The Situation of Genetic Exchange in Duroc Breed and Impacts on Genetic Evaluation (국내 듀록의 종돈장간의 교류현황과 유전능력평가에 미치는 효과)

  • The study was carried to identify the impact on nation-wide genetic evaluation and to obtain basic materials for the development of strategies in Swine Improvement Network Project (SINP). Data consisted of pedigree records of 235,511 and performance records of 70,747 for Duroc from 1987 to 2010 were collected by Korea Animal Improvement Association. Performance traits included three point back fat thickness (Shoulder, Belly, Waist), loin area, days to 90 kg and average daily gain. Exchange of genetic resources cross the breeding farms was not high, and furthermore the sizable farms which can accommodate genetic evaluation within the farm were scarce. Three data sets (individual farm evaluation: I, two sub-group evaluation: S, and whole eight farm evaluation: P) were used for genetic analysis. Genetic variances were larger in subordinate farms than in joiners farms for connectedness, and consequently the heritabilities were generally higher in subordinate farms than in joiner farms with I. The standard errors of heritability were small in the order of I, S and P. Estimated average inbreeding coefficients were 1.12%, 0.95% and 1.53% for joiner and subordinate group with S and population with P, respectively. The estimated correlations of breeding values with I and P were lowest. The correlations of breeding values with I and P for traits ranged 0.22 to 0.45 for moved parent animals and 0.24 to 0.72 for all animals. The results in the study suggest that nation-wide evaluation uses more pedigree information and improves accuracy. Furthermore SINP for connectedness could help to improve the accuracy of evaluation.

  • 현재 시장의 개방화, 사료비 인상, 조류인플루엔자 및 돼지산업에서는 만성소모성질환으로 많은 농가들이 어려움을 겪고 있다. 이러한 시기에 양돈산업이 국제 경쟁력을 갖추기 위해서는 생산비 절감과 차별화가 큰 대안이 될 수 있다. 하지만 생산비의 절반 이상을 차지하는 사료 대부분을 외국에서 수입하는 현실에서 생산비 절감 부분은 그 한계가 있을 수 있기 때문에 수입돈육과 경쟁하기 위해서는 국내에서 생산되는 돈육의 품질고급화 및 차별화에 초점을 맞추어야 할 것이다. 이러한 차별화를 위한 방안으로 유기축산, 무항생재, 기능성돈육, 고급육 등 많은 대안들이 제시되고 있다. 그리고 현재는 2007년 7월부터 축산물등급판정소에서 돈육의 육질등급 제도를 실시하면서 등급에 따른 가격차이가 발생하고 있어 브랜드 업체나 양돈농가 또한 고품질의 돈육생산에 관심을 가지기 시작했다. 현재 농촌진흥청 축산과학원 양돈과에서는 듀록품종의 계통조성을 '축진듀록'을 완성하여 돼지인공수정센터에 공급하였으며 축진듀록과 연계하여 종돈의 육질개량에 대해 알아보고자 한다.

Associations of the Porcine Melanocortin-4 Receptor (MC4R) Gene with Growth Traits in Duroc Pigs (듀록 품종의 Melanocortin-4 Receptor(MC4R) 유전자와 성장형질과의 연관성 분석)

  • The melanocortin-4 receptor(MC4R) is virtually expressed in all brain regions and plays an important role in energy homeostasis in mammals. MC4R has been intensively studied as a trait gene controlling economically important traits, such as growth and feed conversion, etc. Six hundreds and sixty Duroc pigs were genotyped for the MC4R locus and analyzed their relationships with breeding values for average daily gain(ADG), backfat thickness(BF), days to 90kg(D90) and feed conversion(FC). The estimated genotype frequencies for the all Duroc pigs were: 30.8%, 45.2%, 24.1% for AA, AB and BB genotypes, respectively. The mutant A allele was significantly associated with ADG, D90 and BF whereas no significant relationship was found with FC. The change of gene frequencies by generation was shown in both selected and culled groups. These results indicate that the MC4R polymorphism could be integrated in the present selection program to realize a marker-assisted selection for the growth traits of the Duroc population.

