Effects of Catechin on Lipid Composition and Adipose Tissue in Obese Rats Fed High Fat Diet (고지방 식이로 유도된 비만쥐에서 녹차 Catechin이 체지방 조성 및 지방조직에 미치는 영향)

  • The current study examined the effects of catechin on lipid composition of serum and liver and adipocyte of epididymal fat pads in obese rats fed high fat diet. Sprague-Dawley male rats weighing $100{\pm}10g$ were randomly divided into eight groups, four normal diet groups and four high fat diet groups according to the level of dietary catechin supplement. The rats were fed ad libitum experimental diets for 4 weeks and then they were sacrificed. Body weight in HF group was heavier than that of NC group, but HFCM and HFCH groups were significantly reduced compared to HF group. Relative body weight to abdominal weight and relative body weight to epididymal weight in HF group were increased to 103% and 106%, respectively, compared to NC group, but HFCM and HFCH groups were significantly reduced as compared to HF group. The levels of serum triglyceride, total cholesterol, LDL-cholesterol and atherogenic index in HFCH groups were significantly lower than those of HF group, whereas HDL-cholesterol levels were increased. Total lipid contents of liver in HF group was significantly higher than that of NC group, but HFCH group maintained the NC level. There were no significant difference in hepatic triglyceride contents of high fat diet groups. Contents of hepatic cholesterol in HF group was 29% higher than that of NC group, but HFCM and HFCH groups were significantly reduced as compared to HF group. Cell number and cell size of epididymal fat pads in HFCM and HFCH groups were significantly reduced, respectively, compared to HF group. Improved lipid metabolism observed in rats fed catechin may be caused by an alteration of number and size in epididymal fat pad and lipid composition.

Effects of Unilateral and Bilateral Ovariectomy on Reproductive Organs. Adrenal Gland and Serum Level of FSH and LH in Immature Rats (미성숙 흰쥐에 있어서 편측 및 양측난소척출이 생식기관, 부신 및 혈청중 FSH와 LH수준에 미치는 영향)

  • This experiment was carried out to investigate the effects of unilateral and bilateral ovariectomy in immature rats on the weight of body, ovary, uterus and adrenal gland and the change of serum FSH and LH level. Ninty Sprague-Dawley female rats, 23${\pm}$2 days old, were divided into 3 groups with 30 heads per group; control, unilaterally and bilaterally ovariectomized group. Each group was subdivided into 6 groups according to 6 experimental periods; day 4, 8, 12, 16, 20, and 24 after operation. Five rats at every 4 days intervals were measured their body weights and sacrificed for the measurement of their ovarian, uterine and adrenal weights and at the same time blood samples were taken for the determination of serum FSH and LH level by radioimmunoassay. The following results were obtained: 1. Body weights in the unilaterally and bilaterally ovariectomized goups were higher than those of control groups during all experimental periods, even though there were no significant differences among the above 3 groups. 2. A significant hypertrophy of the remained ovary in the unilaterally ovariecto mized group was observed from day 16 till day 24 after operation. The ovarian weight; 22.1${\pm}$1.73mg, at day 16 in control group was smaller than the unilaterally ovariectomized group weighing 50.5${\pm}$8.45mg (p<0.01) and the ovarian weights, 75.9${\pm}$2.25mg and 63.3${\pm}$7.08mg ; at day 20 and 24 in unilaterally ovariectomized group were significantly larger than 29.1${\pm}$2.33mg and 26.3${\pm}$1.76mg in control group, respectively (p<0.01 and p<0.05). 3. The uterus of bilaterally ovariectomized group were remarkably atrophied from day 8 after operation as compared with those of control and unilaterally ovariectomized group. The uterine weight at day 24 was 96.7${\pm}$9.15mg for control group, 139.4${\pm}$1.73mg for unilaterally ovariectomized group and 21.7${\pm}$1.08mg for bilaterally ovariectomized group, respectively and there were significant differences among 3 groups (p<0.01). 4. A statistically significant increase ofthe weight of adrenal gland was observed at day 16 in the unilaterally ovariectomized group with 24.4${\pm}$2.58mg against 15.5${\pm}$3.09mg in control group and 13.9${\pm}$1.38mg in bilaterally ovariectomized group (p<0.05). The adrenal gland weight in unilaterally ovariectomized group with 24.7${\pm}$1.63mg at day 20 and 31.2${\pm}$1.62mg at day 24 increased significantly as compared with bilaterally ovariectomized group with 15.1${\pm}$13.11mg at day 20 and 15.6${\pm}$1.76mg at day 24. 5. Serum FSH level of unilaterally ovariectomized group increased remarkably up to 2.97${\pm}$0.37mIU/ml at day 4 after operaton and then decreased gradually. Serum FSH level of bilaterally ovariectomized group were higher than those of control group throughout all experimental periods. 6. Serum LH level ofunitelarally ovariectomized group with 3.17${\pm}$0.32mIU/iml at day 4 and 3.57${\pm}$0.58mIU/ml at day 24 increased noticeably more than those of control group with 1.79${\pm}$0.16 mIU/ml at day 4 and 2.17${\pm}$0.27mIU/ml at day 24 (p<0.05).

