  • In the study about the logical basis and interpreting methods of animal experiments using rats in traditional medicine, several conclusions are obtained and summarized as following. 1. The logical basis of the animal experiments in traditional medicine is the essential homogeneity between human and rat defined as various transformation of one Qi(一氣) packed the cosmos. 2. Morphologically, the rat has abundant Yin(陰) and less Yang(陽) in most of anatomical characteristics. 3. Physiologically, the rat has unstable heart and mild temper with good fertility, which shows the features of Yin animal. 4. Pathologically, the rat shows the pathology of injury of viscera by stresses(氣激傷臟) and pathological transformations of internal water(痰飮水濕) mainly. 5. Constitutionally, the rat is alike to water type man(水形人) or Soeumin(小陰人) out of Yin Ren(陰人). 6. So, the rat is the proper experimental animal for diseases of sputum and water, emotional stimulations, endocrine system, heart, kidney, Yin syndrome(陰證) etc..

Antinociceptive Efficacy of Korean Bee Venom in the Rat Formalin Test (랫드의 포르말린 시험에서 한국산 봉독의 항통각 효과)

  • The study was performed to assess the antinociceptive efficacy of Korean bee venom(BV) in the rat formalin test. Korean BV was collected using BV collector devices in which an electrical impulse is used to stimulate the worker bee(Apis mellifera L.) to sting and release venom. After collection, whole Korean BV was evaporated until dry using BV collector. Experiments were performed on male Sprague-Dawley rats(weighing 260-270, 6 weeks old). Rats divided into 4 groups, each comprising 8 rats. BV was diluted and amounts of 6 mg/kg body weight(BW), 0.6 mg/kg BW and 0.06 mg/kg BW were tested. BV was subcutaneously injected to produce an antinociceptive effect and the antinociceptive efficacy was evaluated using a rat formalin test. BV was subcutaneously injected into an acupoint(Zusanli, ST36) at 15 min prior to 1% formalin($50{\mu}l$) injection. The antinociceptive effect observed during 60 min following formalin administration. BV produced antinociceptive efficacy from 10 to 60 min after formalin injection. The antinociceptive efficacy of Korean BV showed a dose-dependent response. These results suggest that Korean BV may be a suitable and preferred choice for antinociceptive efficacy in pain management.

  • This study was performed to know the effect of depolymerized alginate obtained by hydrolysis of alginate through a heating process at $121^{\circ}C$ on intestinal environment, Rats were fed with diets containing $1\%$, $5\%$, and $10\%$ of each depolymerized alginate (HAG-10, HAG-50, HAG-100 and alginate) for 35 days, The changes of weight, moisture content, pH and volatile basic nitrogen (VBN) of fecal, and a short chain fatty acids (SCFA) were checked in the rats. The fecal weight and moisture content were the highest in rats fed with alginate diets (p<0.01), followed by HAG-100, HAG-50 and HAG-10 in order. The $5\%$ of HAG-50 diets induced a significant increase in contents of protein and lipid of feces, resulting in the decrease of apparent digestibility of protein and lipid (p<0.01). The pH and VBN content in feces of the rats decreased in $5\%$ and $10\%$ of HAG-50 diets, but $10\%$ of HAG-100 diets; $5\%$ and $10\%$ of alginate diets brought about an increase of fecal pH and VBN (p<0.01), The amount of n-butyric acid in feces was increased while propionic and acetic acid contents decreased significantly (p<0.01) in diets containing $5\%$ and $10\%$ HAG-50. However, the feces of rat fed diet containing $5\%$ and $10\%$ alginate showed a tendency to being opposite in results than that of HAG-50.

