This study was conducted to investigate the effect of dietary supplementation levels of spinach powder and extract on performance, egg quality and lutein content in egg yolk. Laying hens were subjected to one of the following treatments; C, basal diet (BD); T1, BD + 1 ppm lutein from spinach powder; T2, BD + 1 ppm lutein powder from spinach extract; T3, BD + 2 ppm lutein powder from spinach extract; T4, BD + 2 ppm lutein from spinach extract. As a result, performance of laying hens was no significant difference among treatments. T4 had a significantly (p<0.05) higher egg yolk color than the other treatments. As from the lutein content in egg yolk, T4 had a significantly (p<0.05) higher content than the other treatments. Finally, these results suggested that the dietary supplementation for lutein-rich egg production, lutein concentration must be higher than a minimum of 2 ppm. Also, dietary with lutein extract from raw materials was appropriate for lutein-rich egg production than dietary with raw materials.
This experiment was conducted to investigate the effects of dietary supplementation with lutein-containing materials on meat quality and lutein accumulation in broiler tissue. Broilers were subjected to one of the following treatments: C, basal diet (BD); T1, BD + 2.223% lutein from spinach extracted by ethanol fermentation; T2, BD + 2% radish green powder; T3, BD + 0.61% spinach powder; and T4, BD + 1.83% spinach powder. The weight gain, feed intake, and feed conversion did not differ among treatments. An evaluation of the color of the chicken breast meat revealed that the CIE $L^*$ value of the control was significantly (p<0.05) lower than that of the other treatments, whereas T4 had a significantly (p<0.05) lower CIE $a^*$ value and a higher (p<0.05) CIE $b^*$ value than the other treatments. The lipid oxidation (thiobarbituric acid reaction substances, TBARS) value was not significant among treatments. The results of a high performance liquid chromatography analysis revealed that the lutein peak was present only in the T4 liver tissue. These results showed that spinach powder (T4) affected meat color (CIE $a^*$ and $b^*$) however, TBARS and lutein accumulation were not affected.
A new HPLC method for determining malonaldehyde content in lipid peroxidation systems was developed and antioxidant activities of $\alpha$-, $\beta$- carotene, lutein and lycopene were compared by a newly developed HPLC analysis and by TBA value. In addition, malonaldehyde forming ability of rat liver microsome was determined depending on thawing numbers. As results, malonaldehyde was eluted at a retention o f5.60 min and showed a linear relationship between peak area and concentration in standard curve. The MA content of microsome decreased with thawing numbers possible by destruction of cellular membranes. Lycopene, lutein and $\alpha$-carotene showed stronger antioxidant activities than $\beta$-carotene of DL-$\alpha$-tocopherol both in Fe+3-ADP/NADPH and in paraquat/NADPH system. The inhibitory effects of carotenoids and DL-$\alpha$-tocopherol on Fe+3-ADP/NADPH lipid peroxidation system was similar by TBA value and by the HPLC analysis for malonaldehyde.
This study aimed to analyze charantin and lutein from fresh bitter melon (FBM) for their antioxidant and ${\alpha}$-glucosidase inhibitory activities. We subsequently compared our results with charantin and lutein content of pickled bitter melon (PBM11, PBM13, PBM51, and PBM53), obtained by brining (1 and 5% salt) and blanching (1 and 3 min). Charantin, lutein, $\text\tiny{L}$-ascorbic acid, and total polyphenolic content in FBM were $777.92{\pm}27.59$, $16.87{\pm}2.07$, $94.78{\pm}0.61$, and $22.07{\pm}0.74mg$ catechin equivalents/g on dry basis, respectively. Antioxidant activity of 5 mg/L FBM extract was $31.76{\pm}0.42%$ in ABTS assay and $56.19{\pm}2.39%$ in DPPH assay; ${\alpha}$-glucosidase inhibitory activity of 0.5 mg/L FBM extract was $55.60{\pm}1.34%$. Charantin and lutein content of all PBM samples were higher than control without brining and blanching treatments, those in PBM53 being $375.63{\pm}9.03$ and $4.07{\pm}0.40mg/100g$, respectively. These showed that brining and blanching in pickling process were very effective in retaining charantin and lutein in bitter melon.
The Daecha-12 and Yabukita varieties of powdered green teas were grown under 85% shade-cultivated condition, and their physicochemical properties were analyzed. Total catechin content was not significantly different but, non-gallated catechin content in Daecha-12 was significantly higher than that in Yabukita. Theanine (32%), caffeine (14%), lutein (15%) and total chlorophyll (28%) levels were significantly higher in Daecha-12 than those in Yabukita. The results of a color analysis showed that the lightness L-value of Yabukita was higher than that of Daecha-12 but, that the greenness negative a-value and b-values of Daecha-12 were higher than those of Yabukita. The Daecha-12 cultivar had lower catechin content but higher content of theanine, caffeine, theobromine, lutein, chlorophyll, and a negative a-value than those of the Yabukita cultivar. Thus, the Daecha-12 cultivar is suitable to prepare a high-quality powdered green tea product.

