Effect of Collection Times of Rumen Fluid on In vitro Dry Matter Digestibility of Forage Crops (반추위액 채취 시간이 사료작물의 In vitro 건물 소화율에 미치는 영향)

  • Object of this study were to determine the influence of collection times of rumen fluid on in vitro dry matter digestibility (IVDMD) of forage crops. The donor cow was fed concentrate once a day and given free access to grass-legume mixture hay. Main plot was consisted of different collection time of rumen fluid; T1: 1 hour before concentrate feeding, T2: 1 hour after feeding, T3: 4 hour after feeding and T4: 8 hour after feeding. A total of 7 samples of 4 different forage crops (barely, Italian ryegrass, crimson clover, rye) were used for the estimation of IVDMD and tested in three replicates. For the $DAISY^{II}$ incubation, each sample was inserted into each filter bag then heat-sealed and incubated in a digestion vessel for 48 h at $39^{\circ}C$. The times of rumen fluid collection had no significant effect on the IVDMD values over all varieties except for 2 breeds of IRG (Kogreen and Kospeed). IVDMD values with T1 over all varieties were slightly higher than other treatments, however those with both T1 of Kogreen and Kospeed varieties were significantly higher than T2 (p<.05).

  • Over 90 of lysine producing auxotrophs were obtained from Corynebacterium sp. S-27-12, Brevibacterium flavum ATCC 15168 and Micrococcus glutamicus ATCC 13032 by UV light, $Co^{60}$ irradiation and N-methyl-N'-nitro-N-nitrosoguanidine treatment. One of the mutant, Brev. flavum U46-N59, was identified as a leucine auxotroph and accumulated lysine during flask (500 ml) cultivation (180 strokes/min.) up to 21.6 mg per ml of broth at pH 7.5 and $28^{\circ}C$ after 4 days. The medium consisted of glucose, 100; urea, 10; corn steep liquor, 40; $KH_2PO_4,\;2;\;K_2HPO_4,\;0.5;\; MgSO_4.\;7H_2O,\;0.4;\;antifoam\;S-57,\;1g;\;Fe_2(SO_4)_3.XH-2O,\;10;\; MnCl_2,\;4H_2O,\;10mg;\;biotin,\;30;\;thiamine-HCl,\;100{\mu}g$in 1l of distilled water, and 40 U/ml of penicillin was added after 36 hrs fermentation.

Automatic Extraction of Roof Components from LiDAR Data Based on Octree Segmentation (LiDAR 데이터를 이용한 옥트리 분할 기반의 지붕요소 자동추출)

  • The 3D building modeling is one of crucial components in building 3D geospatial information. The existing methods for 3D building modeling depend mainly on manual photogrammetric processes by stereoplotter compiler, which indeed take great amount of time and efforts. In addition, some automatic methods that were proposed in research papers and experimental trials have limitations of describing the details of buildings with lack of geometric accuracy. It is essential in automatic fashion that the boundary and shape of buildings should be drawn effortlessly by a sophisticated algorithm. In recent years, airborne LiDAR data representing earth surface in 3D has been utilized in many different fields. However, it is still in technical difficulties for clean and correct boundary extraction without human intervention. The usage of airborne LiDAR data will be much feasible to reconstruct the roof tops of buildings whose boundary lines could be taken out from existing digital maps. The paper proposed a method to reconstruct the roof tops of buildings using airborne LiDAR data with building boundary lines from digital map. The primary process is to perform octree-based segmentation to airborne LiDAR data recursively in 3D space till there are no more airborne LiDAR points to be segmented. Once the octree-based segmentation has been completed, each segmented patch is thereafter merged based on geometric spatial characteristics. The experimental results showed that the proposed method were capable of extracting various building roof components such as plane, gable, polyhedric and curved surface.

  • For spatial analysis and decision-making based on territorial and urban information, technologies on 3D GIS with digital image data and photo-realistic 3D image models to visualize 3D modeling are being rapidly developed. Currently, satellite images, aerial images and aerial LiDAR data are mostly used to build 3D models and textures from oblique aerial photographs or terrestrial photographs are used to create 3D image models. However, we are in need of quality 3D image models as current models cannot express topographic and features most elaborately and realistically. Thus, this study analyzed techniques to use aerial photographs, aerial LiDAR, terrestrial photographs and terrestrial LiDAR to create a 3D image model with artificial features and special topographic that emphasize spatial accuracy, delicate depiction and photo-realistic imaging. A 3D image model with spatial accuracy and photographic texture was built to be served via 3D image map services systems on the Internet. As it was necessary to consider intended use and display scale when building 3D image models, in this study, we applied the concept of LoD(Level of Detail) to define 3D image model of buildings in five levels and established the models by following the levels.