  • Gram 음성균의 세포벽 성분인 lipopolysaccharide (LPS)는 생체내에서 각종 생리 활성 물질의 생성을 증진 시킴으로써 내독성 shock등의 병변을 유발하는 것으로 알려져 있다. 이중 prostaglandin (PG)합성 증진 작용에 대해서는 많은 보고가 이루어져 있으나 그 자세한 작용 기전이나 특성등에 대해서는 아직 많은 연구가 되어 있지 않다. 따라서 본 연구에서는 랫드의 폐포 macrophage를 대상으로 하여 LPS의 PG 합성 증진 작용에 대한 특성을 확인코자 하였다. 우선 LPS에 의한 PG 합성 profile을 시간별로 확인한 결과 처리 6 시간 이후에 현저한 합성 증가를 관찰하였으며, 이는 주로 cyclooxygenase의 활성 증가에 기인하는 것으로 추정되었다. LPS는 짧은 시간 동안의 처리에 의해서도 PG 합성을 증진 시켰으나, 양적인 면에서, 장시간 처리에 비해 극히 적었고, 이 작용은 phospholipase $A_2$ 활성 증가에 기인하는 것으로 추정 되었다. LPS에 의한 PG합성 증진 작용은, TNF나 PAF의 생성을 매개로 하지는 않았으며, serum의 존재가 필수적임을 확인하였다. EGF, PDGF 등의 growth factor들은 그 자체로는 PG 합성을 유의적으로 증가시키지는 않았으나 LPS와의 병용 처리에 의해 어느 정도의 증진 작용을 나타내었다. 또한, 정상 랫드 serum도 LPS와의 병용 처리에 의해 PG 합성을 현저히 증가 시켰으며, 따라서, LPS는 serum 중의 각종 growth factor 외에도 LBP등의 serum factor들과의 상호 작용을 통하여 PG합성 증진 작용을 나타내는 것으로 추정 되었다.

  • The purpose of this experiment is to investigate the feasibility of extracts (sample extract, MSE) mixed with hot - water extracts as health functional nutritional foods for prevention and treatment of liver damage. The experimental method was to investigate the effects of carbon tetrachloride on liver damage in rats by 60% of liver cirrhosis level. The effect of mixed extracts (MSE) was investigated by measuring the physiological functions, biochemical functions, and molecular biologic TGF - ${\beta}$ and P53 levels in liver - injured rats. with MSE doses administered differently(0.50ml, 0.75ml). A total of 30 rats were used for normal group, control group, positive control group, experimental group 1, and experimental group 2. The rats were given 6 times a week for 6 weeks at a fixed time once a day. The results of the experiment were statistically significantly higher in the control group than in the normal group and the positive control group, and statistically significantly lower in the experimental group (1 and 2) than the control group. There were some significant differences in the comparison between the experimental groups (1 and 2), but no significant changes were found. In conclusion, the results of this study are expected to be useful as functional food, and the variation of physiological activity and the degree of liver molecular biotechnology will be used for the research of the same field.

Beneficial effect of Polygoni Multiflori Radix in high fructose diet-induced metabolic syndrome rat model (고과당식이 랫드모델에서 적하수오 투여에 의한 대사증후군 개선효과)

  • Objectives : Polygoni Multiflori Radix (Jeokhasuo in Korean) is a Oriental traditional herbs widely used in East Asian countries. Overconsumption of fructose results in hypertension, dyslipidemia, obesity and impaired glucose tolerance which have documented as a risk of cardiovascular diseases. This experimental study was designed to investigate the beneficial effects of an ethanol extract from Polygoni Multiflori Radix (PMR) in high-fructose (HF) diet-induced metabolic syndrome rat model. Methods : Sprague-Dawley (SD) rats were divided into three groups; Control group, receiving regular diet and tap water, HF group, and HF + PMR group both receiving supplemented with 65% fructose (n=10), respectively. The HF + PMR group initially received HF diet with PMR (100 mg/kg/day) for 8 weeks. Results : PMR significantly prevented the metabolic disturbances such as hyperlipidemia, hypertension and impaired glucose tolerance. Chronic treatment with PMR significantly decreased body weight, fat weight and adipocyte size, suggesting a role of anti-obesity effect. PMR led to improve the hyperlipidemia through the increase in HDL cholesterol level as well as the decrease in triglyceride and LDL cholesterol level. In addition, PMR suppressed adhesion molecules and endothelin-1 (ET-1) expression in aorta resulting in the decrease of hypertension. In muscle tissue, PMR significantly recovered the HF-induced insulin resistance through increase of insulin receptor substrate-1 (IRS-1), p-$AMPK{\alpha}1/2$, and p-Akt expression. PMR improved HF-induced metabolic disorders and its action was caused by energy metabolism-mediated insulin signaling activation. Conclusions : These results demonstrate that PMR may be a beneficial therapeutic for metabolic syndrome through the improvement of hyperlipidemia, obesity, insulin resistance and hypertension.

