Use of $^{18}F$-FDG PET/CT in Second Primary Cancer (이차성 원발암에서의 $^{18}F$-FDG PET/CT의 이용)

  • This review focuses on the use of $^{18}F-FDG$ PET/CT to evaluate second primary cancers. The emergence of a second primary cancer is an important prognostic factor in cancer patients. The early detection of a second primary cancer and the appropriate treatment are essential for reducing the morbidity and mortality associated with these tumors. Integrated $^{18}F-FDG$ PET/CT, which can provide both the metabolic and anatomic information of a cancer, has been shown to have a better accuracy in oncology than either CT or conventional PET. The whole body coverage and high sensitivity of $^{18}F-FDG$ PET/CT along with its ability to provide both metabolic and anatomic information of a cancer make it suitable for evaluating a second primary cancer in oncology. Whole body $^{18}F-FDG$ PET/CT is useful for screening second primary cancers with a high sensitivity and good positive predictive value. In order to rule out the presence of a second primary cancer or an unexpected metastasis, further diagnostic work-up is essential when abnormal findings indicative of a second primary cancer are found on the PET/CT images. PET/CT is better in detecting a second primary tumor than conventional PET.

Dystrophin Degradation in Skeletal Muscles with Lipid Enrichment in Cattle (지방 침착률이 높은 식용소에서 나타난 골격근의 디스트로핀 소실)

  • This study investigated the muscular dystrophin levels in freely moving Australian cattle mainly fed grass, freely moving Korean cattle fed mainly a grain diet, and Korean cattle fed a grain diet but housed in a relatively limited space of a cow house. The total skeletal muscle specimens of 244 cattle were collected and immediately fixed in 10% neutral formalin. The same area was biopsied from the cattle in both countries. The findings showed that fatty infiltration is highly correlated with membrane-associated protein degradation in skeletal muscle, and that among several membrane-associated proteins, dystrophin showed the most significant reduction in expression in the cattle with fatty infiltration. Similarly, CD36 was more highly expressed in the cattle with fatty infiltration of skeletal muscle. Various breeding factors, such as oxidative stress; the presence of oxidized lipids in the diet; and environmental factors such as exercise, temperature and amount of time spent, may have critical effects on the degradation of normal cytoskeleton proteins, which are required for maintaining normal skeletal muscle architecture. Among the sarcolemma membrane-associated proteins, dystrophin is the most sensitive membrane protein that is involved muscular dystrophy and muscular degeneration. Thus, the present findings may be useful for studies on muscular dystrophy in humans or the pathogenesis of muscular diseases in animal models.

Overexpression of Profilin 1 Inhibited Ovarian Tumor Cell Growth and Migration (Profilin-1 과발현에 의한 난소암 세포 성장 및 이동 저해 효능 연구)

  • Abnormal actin remodeling is a typical characteristic of tumor cells. Thymosin ${\beta}_{10}$ (TB10) and profilin-1 (PFN-1) are actin-binding proteins and essential regulators of actin polymerization. We previously showed that TB10 induced death in ovarian cancer cells by sequestering F-actin, but the underlying mechanisms of this induction have not been explored. In this study, we identified TB10 as a novel regulator of PFN-1 and demonstrated its novel function as a tumor suppressor in ovarian cancer cell lines. The present study investigated protein expression profiles through polyacrylamide gel electrophoresis (PAGE) and liquid chromatography-mass spectroscopy (LC-MS/MS) in SKOV3 cells, an ovarian cancer cell line, that were transiently transfected with TB10. PFN-1 was highly overexpressed in response to TB10, and overexpression of PFN-1 resulted in inhibition of cell proliferation and migration and promotion of cellular apoptosis in ovarian cancer cells. Furthermore, transiently transfected PFN-1 appeared to deactivate the Erk signaling pathway, followed by decreased expression of Elk-1 and Egr-1 in human ovarian cancer cells. Interestingly, PFN-1 did not affect the activation of Akt. The results demonstrated that PFN-1 induced apoptotic cell death and inhibited proliferation and migration in ovarian cancer cells, suggesting that PFN-1 may be valuable in anti-cancer therapy.