Effects of Dietary Green Tea Probiotics, Alisma canaliculatum (Alismatis rhizoma) Probiotics and Fermented Kelp Meal as Feed Additives on Growth Performance and Immunity in Pregnant Sows (사료내 녹차 생균제, 택사 생균제 및 해조류 발효물의 첨가가 모돈의 생산성 및 면역성에 미치는 영향)

  • An investigation was done to evaluate the effects of adding green tea probiotics, Alisma canaliculatum probiotics and fermented Kelp meal on the growth performance and immune response in sows and piglets. A total of 32 pregnant sows were assigned to 4 treatments in 8 replications with 1 sow as a replicate in this experiment from 2 months before parturition to 28 days after parturition. Three hundred nineteen (319) piglets were produced from experimental sows after parturition. The dietary treatments were CON group (without antibiotics), GTP (basal+green tea probiotics, 0.5% of the diet), ACP group (basal + Alisma canaliculatum probiotics, 0.5% of the diet) and FKM group (basal+fermented Kelp meal, 0.5% of the diet). Litter size and body weight at birth and weaning size and body weight at weanling in piglets were greater in additives group compared to CON group but no statistical differences were found (P>0.05). Diarrhea score showed a decreasing tendency with three additives groups compared to CON group. The lymphocytes in blood of sows fed ACP and FKM group were significantly higher compared to CON group (P<0.05), and the MID of GTP and FKM group were significantly lower compared to CON group (P<0.05). The serum IgG concentration in sows tended to increase with additives groups compared to CON group and the serum IgA and IgM concentration in GTP and FKM group were significantly higher compared to ACP group (P<0.05). The cortisol levels showed a decreasing tendency in additives groups without statistical differences among the groups (P>0.05). Based on these observations, it can be inferred that the GTP and FKM may have such efficacy as antibiotics in growing pigs.

Effect of Dietary Supplementation of Alisma canaliculatum (Alismatis Rhizoma) and Viscum album (Mistletoe) on Growth Performance and Immunity in Broiler Chickens (택사와 겨우살이 급여가 육계의 생산성 및 면역성에 미치는 영향)

  • This study was conducted to investigate the effect of dietary supplement of Alisma canaliculatum and Viscum album on the growth performance and immune response of broiler chickens. One hundred and forty Ross broiler chicks (1 day old) were assigned to 4 treatments in 5 replications with 7 birds per replication. The dietary treatments were NC group (without antibiotics), PC group (basal + 50 ppm Oxytetracycline), AC group (basal + Alisma canaliculatum 0.5%), and VA group (basal + Viscum album 0.5%). The results revealed that body weight gain and feed intake were not significantly different among the treatments. Addition of AC to the diet showed significantly higher feed conversion ratios than the VA addition diet and other treatments (p<0.05). The concentration of linolenic acid, ${\alpha}$-linolenic acid and arachidonic acid in the AC and VA treatment were relatively higher than that in the antibiotic treatment, while there was no difference in the concentration of SFA, USFA, and USFA/SFA. The serum IgG concentration in chickens tended to increase in the AC and VA treatment groups compared to NC group. The IL2 concentration in AC and VA groups was significantly higher than in other group (p<0.05). IL6 was not detected, however. GOT, GPT, and T-billirubin in the VA group were significantly higher than in other groups (p<0.05). Thus, the VA additives can result in the damage of liver and kidney. In conclusion, the dietary supplement of a 0.5% of AC and VA showed a similar growth performance and increased immunity response comparable to supplementing a 50 ppm Oxytetracycline. Therefore, it can be said that the AC and VA are possible alternates of antibiotics.