Entrapment of hydrophobic flavonoid luteolin into ethosome was carried out for improving its stability and making practical application in the field of drug and cosmetics. The formation of liquid crystalline phase and its thermal properties were investigated by polarized optical microscopy and DSC. The phase inversion from W/O to W/O/W was detected by conductivity change with the addition of PBS buffer solution into the ethanol-dissolved lecithin mixture. The particle size change of ethosome with constituent composition was examined, which showed that the incorporation of luteolin into lecithin up to 10% had little effect on the size of ethosome. Enhancement of stability of luteolin by entrapment into ethosome was verified through DPPH test. The stabilization efficacy of ethosome was improved further by the addition of tocopherol.
This study was conducted to evaluate dietary effects of Marigold extract on laying performance, egg quality, oxidative stability of egg yolk and lutein transfer into chicken eggs. A total of one-hundred eighty nine 55-wk-old Hy-Line Brown layers were divided into seven groups and fed control diet or each experimental diet containing 0.1, 0.3, 0.5, 1.0, 1.5 or 2.0% Marigold extract. Egg production, egg weight and daily egg mass were not affected by dietary treatments. The yolk colors in groups fed diets containing Marigold extract were significantly higher than that of control. The Haugh unit were tended to be improved by feeding of diets containing Marigold extract although there were no significant difference in egg shell strength and thickness. The MDA (malondialdehyde) contents in groups fed diets containing Marigold extract above 0.5% were significantly reduced than that of control. After 14d of storage, the Haugh unit values in groups feed diets containing 0.3 and 1.0% Marigold extract were significantly higher than that of control (p<0.05). The concentration of lutein in egg yolk increased by feeding of Marigold extract. When 2% Marigold extract was supplemented to the diet, lutein content of egg was increased as much as 1.71 mg/60 g. These results indicated that the use of Marigold extract in layer diets was effective in egg quality and for the production of lutein fortified eggs.
Background: Luteolin, a flavone found in various Chinese herbal medicines is known to possess anti-inflammatory properties through its ability to inhibit various proinflammatory signaling pathways including NF-${\kappa}B$ and p38 MAPK. In this study, we investigated the potential therapeutic effect of luteolin on dextran sodium sulfate (DSS)-induced colitis. Materials and Methods: We used a transgenic mouse model expressing the enhanced green fluorescent protein (EGFP) under the transcriptional control of NF-${\kappa}B$$cis$-elements. C57BL/6 NF-${\kappa}B^{EGFP}$ mice received 2.5% DSS in their drinking water for six days in combination with daily luteolin administration (1mg/kg body weight, 0.1ml vol, intragastric) or vehicle. NF-${\kappa}B$ activity was assessed macroscopically with a Charge-Coupled Device (CCD) camera and microscopically by confocal analysis. Results: A significant increase in the Disease Activity Index (DAI), histological score (p<0.05), IL-12 p40 secretion in colonic stripe culture (p<0.05) and EGFP expression was observed in luteolin and/or DSS-treated mice compared to water-treated mice. Interestingly, a trend toward a worse colitis (DAI, IL-12p40) was observed in luteolin-treated mice compared to non-treated DSS-exposed mice. In addition, EGFP expression (NF-${\kappa}B$ activity) strongly increased in the luteolin-treated mice compared to control mice. Confocal microscopy showed that EGFP positive cells were primarily lamina propria immune cells. Conclusions: These results suggest that luteolin is not a therapeutic alternative for intestinal inflammatory disorders derived for primary defects in barrier function. Thus, therapeutic intervention targeting these signaling pathways should be viewed with caution.

Soybean [Glycine max (L.) Merr.] is grown worldwide for its high protein and oil content. Anthocyanins from black soybean seed coats are known to have many pharmaceutical effects. Soybean cultivars with large seed sizes and black seed coats are needed by soybean farmers. However, antinutritional factors, like protein, stachyose, and Kunitz trypsin inhibitor (KTI) exist in raw mature soybeans. Genetic elimination or reduction of these components is needed in soybean breeding. The objective of this research was to develop new a soybean strain with black seed coats and green cotyledons that was KTI protein free and low in stachyose. Six parents were used. The presence or absence of KTI protein was detected using the Western blot technique. The content of stachyose in mature seeds was detected using HPLC. One new strain was selected from 11 $F_2$ plants with black seed coats and green cotyledons that lacked KTI protein. The new strain had black seed coats and green cotyledons and was KTI protein free and low in stachyose. The plant height of the new strain was 66 cm, and its 100-seed weight was 28.4 g. The stachyose content of the new strain was 2.59 g/kg. The new strain developed in this research will be used to develop new cultivars that are KTI protein free and low in stachyose.