Association between IDH3B Genotypes and Economic Traits in a Crossbred F2 Population between Duroc and Jeju Native Black Pigs (제주재래흑돼지와 듀록 교배 세대에서 IDH3B 유전자형과 경제형질의 연관성)

  • This study examined the association between genotypes of the isocitrate dehydrogenase 3, beta subunit (IDH3B) gene and economic traits in an $F_2$ population of Duroc and Jeju (South Korea) native black pigs (JBPs). The genotypes was determined the presence/absence of a 304-bp insertion/deletion fragment in the promoter region of the IDH3B gene for JBP, Duroc, and their $F_1$ and $F_2$ progeny. Three genotypes (AA, AB and BB) were found in the $F_1$ and $F_2$ populations, but there was no AA genotype found in JBP and no BB in Duroc. Association analysis results showed the significant differences with carcass weights (CW), backfat thicknesses (BFT) and eye muscle area (EMA) (p<0.05), but not with growth traits including body weights and average daily gains at different stages, reproductive traits including teat numbers, and crude fat contents (CFAT) measured in longissimus dorsi (p>0.05). The $F_2$ pigs possessing the IDH3B BB homozygote had heavier CW ($72.92{\pm}11.133kg$), thicker BFT ($25.75{\pm}6.06mm$), and larger EMA ($23.82{\pm}4.825cm^2$) than those from the other genotypes (p<0.05). These results were estimated that there are biological roles related with IDH3B genotypes resulting development of EMA, BFT, and CW but not with intramuscular fat deposition during late period of pig production. Our findings suggest that the 304-bp insertion allele of porcine IDH3B may be a genetic marker for marker assistant selection for improving meat productivity of the Jeju Black pig and Duroc-related molecular breeding systems.

Study on Reproductive and Pork Production Performance for Two-way and Three-way Crosses in Swine (이원교잡종(二元交雜種) 및 삼원교잡종(三元交雜種) 돼지의 산자(産仔) 및 산육능력(産肉能力)에 관(關)한 연구(硏究))

  • The results of a crossbreeding experiment with a total of 315 litters and 325 pigs of Berkshires, Hampshires, Durocs, Landraces, Large Whites, eight different two-breed crosses and twelve different three-breed crosses, produced at Livestock Experiment Station from 1975 through 1979, are summarized as follows. 1. Number born alive per litter was largest in the D♂${\times}$(Lw♂${\times}$L♀) $F_1$ ♀ mating, followed by the D♂${\times}$(H♂${\times}$L♀) $F_1$ mating, and smallest in the Hampshires. The pigs in the 3rd-6th parities had larger litter size at birth than those in other parities. 2. Birth weight of pig was heaviest in L♂${\times}$Lw♀ mating and lightest in the Large White. The total litter weight at birth was heaviest in the D♂${\times}$(Lw♂${\times}$L♀) $F_1$ ♀ mating, followed by D♂${\times}$(H♂${\times}$L♀) $F_1$ ♀ and Lw♂${\times}$L♀ matings, and was smaller in Hampshires and Birkshires. 3. Litter size at weaning was largest in the D♂${\times}$(Lw♂${\times}$L♀) $F_1$ ♀ mating, followed by D♂${\times}$(H♂${\times}$L♀) $F_1$ ♀ and Lw♂${\times}$L♀ matings, and was smaller in Durocs and Hampshires. The pigs in the 3rd-6th parities had larger litter size at weaning than those in other parities. 4. Survival rate at weaning was highest in L♂${\times}$Lw♀ mating, followed by D♂${\times}$(L♂${\times}$H♀) $F_1$ ♀ and D♂${\times}$(H♂${\times}$L♀) $F_1$ ♀ mating, and was lowest in Durocs. 6. The three-breed cross from D♂${\times}$(H♂${\times}$L♀) $F_1$ ♀ mating had the highest average gain and lowest feed requirement per unit gain, followed by the D♂${\times}$(Lw♂${\times}$L♀) $F_1$ ♀ and H♂${\times}$(Lw♂${\times}$L♀) $F_1$ ♀ matings. The Birkshires and Landraces ranked lowest among the 25 mating groups compared for both of the traits. Males had higher average daily gain than females by about 0.06kg and had lower feed requirement by about 0.14. 7. The three-breed crosses from D♂${\times}$(H♂${\times}$L♀) $F_1$ ♀, D♂${\times}$(Lw♂${\times}$L♀) $F_1$ ♀ and H♂${\times}$(Lw♂${\times}$L♀) $F_1$ ♀ matings reached 90kg body weight at younger age than the other groups. The D♂${\times}$(H♂${\times}$L♀) $F_1$ ♀ group reached 90kg at younger age than the Landrace by 39 days. 8. The dressing percentage and lean meat percentage tended to be higher in H♂${\times}$(Lw♂${\times}$L♀) $F_1$ ♀, H♂${\times}$L♀ and L♂${\times}$B♀ matings compared to the other mating groups. The loin-eye area was largest in the Lw♂${\times}$L♀ mating and smallest in the B♂${\times}$L♀ mating. Males had higher dressing percentage, higher lean meat percentage and lion-eye area than females. The backfat was thinnest in purebred Hampshire and was thickest in B♂${\times}$L♀ mating. 9. The results obtained in this study suggest that the two-breed cross from Lw♂${\times}$L♀ mating, and the three-breed crosses from D♂${\times}$(Lw♂${\times}$L-♀) $F_1$ ♀ and D♂${\times}$(H♂${\times}$L♀) $F_1$♀ matings are superior crossbreds for reproductive and pork prodnction performance.