  • 복잡한 전자부품의 조립시에 필요한 열적 디자인에 관한 정보는 오래전부터 실험을 통하여 얻어지고 있다. 실험적 데이터를 이용하여 무차원 파라미터로 표시된 실험결과는 꼭 같지는 않지만 현상적으로는 비슷한 상황에 응용될 수 있다. 여기서는 학술문헌에 나타나 있는 자연대류에 관한 실험적인 상관관계식들과 프레임에 수직으로 꽂혀있는 균일가열 전자회로기판의 모델에서 얻어진 무차원 자료들을 비교하고자 한다. 대부분의 자료들은 수정채널 Rayleigh수(Ra")가 15~100범위에 속하며, 이러한 범위는 부품이 조밀하게 배치된 기관이 서로 좁은 채널을 이루고 있으며, 동시에 상당한 전력을 소비하고 있는 경우에 해당한다. Wirt와 Stutzman, Bar-Cohen과 Rohsenow의 일반상관관계식은 AT'||'&'||'T Bell 연구소에서 개발된 전자기기를 이용하여 수집한 실험데이터를 잘 표현하고 있으며 10 < Ra" <1,000범위에서 추천될 수 있다. 두개의 유사한 상관관계식과 비교할 때 상당히 좋은 예측을 보였으며 또한 Sparrow와 Gregg의 연구결과와도 잘 일치하므로 Ra" < 10인 경우에 Aung의 완전발달층류의 채널유동방식, Ra" > 1,000인 경우에는 Aung등의 단일 수직평판 근사식이 추천될 수 있다. Protective Effects of Curcumin on CCl4-Induced Hepatic Fibrosis with High Fat Diet in C57BL/6 Mice (C57BL/6 마우스에서 고지방 식이와 CCl4로 유발한 간섬유증에 미치는 커큐민의 보호효과)

  • Curcumin, a major polyphenolic compound of turmeric, is well known to prevent non-alcoholic steatohepatitis (NASH) related to obesity. The aim of the study was to investigate the effect of curcumin on hepatic fibrosis induced by carbon tetrachloride ($CCl_4$) in obese mice. $CCl_4$ was administrated in mice fed a normal diet (ND) or a high fat diet (HFD) for 7 weeks together with or without curcumin. It was conducted to examine for metabolic profiles, adipocyte size, and liver fibrosis by serum biochemistry, histology and immunohistochemistry. Also, Apoptosis of hepatic cells was determined by the TUNEL method. Treatment with curcumin significantly lowered the body weight, fasting glucose, serum AST and ALT, and decreased the adipocyte size, the number of macrophage and mast cells in adipose tissue, and collagen deposition in liver tissue in the HFD+$CCl_4$ group compared with the findings of the HFD+$CCl_4$ group. In contrast, treatment with curcumin on the ND+$CCl_4$ group did not show a significant difference except the body weight and mast cell number when compared with the ND+$CCl_4$ group. Furthermore, curcumin significantly reduced the number of parenchymal apoptotic cells, whereas it increased the number of non-parenchymal apoptotic cells, especially resembling an activated hepatic stellate cell in the liver. Taken together, this data suggests that curcumin might be an effective antifibrotic drug for the prevention of liver disease progression in obese mice. Thus, the development of curcumin as a therapy for obesity and liver fibrosis is supported.

A Case of Subacute Necrotizing Lymphadenitis Presenting as Fever of Unknown Origin (불명열로 오인된 아급성 괴사성 림프선염 1례)

  • Kikuchi's disease(histiocytic necrotizing lymphadenitis) is characterized by lymphadenopathy in young patients below 30 years old, and persistent fever, lymphopenia and splenomegaly are concomitantly developed in many cases. So, it has been confused with lymphoma, SLE, and tuberculosis, and has easily led to inappropriate diagnostic procedures and administration of drugs. Many reports have indicated that Kikuchi's disease should be added to the list of causes of FUO in the setting of lymphadenopathy, and recommended early lymph node biopsy to distinguish from lymphoma, SLE, and tuberculosis to avoid unnecessary treatments. We experienced a case of subacute necrotizing lymphadenitis in a 14-year-old boy who presented with persistent high fever, productive coughing and cervical lymphadenopathy for about 1 month. Initially, diagnostic workup was done to look for the causes of FUO in vain. Finally, we confirmed diagnosis by histopathological findings of lymph node biopsy and detected latent gene of EBV in the biopsied specimen using in situ hybridization.