Effect of Lactic Acid Bacteria and Chlorella on Nutritive Values and Quality of Italian Ryegrass-Alfalfa Silage (젖산균과 클로렐라 첨가가 이탈리안라이그라스-알팔파 혼파 사일리지의 사료가치 및 품질에 미치는 영향)

  • To provide the basic data on the utilization of chlorella as a method to improve the quality of forage, we examined the effect of the addition of lactic acid bacteria (LAB) and chlorella on the change of nutritive value, quality and microbes on Italian ryegrass and alfalfa mixture silage (IRG-Alfalfa silage). The nutritive values of IRG-Alfalfa silage were not significantly different in all treatments. The pH of IRG-Alfalfa silage decreased in both LAB and chlorella treatments. Further the content of lactic acid in the LAB treatment increased as compared to the control, and also significantly increased in the treatment of LAB with chlorella (p<0.05). In addition, the number of LAB increased in lactic acid bacteria treatment as compared to the control, and also increased in the treatment of LAB with chlorella (p<0.05). Therefore, we suggest that LAB and chlorella would improve the quality of IRG-Alfalfa silage.

Characteristics of seasoning pastes fermented by Aspergillus oryzae and Bacillus subtilis using edible insects (장류 미생물을 이용한 식용곤충 발효 조미페이스트 제조 및 품질특성)

  • The aim of this study was to develop a seasoning paste fermented by Aspergillus oryzae and Bacillus subtilis using three edible insects: Tenebrio molitor larvae, Gryllus bimaculatus, and Bombyx mori pupa. No significant changes in pH and titratable acidity were observed between the insect pastes and soybean control during a 5-week fermentation process. The amino nitrogen was 0.35-0.50% (w/w) in the early stage of the fermentation and increased to 0.72-0.78%. The total nitrogen was 2.36-3.62% (w/w) in the early stage and was preserved during the fermentation period. Free amino acids of the insect pastes were similar to those in soybean paste. In general, the fermented insect pastes showed a lower flavor and color than soybean paste but B. mori pupa-fermented paste did not show any significant difference in preference from soybean. Glutamate, aspartate, lysine, phenylalanine, arginine, and serine were found to affect taste preference. It was possible to ferment edible insect protein to produce a fermented seasoning paste like Korean doenjang.

Antidiabetic and Antioxidative Effects of Bitter Melon on Streptozotocin-induced Diabetic Rats (당뇨유발 흰쥐에 있어 여주분말의 항당뇨 및 항산화작용에 대한 연구)

  • The purpose of this study was to determine the antidiabetic and antioxidative effects of Bitter melon on streptozotocin-induced diabetic rats. The normal and the control groups were fed an AIG -93M diet, and the Bitter melon groups were fed 1%, 2% and 3% Bitter melon powder. After two weeks, the control and the experimental group were induced to a diabetic state with the administration of streptozotocin. The blood glucose control and antioxidant activity were analyzed after the animals were sacrificed. The blood glucose levels of all the Bitter melon groups were lower than those of the control group, and the 2% Bitter melon group showed significantly lower blood glucose levels than those of the control group. Serum Triglyceride and HDL-cholesterol of the 2%, and 3% Bitter melon groups were significantly lower than those of the control group. The total cholesterol levels of the bitter melon groups were significantly lower than those of the control group. The serum insulin levels of the induced groups were significantly lower than those of the normal group. The HbA1c levels of the 2% and 3% Bitter melon groups were significantly lower than those of the control group. For the level of antioxidant enzymes in the liver tissues, the 2% Bitter melon group was significantly higher than that of the control group. These results show the antidiabetic and antioxidative effects of Bitter melon for the prevention and treatment of diabetes.

Effect of Dietary Herb Medical Stuff on the Non-specific Immune Response of Nile Tilapia, Oreochromis niloticus (나일틸라피아, Oreochromis niloticus의 비특이적 면역반응에 대한 생약재 투여 효과)

  • In order to investigate the immune response induced by supplementation of herb medical stuff in diet on the nile tilapia. Oreochromis niloticus, experiments were performed with feeding of four different experimental diets supplemented with 2% ginseng. Panax ginseng, 3% Kugija. Lycium chinense, 3% Hasuo, Polygonum multiflorum, 2% Omija, Schizandra chinensis, respectively, for 84 days. The non-specific immune responses changed during the feeding period were investigated at 2, 4, 6, 8, 10, 12 weeks in each group. Average body weight of the nile tilapia with supplemented diets was heavier than control group. The fish fed on diet supplemented with 3% Kugija showed the better growth than the other tested groups. Complement activity such as complete hemolytic activity ($CH_{50}$) and bactericidal activity against Escherichia coli tended to be increased by the supplementation of herb medical stuff. The lysozyme activity of serum and adherent phagocyte activity showed higher in the fish fec on diet supplemented with 3% kugija than the other tested groups. In respect to the RPS against experimental Edwardsiella tarda infection, all of the group fed on the herb medical stuff in diet appeared higher response compared with control group. From these results, herb medical stuff (Panax ginseng, Lycium chinense. Polygonum multiflorum, Schizandra chinensis) might be used a additives of diet for the increasing of non-specific immune response or resistance against bacterial fish diseases.