Characterization and Evaluation of Melanocortin 4 Receptor (MC4R) Gene Effect on Pork Quality Traits in Pigs (돼지 Melanocortin 4 Receptor (MC4R) 유전자의 육질연관성 분석)

  • This study aimed to investigate the single nucleotide polymorphisms (SNPs) of the porcine MC4R gene and validate the effect of the MC4R genotype for marker assisted selection (MAS). Six amplicons were produced to analyze the entire base sequences of the porcine MC4R gene and six SNPs were detected (c.-780C>G, c.-135C>T, c.175C>T-Leu59Leu, c.707A>G-Arg236His, c.892A>G-Asp298Asn, and c.*430A>T). Linkage disequilibrium (LD) of the six SNPs was analyzed by performing haploid analysis. There was a perfect linkage disequilibrium in c.-780C>G, c.-135C>T, c.175C>T-Leu59Leu, c.707A>G-Arg236His, and c.*430A>T. Only the c.892A>G (Asp298Asn) SNP showed a very low LD with an $r^2$ value of 0.028 and the D' value of 0.348. As a result, the two SNPs-c.707A>G (Arg236His) and c.892A>G (Asp298Asn)-were selected to extract the genotype frequencies from the 5 pig breeds by using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) genotype analysis method. The SNP frequency of c.707A>G (Arg236His) indicated the presence of the A (His) allele only in Yorkshire, while the G allele was fixed in the KNP, Landrace, Berkshire, and Duroc. Association analysis was carried out in 484 pigs with the c.707A>G (Arg236His) SNP and the meat quality traits of four different pig cross populations: a significant association was noted in crude fat, sirloin moisture, meat color, and the degree of red and yellow coloration. The frequency of the c.892A>G(Asp298Asn) SNP genotype varied among the breeds; while Duroc showed the highest frequency of the A (Asn) allele, KNP showed the highest frequency of the G (Asp) allele. Association analysis was carried out in 1126 pigs with the c.892A>G (Asp298Asn) SNP and the meat quality traits of four pig populations: a highly significant linkage was noted in the back-fat thickness (P<0.002). It was found that the back-fat thickness was higher in individuals with the AA genotype than in those with the AG or GG genotype. Thus, in this study, we verified that the c.892A>G (Asp298Asn) SNP in the pig MC4R gene has a sufficient effect as a gene marker for MAS in Korean pork industry.

Estimation of Genetic Parameters via Gibbs Sampler using Animal Model for Economic Traits in Pigs (Gibbs Sampler를 이용한 돼지 주요 경제형질의 유전모수 추정)

  • Heritability and genetic correlation for growth traits in Duroc pig breed were estimated using Bayesian method via Gibbs sampling. The data set consisted of 3,526 performance records at National Institute of Animal Science. For estimating those parameters using Gibbs sampling, 5,000 cycles of ‘burn-in’ period were discarded among a total of 55,000 samples. Out of the remaining 50,000 samples, 5,000 estimates by each parameter were retained and used for analyses to avoid any correlation among adjacent samples. The growth traits considered in this study were average daily gain at 30kg(ADG1), average daily gain at 90kg(ADG2), backfat thickness(BF), days to 90kg(D90) and feed conversion ratio(FC). The estimated heritabilities and their standard deviation using Gibbs sampler were 0.43±0.04, 0.49±0.038, 0.31±0.040, 0.48±0.039 and 0.62±0.086, respectively. Genetic correlations were -0.02, -0.13, -0.55 and -0.15 between ADG1 with ADG2, BF, D90 and FC, respectively, 0.16, -0.73, -0.32 between ADG2 with BF, D90 and FC respectively, 0.01, -0.08 between BF with D90, FC, respectively, and 0.23 between D90 with FC.

