Hamcho.

In Vitro Culture of Immature Embryo Obtained by Crossing between Tetraploid Grape 'Fujiminori' and Triploid 'Summer Black' (포도 4배체 '후지미노리'와 3배체 '썸머블랙'의 교배로 얻은 미숙배의 기내배양)

  • For the germination and differentiation of immature embryos obtained by artificial crossing between tetraploid grape 'Fujiminori' (Vitis vinifera ${\times}$ V. labruscana) and triploid 'Summer Black' (V. labruscana ${\times}$ V. vinifera), were incubated in vitro using MS medium supplemented with $GA_3$ or coconut water (CW) at various concentrations. The percentage of embryo formation of 'Fujiminori' ${\times}$ 'Summer Black' was 64.3%. Embryo germination percentage was higher than 95% in all the $GA_3$ treatments at the concentrations of 0.01, 0.05, 0.25, and $1.25mg{\cdot}L^{-1}$. However, only 15.8-31.6% of the germinated embryos successfully developed into normal plantlets. At higher concentration of $GA_3$, the plantlets developed infirm hypocotyls with over elongated and less enlarged structure. Among the treatments of CW at the concentrations of 5, 10, 15, and 20% (v/v), 10% and 15% were more effective and plantlet achievement percentage were 68.4 and 66.7%, respectively. The addition of 10% CW was most effective to obtain plantlets with optimal shoot length, node and root numbers. 15% CW was suitable to obtain plantlets with longer roots. Accordingly, the embryo culture using the MS medium supplemented with 10-15% CW was observed to be more efficient for germinating and growing the immature embryos produced from artificial crossing between tetraploid grape 'Fujiminori' and triploid 'Summer Black'.

Development of Female Gametophyte of Six Species of Corydalis (Fumariaceae) (현호색속(현호색과) 6종의 자성배우자체 발달과정)

  • The development of the female gametophyte of Corydalis albipetala, C. ambigua, C. filistipes, C. nobilis, C. solida, C. ophiocarpa have been comparatively investigated using laser scanning confocal microscope (LSCM) and light microscope. An archesporium was originated from one of the outmost parietal cells beneath the one-layered epidermis of protuberant nucellus, and acted directly as a megaspore mother cell (MMC). These species had linear tetrads after successional meiotic division during the megasprogenesis. A functional megasprore developed from one of the tetrad in the chalazal end, and the rest three being degenerated. The developmental type of the female gametophyte was monosporic in accordance with the Polygonum type. Prior to anthesis the female gametophyte was organized. So mature embryo sac was comprised a three-celled egg apparatus, three large antipodals were developed from the apex of each antipodal cell, and extended toward micropylar end to be contacted with egg apparatus. Two synergids were usually observed as degenerated condition, and in this time the apices of antipodal haustoria were connected with the degenerated synergids. The developmental characteristics of seven-nucleate female gametophytes were common in all the species investigated. But the shape of mature embryo sac was ovoidal in C. albipetala, C. filistipes, C. ophiocarpa and C. solida, reflexed in C. ambigua, and rather flattened ovoidal in C. nobilis. Also, the type of megasporangium was anatropous in all the species except C. ambigua with campylotropous ovule.

Characteristic of Glasswort (Salicornia Herbacea L.) Mixture Fermentation Utilizing Aspergillus oryzae (황국균을 이용한 퉁퉁마디(Salicornia herbacea L.) 혼합물의 발효 특성)

  • In this study, we investigated the quality of glasswort (Salicornia herbacea L.) mixture fermented by Aspergillus oryzae at $30^{\circ}C$ for 7 days. Changes of pH, titratable acidity, amino-nitrogen content, reducing sugar content, activities of $\alpha$-amylase and protease and number of mold were determined in the course of the fermentation. Angiotensin converting enzyme (ACE) inhibition activities and electron donating ability (EDA) were measured after 7 days fermentation. The pH lowered from 6.19~6.22 into the level of 5.41~6.08 after fermentation for 7 days. Titratable acidity increased from 0.59~0.68 into the highest level of 0.95~1.13% after 6 days fermentation. Furthermore, the amino-nitrogen content increased from 128.0~167.2 mg% to 159.4~209.0 mg% after fermentation for 7 days. For the reducing sugar content, it decreased from 2.0~5.9% into the level of 0.4~1.1% during 7 days fermentation. The number of molds decreased from $10^7\;CFU/g$ to approximately $10^6\;CFU/g$ after 5 days fermentation. $\alpha$-Amylase activity showed from the beginning of the fermentation in all samplings, but protease activity reached the value of food code standards after 5 days of fermentation. ACE inhibition activities were slightly increased from 9.5~16.6% to 19.3~22.7% and electron donating ability were slightly increased from 55.6~57.8% to 60.9~62.7% after 7 days of fermentation.