The Effects of Gobonyangjeonbang Administration on Antioxidative of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) induced Rats. (고본양정방 투여가 TCDD 유도 랫드의 항산화 변화에 미치는 영향)

  • The purpose of this study was to evaluate the effects of gobonyangjeonbang (GYB) on the endocrine function and the antioxidant efficacy of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD)-induced oxidative stress in rats. In 2017, to evaluate the efficacy of GYB on oxidative stress, 35 male SD rats were divided into five groups and tested. The normal control group was administered saline as a vehicle, while the TCDD-alone group was administered TCDD (2 ㎍/kg per week) intraperitoneally and with physiological saline, and the test group was administered GYB orally by dividing it into three concentrations (75, 150, and 300 mg/kg) for six weeks. Bodyweight decreased significantly after six weeks of TCDD exposure, when compared to rats in the NC group (p<0.001). However, weight loss from TCDD was significantly protected by administration of GYB at 300 mg/kg (p<0.01). The rat liver induced by TCDD showed cytoplasmic vacuole degeneration, and the hepatic sinusoid and weight increased. As a result of measuring MDA and SOD, both items tended to decrease under TCDD administration. On the other hand, there was no change due to GYB administration, and significance was observed in the GYB 300 mg/kg group compared to the NC group in the SOD result (p<0.05). These findings demonstrated that GYB may have a protective effect against TCDD-induced liver toxicity in rats.

  • This study was performed to know the hypocholesterolemic effect of depolymerized alginate obtained by hydrolysis of alginate through a heating process at $121^{\circ}C$. The changes of total cholesterol, HDL-cholesterol, LDL-cholesterol, triglycerides and phospholipid levels in serum, as well as the atherogenic index were elucidated in rats for 35 days. In order to evaluate single-dose toxicity of HAG-50 in rats via oral routes, induction any mortalities and abnormal signs in clinical finding, body weight, gross findings and histopathological finding, and estimation of $LD_{50}$ were elucidated. Total cholesterol, HDL-cholesterol, LDL-cholesterol, triglycerides and phospholipid levels in the serum significantly decreased and resulted in the decrease of atherogenic index in rats fed diets containing $5\%\;and\;10\%$ HAG-50 (p<0.01). HAG-50 did not induce any mortalities and abnormal signs in clinical findings, body weights, gross findings and histopathological findings in single-dose toxicity test via oral routes of rats, after which it was impossible to estimate $LD_{50}$ values.

Acute Toxicity Study on Sipjeondaebo-tang in Rats (SD 랫드를 이용한 십전대보탕의 급성 독성 연구)

  • Sipjeondaebo-tang has been traditionally prescribed a medicine as a restorative. In this study, we investigated the acute toxicity about water-extracted Sipjeondaebo-tang. Thirty rats completed 14 days of oral Sipjeondaebo-tang at the respective doses of 0(control group), 2000 and 5000 mg/kg. We observed survival rates, general toxicity, change of body weight and autopsy. To be confirmed the data for the toxicity and safety problems of oriental medicine prescription. Compared with the control group, we could not find any toxic alteration in all treated groups (2000 and 5000 mg/kg). LD50 of Sipjeondaebo-tang was over 5000 mg/kg and it is very safe to SD rats.