Effects of Ethanol Extracts from Red Pepper (Capsicum annuum L.) Seeds on Cholesterol Adsorption Capacity and UDP-Glucuronyl Transferase Activity (고추씨 에탄올 추출물이 콜레스테롤 흡착능 및 UDP-glucuronyl transferase 활성에 미치는 영향)

  • The purpose of this study was to investigate the effects of ethanol extracts from red pepper seeds on cholesterol adsorption capacity and UDP-glucuronyl transferase activity. In vitro cholesterol adsorption capacity of 2%, 5% and 10% ethanol extracts from red pepper seed groups were significantly higher than that of the control group. Sprague-Dawley strain male rats weighing $100{\pm}10$ g were randomly assigned to one normal diet N group and experimental groups fed high fat and high cholesterol diet, which were divided into HF (0.0%), HEA (0.1%), HEB (0.2%), and HEC (0.5%) groups according to the amount of ethanol extracts from red pepper seeds added to their basal diet. The body weight gain in the HF group was higher than that in the N group, and those in the HEA, HEB and HEC groups were lower than that in the HF group However, there were no statistically significant differences among the all the groups. The hepatic triglyceride and total cholesterol contents in the N group was significantly lower than that in the HF group, and those in the HEA, HEB and HEC groups were lower than that in the HF group. The hepatic UDP-glucuronyl transferase activity in the N group was lower than that of the HF group and those in the HEA, HEB and HEC groups were lower than that of the HF group. The serum total cholesterol and triglyceride contents of the HF group were significantly higher than that of the N group, and those of the HEA, HEB and HEC groups were lower than that of the HF group. The serum HDL-cholesterol contents in all groups supplemented with the ethanol extracts from red pepper seeds were significantly higher than that of the HF group. The serum LDL-cholesterol contents of the HF group were significantly higher than that of the N group, and those of the HEA, HEB and HEC groups were lower than that of the HF group. The fecal total cholesterol contents were significantly higher in the HF group compared to the N group, and those of the HEB and HEC groups were lower than that of the HF group. The fecal triglyceride contents in the N group was higher than that of the HF group, and those of the HEA, HEB and HEC groups were lower than that of the HF group. This study suggested that the ethanol extracts from red pepper seeds have powerful health benefits by the UDP-glucuronyl transferase activity and lipid metabolism.

Protective Effect of Monascus pilosus Mycelial Extract on Hepatic Damage in High-Fat Diet Induced-obese Rats (고지방식이로 유도한 비만 흰쥐에서 Monascus pilosus 균사체 추출물의 간 손상 예방효과)

  • Hepatoprotective effects of Monascus pilosus mycelial ethanol extract (MPME) were examined in high-fat diet induced-obese rats. The rats were randomly divided into 2 groups; normal control (NC) and a high-fat and high cholesterol diet group (HFC). The HFC diet group was fed a 5L79 diet supplemented with 15% lard and 1% cholesterol for 3 weeks for induction of obesity. And then, the rats were divided into 4 groups (n=5); the NC, a HFC diet obesity control group (HF), 0.5% MPME supplemented HFC diet group (MPM), and 2% conjugated linoleic acid (CLA) supplemented HFC diet group for 7 weeks. Whereas the daily weight gain of NC and HFC groups were 3.48 g and 4.48 g, respectively, those of MPM and CLA were 3.09 g and 4.38 g, respectively. Furthermore, activity of serum alanine and aspartic aminotransferase in HF was markedly higher than those of NC group, but, the activity in MPM and CLA was significantly lower than HF. Hepatic reduced glutathione content in MPM and CLA was higher than HF. On the contrary, hepatic lipid peroxide content in MPM and CLA was significantly lower than HF. In conclusion, although the precise mechanisms of the hepatoprotective effects of the MPME in this study are unknown, our study provides experimental evidence that MPME may prevent obesity and hepatic damage by high-fat and high cholesterol diet via inhibition of lipid absorption and induction of reactive oxygen spices scavenging enzyme such as superoxide dismutase.