Status of Paddy Weed Flora and Community Dynamics in Korea (한국(韓國)의 논잡초분포(雜草分布) 및 군락현황(群落現況))

  • Nationwide weed survey of paddy rice field conducted in 1981 and 1971 was compared and determined major dominant weed community types distributed by province. Based on the similarity coefficients between 1971 and 1981 the floristic composition based on the degree of dominance was greatly dissimilar while kind of weed flora were not much differed. Degree of dominance were concentrated to perennial weeds in 1981 while these were at animal weeds in 1971. The moat important 10 weed species and their dominance in 1981 were Monochoria vaginalis Presl.(22.2%), Sagittaria pygmaea Miquel(17.5%), S. trifolia L.(9.0%), Poramogeton distinctus Benn.(9.0%), Cyperus serotinus Rottb.(8.5%), Rotala indica Koehne(6.0%), Aneilema japonica Kunth(4.4%), Lindernia procumbens Philcox(3.9%), Eleocharis kuroguwai Ohwi(3.4%) and Ludwigia proarrara Roxb(3.0%), respectively while these for 1971 were R. indica(34.5%), Eleocharia acicularis Roem, et Schult (11.9%), M. vaginalis(11.1%), Cyperua difformis L.(8.7%), Echinochloa crus-galli Beauv.(6.9%), L. procumbens(3.3%), P. distinctus(3.1%), A. Japonica (2.4%), E. kuroguwai(1.8%) and P. hydropiper(1.8%), respectively. Weed occurrence was also closely related with soil type, cropping pattern and cultural practices. Particularly, the occurrence of P. distincrus was negatively correlated with the degree of land utilization. Weed community types dafined by two-dimensional ordination analysis were 11 for Jeonnam province, 9 for Gyeoaggi, Gangweon, Chungbuk and Gyeongnam provinces, 8 for Jeonbuk and Gyeongbuk provinces, 7 for Chungnam province and 4 for Jeju province, respectively.

Changes in Early Stage Vegetation Succession as Affected by Desalinization Process in Dae-Ho Reclaimed Land (대호 간척지의 제염진해에 따른 초기 식생 변화)

  • In order to obtain the reference data to be used as an vegetation index for degree of desalinization, vegetation successions were surveyed and soil chemical characteristics were analyzed at the naturally maintained plot in Dae-Ho reclaimed land. Seven Groups $(A\;{\sim}\;G\;group)$ were classified as major vegetation; A group (Suaeda asparagoides MAKINO etc.), B group ( Aster tripolium L. etc.), C group (Stearia viridis L. BEAUV), D group ( Phragmites longivalvis STEUD), E group (Limonium tettagonum THUMB A. A. BULLOCK), F group (Trifolium repens L.), G group (Zoysia sinca HANCE etc.). As desalinization process proceeded, the wild vegetation changed in order of $A\;{\to}\;B\;group$, $D\;{\to}\;C\;group$, $E\;{\to}\;F$, G group. Soil texture of the naturally maintained plot was silt loam and soil fertility was very low compared with agricultural cultivated soil. Soil pH ranged from 7.0 to 8.0. Electrical conductivity (ECe) was below 10 and $20\;dS{\cdot}m^{-1}$ at top and subsoil, respectively, except the plot where A group were growing. Resulting from SAR and ECe, The plot where A group was growing was saline-sodic soil and the others were saline soil. The relation between vegetation sucession and soil desalinization showed that vegation appeared under $10\;dS{\cdot}m^{-1}$ of ECe and 15 of SAR except A group.

Genotypic Effect of Watermelon (Citrullus lanatus Thunb.) on Organogenesis from Shoot Tip Culture of Seedlings (수박의 유묘 정단 배양시 유전적 차이가 기관 형성에 미치는 영향)

  • The genotypic (2n, 3n, 4n) response of watermelon in vitro shoot tip culture was evaluated. Different genotypes had similar response in terms of shoot formation and growth. Shoot formation was better at lower concentration of 0.3 mg/L BA and higher concentration of 5-10.0 mg/L 2iP and kinetin, but growth of newly formed shoot was inhibited. With further subculture, kinetin did not promote shoot formation Better shoot formation was observed at 0.3-0.5 mg/L BA. Combination of 0.3 mg/L BA and 0.3-0.5 mg/L BA was effective in shoot multiplication, growth and induction of more internodes. Varrying levels of light intensity and agar concentration did not affect the performance of tetraploid plants. Higher light intensity and agar concentrations decreased the number of shoot formed in triploid plane. Growth in both genotype, however was inhibited. Higher light intensity was found to promote leaf senescence in all genotypes. All growth inhibitors decreased the number of shoots formed and slowed plant growth there by prolonging duration of cultures. Growth inhibitors were to observed to decrease incidence of hyperhydricity in culture. No difference in shoot formation was observed in each of the concentrations used in Ancymidol, TIBA, CCC and PP333. Shoot formation and growth was more inhibited in ABA treatments. Leaf expansion and growth was poor in all treatments.

Effect of Water Soluble Silicate Fertilizers on Stem Strength and Yield of Paddy Rice (수용성 규산질 비료의 시용에 의한 벼 줄기 강도 강화와 수량에 대한 효과)

  • A field experiment was conducted to evaluate effect of water soluble silicate fertilizer (WSS) application on rice plants with respect to comparing with powdery slag-originated silicate fertilizer (PSS) and granular one (GSS). The 30-day seedlings were transplanted on May 10, 2012. The plot size was $25m^2$, and the planting density was 15 hills $m^{-2}$. The standard application level was $2kg\;ha^{-1}$ for WSS, $200kg\;ha^{-1}$ for GSS, $200kg\;ha^{-1}$ for GSS. The application rates were 50 %, 100 %, and 200 % of the standard levels. The soil and plant samples were taken after harvest on September 10. Strength weight of the stem was measured on the center of the 5-cm of the fourth internode. Nutrient contents and yield of grains and were evaluated. The strength weight of the stem was positively correlated with the silicate content of the stem with the highly significant $R^2$ of 0.601. The strength of the stem was satisfactorily enforced by application of 50 % WSS and GSS, and 100 % PSS. Application of 50 % or 100 % of WSS showed little difference in rice yield in comparison with application of 100 % of PSS or GSS. Therefore, application of $20kg\;ha^{-1}$ of WSS would be recommendable for rice cultivation which enforced stem strength, and increased yield of rice.