Effects of diets supplemented with Yuzu Citrus junos Siebold ex Tanaka on disease resistance of olive flounder, Paralichthys olivaceus (유자첨가사료가 넙치의 질병저항성에 미치는 영향)

  • Effects of various concentration of yuzu Citrus junos Siebold ex Tanaka in the diets on growth, blood chemistry and disease resistance of olive flounder were determined. Fifteen hundred fish averaging 200~270g were fed on moist pellet containing yuzu at the concentrations of 0%(control), 2.5%, 5% and 7.5% for 3 months. After feeding trial, weight gain of fish fed 2.5% and 5.0% yuza diet was higher than those of fish fed 0% and 7.5% yuza diet but not significant(P>0.05). No differences in hematological and physiological indices of olive flounder were found among the experimental diets except for total cholesterol. Serum lysozyme activity was not significantly different among fish fed experimental diets but bactericidal activity of fish fed 7.5% yuza diet was significantly different from those of fish fed 0, 2.5 and 5% Yuza diets. Survival rates of fish fed 2.5% yuza diet at experimental infection with Edwardsiella tarda and Streptococcus sp. were higher than those of the other experimental groups but not significant. In conclusion, Dietary inclusion of 2.5~5% yuzu seems to have positive effects for fish health condition and disease resistance.

A Study on the sludge drying using waste heat of cogeneration plant (열병합발전소 보일러 폐열을 이용한 슬러지 건조 연구)

  • 염색폐수의 정화에는 필연적으로 다량의 슬러지 폐기물이 발생한다. 염색폐수 슬러지는 그간 인근 공해 해상에 투기하는 해양 배출로 저렴하게 처리하였으나, 해양오염을 우려하는 국제협약(1972년 런던협약, 1996년 교토의정서)에 의하여 2008년 8월부터 배출기준이 강화되고 2012년 2월부터는 해양배출이 금지 될 예정이다. 염색폐수 슬러지의 해양 배출이 금지되면 대체 처리방법으로는 지정매립장을 통한 매립처리 방법이나 고온 소각시설에서의 소각처리 방법이 거론되고 있다. 그러나 매립처리는 슬러지 내 함유 수분으로 인한 침출수의 문제와 더불어 장기간 안정적으로 저렴하게 사용할 수 있는 대규모 처분장을 확보하기 어려운 실정이며 소각처리는 슬러지의 높은 함수율로 인해 소각 시보조 연료의 투입이 필연적으로 최근 원유가 급등 등 에너지 비용이 지속적으로 상승함을 고려할 때 소각처리비용 또한 상당한 고가가 될 것으로 예측된다. 이와 같이 슬러지 해양배출이 금지되면 섬유 염색업체들은 많은 환경비용 부담을 안을 것이다. 본 연구에서는 대규모 염색산업단지 공동폐수처리장에서 발생하는 염색폐수 슬러지의 효율적인 건조를 위해 산업단지 내의 열병합발전소에서 발생하는 보일러 폐열을 이용하였으며, 조건 특성 및 효율을 파악하기 위해 보일러 폐열의 특성을 고려하여 슬러지 두께 및 체류시간 등 건조공정 운영조건에 따른 변수별 연구를 수행하였다. 열병합발전소 보일러에서 배출되는 폐열은 온도가 $150^{\circ}C$ 정도로 기존의 슬러지 건조에서는 사용되는 $700^{\circ}C$에 비해서는 매우 저온이다. 하지만 보일러 배가스의 경우, 온도에 비해 많은 풍량을 가지고 있으므로 열량으로 환산시 충분히 가치가 있는 것으로 조사되었다. 염색폐수 슬러지의 경우, 함수율 70% 이내의 탈수 Cake 형태이므로 두께가 두꺼울수록 건조효율이 감소하였으며, 체류시간이 길어질수록 건조효율은 증가하나 20mm 이상에서는 건조효율이 급격히감소하였다. 이를 바탕으로 5톤/일 규모 슬러지 건조 Pilot Plant를 제작하여 운영하였는데, 염색폐수슬러지의 투입공정에서 슬러지와 열풍의 접촉면적을 넓혀 건조효율을 높이기 위하여 슬러지를 압출노즐을 이용하여 슬라이스 칩 형태로 제조하여 건조공정에 투입하였으며, 건조실 내에서도 건조효율의 상승을 위하여 내부열풍순환팬을 설치하여 운영하였다. Pilot 운영결과, 체류시간 52분에서 슬러지의 함수율은 70%에서 10%이하로 감소하였다.