Surgical Angioplasty of the Left Main Coronary Artery Stenosis (좌주관상동맥 협착에 대한 수술적 혈관 성형술)

  • Background: Left main coronary artery (LMCA) angioplasty is another option in the surgical treatment for LMCA disease because of its advantages over the conventional coronary artery bypass grafting (CABG). Material and Method: Between July 1994 and December 1997, 15 patients underwent left main coronary angioplasty for the stenoses of LMCA. There were 8 males and 7 females with the mean age of 53.3${\pm}$8.8 years. The locations of the LMCA stenoses were proximal one-third of the LMCA in 9, middle one-third in 1, distal one-third in 3, and the whole length of the LMCA in 2 cases. Nine patients had peripheral coronary lesions in addition to the LMCA stenosis. The LMCA was approached anteriorly with or without transsection of the main pulmonary artery. The angioplasty was performed with onlay patch widening using an autologous pericardium (14 cases) or saphenous vein (1 case). Additional graftings were required in 9 cases, and both LMCA angioplasty and right coronary ostial angioplasty were done in 1 case. Result: There was no operative mortality. One case needed redo CABG due to the stenosis of the angioplasty site which developed 4 months postoperatively. Coronary angiography was performed in 8 cases one year postoperatively, and revealed good patency of the angioplasty site except for one who showed 50% stenosis at the angioplasty site. No patient complained of angina with a mean follow up of 23${\pm}$11 months. Conclusion: Surgical angioplasty of the LMCA stenosis can be performed in selected cases with safety and good mid-term results.

Anti-inflammatory activities of Scolopendra subspinipes mutilans in RAW 264.7 cells (RAW 264.7 세포에서 왕지네 추출물의 항염 활성)

  • Purpose: The dried body of Scolopendra subspinipes mutilans has long been used as a traditional Korean medicinal food, but little is known about its mechanisms of action. In this study, we investigated the anti-inflammatory activities of Scolopendra subspinipes mutilans and possible mechanisms in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Methods: Cytotoxicity of Scolopendra subspinipes mutilans extract (SSME) was measured by MTT assay, anti-inflammatory activities were analyzed by nitric oxide (NO) production, the expression of inducible NO synthase (iNOS) and the mRNA level of pro-inflammatory cytokines such as $interleukin-1{\beta}$ ($IL-1{\beta}$) and interleukin-6 (IL-6). Nuclear translocation of nuclear factor-kappa B ($NF-{\kappa}B$) p65 subunit and degradation of inhibitory kappa B ($I{\kappa}B$) were examined by western blot. Results: SSME inhibited LPS-induced NO production and iNOS expression without cytotoxicity. Up-regulation of LPS-induced pro-inflammatory cytokines, $IL-1{\beta}$ and IL-6 was dose dependently attenuated by SSME. Exposure of pyrrolidine dithiocarbamate, an $NF-{\kappa}B$ specific inhibitor, accelerated the inhibitory effects of SSME on NO production and iNOS expression in LPS-stimulated cells. Moreover, translocation of $NF-{\kappa}B$ from the cytosol to the nucleus and degradation of $I{\kappa}B$ were decreased by treatment with SSME in LPS-induced cells. Conclusion: These results suggest that the SSME might have the inhibitory effects on inflammation, partly through inhibition of the $NF-{\kappa}B$ signaling pathway.

The Clinical Characteristics of Influenza B Infection during the 2011-2012 Influenza Season (2011-2012절기 B형 인플루엔자 감염의 임상 양상)