Phytoestrogenic Effects of Combined Plant Extracts on the Change of Bone Metabolism of OVX Rats (복합 생약추출물의 식물성 에스트로겐 활성이 OVX rat에 미치는 골대사 변화)

  • This study examined the combined plant extracts (FGF271) of Estromon in ovariectomized (OVX) rats to determine whether Estromon's significant clinical improvement effects on menopausal symptoms are predominantly due to the phytoestrogenic action of the combined extracts. The results showed that all three FGF271-treated groups had significantly improved serum osteocalcin levels as compared to the control group (p<0.05). In addition, all FGF271- and Estromon-treated groups had increases in femoral bone mineral density (FBMD) (p<0.05), and the increase in the FGF271 group was dose-dependent. A pairwise comparison of the FGF271- and Estromon-treated groups receiving the same dosage of FGF271 indicated that there was no significant difference between the groups. Therefore, the FBMD increases that occurred in the Estromon groups were solely attributable to the phytoestrogenic effects of FGF271. It was conclude that the phytoestrogenic effects of Estromon, as shown in clinical studies, are predominantly caused by FGF271, the mixed extracts of Cynanchum wilfordii, Phlomis umbrosa, and Angelica gigas.

Transfer of Genes for Antimicrobial Resistance and Toxin of Hemolytic Escherichia coli Isolated from Feces of Pig Suffering Diarrhea to Human Isolates (설사 증상의 돼지 분변에서 분리된 용혈성 대장균의 항생제 내성과 독소의 인체로부터 분리된 균주로의 전이)

  • Between 1997 and 1998 in Korea, 56 isolates of Escherichia coli were obtained from pig suffering diarrhea. Among those, 38 isolates that showed the hemolytic activity, antimicrobial resistance, and toxin production were studied. Among 38 isolates, thirty-six isolates $(94.7\%)$ were resistant to tetracycline, 27 isolates $(71.0\%)$ were resistant to ampicillin, 26 isolates $(68.4\%)$ were resistant to chloramphenicol, and 21 isolates $(55.2\%)$ were resistant to trimethoprim, while none was resistant to aztreonam, amikacin, and norfloxacin. Among these iso­lates, 21 isolates $(55.3\%)$ were multiple drug resistant to at least four different class antimicrobial agents. Extended spectrum $\beta-lactamase$ producing isolates were not detected in the double disk synergy test. In these hemolytic Escherichia coli, heat-stable enterotoxin $(89.5\%)$ was the most prevalent toxin, followed by vero­toxins $(47.4\%),$ and then heat-labile enterotoxin $(31.6\%).$ Except 8 isolates $(21.0\%)$ which produced ST only, 12 isolates $(31.6\%)$ produced ST and LT, 13 isolates $(34.2\%)$ produced ST, VT, and VTe, and 5 isolates $(13.2\%)$ produced VT and VTe. However, none produced all 4 types of toxin, simultaneously. The predominant serotype could not be determined by the agglutination method. Sixteen isolates $(42.1\%)$ were strongly adhered to T-24 bladder cell and 17 isolates $(44.7\%)$ were to Caco-2 intestinal cell. Especially, 11 strains $(28.9\%)$ were evaluated as strongly adhesive to both T-24 cells and Caco-2 cells. Genes for toxin and the antimicrobial resistance were transferred to clinical isolates of Escherichia coli from human urine by the filter mating method. Results suggest the possibility that antimicrobial resistance and toxin can be transferred from animals to humans by direct con­tact of resistant bacteria as well as gene transfer, although there was no correlation between toxin production, adherent activity, and antimicrobial resistance among hemolytic E. coli isolated from pig suffering diarrhea.

