  • 두족류는 연체동물문 두족강에 속하는 무척추동물로 해양에만 서식한다. 조간대에서 심해에 이르기까지 모든 해양 환경에 적응한 두족류는 뛰어난 유영능력과 감각기관을 가지고 활동적인 포식활동을 한다. 이빨고래류, 물개류, 상어류, 어류 등의 먹이가 되며 우리나라를 비롯한 북서태평양 연안국에서는 상업적으로도 중요한 수산자원생물이다. 국내에서는 분류학적 연구와 참갑오징어나 살오징어와 같은 일부 상업성 종류의 자원학적 연구가 이루어져 있으나 두족류 군집 전체를 대상으로 한 연구는 보고된 바 없다. 본 연구에서는 가덕도 주변 해역에 서식하는 두족류 군집의 종조성과 계절변동을 파악하고자 하였다. Korean Name of Cephalopods in Korea (우리나라에 출현하는 두족류 이름)

  • Korean cephalopods only recorded with Korean name in the literature were reviewed to solve confusions in use of their Korean name for taxonomic studies of cephalopods in Korea, and to prepare a basic check list for standardizing total cephaolopods fauna of Korea in the future. A total 38 cephalopods species with Korean names were listed in this study; 14 species in order Sepioidea, 17 species in Teuthoidea, 7 species in Octopoda. The etymology and some problems of Korean names of cephalopods occurring in Korea, and the useable rules for naming cephalopods with Korean words are recommended and discussed.

Development and Validation of Quick and Accurate Cephalopods Grouping System in Fishery Products by Real-time Quantitative PCR Based on Mitochondrial DNA (두족류의 진위 판별을 위한 Real-time Quantitative PCR 검사법 개발 및 검증)

  • In this study, an approach for the analysis of the five cephalopod species (octopus, long-arm octopus, squid, wet-foot octopus, beka squid) consumed in the Republic of Korea is developed. The samples were collected from the Southeast Asian countries Thailand, Indonesia, Vietnam, and China. The SYBR-green-based real-time qPCR method, based on the mitochondrial DNA genome of the five cephalopods was developed and validated. The intergroup variations in the mitochondrial DNA are evident in the bioinformatic analysis of the mitochondrial genomic DNA sequences of the five groups. Some of the highly-conserved and slightly-variated regions are identified in the mitochondrial cytochrome-c-oxidase subunit I (COI) gene, 16s ribosomal RNA (16s rRNA) gene, and 12s ribosomal RNA (12s rRNA) gene of these groups. To specify each five cephalopod groups, specific primer sets were designed from the COI, 16s rRNA and 12s rRNA regions. The specific primer sets amplified the DNA using the SYBR-green-based real-time PCR system and 11 commercially secured animal tissues: Octopus vulgaris, Octopus minor, Todarodes pacificus, Dosidicus gigas, Sepia esculenta, Amphioctopus fangsiao, Amphioctopus aegina, Amphioctopus marginatus, Loliolus beka, Loligo edulis, and Loligo chinensis. The results confirmed by a conveient way to calculate relative amplification levels between different samples in that it directly uses the threshold cycles (Ct)-value range generated by the qPCR system from these samples. This genomic DNA-based molecular technique provides a quick, accurate, and reliable method for the taxonomic classification of the animal tissues using the real-time qPCR.

Soluble Proteins Analysis of Class Cephalopoda in the Yellow Sea(I) (황해산 두족류의 가용성 단백질에 대한 연구 (I))

  • To investigate a possibility of the species genetic relationship for the soluble proteins analysis on the class Cephalopoda in the Yellow Sea, the isolate eye, muscle and liver proteins from five species (Sepia esculenta, Sepiella japonica, Loligo chinensis, Loligo beka and Octopus minor) were analysed using different electrophoretic techniques (Davis-polyacrylamide gel electrophoresis, SDS-PAGE, exponential gradient SDS-PAGE, thin-layer isoelectro-focusing and two-dimensional PAGE). The average molecular weight of the soluble eye and muscle proteins was estimated at 35-50 KDa, separated b the exponential gradient SDS-PAGE. It was corresponds to that of electrophoretic patterns by t재 dimensional PAGE. By which the thin layer IEF, the target proteins showed a reasonable specificity based on their isoelectric points (pI) 7.5-8.5.

  • Distribution of Cephalopods was investigated in the Yellow Sea, Korea from February, 2006 to November, 2008. The species composition of cephalopods, consisting of 14 species and 19,789 individuals has been identified. The dominant species of the Yellow sea was Euprymna morsei. The dominant species of the northern part, middle part and southern part were Todarodes pacificus, Loligo spp. and Loligo beka.

  • The isolated soluble eye proteins from six species (Sepia esculenta, Sepiella japonica, Loligo chinensis, Todarodes pacificus, Loligo beka and Ocotopus minor) of class Cephaloopoda were compared by crossed immunoelectrophoresis methods using antibodies directed against total soluble eye proteins antigens. It showed a distinct antigen-antibody reaction between the species of order Sepioidea and various antiserum of class Cephalopoda. Our results suggested that the common precipitation lines of Sepia and Loligo species which were different from that of Octopus minor. By which obtained from elution of gel filtration chromatography and 12.5% SDS-polyacrylamide gel electrophoresis, the molecular weight of Todarodes pacificus eye protein (crystallin) was determined in abut 20-35 KDa.

  • Stomach contents of the common squid, Todarodes paciかcus in the Korean waters were analyzed using the samples taken from February, 1991 to July, 1992. Stomach contents of T. pacificus consisted of pisces, cephalopods, crustaceans, algae and unidentified things. Pisces were the most selected prey items. Major food items of summer and autumn populations were pisces, but that of the winter population was cephalopods. Algae and crustaceans were minor food items of the squid, T. pacificus mainly fed on pisces and three populations of the squid did not have different stomach contents.