  • Purpose: This retrospective study was performed to identify the clinical characteristics of influenza B infection and compare to influenza A infection. Methods: Medical records of patients diagnosed with influenza using a multiplex PCR test, admitted to Seoul St. Mary's Hospital, during the 2011-2012 influenza season were analyzed. Clinical and laboratory characteristics of influenza B patients were investigated and compared with those of influenza A patients. Results: A total of 145 influenza patients were enrolled during this study period. Among these, 66 and 78 patients were diagnosed with influenza A and B, respectively, and 1 patient was diagnosed with co-existing influenza A and B. Cough (88.2%), rhinorrhea (77.1%) and sputum (60.4%) were the most common symptoms among these influenza patients, and most were diagnosed with upper respiratory infection (31.9%) or lower respiratory infection (49.3%). In comparison to influenza A patients, influenza B patients were older ($4.7{\pm}4.1$ years vs. $3.3{\pm}2.5$ years, P=0.016), and the number of fever days before hospitalization were longer (3.0 days vs. 2.5 days, P=0.043). While sore throat (10.3% vs. 1.5%, P=0.039) and vomiting (20.5% vs. 6.1%, P=0.012) were more common in influenza B patients than in influenza A patients, other clinical and laboratory characteristics were not significantly different between the two groups. Conclusions: No significant differences in clinical and laboratory perspectives were manifested in influenza A and B infections. Preventive measures should be emphasized over treatment in influenza B due to prolonged fever duration before admission.

  • 최근 UV LED는 생화학 및 의료 산업에서 많은 각광을 받고 있다. 특히, 360nm 이하의 파장대를 갖는 UV LED는 치료 기술, 센서, 물이나 공기 등의 정화와 같은 목적으로 특별한 관심이 쏠리고 있다 [1]. 이러한 지속적인 연구를 통하여 현재까지 UV LED는 거대한 성장을 이루어 왔다. 하지만 이러한 노력에도 불구하고, 360 nm 이하의 UV LED는 여전히 오믹 접촉과 전류 분산이 원활하지 못하다는 문제점을 가지고 있다. 이것은 UV LED의 외부 양자 효율을 감소시키고, 더 나아가 극도로 낮은 광 추출 효율을 초래한다. 최근 이러한 문제를 해결하고자, 투명 전도성 산화물(TCO)을 금속 전극과 p-AlGaN 사이에 삽입해주는데, 현재 가장 널리 사용되는 TCO 물질은 ITO 이다 [2]. 하지만 ITO 물질은 상대적으로 작은 밴드갭(3.3~4.3 eV)과 단파장 빛이 가지는 큰 에너지로 인하여 deep-UV 영역에서는 빛이 투과하지 못하고 대부분 흡수된다 [3]. 따라서 본 연구에서는 기존의 박막형 ITO 투명 전극에 비해 투과도 손실을 최소화할 수 있는 mesh, grid 기반의 투명전극을 연구하였다. Fig. 1과 같이 $5{\mu}m$, $10{\mu}m$, $20{\mu}m$ 간격으로 이루어진 mesh, grid 구조의 투명전극을 구현하여 투과도 손실을 최소화하면서 우수한 전기적 특성을 확보하기 위한 구조 최적화 연구를 진행하였다. 본 연구를 위해 mesh, grid 구조의 ITO 전극 패턴을 photolitho 공정으로 형성하였으며, e-beam 증착법으로 60 nm 두께의 ITO 전극을 형성 후 질소 분위기/$650^{\circ}$에서 30초 동안 RTA 공정을 진행하였다. Fig. 1에서 볼 수 있듯이 mesh, grid의 간격이 증가할수록 투명 전극이 차지하는 면적이 감소하여 투과도는 향상되는 반면, 투명 전극과 p-GaN과의 접촉 면적 또한 감소하므로 오믹 특성이 저하된다. 따라서 투과도 손실을 최소화하면서 우수한 전기적 특성을 확보하기 위해 mesh는 $20{\mu}m$, grid는 $10{\mu}m$ 간격의 구조로 각각 최적화하였다. 그 결과 박막 기반의 ITO 투명전극 대비 최대 약 10% 향상된 투과도를 확보하였으며, I-V Curve 결과를 통하여 p-GaN 기판과 mesh 구조의 ITO 전극 사이에 박막 기반의 투명 전극과 비슷한 수준인 $0.35{\mu}A(@5V)$의 전기적 특성을 확보하였다. 결과적으로 mesh, grid 기반 투명전극의 구조 최적화를 통하여 p-GaN과 원활한 오믹 접촉을 형성하는 동시에 기존 박막형 ITO 투명 전극 구조보다 높은 투과도를 확보할 수 있었다.