Growth performance and, blood metabolites and leukocytes of growing pigs fed with fermented hulless barley (발효 쌀보리의 급여가 육성돈의 혈중 대사산물, 혈구조성 및 생산성에 미치는 영향)

  • This study was conducted to investigate effect of feeding fermented hulless barley (FHB) on growth performance and blood metabolites in growing pigs. Forty-five pigs (LYD; initial body weight, $30.33{\pm}0.05kg$) were randomly allotted into three dietary treatments that consisted of 0, 0.5 and 1.0% of the FHB in the basal diets. The pigs fed 0.5% FHB showed higher average daily gain than the 0 and 1% FHB treatments, although there was not significant among the treatments. Similarly, average daily feed intake and feed conversion ratio were not different among the treatments. Blood white blood cells, neutrophil, lymphocyte, monocyte, eosinophil and basophil were ranged to reference values, but not difference among the treatments. Serum glucose was increased in the control compared with 0.5 and 1.0% FHB. However, parameters related to protein, lipid and mineral also were not different among the treatments. These results indicate the FHB has no significant effect of growth performance and metabolizable responses in growing-finishing pigs.

Genetic and of environmental effects for economic traits in pigs (돼지 주요 경제형질들에 영향을 미치는 유전 및 환경효과 추정)

  • This study was conducted to estimate the effect of sex, breed and on the basis of the records on 300 pigs of Duroc, Landrace and Yorkshire breeds raised at the National Institute of Animal Science. Yorkshire was observed to be superior to other breeds between 90 to 110 kg of body weight. Thicker backfat was estimated in the B-mode than in A-mode of 110 kgs range than in 90 kgs. Loin eye muscle area was larger in the final weight of 110kg than in 90 and the Duroc had significantly (p<0.05) larger loin eye muscle area than other breeds. Thinner backfat was observed in males than in females in both modes of backfat measurements and in both of the final weights of test. Females had significantly larger loin eye muscle area than males, and the difference between the two sexes were larger in the final weight of 110 kg than in 90 kg. Analysis models concluded that backfat thickness certainly influences the body weight gain when at 90 kgs and 110 kgs analysed with A-mode and B-mode respectively.

Characteristics of Carcass and Meat Quality for Landrace, Yorkshire, Duroc and their Crossbreeds (랜드레이스, 요크셔, 듀록 및 교잡종에 대한 도체 및 육질특성)

  • In this study, changes of ammonia, alcohol and volatile fatty acid(VFA) concentrations or pH in in vitro ruminal fluid were studied to determine the effects of alcoholic feeds on in vitro ruminal fermentation characteristics. To formulate the alcoholic feeds, alcohol was added to commercial formulated feed at the levels of 1, 3, and 5 %. Experiments were done with four treatment groups, control(commercial feed), AF-1(commercial feed+1% alcohol), AF-3(commercial feed+3% alcohol), and AF-5(commercial feed+5% alcohol). Ammonia concentrations of AF-1 and AF-5 were significantly lower than that of control for the 12h incubation(p<0.05). Ruminal alcohol concentration was increased with the addition level of alcohol increased(p<0.05). TVFA concentrations of AF-1, AF-3 and AF-5 were significantly higher than those of control at 12h(p<0.05). Significant decrease of molar percentage of acetate was observed in control from 8 to 12h incubation, but molar percentage of acetate for AF-1, AF-3 and AF-5 was constant. Molar percentage of propionate was increased in control compared with AF-1, AF-3 and AF-5 from 8 to 12h incubation(p<0.05). Molar percentages of butyrate and valerate were higher in AF-1, AF-3 and AF-5 than in control(p<0.05). Molar percentage of caproate for AF-1, AF-3 and AF-5 was 0.05, 0.58 and 0.47M% at 8h, respectively, but that was not detected for control. Present results may indicate that the alcoholic feeds show positive effects on in vitro ruminal ammonia, alcohol and VFA concentrations or pH. Furthermore, the results of this study implies that the addition level of 5% could be more effective to ruminal fermentation than other addition levels.