Effects of Characteristics of Ovarian follicular Fluid and Ant-Inhibin Serum on Steroid Hormone Secretion by Hanwoo Granulosa Cells In Vitro (한우 난소의 Follicular Fluid의 특징과 과립막 세포의 스테로이드호르몬 분비에 대한 Anti-Inhibin Serum의 첨가효과)

  • This study was performed to investigate the effects of the peptide to carrier ratio on the immune and biological functions to inhibin immunization in Hanwoo. A peptide sequence kom the alpha -subunit (19~32 peptide) of porcine inhibin was synthesized for antigen and conjugated to human serum albumin(HSA) for carrier protein. Anti-inhibin sera(AI) were produced 52 day later from rabbit after injection of inhibin-$\alpha$ -subunit peptide conjugator for antigen with the interval of 2 weeks. Immune-blotting analysis using antibody specific fur inhibin-$\alpha$ subunits revealed that the inhibin was detected at 1.0 cm bovine follicular fluid(bFF). However, each stage of corpus lutea and 0.1 cm of follicular fluid were not detected. The maximal contents of estradiol-17 $\beta$ in Hanwoo ovarian follicular fluid were detected at 2.0 cm of follicular size(diameter), but the mean total contents of these hormone decreased significantly with decreasing diameter of follicles. However, progesterone contents of follicular fluid were high at 1.0 cm of follicle. Progesterone secretion by Hanwoo granulosa cell cultured for 48 hr in vitro was significantly (p<0.05) inhibited in 5% bFF and 5% bFF + 5% AI addition group compared with control group. Estradiol-17 $\beta$ secretion by Hanwoo granulosa cell cultured for 48 hr in vitro was significantly (p<0.05) increased in 5% AI and 5% AI + 5% bFF addtion group compared with control group. However, the groups added 5% AI were not changed compared to control groups in progesterone and estradiol-17 $\beta$. Taken together, we suggested that inhibin in the mature FF plays a pivotal role on the biosynthesis of steroid hormone of follicular cells during follicular development.

Effect of the Supply of Natural Water from Deep Sea Rock on the Immune Response and Antioxidant Activity in Rats (천연 암반 심해수 공급이 흰쥐의 면역반응 및 항산화 활성에 미치는 영향)

  • This experiment was conducted to study the effects of the natural deep sea water, which contained approximately 2.3% salt, and various minerals of K, Mg, Ca, Na, Fe, Mn, Zn, Cu etc, on the immune response and antioxidant activity in rats. 24 Sprague Dawley rats were randomly allotted to a control group and 3 treatment groups. Control rats were supplied with filtered tap water, and each treatment group rats were supplied with 0.5% deep sea water, 1% deep sea water and Jijangsoo, respectively, which is upper clear water separated from sediment by the clay. Feed and water were provided ad libitum throughout the experiment that lasted for 4 weeks. The results showed that 1% deep sea water group showed the highest values in weight gain, feed intake, and feed efficiency than those of other groups. The levels of water intake of 1%- and 0.5%-deep sea water, and Jijangsoo group were 49.1%, 22.8%, and 40.5% higher than that of control group, respectively. The Jijangsoo group rats showed that perirenal and epididymal adipose tissue weights were decreased by 32% and 25%(p<0.05), respectively, when compared to control group rats. There were no remarkable differences of serum glucose concentration among all experimental groups. However, insulin concentration of experimental groups were remarkably increased in order of Jijangsoo (4.54), 1% deep sea water (3.70), 0.5% deep sea water (3.25)(p<0.05). B cell and T cell stimulation were increased about 44.7% and 207%, respectively, by 0.5% deep sea water in comparison with control (p<0.05). TBARS values of 0.5 % deep sea water group were significantly lower than that of control(p<0.05). Catalase and SOD activities of 0.5 % deep sea water group were 200% and 47% higher than that of control, respectively. From the results, it can be concluded that the supply of natural deep sea water can slightly improve the physiological activity which modulates immune response and antioxidant activity in rats.
